Expression of Proteinase Inhibitor II gene in Transgenic Flowering Cabbage, Brassica oleracea var. acephala DC. (형질전환된 꽃양배추에서 Proteinase Inhibitor II 유전자의 발현)

  • Hypocotyl explants of flowering cabbage were cocultured with Agrobacterium tumefaciens LBA4404;;pGA875 harboring proteinase inhibitor II(PI-II) cDNA and then regenerated into plants. Sucessful transcripts of PI-II gene were detected by RNA dot blot analysis. Bioassay was conducted on transgenic flowering cabbage. It was confirmed that insecticidal activities of transformants were much higer than that of control plants. In progeny test of hansformants, 27.4% of T$_1$ seeds was resistant on MS medium containing 20 mg/L kanamycin.

Occurrences of Major Pests in Japanese Apricot, Prunus mume Siebold & Zucc. in Gyeongnam Province (경남지역 매실에 발생하는 주요해충의 발생소장)

  • Twenty-five species of pests had occurred at orchards of Japanese Apricot, Prunus mume Siebold & Zucc., in Hadong and Jinju (Gyeongsangnam-do province) from 2005 to 2007. Rhopalosiphum rufiabdominale (Sasaki), Singapora shinshana M., Tetranychus viennensis Zacher, and Synanthedon hector B. occurred as major pests and Anthonomus persicae sp. nov., Grapholita molesta (Busk), Spodoptera litura F., and Myzus persicae (Sulzer) as a sporadic pests. The population of S. shinshana had increased continuously during the season, from late April to late October. The population of T. viennensis occurred early May, reached the highest peak in late June to early July, and then declined with worsening consequences of nutritional conditions in the apricot leaves. A. persicae was observed to occur before breaking flower-bud of apricot and emerged adults were found in April to early May.

Pest Biodiversity and their Characteristic Damage caused to Greenhouse Strawberries in Jeju (제주지역 시설딸기에 발생하는 해충의 종류와 피해특징)

  • This study was carried out to investigate pests and their damage caused to commercial greenhouse strawberries in Jeju from 2013 to 2016. A total of 8 orders, including 31 species, of pests were found. Among them, the major pests were Tetranychus urticae, Frankliniella occidentalis, and Aphis gossypii. Although Trialeurodes packardi and Empoasca vitis were sometimes severely damaged strawberries, their frequency of occurrence and damage caused increased gradually. Of lepidopteran pests, Spodoptera litura showed the highest occurrence and damage. Although Helicoverpa armigera was showed a low occurrence frequency, it caused severe damage. We discussed the seasonal occurrence patterns of four pests-F. occidentalis, the alate A. gossypii, T. packardi and E. vitis-monitored by yellow-colored sticky traps.

Seasonal Occurrence of Insect Pests and Related Yield Loss in Amaranth Crop in South Korea (국내 아마란스(Amaranthus spp.)에 발생하는 주요 해충의 발생소장과 수량 감소율)

  • This study was carried out to investigate insect pests and their damage caused to commercial amaranth in Gangneung, Gangwon-do, Korea, during 2019-2020. A total of seven families and 18 species were identified including three species of aphids, twelve species of stink bugs, and four species of moths. Among them, five species caused significant economic damage due to a large number of occurrences; Aphis fabae, Nysius plebejus, Polymerus cognatus, Spoladea recurvalis and Spodoptera exigua. Aphids started to occur from the end of April, and showed the greatest incidence in early June. Three Nysius species occurred from mid-May and showed the maximum density in late August, and then its population decreased from the beginning of September. Two species of moth caterpillars began to occur from mid-August and damaged amaranth until mid-October. Comparing the yields between amaranth plants with and without a spray of pesticides for each pest, the yield loss of seed by aphids and stink bugs were 51.9% and 69.8%, respectively. Also, the reduction rates of leaf biomass by S. recurvalis and S. exigua were 72.5% and 36.5%, respectively.

  • Extracts of 43 species of plants were tested for their insecticidal and acaricidal activities against six species of insect pests and one mite species. The methanol extract of Ginkgo biloba leaves selectively was found to have potent insecticidal activity against Nilaparvata lugens (Stal), whereas steam distillate of Thujopsis dolabarata var. hondai sawdust showed potent insecticidal activities with a broad spectrum. The methanol extract of Pinus densiflora leaves and steam distillate of T. dolabrata var. hondai sawdust exhibited potent activities against Tetranychus urticae (Koch).

  • This experiment was conducted to select prominent microorganisms with a good insecticidal activity among the ten species, which isolated from soil at the near of Chung-buk, Chung-nam, and Gang-won provinces and made protein crystal endotoxin. As a result, GB-413 strain was finally selected, which showed the high insecticidal activity against susceptible diamondback moth (Plutella xylostella), beet army worm (Spodoptera exigua) and tobacco cutworm (Spodoptera litura) as well as resistant diamondback moth strains. By modifying the cultivation process f.g. lowing the glucose concentration at early cultivation stage and adding the carbon after inducing the spores, the percentage of making spore as well as the number of active spore were increased and the time for cultivation and spore forming was reduced without a reduction of insecticidal activity. These results were not only applied successfully for the optimized cultivation process for a fermentation tank containing five tons capacity, but also improved the possibility of mass cultivation for commercial production.

