Restoration of gold guilding on Geumdong-samjonpanbul excavated from Walji, Gyeongju using plum acid & gold(Au)-mercury(Hg) amalgam (매실산과 금아말감을 이용한 월지 출토 금동삼존판불의 금도금법 복원)

  • 본 연구에서는 고문헌에 기록된 도금 재료와 도금법 등을 확인하고 이를 기초자료로 활용하여 매실산을 사용한 금(Au)-수은(Hg) 아말감기법으로 고대 도금(鍍金)기술을 되살리고, 이를 토대로 경주 월지에서 출토된 금동삼존판불을 복원하였다. 먼저, 전통 도금법을 되살리기 위해서, "오주서종박물고변", "확지신편", 조선시대 각종 의궤 등에 공통적으로 기록되어 있는 매실에 주목하고, 매실을 3~4개월 발효 숙성 후 착즙한 뒤 그것을 농축하여 만든 매실산을 도금 실험에 적용하였다. 금아말감 도금을 위하여 월지 출토 금동삼존판불의 바탕소지금속인 청동삼존판불을 구리와 주석 89:11(Cu:6kg, Su:750g)로 합금하여 주물사 주조법으로 복원하였으며, 동일한 합금비로 제작된 $2.3cm{\times}3.5cm$(가로${\times}$세로)의 시편에 사전 실험을 실시하였다. 현대적 산처리 방식에 사용되는 질산과 전통방식으로 사용되는 매실산으로 시편에 산처리 한 후 각각 비교해 보고, 금분과 금박, 상온과 가온에 따른 아말감상태를 비교하는 실험을 진행하였는데, 실험에 사용된 매실산 70%는 pH가 1.94로, 오늘날 산처리에 사용하고 있는 질산 20%와는 차이가 있지만, 청동 시편 실험을 통해 매실산에 20분 정도 담근 뒤 금아말감을 도포 후 24시간 지나 가열($380{\sim}400^{\circ}C$) 했을 때 금도금이 잘 되어, 현대적 방법인 질산처리로 도금을 한 시편과 큰 차이가 없는 것으로 관찰되었다. 사전 실험을 통한 결과를 적용한 월지 출토 금동삼존판불 복원은 청동삼존판불 표면처리, 금-수은 합금 및 도금하기, 도금 후 표면처리의 순서로 진행되었는데, 금과 소지금속의 밀착력을 높이기 위해 표면을 숯을 이용해 탈지한 후 물로 씻어내고 매실산을 도포하여 20분 동안 두어 부식 및 세척을 시행하였다. 금도금을 위한 금-수은아말감은 가온할 때 수은이 증발하는 양을 고려하여 금1 : 수은10 비율로 합금하여 완성하였으며, 금아말감 도포 후 약 24시간 지난 다음, $380{\sim}400^{\circ}C$에 가열하여 수은을 기화시켜 도금작업을 완성하였다. 금아말감도금은 평균적으로 6~7차례 시행하여야 완벽히 도금되지만, 본 연구에서는 단 4차례의 도금만으로 금아말감도금을 완성시켰는데, 이것은 금아말감을 바탕소지인 청동에 도포한 후 24시간 동안 금아말감과 청동과의 반응 시간을 두게한 것이 큰 역할을 한 것으로 보이며, 이는 청동시편을 이용한 실험과 과학적 분석을 통하여 입증하였다. SEM으로 표면을 관찰한 결과 아말감 도포시간이 즉시인 경우 도금이 거의 되지 않은 것을 확인할 수 있었고 36시간이 넘어갈 경우 금 도금층이 불균일하게 관찰되었으므로 도금시간은 12시간~24시간 이내가 적절함을 확인할 수 있었다. EDS로 성분을 분석한 결과 산처리 시간이 20분인 시료의 경우 5 wt% 내외로 수은의 비율이 다른 시료에 비해 낮은 것을 확인할 수 있었다. 실험 및 분석결과 산처리 시간이 20분이고 아말감 도포시간이 24시간일 때 도금이 잘 이루어지므로 이 결과를 토대로 금동삼존판불을 복원하였다. 이번 연구를 통해 도금법에 표면을 세척하고 부식시키기 위해 사용한 물질이 매실산임을 찾아내어 확인할 수 있었는데, 이러한 점 에서 이 연구의 가장 큰 의미는 전통 소재와 기술을 복원한 것으로, 앞으로 매실산을 이용한 금 도금기술은 관련 학계에도 큰 기여를 할 것으로 기대된다.

