Optimum cultivation conditions for mass production of antagonistic bacterium Pseudomonas azotoformans HC5 effective in antagonistic of brown blotch disease caused by Pseudomonas tolaasii (버섯 세균갈색무늬병균(Pseudomonas tolaasii)에 대한 길항세균 Pseudomonas azotoformans HC5의 대량배양을 위한 최적 배양조건)

  • This study was conducted to investigate optimum conditions for mass production of antagonistic microbes Pseudomonas azotoformans HC5. P. azotoformans HC5 is a potent biological control agent to control brown blotch disease caused by Pseudomonas tolaasii. This markedly showed the antagonistic activity against P. tolaasii, the most destructive pathogen of cultivated mushrooms. To define the optimum conditions for the mass production of the P. azotoformans HC5, we have investigated optimum culture conditions and effects of various nutrient source on the bacterial growth. The optimum initial pH and temperature were determined as pH 6.0 and $15^{\circ}C$, respectively. The optimal concentration of medium elements for the growth of pathogen inhibitor bacterium was determined as follows: 0.6% adonitol, 1.5% yeast extract, 0.8% $NH_4H_2PO_4$, 5mM $MgSO_4$, and 0.2% asparagine.

  • This study was conducted to investigate optimum conditions for mass production of ntagonistic microbes Alcaligenes sp. HC12. Alcaligenes sp. HC12 had a potent biological control agent to control browning disease caused by Pseudomonas agarici. Alcaligenes sp. HC12 markedly showed the antagonistic activity against Pseudomonas agarici, the most destructive pathogen of cultivated mushrooms. To define the optimum conditions for the mass production of the Alcaligenes sp. HC12, we have investigated optimum culture conditions and effects of various nutrient source on the bacterial growth.   • In vitro culture system was established to induce multiple shoots of Albizzia julibrissin Duraz. by investigating the effects of cytokinins. Cotyledon, hypocotyl and root explants were cultured on MS media supplemented with either three different plant growth regulators or their combinations. The most effective cytokinin sources were zeatin 2.0 + TDZ 0.5 mg/L in cotyledon, zeatin 1.0 mg/L in hypocotyl, and BA 0.2 + TDZ 0.01 mg/L in root explant for producing shoots ($5.67\;{\pm}\;1.20$, $19.50\;{\pm}\;3.50$, and $20.50\;{\pm}\;2.47$, respectively). Also, zeatin treatment was tended to induce more shoots rather than the combinations of other cytokinins. In addition, the root induced in 1/2 MS medium without any plant growth regulators was longer and thicker than treatments of IBA, NAA, IAA and 2.4-D as auxins. Overall, the highest average percent of in vitro shoot formation was 73% from three different types of explants with treatment of zeatin (1.0mg/L).

Establishment and Using of mass propagation system for Calanthe discolor Lindle (새우난초(Calanthe discolor Lindley)의 대량증식 시스템 확립 및 이용)

  • This experiment was conducted to establish the mass propagation system of Calanthe discolor Lindley. When the Calanthe discolor seeds were sown in Murashige and Skoog medium, the percentage of germination was 65%. Seedlings grew more rapidly in the liquid medium than the solid medium. All regenerated plantlets were survived in acclimatized condition of 70% shade and more than 80% humidity. Also, we found out that the 88% of survival ratio could be achieved in containing soil mixture of vermiculite and perlite as same as amount.

  • Taxiods inclusive paclitaxel were produced isolated and purified from plant cell cultures of Taxus chinensis in large-scale process. their structures were elucidated by spectroscopic analyses. These compounds were exactly identical as those in previous studies from the other biomasses of Taxus chinensis and also other species. Also the concentrations of these compounds were compared with the concentration of the paclitaxel in various batches of plant cell cultures. As paclitaxel concentration increased at the end of cell cultures. the concentrations of the other paclitaxel derivatives decreased. The profile of these taxoids production can provide information for better understanding of structure-activity relationships and biosynthesis Importantly it can be utilized as an useful parameter for the quality control of paclitaxel production.

