Bioconcentration of Perfluorinated Compounds in Fish from Gulpo Stream (굴포천에 서식하는 어류체내 과불화화합물 농축특성)

  • During the last decade, perfluorinated compounds (PFCs) have gained more attention due to their toxicity and global distribution. The aim of this study was to examine the distribution and bioaccumulation of perfluorinated compounds (PFCs) in aquatic wildlife effected from a sewage treatment plant. The concentrations of 12 PFCs were determined in water, sediment and fish samples. PFOS were predominantly detected in both ambient environment and fish. In fish, the concentration of PFCs in blood was the highest (i.e., 112.47 ng/mL wet-wt. PFOS) in comparison to other tissues. However, PFOA and PFHpS were highly detected in gonad as 3.87 and 4.58 ng/g wet-wt., respectively. The bioconcentration factor (BCF) of PFCs was greatest in the blood > liver${\cong}$gonad > kidney > gill, and lowest in the muscle tissue. The BCFs of PFUnDA (39,000), PFDA (2,700) and PFOS (1,100) were rated as high values based on wet weight concentration. BCFs increased with increasing the length of the perfluoralkyl chain.

  • In recently, a climatic change(such as subtropical climate and frequent unusual high temperature) and the open-trade policies of agricultural & livestock products are increasing the outbreak risk of highly pathogenic avian influenza(HPAI) and foot and mouth disease(FMD), and accordingly the socio-economic damage and impacts are also increasing due to the cases such as damage from the last 5 times of FMD outbreak(3,800 billion won), from 10 years public control cost of Pine Wilt Disease (PWD)(238.3 billion won), and from the increased invasive pests of exotic plant like isoptera. Therefore, the establishment of new operation strategy of IoT(Internet of Things) based satellite early warning system(SEWS) for plants and animals as a subsystem of national disaster response and management system is being required, where the forensic technology & measures should be applied as a government policy to estimate the post compensation and to carry out the legal responsibility.

Hepatic Detoxification activity and reduction of Serum Alcohol concentration of Hovenia dulcis $T_{HUNB}$ from Korea and China (중국산과 국내산 헛개 나무 열매의 체내 알콜 분해능 및 간 해독 작용)

  • There was not noticeable differences in decreasing blood alcohol concentrations between Korea and China-produced Hovenia dulcis $T_{HUNB}$, showing only 1-2 % higher decreasing rate for Korea-produced seed extracts than those from China. It was also found that the blood alcohol decreasing ability was greatly enhanced by partitioning the crude extracts produced from both places. The both extracts (crude and partitioned) accelerated the reducing rate of blood alcohol concentrations down to 1-2 hours, compared to that of control (taking only ethanol). The crude extracts from imported seeds seemed to have slightly better effect on improving in vivo ADH and ALDH activities than domestic ones; however, not for partitioned extracts. It was interesting that the partitioned extracts from both countries enhanced ADH enzyme activity up to 60% than the crude, compared to the control, while ALDH activity was not much affected by the partitioned extracts. It was also confirmed that both ADH and ALDH activities were well balanced in controlling blood alcohol concentration maintaining 28-29% of enzyme activities in vivo. The extracts proved to have better effect on enhancing ALDH activity than ADH activity, which is one of possible explanation that Hovenia dulcis $T_{HUNB}$ can effectively relieve the hangover by fast decreasing acetaldehyde concentration in the liver and blood. GST activity was also increased in the range of 120 to 300% by adding crude or partitioned extracts from both countries; however, there was no difference in enhancing GST activity between the extracts from two countries. The extracts showed competitive inhibition with GST activity, showing the reduction of enzyme activity at higher than 0. 6 (g/L) of the imported extracts.

Method for Supplementing Lecithin to Ginseng Extract (레시틴이 강화된 인삼 추출물 제조 방법)