Mucilage Separation of Korean Yam Using Microparticulation/Air Classification Process (초미세분쇄/공기분급을 이용한 마의 점질물 분리)

  • To separate and concentrate mucilage from yam(Dioscorea batatas DECNE), yam was dried, microparticulated using impact mill and air-classified at different air classifying wheel speed(ACWS) in classifier. As ACWS increased from 5,000 rpm to 22,500 rpm, the contents of dietary fiber, protein and lipid of air classified microparticles(ACM) increased remarkably. Especially the ACM with ACWS over 15,000 rpm showed 36.41% dietary fiber and 16.66% protein. The dietary fiber and protein components were concentrated to $2.5{\sim}9.0$ times as compared with whole yam powder. Concomitantly the non-fibrous carbohydrate decreased from 88.31% to 16.84. The damaged starch(%), WSI and WAI of ACM of ACWS over 15,000 rpm were $1.5{\sim}3.0$ times higher than those of ACM under ACWS 15,000 rpm. The apparent viscosity of ACM was 0.0800 Pa s over ACWS 15,000 rpm and 0.0080 Pa s under ACWS 15,000 rpm. Judging from viscosity of ACM, the mucilage component of yam was concentrated to 10 times. In conclusion, the optimum process to separate and concentrate the mucilage from yam consisted of the microparticulation to $5{\sim}30{\mu}m$ and the air-classification at ACWS over 15,000 rpm.

  • TEvaporator is key component in food, seawater distillation and waste water treatment system, which is basically to concentrate the raw liquid by evaporating the pure water under vacuum condition. The liquid concentration is performed through the membrane, electro-dialysis and evaporation. In this study, only the evaporating type was treated for evaluating the economic analysis with the various operating conditions. The results of this study showed that the performance of the OT-MSF desalination system is increased with decreasing the temperature difference between the neighboring evaporators, which means that the number of evaporators is increased, under the determined design conditions.

Acid Production by Lactic Acid Bacteria in Soy Milk Treated by Microbial Pretense or Papain and Preparation of Soy Yogurt (미생물 Protease 또는 Papain으로 처리된 두유에서 젖산균의 산생함과 대두요구르트의 제조)

  • The soy milk prepared from soy protein concentrate was treated with microbial protease or papain. Growth and acid production by Lactobacillus acidophilus in soy milk containing partially hydrolyzed proteins were investigated. Sensory evaluation of yogurt beverage prepared from protease treated soy milk was also performed. Protease treatment of soy milk enhanced acid production by lactic acid bacteria, particularly in case of microbial pretense and simultaneous treatment by two types of protease showed synergistic effect. pH and number of viable cells were not affected markedly by pretense treatment. Microbial pretense treatment up to 15 minutes or papain treatment up to 45 minutes enhanced acid production, but further treatment up to three hours did not affect the acidity markedly. rho sensory evaluation showed that overall acceptability and taste of soy yogurt beverage were slightly improved when soy milk was treated with microbial pretense of 0.2% or papain of 0.2%. The amount of non-protein nitrogen considerably increased by pretense treatment of 15 minutes and it increased gradually by further treatment up to three hours.

