Development of One Layer Loading Hanger Device for Flue-cured Tobacco (수확 담배잎을 엮는 단선형 행거 엽편장치 개발)

  • In order to improve the loading efficiency in bulk dryer for the flue-cured tobacco leaves, a new hanger device was developed. It consists of two layers frame, in which large volume of the harvested leaves could be simply loaded. As the results of experiments with new hanger device, labor hours for loading could be saved by 34 % as compared with those in conventional 3 layers hanger, and loading capacity increased by 36%. The weight of hanger could be also reduced by 0.6kg compared to conventional 3.4kg, showing possibility of cost reduction for the developed hanger device with saved number of punching the rolled steel panels.

Expression of an artificial gene encoding a repeated tripeptide lysyl-g1utamyl-tryptophan in Tobacco Plant (담배식물체에서 필수아미노산인 lysyl-glutamyl-tryptophan을 암호화하는 인공유전자의 발현)

  • To investigate expression of the artificial gene encoding a repeated tripeptide lysyl-glutamyl-tryptophan in tobacco plant, the plant binary vector, pART404 has been constructed, which contains the duplicated CaMV 35S promoter, an artificial gene coding for repetitive polymer (Lys-Glu-Trp)$_{64}$, and nopaline synthase (nos) terminator. The recombinant expression vector was introduced in Nicotiana tabacum (var. Xanthi) via Agrobacterium tumefaciens-mediated trans-formation. The transgenic calli selected by kanamycin containing medium were then regenerated to whole plants. Southern blot analysis indicated that five transgenic plants (No. 1, 7, 9, 43, 45) showed the hybridizing signals at 1.1 kb of the expected size on EcoRI digestion and each of the transgenic plants contained 1 or 3 copies of the artificial gene inserted into its genome. By northern blot analysis, the size of the hybridized total RNA was estimated to be approximately 1.2 kb and the RNA appeared generally to have the integrity. Western blot indicated that the protein was detected at the position of 33 kDa and the expression level of the polypeptide in the transgenic plant (No. 45) was measured to approximately 0.1% of the total protein.

  • Transgenic tobacco plants expressing calmodulin derivative($lys{\rightarrow}ile$ 115 calmodulin) and hygromycin resistance genes or hygromycin resistance gene alone(control) were generated by Agrobacterium-mediated DNA transfer. Seeds obtained from the transgenic plants($F_o$) were tested for resistance to hygromycin and the expected 3 : 1 ratio was observed. The expression of calmodulin derivative in the tobacco plants was identified by a combined method of Western blot and Chemiluminescence. The effects of surface sterilizers on the germiation of seeds from transgenic tobacco plants were tested in Murashige and Skoog agar medium. Seeds obtained from transgenic tobacoo plants expressing the calmodulin derivative showed no fungi contamination with normal germination by treating with sterilized water alone or sodium hypochlorite(2% effective chlorine). In contrast, seeds from the control transgenic tobacco plants showed severe contamination with fungi by treating with sterilized water alone and showed no contamination with normal germination by treating with sodium hypochlorite(2% chlorine). The effects of calcium concentration on the germination of seeds from transgenic tobacco plants were tested in Murashige and Skoog agar medium. Seeds obtained from transgenic tobacco plants expressing the calmodulin derivative showed better germination frequency than that of the control transgenic tobacco seeds in the medium containing 30 mM $CaCl_2$. The data raise the possibility that the expression of calmodulin derivative gene in tobacco plants could increase adaptability of the seeds to environmental stresses.

Economic Injury Level of Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) on Oriental Melon (Cucumis melo L.) in Greenhouse (참외에서 담배가루이의 경제적 피해수준 설정)

  • This study was conducted to develop economic injury level (EIL) of sweet potato whitefly, Bemisia tabaci, on oriental melon. In greenhouse, seedlings of oriental melon were transplanted at June 8, 2010 and we inoculated adult B. tabaci with the density of 0, 1, 5, 10, or 20 per ten leaves at July 14. Adult of B. tabaci increased approximately twenty five-fold at 60 days after inoculation in the plot of 20 adults per ten leaves. The damages on leaves and fruits by B. tabaci were started to appear at 20 days after inoculation, and the damage rates of leaves or fruits were 28.5 or 31.5 percent at 60 days after inoculation, respectively, in the plot of inoculation with 20 adults per ten leaves. The yield of oriental melon was reduced as the inoculation density of B. tabaci increased, and the relationship between inoculation density of B. tabaci and the rate of damaged fruit could be described by a linear regression Y = 0.961x + 0.0562 ($R^2$ = 0.976). Based on the relationship, the economic injury level was 5.1 adults of B. tabaci per leaf and the control threshold estimated by 80% level of economic injury level was 4.1 adults per leaf for control of sweet potato whitefly.

