Chemical Structures of the Compounds Isolated from the Edible Mushroom Sarcodon aspratus (능이버섯(Sarcodon aspratus)으로부터 분리한 저분자 화합물의 화학구조)

  • In our investigation on chemical constituents of edible mushrooms, seven compounds I-VII have been isolated from the methanolic extract of the fruit body of Sarcodon aspratus. The methanolic extract was separated by solvent partition, silica gel column chromatography and Sephadex LH-20 column chromatography, and compounds I-VII were finally purified by preparative HPLC or TLC. Their structures were assigned as 4-hydroxybenzoic acid methyl ester, 4-hydroxybenzaldehyde, cyclo(Ala-Pro), adenosine, nicotinamide, BL V, linoleic acid, respectively, on the basis of mainly $^1H\;NMR$ spectra.

  • Drug resistance is one of the most significant impediments to successful chemotherapy of cancer. Multidrug-resistance (MDR) is characterized by decreased cellular sensitivity to anticancer agents due to the overexpression of P-glycoprotein. By employing adriamycin-resistance CL02 cancer cells, we undertook the screening for agents which were effective to multidrug-resistant cancer cells. As a result, a myxobacterial strain JW117 was selected for study since the solvent extract of cell mass of the strain was found to exhibit significant activity against the CL02 cancer cells. Cytotoxicity-guided chromatographic fractionation led to the isolation of phenalamides $A_2$ and $A_3$. The producing organism was identified as Myxococcus stipitatus by taxonomic comparison with type strains of Myxococcus sp. as well as its morphological and physiological characteristics. Phenalamides$ A_1$,$ A_2$ and $A_3$ were as active against drug-resistant cancer cells CL02 and CP70 as against the corresponding sensitive cells with $IC_{50}$ values ranging from 0.23~0.57 $\mu\textrm{g}$/ml.

Medium Optimization for Cell Growth and Metabolite Formation from Haematococcus sp. under Mixotrophic Cultivation (혼합영양 조건하에서 Haematococcus sp.의 배지 최적화 및 대사산물 생산)

  • In this study, the medium optimization for cell growth and metabolite formation of Haematococcus sp. under mixotrophic cultivation was investigated. As a result, modified MS medium was selected as the basal medium; glucose was selected as the carbon source, with an optimum concentration of 10 g/l, and potassium nitrate was chosen as the nitrogen source, with an optimum concentration of 1.9 g/l. Under optimum conditions, Haematococcus sp. demonstrated an increase in biomass concentration from 0.18 gDW/l to 5.58 gDW/l in 14 days, after which there was a 31-fold increase in its growth. At the same time, the concentrations of chlorophyll and carotenoids were 172.16 mg/l and 42.33 mg/l, respectively. This work will contribute to the basic data for mass cultivation of microalgae.

Nutritional and Bioactive Compounds of Yellow Cherry Tomato (황색 방울토마토의 영양성분 및 생리활성 물질분석)

  • This study was carried out to investigate the value of yellow cherry tomato as a food material. Contents of nutritional and bioactive compound were determined from yellow cherry tomato. 17 of amino acids occurred. L-Glutamic acid (45.15%), L-glutamine (22%) and L-aspartic acid (11%) were the main amino acids. 8 of essential amino acids were found except tryptophan. γ-aminobutyric acid (GABA) was found in high contentration (258.58 mg/100g). Contents of Lycopene and β-carotene were 2.18 mg/100 g and 9.90 mg/100 g, respectively. Yellow cherry tomato contained naringenin chalcone, quercetin-3-rutinoside(rutin), 5-caffeoylquinic acid, 3-caffeoylquinic acid, and quercetin-3-apiosylrutinoside that have various bioactivities. These results revealed that yellow cherry tomato would be very useful and valuable food material.

Inhibitory Effects of Bile Acids on the Cholesterol Biosynthesis in Cultured Hepatocytes (배양 간세포내에서의 콜레스테롤 합성에 대한 담즙산의 저해효과)

  • The present work tested the inhibitory effects of bile acids on the cholesterol biosynthesis and the activity of HMG-CoA reductase in cultured rat hepatocytes. The uptake of bile acids in hepatocytes were increased in according to the different bile acid concentrations and culture times. The rate of cholesterol synthesis in cells were inversely decreased to the bile acid concentrations and culture times. As expected, insulin injection (4 units/100g body weight) showed an enhancing effect of the cholesterol synthesis and the HMG-CoA reductase activity. The addition of bile acids in medium of insulin-treated hepatocytes also showed the suppressing effect. This effect was directly confirmed in isolated hepatic icrosomes by the test of HMG-CoA reductase activity. In the test of $Na^+$,$K^+$-ATPase activity in the isolated hepatocyte membrane, only the cholic acid did not stimulate the enzyme system. The reason of such difference is not obvious, but this result indicates that the cholic acid could be absorbed by simple diffusion.

Complete genome sequencing of Pseudomonas fluorescens NBC275, a biocontrol agent against fungal pathogens of plants and insects (식물 및 곤충의 곰팡이 병원균에 항균력을 가진 Pseudomonas fluorescens NBC275 균주의 유전체 염기서열)

  • Pseudomonas fluorescens NBC275 (Pf275) isolated from soil sample collected at riverside of Nakdonggang showed antagonistic activity against fungal pathogens of plants and insects. Here we present complete genome sequence of Pf275. The genome comprises of 6,610,362 bp with GC content of 60.9%, which includes 5,869 predicted protein-coding genes, 16 rRNAs, and 65 tRNAs. Genome analysis revealed gene clusters encoding antimicrobial secondary metabolites such as pyoverdine, 2, 4-diacetylphloroglucinol, and phenazine, which are known to play essential roles in biocontrol of diseases.

