Hypoglycemic Effects of Atractylodis Rhizoma in Rats with Streptozotocin-Induced Hyperglycemia (Streptozotocin에 의해 고혈당을 유발시킨 흰쥐에 미치는 Atractrylodis Rhizoma의 영향에 관한 실험적 연구)

  • A single i.v. dose of streptozotocin (65 mg/kg) given to rats has produced a marked hyper-glycemia (>500 mg serum glucose/dl). Since the Atractylodis Rhizoma is known to have hypo-glycemic action, the water extracts of Atractylodis Rhizoma (ARWE) was given to the streptozotocin-induced (SZ) hyperglycemic rats. To determine whether ARWE has the anti-hyperglycemic effects, two different daily doses of ARWE (i.e.0.2 g/kg and 2.0 g/kg) were given orally to the SZ rats for up to 8 days. Thereupon, serum levels of glucose, insulin, amylase and cholesterol were determined on days 1, 3 and 8, following the initial and repeated daily administrations of ARWE. On day 8, glycogen content and glucose-6-phosphatase activity in the liver were assayed. Results showed that ARWE decreased the serum glucose levels, which had been markedly elevated by the SZ pre-treatment. In support of this, the serum insulin level, which had been quickly lowered by the SZ pre-treatment $(20{\mu}U/ml)$, was quickly elevated in the ARWE dose dependent manner that, at 2.0 g/kg ARWE, the serum insulin level was increased $(20{\mu}U/ml)$ above the normal level $(42{\mu}U/ml)$. Also, the serum amylase level, which was steadily decreasing after the SZ pre-treatment, was restored to the normal level folowing 8 day of ARWE (2.0 g/kg) treatment. Hepatic glycogen content and glucose-6-phosphatase activity, which decreased and increased, respectively in the 52 treatment group, were restored toward the normal level in SZ plus ARWE group.

Anti-diabetic Effects of Mixed Extracts from Lycium chinense, Cordyceps militaris, and Acanthopanax senticosus (지골피, 동충하초, 가시오가피 혼합 추출물의 항당뇨 작용)

  • In this study, we examined the anti-diabetic activity in vitro by the mixed Korean herb water extracts(JDG) composed of Lycium chinense, Cordyceps militaris, and Acanthopanax senticosus on hepatic glucose-regulating enzyme activities such as glucokinase(GK), acetyl-CoA carboxylase(ACC), and inhibition activity of $\alpha$-glucosidase. The hepatic cytosol fraction of a type II diabetic animal(Goto-Kakizaki rat) was used in GK and ACC activity assays. JDG mixed water extracts significantly increased the hepatic GK and ACC activity. The highest anti-$\alpha$-glucosidase activity was observed in JDG 5 water extract when compared to the acarbose control, inhibitor of $\alpha$-glucosidase. We suggest that Lycium chinense, Cordyceps militaris, and Acanthopanax senticosus mixed water extracts may exert an anti-diabetic activity by enhancing the glucose metabolism and may be used as natural $\alpha$-glucosidase inhibitors in type 2 diabetic conditions.

A Study on the Glucose-regulating Enzymes and Antioxidant Activities of Water Extracts from Medicinal Herbs (한약재의 물 추출물이 당대사 관련 효소와 항산화 활성에 관한 연구)

  • The anti-diabetic effects of water extracts (WE) from medicinal herbs on hepatic glucose-regulating enzymes, such as glucokinase (GCK), pyruvate dehydrogenase (PDH), acetyl-CoA carboxylase (ACC) and ${\alpha}$-glucosidase, were studied using the cytosol fraction in liver and mitochondia fraction in heart of a type II diabetic animal (GK rat, Goto-Kakizaki). The free radical scavenging activity of water extracts by DPPH method was also tested. We found that free radical scavenging activity was strong in Corni fructu (CF), Mokdan Bark (MDB), Chenhwabon (CHB) and Sanyack (SY), while that of Backbocreng (BBR), Shuckgihwang (SGH) and Taecsa (TS) was lower. For GCK activity in cytosol of liver, CF and CHB had a more effective activity than other extracts. PDH activity in mitochondria fraction of heart was higher in all of the extracts, expect for the TS extract, than in the control. ACC activity in cytosol fraction of liver was significantly higher in the CF, CHB, SGH, TS and SY extracts than in the control. CF, BBR and MDB led to a decrease in the ${\alpha}$-glucosidase activity. Therefore, these results suggest that all of the extracts may be used as functional material in the development as anti-diabetic functional food and medicine.

