Close relatedness of Acanthomoeba pintulosa with Accnthcmoebc palestinensis based on isoenzyme profiles and rDNA PCR-RFLP patterns (Acanthamoeba pustulosa와 A. palestinensis의 동위효소 및 rDNA PCR-RFLP 양상의 유사성)

  • The taxonomic validity of morphological group III Accnthamoeba app. is uncertain. In the present study. six type strains of group III Aconthamoeba spry. , A. culbertsoni, A. heniyi, A. pustulosc, A. palestinensis, A. royrebn and A. lenticulnto were subjected for the evaluation or their taxonomic validity by comparison of the isoeneyme patterns by isoelectic focusing on polyacrylamide gels, mitochondrial DNA (Mt DNA) restriction fragment length polymorphism (RFLP) . and small subunit ribosomal DNA (ssu rDNA) PCR-RFLP patterns. The Mt DNA RFLP patterns were heterogeneous between the species. The type strains of A. pclestinensls and A. pustulosc showed almost identical patterns of isoenrymes and rDNA PCR-RFLP with an estimated sequence divergence of 2.6%. The other species showed heterogeneous patterns of isoenxymes and rDNA PCR- RFLP. It is likely that A. pustuLosc is closely related with A. palestinensis and that the former may be regarded as a junior synonym of the latter.

  • 최근 천연물을 중심으로 한 학문이 발전하면서 천연물이 가지는 생리활성 물질에 대한 관심이 중대되고 있다. 또한 인공합성품의 일부가 안정성의 문제가 제기되면서 천연물의 이용분야는 더욱 확대되고 있다. 급속한 경제발전과 생활수준의 향상으로 식생활을 포함한 생활방식의 다양화로 인하여 과거 감염 위주의 질병이 감소하고 선진국형의 만성퇴행성 질환이 증가하는 추세이다. 혈소판은 혈전증(thrombosis)과 지혈증(haemstasis)에서 중요한 역할을 담당하고 있는 인자로서 현관 내 병적 이상으로 인한 과도한 혈전의 생성은 뇌 -심혈관계 질환의 중요한 유발인자로 작용하므로 뇌ㆍ심혈관계 질환이나 항고지혈증을 연구하는데 유용하게 이용되고 있다. 본 연구에서는 토끼 혈액에서 분리한 세정 혈소판 부유액을 이용하여 작약 MeOH 추출물에서 분리한 천연생리활성물질들을 대상으로 혈소판 응집억제활성에 대하여 연구하였다. 작약 MeOH 추출물의 혈소판 응집억제활성 측정에서 강한 혈소판 응집억제활성 작용을 보였다. 따라서 작약 MeOH 추출물을 크로마토그라피법을 이용하여 분리하였고, NMR을 이용한 분광학적 방법으로 지금까지의 분리한 자료와 비교ㆍ분석하였다. Monoterpene glycoside 계열의 성분들인 15개의 compound와 다수의 fraction들을 HPLC를 이용하여 분리하였으며, collagen으로 유도된 혈소판 응집억제활성측정 방법에서 뛰어난 응집억제활성을 보였다. 표준물질을 이용한 HPLC 분석과 ¹H-NMR 관련 자료의 검색을 통하여 최종적으로 compound 1b가 benzoyloxy-paeoniflorin(2.6%). compound 1d가 paeond(1.3%), compound 2c가 albiflorin(3.2%), compound 2e가 paeoniflorin(33.6%)임을 확인할 수 있었다. Compound 3a의 분석결과 benzoyloxypaeoniflorin과 구조적 유사성은 있으나 동일한 구조식으로 확인할 수 없었다. 그러나 collagen에서 응집억제활성이 90% 이상으로 뛰어난 활성을 나타내므로 benzoyloxypaeoniflorin과 유사한 구조에서 benzoyl group이 다른 작용기로 치환되었거나 R₁ group이 다른 작용기로 치환된 형태로 추측하였다. Benzoyloxypaeoniflorin은 collagen>thrombin>U46616>A.A(arachidonic acid)>PAF의 순으로 활성을 보였다. 이는 paeoniflorin의 glycoside 5-carbon위치에 위치한 OH기 대신에 benzoyl기로 치환된 benzoyl 기가 혈소판 억제 산물로 작용한 것으로 추측했다. Paeoniflorin.은 U46619>thrombin>collagen>A.A>PAF순으로 억제를 보였다. Paeoniflorin이 collagen보다 thrombin에서 강한 억제를 보이는 것으로 Ca/sup 2+/ chelate를 형성함으로 인해 calciu 대사를 저해하는 것으로 추축했다. Compound 3a는 U46619>collagen>A.A>thrombin>PAF순으로 억제율을 보이므로 이 화합물은 paeoniflorin의 benzoyl기에 OH기가 다른 치환기로 바뀌거나 paeoniflorin의 glycoside 5-carbon 위치의 OH기 대신에 다른 작용기로 치환된 것으로 추정하였다. 이러한 결과로 작약의 주성분인 paeoniflorin과 유사한 구조를 가진 다른 monoterpene glycoside 계열의 화합물들과 비교 분석하고 구조를 화인하고 이들 성분이 어떻게 혈소판 응집억제활성에 작용하는지를 연구하였다.