Development of a Binomial Sampling Plan for Bemisia tabaci in Paprika Greenhouses (파프리카온실에서 담배가루이의 이항표본조사법 개발)

  • Infestation of adults and pupae of sweetpotato whitefly, Bemisia tabaci, on paprika (Capsicum annuum var. angulosum) grown in greenhouses in Jinju, Gyeongnam province during 2014was determined by counts of the number of target stage of B. tabaci per leaflet. Binomial sampling plans were developed based on the relationship between the mean density per leaflet (m) and the proportion of leaflets infested with less than T whiteflies ($P_T$), according to the empirical model $(({\ln}(m)={\alpha}+{\beta}({\ln}(-{\ln}(1-P_T))))$. T was defined as the tally threshold, and set to 1, 2, 3, 4, 5 (adults) and 1, 3, 5, 7 (pupae) per leaflet in this study. Increasing the sample size, regardless of tally threshold, had little effect on the precision of the binomial sampling plan. Based on the precision of the model, T = 1 was chosen as the best tally threshold for estimating densities of B. tabaci adults and T = 3 was best tally threshold in B. tabaci pupae. Using the results obtained in the greenhouse, a simulated validation of the developed sampling plan by RVSP (Resampling Validation for Sampling Plan) demonstrated the plan's validity. Above all, the binomial model with T = 1 and T = 3 provided reliable predictions of the mean densities of B. tabaci adults and pupae on greenhouse paprika.

  • This study was conducted to determine the appropriate culture conditions for developing photoautotrophic cells from tobacco using plant growth regulators, sucrose and light condition in LS medium. The greatest callus induction was observed in the LS medium supplemented with $10^{-5}$ M NAA and $10^{-4}$ M BA, showing 3.08 g of callus fresh weight determined at 30 days after incubation. The highest chlorophyll content of callus was obtained in the LS medium supplemented with $10^{-5}$M NAA and $10^{-6}$ M BA, showing 28.42${\mu}g/g$. In both light and dark conditions, callus induction increased as the concentration of sucrose increased from 0.5%to 3.0%. In particular, in the light condition, the greatest callus induction was made in the LS medium supplemented with 2% sucrose. On the other hand, the heighest chlorophyll content was observed at 0.5% sucrose and chlorophyll wasn't induced in dark condition. The chlorophyll content of callus cultured in LS medium containing 0.75% sucrose was similar to 1% sucrose, as far as the chlorophyll content of callus was concerned.

Biological Control of Insect Pests with Arthropod Natural Enemies on Greenhouse Sweet Pepper in Winter Cropping System (파프리카의 겨울작형 시설재배에서 천적을 이용한 해충 밀도억제 효과)

  • This study reports the results of natural enemy application against four insect pests on greenhouse sweet pepper in winter cropping system. Orius laevigatus ($3.3/m^2$) was released at two different times (Nov. 2005 & Feb. 2006) to control Frankliniella occidentalis, western flower thrips (WFT). Throughout the cropping season, the lowest level recorded of WFT was less than 2.0 individuals per yellow sticky trap. Amblyseius swirskii ($232.3/m^2$) was released four times (Nov. 2007 ~ Aug. 2008) to control Bemisia tabaci, tobacco whitefly (TWF). Until July 2008, TWF population had been suppressed lower than 6.6/trap. Phytoseiulus persimilis ($44.5/m^2$) was released seven times (Apr. 2008 ~ Aug. 2008) to control Tetranychus kanzawai, tea red spider mite (TRSM). As a result, TRSM population was suppressed lower than 1.7/1eaf. To control aphids, Aphidius colemani ($9.5/m^2$) was released seven times (Oct. 2004 ~ Jan. 2005), with the transplantation of banker plants ($5.5pot/660m^2$). As a whole, aphid's population has been successfully reduced to less than 2.0/leaf.

