Biodegradation Pathways of Polychlorinated Biphenyls by Soil Fungus Aspergillus niger (Polychlorinated Biphenyl의 토양 미생물 Aspergillus niger에 의한 생분해 경로)

  • As of many organochlorine pesticides, polychlorinated biphenyls are ubiquitous organic contaminants, which can be found in the most environmental matrices. Their toxic effects include endocrinedisrupting activity. Most researches with these toxicants performed with mixtures of congeners, namely Aroclor and related study has been done in complex environmental matrix, rather than single biosystems or pure congeners. 5 congeners were synthesized and their fates in pure microbial culture (Aspergillus niger) were determined in this study. Among biphenyl and synthetic congeners, biphenyl, PCB-1 (2-chlorobiphenyl), and PCB-3 (4-chlorobiphenyl) were rapidly transformed to hydrophilic metabolites, followed by PCB-38 (3,4,5-trichlorobiphenyl), while the degradation of PCB-126 (3,3',4,4',5-pentachlorobiphenyl) was not observed. The amounts of transformation for biphenyl, PCB-1, PCB-3, and PCB-38 were 65, 38, 52, and 2% respectively. The major metabolites of the above congeners were identified as mono- and di-hydroxy biphenyls, which are known to give adverse endocrinological effects.

Comparison of analytical methods of hippuric acid in urine (분석 방법에 따른 요중 마뇨산의 분석값 비교)

  • The aim of this study is to assess the interference effect of other organic acids to the values of hippuric acid analysed by UV method. We calculated the interference effect of several metabolites of styrene and xylene, i.e., methylhippuric acid, phenylglyoxylic acid, and mandelic acid to hippuric acid, respectively. The result of interlaboratory quality control program of urinary hippuric acid showed that there was no significant difference between the results by UV and HPLC if there were no other organic acids in urine. However, 0.5-2.0 g/L methylhippuric acid showed positive interference of 64-82% to 0.33 g/L urinary hippuric acid while mandelic acid or phenylglyoxylic acid did not show this positive effect. We suggest that HPLC or GC method is more acceptable than UV method to analyse urinary hippuric acid for biological monitoring when the worker was exposed to mixture of toluene and xylene.

Effect of Cimetidine on Theophylline Disposition and Metabolic Pathways (Cimetidine의 Theophylline 약동학 및 대사과정에 미치는 효과에 관한 연구)

  • The effect of cimetidine on theophylline metabolism was examined in dogs. Single dose intravenous theophylline kinetic studies were performed in cross-over manner before and after one week intravenous cimetidine (30 mg/kg/day) treatment. Cimetidine decreased theophylline clearance by an average of 31% (p<0.05) and prolonged theophylline half-li fe by an average of 29% (p<0.01) compared to those in control peirods. However, steady-state volume of distribution and protein binding of theophylline were not changed significantly. Twenty-four hours urinary excretion of 3-methylxanthine, 1-methyluric acid and 1,3-dimethyluric acid, which are the major metabolites of theophylline, were all decreased after cimetidine treatment, whereas the excreted fractions of individual metabolites were unchanged by cimetidine. From the above data, it could be susggested that cimetidine decreases theophylline clearance and prolongs the half-life by non-specific inhibition of the demethylations and 8-hydroxylation pathways.

영양소로서의 물

  • 영양소로서의 물의 중요성에 대해서는 앞에서 이미 충분히 강조한 바 있다. 물은 동물체의 주성분일 뿐아니라 며칠간 사료의 급여를 중단했을 때 가축이 받는 스트레스 보다 물의 급여를 중단했을 때 받는 스트레스가 훨씬 크다는 사실로 보더라도 물을 제외한 모든 영양소를 합한 것보다 물이 더 중요하다는 사실을 말해준다. 오늘은 물의 물리적 및 화학적 성질을 고찰하고 닭의 체내에서 물의 생리적 기능 및 물의 체내 대사에 대해서 알아보기로 한다.