Estimation of Genetic Parameters for The Growth Traits of Performance and Progeny Test in Hanwoo(Bos taurus Coreanae) (한우 당대검정우와 후대검정우의 성장형질에 관한 유전모수 추정)

  • This study was conducted to estimate the genetic parameters and their relationships with weight traits of the steers and bulls in the Hanwoo population. The data used were weights and weight gain of performance and progeny test from 6,024 heads of Hanwoo. Data of performance test consisted of total 3,737 heads raised from August, 1989 to September, 2005. The number of the records of progeny test was total 2,287 heads from August, 1996 to June, 2004. The heritabilities and correlations for the body weights at the ages of 6 months, 12 months and 24 months and average daily gain were estimated by DFREML. Overall means and standard deviations of body weights at 6 and 12 months of age and average daily gain(ADG) from the data of performance test were 181.72±30.22kg, 351.48±40.24kg, 998.07±153.84g, respectively. Overall means and standard deviations of body weights at 6, 12, and 24 month of age and ADG from the data of progeny test were 169.18±32.82kg, 229.37±44.57kg, 570.45±64.36kg and 739.41± 172.14g, respectively. The heritability estimates of the body weight at 6, 12 month and ADG from the performance test were 0.54±0.06, 0.60±0.06 and 0.23±0.04, respectively. The heritability estimates of the body weight at 6, 12, 24 month and ADG from the progeny test were 0.80±0.08, 0.50±0.07, 0.46±0.07 and 0.07±0.03, respectively.

Between Text and Image, The Audience and Film -The Weekly Newsletters and Leaflets of Dansungsa as Media (1926-1937) (문자와 영상, 관객과 영화의 사이에서 -미디어로서의 단성사 주보와 전단(1926-1937))

  • This paper examines printed materials such as weekly newsletters and leaflets issued by Dansungsa, a movie theater in Colonial Korea for a promotional purpose as independent modern media. During the 1920s and 1930s, in tandem with the development of the incipient printing houses in Namchon, Gyeongseong, including Suyeongsa, Dansungsa published promotional prints including weekly newsletters and leaflets in a serial manner to compete with Joseon-gukjang and Umigwan. As these materials contain various information including movie programmes, spectatorship, distributional channels, and promotional strategies that bears witness to theater culture of this time, this paper focuses on the dynamics where not only text and image but also audiences and filmic texts are mediated one another. To this end, the paper has three objectives. First, I argue that weekly newsletters and leaflets can be considered as 'flickering media' that meddles in text and image culture. Second, Dansungsa's promotional prints interpellated film audiences as a loyal fan group while mediating audiences and filmic texts. In doing so, I suggest that these print materials established its own cultural domain differentiated from filmic culture itself. Third, these ephemeral materials contributed to narrowing the gap between colonial Joseon and the World in its imaginary geography through the function of mediation.

역사속 과학인물 - 중국의 '산학계몽' 맥 이어준 조선조 수학자 "김시진(1618~1667년)"

  • 조선조 현종때 관리였던 김시진(1618~1667년)은 중국판 수학책 "산학계몽"을 우리나라서 인쇄 보급해 수학자 양성에 큰 공을 세운 인물이다. 중국 송나라때인 1299년 주세걸이 쓴 이 책은 중국의 수학사에서 빼놓을 수 없는 귀중한 책이었다. 그런데 이 책이 5백년의 세월이 흐르는 동안 중국에서는 사라져버려 조선에서 구해다가 다시 찍어낸 것이 지금의 "산학계몽"이다. 사무에 통달하고 산법이 밝았던 김시진은 현종때 경기 좌균전사의 벼슬을 지내고 경상감사까지 오른 당대의 수학자였다.

