Optimization of Proteolytic Enzyme Treatment for the Production of Spirulina Extract (단백질 분해 효소를 이용한 스피루리나 추출물 제조 공정 최적화)

  • An efficient production method of spirulina extract was developed by enzymatic treatment using proteolytic enzymes. The suitable dosage of Tunicase, a cell lytic enzyme, was used to be 2.0% (w/w). To maximize solid recovery and spirulina extraction (SE) index, which indicates nucleic acid-related substances content, the dosage of Alcalase, commercially available pretense, was found to be 1.0% (w/w). By simultaneous treatments using optimal dosages of Tunicase and Alcalase, the highest SE index and solid recovery were obtained. The SE index and solid recovery of simultaneous treatments were notably enhanced by 100% ($11.4%\;{\rightarrow}\;22.8%$) and 56% ($45.2%\;{\rightarrow}\;70.7%$), respectively, than those of the non-treated extracts.

  • 분자 생물학의 발전염기서열, 단백질 서열, 지놈 서열 등의 서열데이터베이스와 단백질 3차구조를 제공하는 구조 데이터베이스등이 구축되어서 웹을 통해 많은 정보를 제공하고 있다. 전세계적으로 분산되어 있는 다양한 생물정보 데이터베이스의 효율적인 검색을 위해서 통합 검색 시스템의 개발이 필요하다. 이 논문에서는 전세계의 생물정보 데이터베이스의 개발 현황을 보이고 분산되어 있는 생물정보데이터베이스로부터 통합검색을 위한 생물정보 통합검색시스템(GenPlus)를 제안하였다. 제안한 GenPlus 에서는 염기 서열, 단백질서열, 그리고 키워드를 이용한 서열정보, 구조정보,완전한 지놈 정보, 그리고 문헌정보의 통합 검색을 제공한다.

  • 점농어, Lateolabrax maculatus는 옛부터 우리나라의 횟감으로 이용되던 어종으로 대 크기가 100cm를 넘는 대형종이다. 그리고, 성장이 빠르고 담수적응력이 강할 뿐만 아니라 수온변화에도 강해 해상가두리 및 축제식 양식대상어종으로 선호하고 있는 실정이다. 이러한 점농어의 양식활성화를 위해서는 전용 배합사료의 개발이 시급하다. 그러나 배합사료개발에 관한 연구는 거의 없어 효율적인 해면양식이 이루어지고 있지 않다. 본 연구에서는 점농어의 생산량 증대를 위한 일련의 연구로 점농어 배합사료 개발에 필요한 기초자료를 제공하기 위하여 단백질 수준을 달리한 사료를 제작, 공급하여 점농어의 사료내 단백질 요구량 수준을 알아보고자 실시하였다.   • This article evaluated levels of Estimated Average Requirements (EARs), Reference Nutrient Intakes (RNIs), and Acceptable Macronutrient Distribution Ranges (AMDRs) of protein using the recently revised Dietary Reference Intakes (DRIs) for Koreans (2020). Dietary protein requirements are based on amounts sufficient to satisfy physiological demands to accomplish nitrogen equilibrium. The same principle was applied to estimate EARs and RNIs, for adults in DRIs conducted in 2015 and 2020 in Koreans. EAR was estimated to be 0.73 g/kg body weight/day, according to data (0.66 g/kg body weight/day) obtained using the nitrogen balance method and adjusted by efficiency of protein use (90%). RNI was calculated as EAR increased by an amount equal to twice the standard deviation of an age group so as to cover 97.5% of the group and was found to be 0.91 g/kg body weight/day. For weaned infants, children, and adolescents, growth requirement was added to estimate EAR. In particular, growth requirement was adjusted by efficiency of protein use in the revised EAR, which led to higher EARs for weaned infants, children, and adolescents of both genders as compared with 2015 DRIs. The AMDR for protein was set at 7%-20% of energy intake, which was the same as 2015 DRIs. Current, average protein intake by the Korean population is almost twice times the EAR, which suggests it might be better to increase the minimal margin for AMDR. However, it was not adjusted in this revision due to lack of evidence.

