The Anaesthetic Effect of MS-222 for Longtooth Grouper, Epinephelus moara and the Hybrid Grouper, E. moara ♀ × E. lanceolatus ♂ (자바리(Epinephelus moara)와 대왕자바리(E. moara ♀ × E. lanceolatus ♂)에 대한 MS-222의 마취효과)

  • The anaesthetic effect of tricaine methanesulfonate (MS-222) concentrations and water temperatures for longtooth grouper (Epinephelus moara) and hybrid grouper (E. moara ♀ × E. lanceolatus ♂) were investigated. Anesthetic induction and recovery time were measured at 18, 22, 26 and 30℃ of Cwater temperature and 100, 150, 200 and 250 ppm of anesthetic concentrations. Anesthetic induction time tended to decrease with increasing concentration and water temperature. Recovery time was proportional to concentration, but inversely proportional to water temperature. However, there was no significant differences in recovery time at 22℃ or lower. The optimal anesthesia condition was 30℃ and 100 ppm, which was the shortest recovery time for longtooth grouper, and 150 ppm at 30℃ in the case of hybrid grouper because anesthetic time is significantly different with 100 ppm in spite of no significant differences with 100 ppm for recovery time. As a results of two-way ANOVA test, there was a significant difference between the species of longtooth and hybrid grouper. On the other hand, there was no interaction effect between concentration and species. Also, there was no interaction effect among species, concentration, and water temperature.

Design of a Variable Resonator for the Sacred Bell of the Great King Seongdeok (성덕대왕신종을 위한 가변형 명동의 설계)

  • This study proposes a design model of the variable type resonator which corrects the temperature variance according to the season, in order to maximize the resonance effect in the Sacred bell of the Great King Seongdeok. In the bell, the 1st natural frequency (64 Hz) and the 2nd natural frequency (168 Hz) are the most important partial tones. Resonance conditions of the two components are determined for the internal acoustic cavity system, which consists of bell body cavity, gap and the resonator. Acoustic frequency response characteristics of the internal cavity are determined by the boundary element analysis using SYSNOISE. As an external factor, temperature variance according to the season largely influences the resonance condition and the length of the resonator should be controlled to maximize the resonance effect. As a measure, this study proposes a design model of the variable type resonator for the Sacred Bell of the Great King Seongdeok, which can control the length at the belfry according to the season.

  • the famous King Song-dok Bell which was cast in A.D. 771, during Silla dynasty has significant xize of concave cavity which acts as Helmholtz resonator on the ground. To see the effect of this cavity and the bell's internal acoustic cavity on radiating sound of bell, three dimensional measurement of internal sound field was performed. In this paper, the dtail measurement procedure and results are carefully addressed. The effect of the cavity on the radiating sound of bell is investigated using simple and physically representative model. A new size of cavity is proposed.

Allylisothiocyanate Content in Different Plant Parts of Wasabia japonica Mastum (고추냉이 부위별 Allylisothiocyanate 함량)

  • Essential oil extracted by solvent, n-pantane and diethyl ether for 2 hours was 0.1∼0.5% in Wasabia japonica Mastum. and it was showed the highest content in root and the lowest content in petiole. Allylisothiocyanate detected by gas chromatography contained in all of the plant part in wasabi. Its content in rhizome was 0.687∼1.339mg /g FW showing the highest content in rhizome. Allylisothiocyanate content was varied from culture site, variety and position of rhizome in wasabi.

  • Silla Great Bell was manufactured in order to ring for the new millennium in succession to King Seongdeok Divine Bell. This study investigates how Silla Great Bell is similar to King Seongdeok Divine Bell in respect of structure and sound. First, design and manufacturing processes are introduced and dimensions of the two bells are compared and the similarity is estimated. Three main characteristics of the sound of a Korean bell are magnificent and harmonic striking sound, dynamic beat, and long lasting hum tone. These three features are compared and objectively estimated using physical parameters. The purpose of the study is to provide useful information to manufacture Korean bell which has beautiful appearance and excellent sound.

Effectiveness of Microsatellite Markers for Parentage Analysis of Giant Grouper (Epinephelus lanceolatus) Broodstock (Microsatellite 마커를 이용한 대왕바리(Epinephelus lanceolatus) 친어 집단의 가계도 분석 효율)

  • Giant grouper (Epinephelus lanceolatus) is a endangered species considered as a vulnerable grade-organism in the International Union for Conservation of Nature (IUCN) red list. As a fundamental baseline study for establishing a giant grouper broodstock management system, the efficiency for parentage analysis was evaluated by using microsatellite makers previously available in this species. The eight microsatellites generated a total 52 alleles from 32 individuals, the mean expected heterozygosity was 0.663, and mean inbreeding coefficient was 0.011, consequently suggesting that the present broodstock has retained the high level of genetic diversity. However, our analysis also recommended the collection of more broodfish for more stable brood line, since the estimated value of the effective population size was proven to be 35. The average probability of identity was $6.85{\times}10^{-11}$. NE-2P and NE-PP of paternity non-exclusion probabilities were 0.00835 and 0.00027, respectively. As the result of principle coordinate analysis, the genotype of broodstock was not overlapped, suggesting that the management system of giant grouper based on eight selected microsatellite markers might be effective, although further validation with extended number of broodfish might also be needed in future. Data of present study could be a useful basis to avoid the unwanted selection of broodfish that possess close genetic relationship with current broodstock, and consequently to establish effective broodstock management system allowing the production of progeny with high genetic diversity.

Growth and Yield of Wasabi Cultivated in Cold Water Drained from Trout Nursery (송어양식장 배출냉수 이용 재배시 고추냉이의 생육 및 수량)

  • We investigated rhizome yield and growth characteristics of wasabi variety, Daioichigo and Daruma cultured by flowing water from Trout nersery in Pyong Chang. Average temp. of air and water during a year was 14.3, 12.3$^{\circ}C$, respectively in PE film house. Wasabi growth was good and healthy in May and October. Rhizome weight of main stem in Daioichigo was 78.78g per plant and 608kg per l0a, while those of the Daruma was 37.1g per plant and 287kg per l0a. Rate of marketable rhizome was 90% in Daioichigo and 37.5% in Darurna. Rhizome weight of main stem in Daioichigo was increased, up to 21% of the control by the remove of floral axis with two times a year.