Complete genome sequencing of Pseudomonas parafulva PpaJBCS1880, a biocontrol and plant growth promoting agent (식물 병 방제 및 생육촉진 효과를 나타내는 Pseudomonas parafulva PpaJBCS1880균주의 유전체 염기서열)

  • Pseudomonas parafulva PpaJBCS1880 (PpaJBCS1880) isolated from rice seeds showed strong antagonistic activity against bacterial plant pathogens by producing lipopeptide. Furthermore, the strain controlled the incidence of bacterial pustule in soybean plants and promoted the growth of rice plants. Here we present complete genome sequence of PpaJBCS1880. The genome comprises of 5,208,480 bp with GC content of 63.4%, which includes 4,487 predicted protein-coding genes, 19 rRNAs, and 74 tRNAs. Genome analysis revealed genes encoding antimicrobial secondary metabolites such as lipopeptide, pyoverdine, phenazine, and hydrogen cyanide, which are known to play essential roles in biocontrol of plant diseases.

Manipulation of Mini-Yeast Artificial Chromosome Containing Xylan Metabolism Related Genes and Mitotic Stability Analysis in Yeast (Xylan 대사유전자를가진미니효모인공염색체의가공및 Mitotic Stability 분석)

  • In this study, yeast artificial chromosome Insert (YAC) harboring genes which related xylan metabolism was constructed by using chromosome manipulation technique. For efficient chromosome manipulation, each splitting fragment (DNA module) required for splitting process was prepared and these DNA modules were transformed into Saccharomyces cerevisiae strain YKY164. By two-rounds chromosome splitting, yeast chromosome VII (1,124 kb) was split 887 kb-YAC, 45 kb-mini YAC and 198 kb-YAC and YKY183 strain containing 18 chromosomes was constructed. Splitting efficiency for chromosome manipulation was 50- 78% and expression level of foreign genes on 45 kb-mini YAC and enzyme activity were indistinguishable from that of the YKY164 strain. Furthermore, xylan-degraded products by recombinant enzymes were confirmed and mini-yeast artificial chromosome maintained stable mitotic stability without chromosome loss during 160 generations.

Relationship between Environmental Exposure and Biological Monitoring Values in Workers Exposed to Styrene (스타이렌 폭로 근로자의 환경중 폭로농도와 생물학적 모니터링에 관한 연구)

  • This is an effort to confirm changes biological monitoring according to changes in levels of exposure to styrene for industrial workers. This study was conducted on 108 workers, including male of 64 and female 44 who were working at factories of FRP, dipping, and coating. An improved passive monitor method(organic vapor monitor; OVM) was employed to determine levels of exposure. The biological monitoring include blood styrene concentration, urinary mandelic acid(MA), and urinary phenylglyoxylic acid(PGA). Biological monitoring were made through the Collection of blood and urine. The mean value of exposure to styrene was 21.0ppm, which is measured by organic vapor monitor, one of improved passive monitors. The highest exposure level was observed among workers in boat factories, laminating procedure workers, processing workers, respectively(p<0.01). For exposure level, 11% of subjects under study showed over 50ppm which is time weighted average(TWA). The correlation coefficient between biological specimens and the exposure level was 0.62 for blood styrene concentration, 0.58 for MA corrected by creatinine, and 0.70 for PGA corrected by creatinine, respectively(p<0.01). The regression analyses found exposure level relative importance in explaining variance in biological monitoring. In additional to that, gender was a significant factor in explaining variance of MA and MA+PGA. Almost half of variance(49%) in blood styrene concentration was explained by predictors, including exposure level, age, gender, duration, and drinking volume during the last week(p<0.01). The very high correlation(higher than 0.95 was found when a comparison was made among three types of corrected methods, including uncorrected specific gravity and creatinine. In conclusion, these findings suggest OVM to represent levels of exposure to styrene for industrial workers. A discussion was made on possible use of specific gravity sample for biological monitoring. Exposure level may be predicted on MA, PGA in urine, which could be applied to represent biological monitoring.

Development of a Simultaneous Analytical Method for Determination of Herbicide Fenquinotrione and KIH-3653-M-2 Residues in Agricultural Crops using LC-MS/MS (LC-MS/MS를 이용한 농산물 중 제초제 fenquinotrione 및 대사산물 KIH-3653-M-2 동시시험법 개발)

  • An analytical method was developed for the determination of fenquinotrione, a triketone herbicide, in agricultural products. Fenquinotrione was metabolized to KIH-3653-M-2 in plants. Analyte extraction was conducted using 2% formic acid in acetonitrile and cleaned up using a hydrophillic-lipophillic balance (HLB) cartridge. The limits of detection (LOD) and quantification (LOQ) were 0.004 and 0.01 mg/kg, respectively. Matrix-matched calibration curves were linear over the calibration ranges ($0.001{\sim}0.1{\mu}g/mL$) into a blank extract with $r^2>0.99$. The recovery results for fenquinotrione and KIH-3653-M-2 ranged between 81.1 to 116.2% and 78.0 to 110.0% at different concentration levels (LOQ, $10{\times}LOQ$, $50{\times}LOQ$) with relative standard deviation (RSD) less than 4.6%. All values were corresponded with the criteria ranges requested in both the Codex (CAC/GL 40-1993, 2003) and MFDS guidelines (2016). Therefore, the proposed method can be used as an official analytical method for determination of fenquinotrione in the Republic of Korea.