  • This study was carried out to develop the method of preparing lecithin-fortified ginseng extract. Firstly, soybean lecithin was mixed with soybean oil (LCS) in varying ratio (2.5%, 5%, 10% and 20%). Then, one part volume of LCS was mixed with three parts volume of ginseng extract with 10% solid matter content and the mixture was vortexed vigorously. Finally, the mixture was spinned at the speed of 3,000 rpm for 30 minutes to separate oil and aqueous ginseng extract layer (AG). AG was then subjected to qualitative and quantitative analysis of phospholipids and ginsenosides. Fatty acid composition and crude fat content before and after LCS was determined. Stability of lecithin in ginseng extract was determined by analyzing phospholipid content in the one third upper and lower layer of the concentrated AG in Falcon tubes while storing the LCS treated concentrated AG in 4, 25 and 40oC for 6 months. Ratio of lecithin transferred to AG increased with the increase in lecithin content of soybean oil. There was no significant change in fatty acid composition and crude fat content, and ginsenoside content in the ginseng extract before and after LCS treatment. TLC and HPLC pattern of saponin fraction before and after treating the ginseng extract with LCS demonstrated no observable difference. There was no change in lecithin content in the upper and lower one third layer of ginseng extract in the tubes after storing the concentrated AG in 4, 25 and $40^{\circ}C$ for 6 months. Ginsenosides HPLC pattern was not changed when stored the LCS-treated ginseng extract in those conditions for six months, indicating satisfiable stability of the LCS-treated concentrated ginseng extract. From these results, it can be concluded that treatment of the ginseng extract with lecithin containing soybean oil is a labor effective method with satisfiable stability to fortify lecithins to ginseng extract.

Antimicrobial Activities and Phenolic Compounds of Pyroligneous Liquor (목초액의 항균활성과 페놀화합물의 함량)

  • Antimicrobial activities of pyroligneous liquor were investigated by determining Minimal Inhibitory Concentration (MIC). The solvent extracts of pyroligneous liquor, which were extracted by using solvents with different polarities such as hexane, ethylacetate, or butanol. The activities were examined by disc diffusion method using MIC against 7 food poisoning microbe type strains. Antimicrobial activities were shown in hexane, ethylacetate, butanol, and aqueous fractions of pyroligneous liquor. Among the four fractions, ethylacetate fraction showed the highest inhibitory effect on the microorganism such as Shigella sonnei, and Yersinia enterocolitica at the concentration of 2.0 mg/disc. The purified P-1 and P-2 fractions isolated by silica gel column chromatography from ethylacetate fraction of pyroligneous liquor had the highest antimicrobial activity. The total phenolic compounds content in ethylacetate, hexane, butanol, and aqueous fraction was 488.3 mg/g, 403.8 mg/g, 83.6mg/g, and 74.5 mg/g, respectively. Taken together, these results suggest that the ethyl acetate fraction could be suitable for the development of isolation and identification of antimicrobial compound from pyroligneous liquor, resulting from the above antimicrobial activity.

ACE Inhibitory Materials from Raja kenojei (홍어의 항고혈압 활성물질)

  • This study was carried out to investigate the ACE inhibitory materials of Raja kenojei. Raja kenojei was sperated to fillet and viscera, and these were extracted with hot water. Antihypertensive activity was examined by mesearing angiotensin converting enzyme ACE inhibitory activity. ACE inhibitory activity of viscera at the concentration of 2% for Day 0 showed the highest value by 71.0%. But ACE inhibitory activity of fillet at 2% showed by 29%, which was lower antihypertensive activity than viscera. The protein content of viscerial hot water extracts in proximate composition showed the highest. And also, there was a large amount of aromatic and branched aliphatic amino acids in viscera than those in fillet. For the purification of antihypertensive material in visceral hot water extracts, it was separated and collected by Sephadex G-25 gel chromatography. The fraction (B) of 111 to 160 showed the highest ACE inhibitory activity by 65.1% at the concentration of 0.05%. But the other fractions (A and C) showed lower activity than B. These results demonstrate that crude hot water extracts of viscera from Raj kenojei may be useful as functional food ingredient with antihypertensive property.

Structure Analysis of Water-soluble Polysaccharides Extracted from The Unripe Fruit of Cudrania tricuspidata (꾸지뽕나무 열매에서 추출한 수용성 다당류의 구조분석)

  • The unripe fruit of cudrania tricuspidata was extracted with 50% ethanol. The crude water-soluble extracts were separated by liquid-liquid separation with n-hexane, ethyl acetate and butanol followed by precipitation with ethanol, and then the water-soluble polysaccharide (F1) was isolated by the fractionation through gel permeation chromatography using preparative PLaquagel-OH column with water. The structure was characterized by monosaccharide composition with HPAEC-PAD, methylation analysis with GC-MS, FT-IR and HPLC. According to the data, F1 was com posed of glucose (22.84 mM), galactose (13.75 mM), arabinose (45.87 mM), xylose (7.49 mM). It was revealed which uronic acid and acetyl group were not attached in F1. And it is constituted of 1-linked arabinose, 1,4-linked arabinose, 1,3-linked glucose, 1,4-linked galactose, 1,4-linked glucose, 1,3,6-linked galactose, 1,3,6-linked glucose and the ratio was showed 1.1 : 1.0 : 4.9 : 7.5 : 3.0 : 3.1 : 1.4 : 1.5.