  • The common harpacticoida copepod species Tigriopus japnnicus was often obseued in the mass culture of marine rotifer tanks of sea farming stations. In this study, we investigated the co-existing effect of T. iaponicus to the rotifer Brachionus rotundiformis population growth. The culture conditions as temperature, salinity, culture volume, photo period, culture period and observation interval were 25{\circ}C, 22ppt, 40ml, 12L:12D, 16 deys$ and every two days during the experimental period, respectively. Nannochloropsis oculata was used as the food for the two testing organisms. After counting the growth of rotifer and copepod, they were transferred to new culture tank with N. oculata at the density of $7{times}10^5\;cells/ml$. The population growth of B. rotundiformis was suppressed by T. japonicus when these two organisms were mix-cultured comparing to single culture of B. rotundiformis. But population growth of T, japomcus was not affected by the presence of B. rotundiformis. The interspecific relationships of prey-predation and food competition were not observed in the combination culture of rotifer B. rotundiformis and copepod T japonicus.

  • An isolate of Ampelomyces quisqualis 94013(AQ94013) was selected as an effective agent for biological control against cucumber powdery mildew. In order to develop mass production technique, six cereal media made with barley, rice, mille and brown rice, sorghumand rice seed were tested. Among them, barley medium was the best for the growth and conidial production of AQ94013. Optimum temperature for the mass production of AQ94013 was $25^{\circ}C$ and followed by $20^{\circ}C$ and $30^{\circ}C$. Light radiation inhibited conidial production of AQ94013 since number of conidia formed on barely medium under continuous light or 12 hrs alternative light were much less than cultured in darkness. Tthe conidia produced on the medium at $30^{\circ}C$ maintained the parasitic ability to Sphaerotheca fusca for 30 days. A culture method of AQ94013 in barley liquid medium with adding barely power(40 g/l) in darkness for five days at $25^{\circ}C$using a 30 l-fermenter was very effective for mass production of conidia.

  • Several bacteria are known as the causal agents of diseases of the cultivated button mushroom(Agaricus bisporus) and oyster mushroom(Pleurotus ostreatus). Pseudomonas tolaasii is the causal agent of brown blotch disease of commercial mushrooms. Pseudomonas sp. HC1 is a potent biological control agent to control brown blotch disease caused by Pseudomonas tolaasii. This can markedly reduce the level of extracellular toxins (i.e., tolaasins) produced by Pseudomonas tolaasii, the most destructive pathogen of cultivated mushrooms. To define the optimum conditions for the mass production of the Pseudomonas sp. HC1, we have investigated optimum culture conditions and effects of various nutrient source on the bacterial growth. The optimum initial pH and temperature were determined as pH 5.0 and $20^{\circ}C$, respectively. The optimal culture medium for the growth of tolaasin inhibitor bacterium was determined as follows: 0.9% dextrin, 1.5% yest extract, 0.5% $(NH_4)_2HPO_4$, 4mM $FeCl_3$, and 3.0% cysteine.

Selection of Carbon, Nitrogen Source and Carrier for Mass Production of Beauveria bassiana (Beauveria bassiana 대량배양을 위한 탄소원, 질소원 및 고체 기질 선발)

  • For mass production of entomopathogenic fungus Beauveria bassiana 149, isolated from moth larva, by two-phase fermentation, we performed selection of carbon and nitrogen sources for liquid culture and examined solid fermentation on carrier, ingredient, temperature, and water content. Spore production with rice powder, corn powder, and starch from sweet potato was higher than that of sucrose and dissolvable starch for liquid fermentation as first-phase fermentation. As a nitrogen source, addition of peptone and yeast powder showed higher spore production than $NaNO_3$, fish powder, and soybean powder. The isolate produced more conidia in sawdust + wheat bran + corn powder, sawdust + wheat bran and rice shell + wheat bran as carrier and ingredient than vermiculite as carrier. Conidia production of B. bassiana 149 in solid-phase fermentation was twice higher at 30 than 20. Conidia yield was higher at 60% and 70% water content ($26.9{\times}10^8$ and $38.6{\times}10^8conidia/g$) than 40% and 50% ($13.9{times}10^8 $and $11.6{\times}10^8conidia/g$), respectively.