  • The CryIIA gene encoding the insecticidal crystal protein of Bacillus thuringiens!s subsp. kurstalri HD-l has been cloned in Escherichia col!, and its nucleotide sequences were determined completely. 5kb Hindlli fragment harboring CryIIA gene was screened in the large ca. 225kb plasmid DNA by southern blot. HindlIT digested 5kb fragment was ligated into pUC19 and transformed in E. coli. The 4kb BamHI-HindlIT fragment containing the CryIIA gene was subcloned and named pSKIIA. DNA sequence analysis demonstrates that pSKIIA is the gene of an operon which is comprised of Lhree open reading frames (designated orn, orf2 and or£3). The CrylIA gene is composed of 3,952bp-long BamHI-Hindill DNA restriction fragment. The orf3 code for a polypeptide of 633 amino acid residues. The protoxin protein has a predicted molecular weight of 70,780. The E. coli derived protoxin gene product is biologICally active against three species of Lepidopteran (Plu.lelia maculipennis, He/iolhis assulta, Spodoptera litura) and a species of Dip Leran( Culex pipines) larvae in bioassay.

Phytophagous Insect Fauna of Dicotyledoneae(Tracheophyta : Angiospermae) Weeds (쌍자엽 식물아강(관속식물문 : 피자식물강)의 잡초가해 곤충상)

  • Phytophagous insects associated with Dicotyledoneae weeds and host specificities in the field populations were investigated for the survey of biological control agents of weeds in Korea. Fifty four weed species in 39 genera were collected during the survey. The most insects were collected from Polygonales by 24 species in 22 genera and followed by Urticales and Centrospermales by 17 species of 17 genera. The insects collected in the other weed orders were ranged from 1 to 12 species. Out of 17 insect species collected in Urticales, Baris sp. damaged the leaves of Hamulus japonicus in Cannabinaceae as scattered holeshape and showed host specificity. In Polygonaceae, Rumex japonicus and R. crispus were severely damaged by Aphis rumicis and Gastrophysa atrocyanea. G. atrocyanea leaf beetle had host specificity on R. japonicus and ate all the leaves except veins. The leaf beetle, Lypesthes japonicus was a potential biological control agent by feeding leaves of Persicaria spp .. And Lixus spp. were also often collected from Persicaria spp .. Liothrips vaneeckei was first collected from weed, P. modosa. P. senticosa was damaged by unidentified geometrid moth larvae and P. perfoiliata by Miarus atricolor snout beetle. Cassida piperata damaged leaves of Chenopodium album of Centrospermales and showed host specificity. In a soybean field, C. album and Amaranthus mangostanus were severely damaged by Spodoptera litura larvae which were eating soybean leaves. This phenomenon indicates that the presence of weed in cultivated land influences the outbreak of insect pests. Altica oleracea leaf beetle was frequently collected from Oenothera spp. of Onagraceae in Myrtales. Aphis gossyphi was outbroken on Solanum nigrum and Phylliodes brettinghami leaf beetle was first recorded on the same plant. Leaf beetles, Longitarsus scutellais and Hemipyxis plagioderoides were first collected from Plantago asiatica of Plantaginaceae in Plantaginales. They showed host specificities in the fields. The hemipterans were collected from many weeds during the survey and their roles on weeds should be investigated. A tractomorpha bedeli was also collected from many kinds of weeds in forest areas.

Synthesis and biological activities of Chloronicotinyl derivatives (Chloronicotinyl 유도체의 합성 및 생물활성 검정)

  • Chloronicotinyl derivatives were synthesized by substitution of amino in 3-pyridylmethylamine with phosphite groups and their insecticidal and fungicidal activities were determined. At 500 ppm, compound 4 with methyl and butyl group in phosphonate and compound 5, 6, 7, and 8 with two butyl, 2,2,2-trifluorotehtyl, 2-ethylhexyl, phenyl, respectively, in phosphonate showed 90% insecticidal activities against brown plant-hopper (Nilaparvate lugens). These compounds showed, however, poor insecticidal activities against diamond-back moth (Plutella xylostella) and two-spotted spider mite (Tetranychus urticae) (<65%), suggesting that insecticidal activity of chloronicotinyl derivatives containing phosphorus moieties are species-dependent. Newly synthesized chloronicotinyl derivatives with halogen and/or heterocycle (compound $10{\sim}21$) did not show insecticidal activities. We also determined fungicidal activity of the synthesized chloronicotinyl derivatives against rice sheath blight (Pyricularia grisea), cucumber gray mold (Bortytis cinerea), tomato late blight (Phytophthora infestans), wheat leaf rust (Puccinia recondita), and barley powdery mildew (Erysiphe graminis). Compound 10 with butyl and 4-nitrophenyl in phosphonate at 10 ppm showed 85% fungicidal activity against rice blast, suggesting that chloronicotinyl derivatives containing phosphorus moieties could be developed as a fungicidal agent of a novel chemical structure.

Applications of Agro-Based Materials for Water Dropwort (Oenanthe stolonifera DC) Organic Farming (미나리 유기재배를 위한 활용자재 시용효과)

  • Organic farming is a type of agricultural practices based on naturally occurring processes excluding or strictly limiting the use of synthetic fertilizers, pesticides, and other chemicals. This study was conducted to investigate the influences of agro-based materials, effective microorganisms (EM), liquid silicate (LS), and organic liquid fertilizer (OLF) for water dropwort (Oenanthe stolonifera DC.) cultivation. Soil pH, soil organic matter, and plant available phosphorous decreased with LS application. Exchangeable Ca and Mg decreased with EM application, and electrical conductivity and exchangeable Ca and K decreased with OLF application. Most of essential nutrient contents in water dropwort were reduced with the treatments of LS, EM, and OLF as compared with those in control plot, except nitrogen and phosphorus. However, diseases and insect pests were almost not observed in the water dropwort in the agro-based material application plots, except cluster caterpillar (Spodoptera litura). Productivity of water dropwort tended to be reduced: its higher productivity in the OLF and EM+LS plots and lower in the LS and control plots.