In vivo metabolism of carbofuran in resistant and susceptible brown planthoppers, Nilaparvata lugens $St{\aa}l$ (저항성 및 감수성 벼멸구 체내에서의 카보후란 대사)

  • This study was conducted to find out the biochemical or metabolic resistance mechanism of brown planthopper (BPH) to carbofuran. Differences between resistant ($LD_{50};\;20.3{\mu}g/g$) and susceptible strains($LD_{50};\;0.3{\mu}g/g$) were shown. The amounts of carbofuran metabolite, benzofuranol, and the origin, not developed by Thin Layer Chromatography, were much more in the susceptible strain. But the mother compound, carbofuran, was much more in the resistant strain. The tendencies of metabolism one and three hours after treatment were similar in both strains except for the amounts of metabolites described above. From the study, it is supposed that hydrolytic enzyme, esterase, changes its role from cleaving the esteric bond of carbofuran to making conjugates with carbofuran. This seems to be the main resistance mechanism of BPH to carbofuran. Oxidase and transferase may play little or no role in resistance mechanism. Oxidative and transferring enzymes gave no effects on the metabolism of carbofuran in the resistant strain compared with the susceptible strain.

Determination of Pyribencarb and Its Metabolite KIE-9749 in Agricultural Commodities by High-Performance Liquid Chromatography (액체크로마토그래피를 이용한 농산물중 pyribencarb와 대사물 KIE-9749의 잔류 분석법)

  • The aim of this study was to develop an analytical method for the determination of pyribencarb and its metabolite KIE-9749 in agricultural commodities. The experiment was performed with a range of concentrations $0.05{\sim}2.5{\mu}g/g$ in apple, green pepper, potato, hulled rice, soybean, pear, peach, grape and cucumber. Each samples were extracted with acetone and cleaned by dichloromethane/saline water partition and purified with Florisil solid phase extraction (SPE) cartridge and aminopropyl SPE cartridge. Pyribencarb and KIE-9749 were separated and quantified by HPLC/UVD at 265nm using acetonitrile and water as mobile phase. The recoveries of pyribencarb and KIE-9749 were within 78.3~108.4% and 73.9~113.7% with RSD below 12.2% and 15.0%, respectively. The limits of quantification (LOQ) were both $0.05{\mu}g/g$. LC/ESI-MS/MS was optimized for confirmation of residue identity.

A Study on the Mechanism of Calcium Binding Inhibition of Cardiac Sarcoplasmic Reticulum by Oxygen Free Radicals (산소대사물에 의한 심장근 Sarcoplasmic reticulum의 칼슘운반 억제 기전에 관한연구)

  • Mechanism of calcium transport inhibition of cardiac sarcoplasmic reticulum (SR) by oxygen free radicals was examined. Effects of oxygen free radicals generated by xanthine/xanthine oxidase (X/XO) system on isolated porcine ventricle SR were studied with respect to its calcium binding, lipid peroxidation, SH-group content and alteration of membrane protein components. The results are as follows. 1) Calcium binding of isolated SR was markedly inhibited by X/XO. 2) During the incubation of sarcoplasmic reticulum with xanthine/xanthine oxidase, there were marked inclose in lipid peroxidation and reduction of SH-group content. 3) An antioxidant, p-phenylenediamine effectively prevented the lipid peroxidation but partially prevented the calcium binding inhibition of X/XO treated SR. 4) The reduction of SH-group content of SR treated with X/XO was partially prevented by p-phenylendiamine. 5) When modifying SH-group of SR by treatment with DTNB, the inhibition of calcium binding activity was partially prevented. 6) On gel-permeation chromatography of X/XO-treated sarcoplasmic reticulum, there was an increase of small molecular weight products, probably protein degradation products. 7) Semicarbazide, which prevents the cross-linking reaction of protein components, did not affect the calcium binding inhibition of X/XO-treated SR. From these results, it is suggested that the inhibition of calcium binding of SR by oxygen free radicals results from the consequence of multiple changes of SR components, which are lipid peroxidation, SH-group oxidation and degradation of protein components.