Estimation Error and Reliability of Measuring Unit Water Content Test Methods for Fresh Concrete Depending on Mix Design Factors at the Laboratory Level (실험실 수준에서 배합변수별 굳지 않은 콘크리트 단위수량 실험방법의 추정오차 및 신뢰성 검토)

  • In this study, water content tests were performed on various fresh concretes subjected to different binder compostions to review the estimation errors and reliability of water content test methods. Micro-oven drying method, air-meter method, capacitance method and microwave penetration method were used to estimate water content of fresh concrete. Errors in water content estimation were analyzed by each test method. Regardless of the test method of water content, the estimation error was less than 5 %, and in the case of the test using mortar, the error in the estimation value was relatively large. In addition, based on the test results of water content of various concrete, the probability density function in which the estimation error for each test method becomes the population was analyzed. Water content test methods of fresh concrete which using concrete samples showed high estimate reliability of 97 % within the estimation error range of ± 10 kg/m3. On the other hand, the reliability of water content test method using mortar samples was lower.

실리콘 나노와이어의 산소 흡착 표면 처리를 통한 초소수성 구현

  • 최근 나노기술의 비약적인 발전을 바탕으로 그 동안 구현이 쉽지 않았던 마이크로-나노 단위의 생체모사(biomimetics) 기술이 큰 각광을 받고 있다. 그 중에서도 특히 연잎 효과(lotus-effect)로 대표되는 접촉각 $150^{\circ}$ 이상의 초소수성(superhydrophobicity) 표면 구현은 생물, 화학, 물질 등의 다양한 분야에 있어 큰 사용가치를 가지기 때문에 연구가 전세계적으로 활발히 진행되고 있다. 초소수성을 가지는 표면을 구현하기 위해서는 표면의 화학적인 조성을 변화시켜 표면의 거칠기를 증대시키는 방법과 표면에너지를 낮추는 방법으로 구분될 수 있으며, 이를 위해 표면에 나노구조체를 형성시켜 표면 거칠기를 증대시키는 방법과 silane 계열의 자가-형성 단일막(Self-assembled monolayer)을 코팅하여 표면에너지를 낮추는 방법이 사용되어 왔다. 그러나 표면에 나노구조체를 형성시키는 과정에서 비싼 공정 비용이 발생하며, 대면적 구현이 쉽지 않다는 단점이 있으며, silane 계열의 자가-형성 단일막의 경우에는 제거가 쉽지 않아 추후 다양한 소자에의 적용이 어렵다는 단점을 가지고 있다. 본 연구에서는 무전해 식각법(Aqueous Electroless Etching)을 이용하여 대면적으로 합성시킨 실리콘 나노와이어의 표면 산소 흡착 처리를 통해 $156^{\circ}$ 이상의 초소수성 표면을 구현하였다. 액상 기반으로 형성된 실리콘 나노와이어의 표면은 열처리 공정을 통해 OH-기에서 O-기로 치환되어 낮은 표면에너지를 가지게 되며, 낮아진 표면에너지와 산화과정에서 증대된 표면 거칠기를 통해 Wenzel-state의 초소수성 표면 성질을 보였다. 변화된 나노와이어의 표면 거칠기는 주사전자현미경 (FE-SEM)과 주사투과현미경 (HR-TEM)을 통해 관찰되었다. 또한, 나노와이어의 길이와 열처리 공정 조건에 따라 나노와이어의 표면을 접촉각 $0^{\circ}$의 초친수성(superhydrophilicity) 특성부터 접촉각 $150^{\circ}$ 이상의 초소수성 특성까지 변화시킬 수 있었으며, 나노와이어의 길이에 따라 표면 난반사율을 조절하여 90% 이상의 매우 높은 흡수율을 가지는 나노와이어 표면을 구현할 수 있었다. 이러한 산소 흡착법을 이용한 초소수성 표면 구현은 기존 자가-형성 단일막 코팅을 이용한 방법에 비해 소자 제작 및 활용에 있어 매우 유리하며, 바이오칩, 수광소자 등의 다양한 응용 분야에 적용 가능할 것으로 예상된다.