  • 고추에 피해를 주고 있는 병으로 잎오갈병, 더뎅이병, 탄저병, 역병등 19종이 우리나라에 보고 되어 있다. 이러한 고추의 병해를 병원균별로 보면 담배모자이크바이러스등 바이러스가 5종, 더뎅이병균등 세균이 3종이 있으며 곰팡이는 역병균등 14종이 고추에 병을 일으키고 있다. 이와같은 고추병들이 고추에 피해를 주고 있지만 그중에서도 특히 고추를 이어짓기 하는 밭이나 비닐하우스에서 고추의 작황을 좌우하는 것은 대부분이 역병에 의한 피해이다. 역병의 피해는 물론 고추를 이어짓기함으로써 토양 전염되는 역병균의 전염원이 매년 증가하기 때문이기도 하지만 역병이 발생하기 쉬운 여름철에 장마가 겹치므로 물빠짐이 나빠지거나 침수가 되기 때문에 피해가 증가하는 요인이 되고 있다.

Comparison on the Capability of Four Predatory Mites to Prey on the Eggs of Bemisia tabaci (Hemiptera: Aleyrodidae) (포식성 이리응애 4종의 담배가루이 알 포식능력 비교 평가)

  • On the leaf disc of fruit vegetables (cucumber, sweet pepper, tomato, egg plant, oriental melon), it has been evaluated that the capability of four predatory mites (Amblyseius swirskii Athias-Henriot, Neoseiulus barkeri (Hughes), N. cucumeris (Oudemans), N. californicus (McGregor)) to prey on the eggs of Bemisia tabaci (Gennadius) and to lay own eggs. The most aggressive mite was A. swirskii which had consumed about $9{\sim}12$ eggs in a day. The quantity of eggs consumed by N. barkeri, N. cucumeris and N. californicus was about $6{\sim}9,\;4{\sim}6$ and $1{\sim}4$, respectively. The most productive mite was also A. swirskii which had laid about $1.2{\sim}1.9$ eggs in a day. The others could not lay more than 0.5 eggs in a day.

Phytophagous Insect Fauna of Dicotyledoneae(Tracheophyta : Angiospermae) Weeds (쌍자엽 식물아강(관속식물문 : 피자식물강)의 잡초가해 곤충상)

  • Phytophagous insects associated with Dicotyledoneae weeds and host specificities in the field populations were investigated for the survey of biological control agents of weeds in Korea. Fifty four weed species in 39 genera were collected during the survey. The most insects were collected from Polygonales by 24 species in 22 genera and followed by Urticales and Centrospermales by 17 species of 17 genera. The insects collected in the other weed orders were ranged from 1 to 12 species. Out of 17 insect species collected in Urticales, Baris sp. damaged the leaves of Hamulus japonicus in Cannabinaceae as scattered holeshape and showed host specificity. In Polygonaceae, Rumex japonicus and R. crispus were severely damaged by Aphis rumicis and Gastrophysa atrocyanea. G. atrocyanea leaf beetle had host specificity on R. japonicus and ate all the leaves except veins. The leaf beetle, Lypesthes japonicus was a potential biological control agent by feeding leaves of Persicaria spp .. And Lixus spp. were also often collected from Persicaria spp .. Liothrips vaneeckei was first collected from weed, P. modosa. P. senticosa was damaged by unidentified geometrid moth larvae and P. perfoiliata by Miarus atricolor snout beetle. Cassida piperata damaged leaves of Chenopodium album of Centrospermales and showed host specificity. In a soybean field, C. album and Amaranthus mangostanus were severely damaged by Spodoptera litura larvae which were eating soybean leaves. This phenomenon indicates that the presence of weed in cultivated land influences the outbreak of insect pests. Altica oleracea leaf beetle was frequently collected from Oenothera spp. of Onagraceae in Myrtales. Aphis gossyphi was outbroken on Solanum nigrum and Phylliodes brettinghami leaf beetle was first recorded on the same plant. Leaf beetles, Longitarsus scutellais and Hemipyxis plagioderoides were first collected from Plantago asiatica of Plantaginaceae in Plantaginales. They showed host specificities in the fields. The hemipterans were collected from many weeds during the survey and their roles on weeds should be investigated. A tractomorpha bedeli was also collected from many kinds of weeds in forest areas.