Effects of Heat Treatment of Soy Milk on Acid Production by Lactic Acid Bacteria and Quality of Soy Yogurt (두유(豆乳)의 가열처리가 젖산균의 산생성(散生成)과 대두요구르트의 품질에 미치는 영향)

  • The effects of various heat treatments of soy milk prepared from soy protein concentrate on growth and acid production by five species of lactic acid bacteria were investigated. Sensory property and sedimentation characteristics of soy yogurt prepared from heat-treated soy milk were also evaluated. Heat treatment of soy milk stimulated acid production by all cultures. Acid production was generally proportional to degree of heat treatment and acid production by all cultures except Streptococcus lactis was maximum in soy milk heated at $121^{\circ}C-1min$. However, viable cell count was not changed markedly by heat treatment of soy milk. Sensory property of soy yogurt beverage (SYB) prepared from soy milk heated at $95^{\circ}C-30min$ was better than that of unheated sample while sensory property of SYB prepared from soy milk heated at $121^{\circ}C-15min$ was inferior to that of unheated sample. Heat treatment of soy milk generally retarded sedimentation of curd in SYB.

Comparative analysis of nitrite scavenging activity and anti-inflammation effects in the fruiting bodies of medicinal mushrooms (약용버섯류 자실체의 아질산염 소거능 및 항염증 효능 분석)

  • This study was carried out to compare the anti-inflammation effects of various fruiting body of Ganoderma species and Cordyceps militaris, Phelinus linteus extracts. We concentrated Ganoderma species and other medicinal mushrooms by extracting with ethanol. And We made it $100{\mu}g/ml$ concentration. As a result of nitrite scavenging activity, in the contrast to the positive control; Ascorbic acid was 25%, ASI 7080 of Ganoderma species was disappeared up to around 40%. And in the contrast to Ascorbic acid was 55%, ASI 7002 was 78.5% that was the highest anti-inflammation effect in the result of "No assay test". The Cordyceps militaris showed 75% and Hericium erinaceus showed 59.7% of anti-inflammation effect. As a result of the fungus yield control test of $TNF-{\alpha}$ through ELISA method to ASI 7002 of Ganoderma species that showed the highest anti-inflammation, it was reduced as same as LPS non-treatment. We extracted RNA from ASI 7002 Ganoderma species 10, 50, $100{\mu}g/ml$ concentration and LPS $10{\mu}g/ml$ of Raw 264.7 cell. And we tested the expression of iNOS, COX-2 and TNF-a that are kinds of inflammation gene after synthesizing RNA with cDNA. Finally we could find that iNOS, COX-2 and TNF-a were all controlled expression in the result of above experiment.

  • In this study, biological treatment process of MWWT(Municipal wet-waste Treatment) has been developed through a moduling of the containerized closed ATAD(Auto thermal aerobic digestion) system & closed vertical dynamic acerator, which were used for food waste and cattle manure, respectively. Though biological process has several advantages such as low concentrations of heavy metals and salts, proper and stable C/N ratio and constant reaction rate against the process treating two wastes separately, it has a obstacles of salt concentration and much usage of bulking agent such as wood chip. After rapid oxidation in the boxed tower reactor for 5 days, the content of sewage sludge would be reduced 65% on around, might be mixed with the food waste that had been treated in the static closed reactor during 6 days and put in the secondary static reactor for curing. During composting process, the odor contained in the gas generated from the reactor was removed by passing it through a biofilter as well as the leachate was treated in the wastewater treatment facility. Consequently, it seemed to be possible to compost sewage sludge at mild and stable operating condition and at low cost through the biological ATAD process resulting in the production of organic compost satisfying the specifications regulated by itself.