Mitochondrial DNA Swquence Variation of the Firefly, Pyrocoelia rufa(Coleoptera: Lampyridae), in Korea (늦반딧불이 Pyrocoelis rufa(딱정벌레목: 반딧불이과)의 미토콘드리아 DNA 염기서열 변이)

  • We have sequenced a portion of mitochondrial CO! gene (403 bp) of the firefly, Pyrocoelia rufa, to investigate genetic diversity within population, geographic variation, and phylogenetic relationships among haplotypes. A total of seven mtDNA haplotypes ranging in sequence divergence from 0.2% to 1.2% were obtained from 26 fireflies collected at four localities in Korea: Namhae, Pusan, Muju, and Yongin. The samples collected at the urban area, Pusan, were all fixed with one haplotype, differently those collected at the forest and/or agricultural areas. This appears to suggest that habitat fragmentation and population bottleneck caused by urbanization might have been severe in Pusan. On the other hand, from Muju known as the largest habitat and sanctuary for the firefly, four haplotypes with the maximum sequence divergence of 1.0% were obtained, and this estimate was the highest among the areas studied. The fireflies collected at the isolated islet, Namhae, revealed relatively low haplotype diversity(H=0.25), but one haplotype (PR7) was phylogenetically differentiated from others. This phenomenon was explained in terms of biogeographic history of the island and gene flow in the recent past. Grouping of Muju- Y ongin and Pusan-Namhae, respectively, in the hierarchical genetic analysis suggests the presence of historically occurred, biogeographic barrier against gene flow between them.

Dimensional Change of PEG-Freeze Dried Waterlogged Woods Exposed at Various Humidity Conditions (PEG처리 후 동결건조한 수침고목재의 습도조건에 따른 상태변화)

  • In order to evaluate two-step treatment of PEG-freeze drying for highly-degraded waterlogged ash woods (Fraxinus PP.; ca. 5,700 BP), which were excavated from peat lands in western Korea, dimension stability was examined during 45 months after complete treatment. The samples pre-treated with PEG in water solution showed better dimensional stabilities than the ones with PEG in t-butanol(TBA) solution. It suggests that TBA reduced the flexibility of wood cells and overflying by TBA induced micro-checks during freeze drying. Micro-checks results in fragile wood structures and consequently, large shrinkage by moisture absorbances of high PEG contents during exposure in humid condition. The results suggest that PEG in water-solution treatment is better than PEG in t-butanol as pretreament for freeze drying of highly-degraded waterlogged ash woods.