Study on Concentrations and Mass Flows of Perfluorinated Compounds (PFCs) in a Wastewater Treatment Plant (폐수처리장의 과불화화합물 검출수준 및 처리공정 중 물질흐름 해석에 관한 연구)

  • To determine the concentrations and the mass flow of selected 10 perfluorinated compounds (PFCs), a field study was conducted in a wastewater treatment plant. Raw influent, primary influent, primary effluent, aeration tank effluent, secondary effluent, final effluent, dehydration liquor, primary sludge, thickened sludge, final sludge were collected over 3 days in the summer and the winter respectively. Collected samples were equally mixed and then served as an analytical sample. Total 10 compounds were analyzed. In terms of treated water, the concentration of perfluorooctanesulfonate (PFOS) and perfluorooctanoate (PFOA) were in range of N.D.~26.29 ng/L and N.D.~38.15 ng/L respectively. Perfluorononanoate (PFNA) and perfluorohexanesulfonate (PFHxS) were ranged from N.D. to 36.79 ng/L and from N.D. to 24.36 ng/L. In terms of sludges, a concentration of PFOS, PFOA, and perfluorodecanesulfonate (PFDS) were detected from 6.82 to 59.37 ng/g, from 0.13 to 0.37 ng/g, from N.D. to 0.83 ng/g respectively. Mass loading for PFCs increased during wastewater treatment except for PFNA. The observed increase in mass flow of PFCs may have resulted from biodegradation of precursor compounds.

Quality characteristics and antioxidant activity of yogurt dressing containing black currant extract (블랙커런트 요구르트 첨가 드레싱의 품질특성 및 항산화성)

  • This study was carried out to investigate the quality characteristics and antioxidant activity of yogurt dressing prepared with different amounts (0, 5, 10, 15, and 20%) of black currant (Ribes nigrum L) extract. All samples were evaluated for chemical composition, pH, viscosity, sugar content, color, organic acid content and antioxidant activity. Our findings revealed that with increasing amounts of black currant extract in yogurt dressing, the moisture, crude protein and crude lipid contents decreased. The pH and viscosity gradually decreased, whereas the sugar increased with increase in black currant extract. Further, the lightness (L) value decreased, while the redness (a) and yellowness (b) values increased with increase in added black currant extract. Among the organic acids, the contents of citric acid, malic acid, succinic acid and acetic acid increased, while that of tartaric acid and lactic acid decreased with increasing levels of added black currant extract. The total polyphenol and total anthocyanin contents were 5.90-245.97 mg/100 g and 0.62-190.09 mg/100 g, respectively. In addition, the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-thylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities significantly increased with increase in added black currant extract.

Isolation and Structural Identification of Antioxidant Substances from Ethyl Acetate Extract of Conyza canadensis (망초(Conyza canadensis) Ethyl Acetate 추출물의 항산화성 물질의 분리와 동정)

  • Background: As a result of analyzing the components of wild Conyza canadensis, it contains physiologically active ingredients, so it is necessary to identify the compound. Purposes: It was to study the compound's molecular structure; a previous study showed that C. canadensis contains antioxidant substances. Methods: The ultrasonic pulverized lysate of C. canadensis stem and leaves was first extracted with 90% methanol and then five organic solvents. Next, the extracts was fractionated by HPLC, LC/MS chromatography, and NMR analyzers identified the molecular structure. Results: 100 g of dry C. canadensis was sonicated in 90% methanol and concentrated under reduced pressure to 11.96 g of a crude extract. Then, this crude was extracted with five types of solvents to obtain 123.8 mg of n-hexane, 448.2 mg of dichloromethane, 1047.7 mg of ethyl acetate (EA), 2563.8 mg of butanol, and 7.04 g of water. The EA extracts were fractionated by LC-MS and then re-fractionated to obtain F1 to F20. Next, the F15 was further fractionated to obtain nine fine fractions. Finally, the F17 fraction was re-fractionated to obtain ten fine fractions. As a result of LC-MS and NMR spectrometer analysis of the F15-7, the structure of this compound was confirmed as 3,5-dicaffeoylquinic acid. As a result of examining the structures of the F17-4 and F17-5 fractions, Quercetin-3-o-β-galactose was identified. In addition, the form of the F17-10 was confirmed to be 1,3,4-tri-caffeoylquinic acid. Conclusions: This study demonstrated that C. canadensis contained phenolic antioxidants, and its utilization may be expected.