Effect of Panax Ginseng on the Carbohydrate Metabolisin of Rat(I) (인삼의 당대사에 미치는 영향 (제 1 보))

  • We have studied on the effects of Panax Ginseng on the carbohydrate metabolism of rat. Ginseng does not so much effect on the body weight of the nutrition diet groups, but effects much on the carbohydrate deficient diet groups; it remarkably inhibited the decrease of body weight. Liver glycogen, both in the nutrition and carbohycdrate deficient diet groups, is higher than that of the control. Particularly, the increase of the glycogen is eminent is the nutrition diet-ginseng administered group. In the nutrition diet group, the blood sugar level of ginseng-adminstered rats is the higher, while, in the carbohydrate deficient diet groups, the blood sugar level of ginseng-administered rats is decreased compared with that of control. the oxidation rate of glucose by rat liver slice indicates that only in the carbohydrate deficient diet groups, from the 6th to 15th hour, the oxidation rate of glucose in ginseng-administered group is increased, compared with that of the control. But there is no statistical significance between them. It is thought that Panax Ginseng would effect more on the enzyme system involved in gluconeogenesis and glycogenesis rather than that of carbohydrate oxidation.

  • The oligosaccharide metabolism in soymilk was investigated by mixed culture with Lactobacillus acidophilus and Saccharomyces uvarum. When Saccharomyces uvarum was cultured in soywhey, change of oligosaccharide could be shown apparently. However, Lactobacillus acidophilus could not utilize oligosaccharide in soywhey for growth and lactic acid production. During the fermentation of mixed culture with Lactobacillus acidophilus and Saccharomyces uvarum, Saccaharomyces uvarum was supposed to convert oligosaccharide to monosaccharide first and then Lactobacillus acidophilus to convert these produced monosaccharide to lactic acid.

  • The purpose of this study was to investigate the effect of dietary zinc on the carbohydrate metabolism and the serum content of magnesium and chromium in rats fed normal diet. Animals were divided into three groups by different dietary zinc levels which were low(15ppm), normal(30ppm) or high(60ppm). Serum glucose and Insulin concentrations were assessed by the glucose method and the radio immuno assay respectively. Serum zinc. magnesium and chromium contents were measured by Indectively Coupled Plasma(ICP). Results of the study were as follows : 1. Feed intake in a zinc deficiency group was significantly higher than that in other group, but the weight gain in high zinc diet group was significantly lower than in other groups. 2. There were no significant differences in liver, kidney and spleen weight. 3. Serum glucose, insulin and zinc concentrations were not significantly different among different dietary zinc groups, However serum magnesium and chromium concentrations were significantly decreased as the level of dietary zinc was increased.

  • Objectives: This study investigated the effects of water extract of Aconitum carmichaeli Debx (ACD) on obesity and glucose tolerance in high fat diet induced obese mice. Methods: Five-week-old C5BL/6 mice were fed a high fat diet (HFD) containing or not containing ACD (100 or 300 mg/kg) for 16 weeks. Body weight, food intake, fasting blood glucose, and body temperature were checked every week and then organs, blood serums were collected after 16-week treatment. Furthermore oral glucose tolerance test (OGTT) was carried out after treatment. Results: ACD treated mice showed no significant decreases in body weight and adipose tissue weight as compared with HFD mice. Lipid accumulations in liver and serum lipid levels were not different between ACD treated and HFD mice. However, ACD extract administration significantly and dose-dependently reduced fasted plasma glucose and glucose tolerance as determined by OGTT. Conclusions: The present study demonstrates that ACD might prevent diet-induced glucose tolerance in mouse models of obesity.