Shear Strength of Interface between Natural Aggregate Concrete and Recycled Aggregate Concrete (천연골재 콘크리트와 순환골재 콘크리트 접합면의 전단강도)

  • Concrete recycling is becoming mandatory rather selective due to depletion of constructional materials and increase of concrete waste. Studies on recycling concrete are conducted in various point of view for long time. However, standard or guideline of many countries for the application of recycled aggregate concrete(RAC) has restrictions such as low replacement rate of coarse aggregate and no fine aggregate allowed due to inferior material properties of recycled aggregate. This study intends to figure out the feasibility of casting natural aggregate concrete(NAC) and RAC separately in a structural member. In making RAC, replacement rate of coarse aggregate was 50, 100% in RAC and treatment of interface of two concretes is introduced. RAC treatment of recycled aggregate or inclusion of additives was not done as it can increase embodied energy of concrete work. Double-shear test with uniformly distributed loading was adopted to evaluate shear strength at the interface of two concretes. After curing it was hard to distinguish interface of two concretes. Experimental result revealed that specimen with higher replacement rate showed higher shear-to-compressive strength ratio, which is possibly attributed to coarse aggregate size and roughness of sheared section. Further study on the effect of various parameters is required and subsequent research activity is on-going.

The Prediction of Durability Performance for Chloride Ingress in Fly Ash Concrete by Artificial Neural Network Algorithm (인공 신경망 알고리즘을 활용한 플라이애시 콘크리트의 염해 내구성능 예측)

  • In this study, RCPTs (Rapid Chloride Penetration Test) were performed for fly ash concrete with curing age of 4 ~ 6 years. The concrete mixtures were prepared with 3 levels of water to binder ratio (0.37, 0.42, and 0.47) and 2 levels of substitution ratio of fly ash (0 and 30%), and the improved passed charges of chloride ion behavior were quantitatively analyzed. Additionally, the results were trained through the univariate time series models consisted of GRU (Gated Recurrent Unit) algorithm and those from the models were evaluated. As the result of the RCPT, fly ash concrete showed the reduced passed charges with period and an more improved resistance to chloride penetration than OPC concrete. At the final evaluation period (6 years), fly ash concrete showed 'Very low' grade in all W/B (water to binder) ratio, however OPC concrete showed 'Moderate' grade in the condition with the highest W/B ratio (0.47). The adopted algorithm of GRU for this study can analyze time series data and has the advantage like operation efficiency. The deep learning model with 4 hidden layers was designed, and it provided a reasonable prediction results of passed charge. The deep learning model from this study has a limitation of single consideration of a univariate time series characteristic, but it is in the developing process of providing various characteristics of concrete like strength and diffusion coefficient through additional studies.

  • Since the cement industry generates more than 60 % of CO2 during the clinker production process, supplementary cementitious materials are used worldwide to reduce CO2 efficiently. Mainly used supplementary cementitious materials such as blast furnace slag and fly ash, which are used in various industries including the cement industry, concrete admixtures, and ground solidification materials. However, since their availability is expected to decrease in the future according to the carbon neutrality strategy of each industry, new supplementary cementitious materials should be used to achieve the cement industry's goal for increasing the additive content of Portland cement. Limestone is a material that already has a large amount in the cement industry and has the advantage of high grinding efficiency, so overseas developed countries established Portland limestone cement standards and succeeded in commercialization. This study was an experimental study conducted to evaluate the possibility of utilizing domestic PLC, the effect of fineness and replacement ratio on the physical properties of cement was investigated, and the environmental impact of cement was evaluated by analyzing CO2 emissions.

A Study on the Stress Evaluation Equations for Steel Circular Column-to- Box Beam Connections (강재 상자형보-원형기둥 접합부의 응력평가식에 관한 연구)

  • This paper presented equations on the stress evaluation of steel frame pier connections that were composed of a box beam and a circular column. The existing equations, which transformed the circular column into an equivalent box column had some problems; they underestimated a shear lag stress as the joint angle decreased, and overestimated a shear stress as the joint angel increased. Therefore, FE analyses were performed with various parameters, such as joint angle(${\alpha}$), span length-width ratio(L/B), and circular column-to-box beam stiffness ratio(${\alpha}$), and new equations on stress evaluation were proposed based on FE analyses. Furthermore, material and geometric nonlinear analyses were performed to estimate ultimate strength and to confirm the validity of the proposed equations.