Anti-obesity Effect of Chopped Sauce with Medicinal Plant Extracts (약용작물 추출물을 첨가한 다진양념의 비만 억제 효과)

  • 본 연구는 항비만 기능성을 가지면서 맛과 향이 증진된 분말형 다진양념(다데기)을 개발하기 위해 첨가할 수 있는 약용작물들의 선정 및 그 기능성을 평가하기 위해 실시되었다. 오미자, 진피, 감초, 녹차엽 등을 첨가군으로 선정하여 각각의 시료들을 8시간, $80^{\circ}C$ 증류수에서 추출 후 감압농축하여 추출물을 제조한 뒤 마우스 유래 adipocyte (3T3-L1)에 처리하여 세포내 지방 축적 억제 정도를 실험하였다. 시료 처리 배양 5일 후 오미자, 진피, 감초, 녹차엽 추출물을 처리한 모든 군에서 대조군과 비교시 20% 이상 세포내 중성지방 농도가 감소하였으며 특히 오미자 군에서는 30% 이상 감소를 보였다. C57BL/6 생쥐를 이용하여 고지방식이로 비만을 유도하면서 분말형 다진양념, 오미자, 진피, 감초, 녹차엽 추출물을 경구투여한 뒤 체중증가를 조사한 결과 8주 후 고지방식이와 다데기만을 경구투여한 군은 고지방식이군과 비교하여 체중감소경향만을 보였으나 분말형 다진양념과 추출물을 함께 경구투여한군에서는 유의적으로 체중이 감소하는 결과를 보였다. 이들 추출물을 혼합하여 제조한 분말형 다진양념은 관능검사에서도 고춧가루 특유의 풋내를 감소시켜 향미가 증진되는 것으로 평가되어 오미자, 진피, 감초, 녹차엽을 첨가한 다양한 항비만 기능성 양념 개발이 기대된다.

Comparative Study on the Control and Removal of Formaldehyde for the Urea-formaldehyde Resin Bonded Plywood -Adhesive control- (요소수지접착합판(尿素樹脂接着合板)의 유리(遊離)포름알데히드 방산(放散)제거 및 조절방법에 대한 비교연구(比較硏究))

  • This study has been carried out to make a comparative study for the adhesive control methods specifically developed for application to formaldehyde. The method for formaldehyde determination used in this report is the improved chromo tropic acid determination. The results are summarized as follows: 1. The soaking treatment in aqueous solution of urea wok the most scavenging effect on the formaldehyde release from a plywood sample glued with a urea formaldehyde adhesive, and other removal treatment such as resorcinol, albumine-, and hardener-treatment gave significant reduction too. 2. In glue shear strength of dry test, 2% of resorcinol treatment and soaking treatment showed the highest strength and all the other treatment met the standard, but in hot water soaking test, 2% of resorcinol treatment gave the best results, on the other hand, adding the hardener showed the lowest strength and failed in meeting the standard. 3. Air dried moisture content of all treated plywood met the standard which calls for 13% or bellow. 4. In this comparative study, we can make a strong combination each other or go into the details of one treatment for the best result through the more study.

Testing the Potential of Sewage Sludge Gasification Solid Residues as a Circulating Resource by Physical Separation (하수슬러지의 가스화 고형 잔재물의 순환자원으로서 물리적 선별에 의한 잠재성 검토)

  • In this study, physical property evaluation and physical separation of the target product were performed to investigate the possibility of using sewage sludge gasification solid residue (GSRs) as a circulating resource. Firstly, the GSRs used in this study was supplied by Sudokwon Landfill Management Corporation, and generally the GSRs was in the form of porous pellets with a particle size of several millimetres. In addition, the partially black areas were confirmed to be unburned and ungasified carbon, and the average carbon content was 5%. In addition, the content of silica, alumina and phosphorus oxide was more than 70% of the total content. It was confirmed that the metallic components of the wet grinding product were separated into individual elements. As a physical separation of metallic and non-metallic components was required, it was finally found that flotation screening was suitable. Accordingly, cationic and anionic surfactants were selected to separate metallic components in which a relatively large amount of non-metallic components were concentrated, and the separation characteristics were confirmed. As a result, it is expected that the concentration of non-metallic components such as silica, alumina and phosphorus will be easier than the separation of metallic components. Therefore, since it is possible to physically treat the gasified sludge residue, it is judged to have potential as a circular resource according to the proposed recycling method for the separated product.