Superhongmi bran extract improves lipid profile and menopause symptoms: a randomized, placebo-controlled clinical trial (슈퍼홍미 미강 추출물의 폐경 후 여성의 혈중 지질 농도 및 대사성 질환 개선 효과)

  • Women who undergo natural menopause transition have increased numbers of risk factors relating to metabolic syndrome due to estrogen deficiency. This study was conducted to investigate the effect of Superhongmi bran extract on metabolic syndrome improvement in menopausal women. Thirty women, who participated in a randomized, double-blind, placebo-controlled trial, were assigned to placebo-control (n=15) or Superhongmi bran extract (n=15) groups and were asked to consume two tablets (350 mg per extract per tablet) per day. After 12 weeks, weight, body mass index (BMI), plasma triglyceride (TG) levels, and total cholesterol (TC) levels were significantly decreased, whereas HDL-cholesterol (HDL-C), apolipoprotein A1 (ApoA1), adiponectin, superoxide dismutase 1 (SOD1), and GSH (glutathione) concentrations were significantly increased in the Superhongmi bran extract group. Moreover, $17{\beta}-estradiol$, and progesterone levels in the Superhongmi group were significantly higher than those in the placebo-control group. These results suggest that Superhongmi bran extract alleviates metabolic symptom in menopausal women.

Effects of Paecilomyces tenuipes Water Extract on the Alcohol Metabolism of Rats (눈꽃동충하초 물추출물이 흰쥐의 알코올대사에 미치는 영향)

  • The effect of Paecilomyces tenuipes water extract (PTWE) on the alcohol metabolism was examined on rats. PTWE of 0, 30, 100 mg/kg body weight was administrated orally to the rats 30 min before oral treatment of 3 g/kg body weight of alcohol. Blood alcohol and acetaldehyde concentrations were measured 0.5, 1, 3, 6, 9 hr after alcohol treatment. Hepatic alcohol dehydrogenase (ADH), aldehyde dehydrogenase (ADH), microsomal ethanol oxidizing system (MEOS) activities were measured 9 hr after alcohol treatment. There were no differences in blood alcohol concentrations and area under the curve (AUC) of alcohol. PTWE decreased acetaldehyde concentration and there were significant differences after 6 hr in 30 mg/kg PTWE and after 3 and 9 hr in 100 mg/kg PTWE, respectively. In particular, 100 mg/kg PTWE decreased AUC of acetaldehyde by 44%. However, there were no changes in the hepatic ADH, ALDH, and MEOS activities.

Effects of Foeniculi fructus Water Extracts on Activities of Key Enzymes of Lipid Metabolism Related with Obesity (회향종자(Foeniculi fructus)의 물 추출물이 비만과 관련된 지질대사 효소의 활성에 미치는 효과)

  • This study was carried out to estimate beneficial effects of Foeniculi fructus water extract on activities of key enzymes such as lipoprotein lipase (LPL), acyl-CoA synthetase (ACS), and hormone sensitive lipase (HSL) on lipid metabolism related with obesity. LPL and ACS were extracted from the epididymal adipose tissue and liver of C57BL/6J normal and obese mouse. Foeniculi fructus water extract treatment significantly reduced the activity of normal and obese LPL. When 100 ppm of Foeniculi fructus water extracts were tested, they decreased obese LPL activity by 12.0%. Foeniculi fructus water extract activated obese ACS activity by 7-fold compared with control at 1,000 ppm concentration. Expression of HSL mRNA was increased in Foeniculi fructus water extracts treated cells compared with non treated cells. All things considered, Foeniculi fructus water extract efficiently inhibits the influx of fatty acid into the cell, and activates metabolic process that uses fatty acids flowing as an energy source. Thus, it suggest that Foeniculi fructus water extract may have great potential as a novel anti-obesity agent.