  • PDB에서 제공하는 단백질 3차원 고분자결정 구조에 대한 플랫파일은 인자들의 좌표, 서열정보, 실험정보 및 참조 정보가 포함된다. 이러한 정보를 포함하고 있는 플랫파일로부터 필수적인 구조정보 및 서열정보 등의 효율적인 검색을 위해서는 이러한 데이터를 추출하여 데이터베이스 구축이 요구되며 이 때 단백질 구조 및 서열 정보와 실험 및 탐조 정보의 관계에 대한 모델링이 중요하다. 따라서 이 논문에서는 PDB에서 제공하는 플랫파일들의 엔트리들을 분석하고 3차원 공간 객체의 기하적 특성을 갖는 단백질 3차 구조를 공간객체로 표현하고 공간객체 모델을 적용하여 모델링한다. 이렇게 함으로써 단백질 3차 구조 분자를 구성하는 인자 및 구조 정보 검색이 가능하며 위상 및 기하 연산자글 이용하여 단백질 구조 분석에 활용할 수 있다.

Nutritive Effects of Feeding Rice Diet Mixed with Barley and/or Millet on the Growth Rate and Biological Availability of Some Nutrients by Albino rats (보리와 속(粟)의 혼식(混食)이 성장기(成長期) 백서(白鼠)에 미치는 영양효과(營養效果))

  • This study was carried out to observe the nutritive effects of feeding rice diet and rice diet mixed with barley or millet, or both on the growth rate and biological availability of some nutrients by albino rats. The experimental diets were prepared on the basis of isocaloric and isonitrogenous containing 357 kcal of energy and 12g of protein per 100g of diet. The experimental animals weighing about 66g of both sexes were fed on 7 kinds of diets such as control diet, rice (100%) diet, rice (70%)+barley (30%) diet, rice (70%)+millet (30%) diet, rice (70%)+barley (20%)+millet (10%) diet, rice (70%)+barley (15%)+millet (15%) diet, and rice (70%)+barley (10%)+millet (20%) diet for 7 weeks. The results obtained are summarized as follows; 1. The gain in body weight was higher for barley and/or millet mixed with rice diet groups than rice diet group with no statistical difference. 2. Although there was no difference in the amount of food consumed by experimental groups, the food efficiency ratio was sightly higher for the miked diet groups than rice atone diet group. 3. The protein efficiency ratio was also higher for barley and millet miked with rice diet groups than rice diet group, although statistical significance was not found. 4. Apparent digestibility of protein of rice diet group was significantly (P<0.01) higher than any other diet group. Although there was no remarkable difference between mixed diet group was found, the apparent digestibility of protein tended to increase when rats were fed on the barley and millet mixed with rice diet. Apparent biological value (p<0.05) and net protein utilization (p<0.01) were also significantly higher for the groups fed mixed diet with barley and/or millet than rice diet group, and those for millet alone mixed diet were slightly lower. 5. The content of total nitrogen in the liver and of protein in serum were not significantly different among experimental groups. It may be concluded from the above results that an adequate supplementation of rice with other cereals and mixing ratio of other cereals to rice were important for the efficient utilization of protein in total diet.

  • Feeding trials were conducted for a test of the optimum dietary protein levels for the fingerling Korean catfish, Parasilurus asotus. The growth response was examined in terms of weight gain, feed coefficient. protein efficiency ratio and net protein utilization for 8 weeks at $23\~27^{\circ}C$. Within a range of 25 to 55\%$ crude protein levels in the diet, the body weight increased while the dietary protein level increased. Accumulation of protein in the body reached the maximum when the crude protein level in the diet was at $45\%$. These results indicate that the optimum dietary protein level of fingerling Korean catfish was about $45\%$ when anchovy meal was used as the protein source.
