Assessment of Immune Quality and Pathogen Contamination of Colostrums Collected from Colostrum Banks in Korea (초유은행에서 수거한 초유의 병원체 오염과 면역수준의 평가)

  • Because colostrum is considered to be the sole source of passively acquired maternal antibodies for calves, newborn calves must consume colostrum to gain disease resistance during their early years of life. Storage of surplus colostrum from dairy cows right after calving and feeding newborn calves in deficiency of colostrum to assure adequate uptake of IgG for protection of the calf has been a common practice in the bovine production. In the current study, 35 colostrums were randomly collected from 3 colostrum banks located in different regions of Korea and monitored for general bacterial contamination and major bovine pathogens. Immunoglobulin concentrations and BVDV-specific antibodies were also determined to evaluate the immune quality of the colostrums. Moderate to severe bacterial contamination (up to 72,000,000 CFU/ml) was observed in most of the colostrums collected from colostrum banks. General immune quality of the colostrums was under the satisfactory level since most of the colostrums contained less than 50 g/L of IgG, which is the minimum concentration for good quality colostrums. Therefore, colostrum for colostrum bank should be collected at the first 2-3 post-partum milkings according to proper harvesting and handling procedures to guarantee the safety and quality of colostrum. In addition, it was recommended that colostrum should be heat-treated before frozen and stored in the bank because pasteurization at $63^{\circ}C$ for 30 min was very effective reducing the risk of disease transmission without causing significant degradation of immunoglobulins.

  • Colostrum provide a complete diet for the newborn calves. In ruminants, colostrum is also the sole source of initial acquired immunity for the offspring. Colostrum, therefore, plays an important role in ruminants host defense. Bovine colostrum imparts passive immunity to newborn calves during the first 24h of life and generally has been fed for the first 3 days after birth. Colostrum can be preserved conveniently for future use by brief refrigeration, freezing, or storage at ambient temperatures (fermentation or chemical treatment). Colostrum management were surveyed via data collection from 67 dairy farms in Gyeonggi and Chungnam provinces. After parturition for five days the total amount of produced colostrum was 80.4 kg on average, and the average amount of lactation fed to a calf was 20.9 kg. According to the survey results, identified treatment methods for surplus colostrum are as follows; disposal 17.9%, other calf feeding 37.3%, storage 22.4%, other livestock feeding 20.9% and other treatment 1.5%. For improvement in the quality of colostrum, there must be controlled usage of medicines (such as antibiotics) before and after parturition, proper choices of milk-drying agents, and increased milking yields during the early stage of the milking period.

The Effect of Chitosan Coating Combined with Carvacrol and Thymol on Microbial and Quality Characteristics of Litopenaeus vannamei during Cold Storage (Carvacrol과 thymol을 병행처리한 키토산 코팅이 냉장저장 시 흰다리 새우의 미생물 및 품질 특성에 미치는 효과)

  • In this study, we stored frozen shrimp (Litopenaeus vannamei) at $4^{\circ}C$ during 12 days and investigated the effect of chitosan coating with natural preservatives (0.05% carvacrol, 0.05% thymol) on the growth of microorganism (mesophilic bacteria, psychrophilic bacteria, Pseudomonas spp., $H_2S$ producing bacteria) and physiological characteristics (total volatile basic nitrogen and pH) and sensory evaluation (appearance, odor and general acceptance). Chitosan coating with natural antimicrobial compounds (0.05% carvacrol and 0.05% thymol) had inhibited the growth of all the target microorganism and showed the significant antimicrobial activity (p < 0.05) to mesophilic bacteria, psychrophilic bacteria and $H_2S$ producing bacteria until 12 day (the last day of this study). These treated groups had showed the significant difference (p < 0.05) in total volatile basic nitrogen and all the sensory characteristics but not in pH. Therefore, chitosan coating combined with natural antimicrobial compounds (0.05% carvacrol and 0.05% thymol) showed the effective antimicrobial activity on major spoilage microorganism on shrimp and could be used to elongate the shelf life of refrigerated shrimp.

  • Chicken patties and frankfurters were manufactured by varying the relative proportion of MDM to HDM as raw materials, and their palatability, shelf-life and textural properties were evaluated. The obtained results were as follows: 1. Chicken patty containing MDM slowed gradual increase in TBA value during frozen storage, but its storage up to g weeks presented no problems in flavor stability. 2. Color score and total palatability of chicken patty were best for the product containing 30% MDM. It was also concluded that MDM can be included in the patties up to 50% of total meat with good results, but more than 70% was not recommended. 3. The formulation of MDM up to 50% in frankfurter gave a quite satisfactory acceptability and textural properties compararable to frankfurter made of 100% HDM, but the inclusion of more than 70% MDM was not recommended. 4. The TBA value of frankfurter containg MDM did not increase to any great extent until 4 weeks of storage at $4^{\circ}C$, indicating no unique problems in flavor instability compared to regular frankfurter. 5. It was concluded that processed meat products such as patties and frankfurters containing MDM up to $30{\sim}35%$ of total meat ingredients gave satisfactory results in color, texture and palatability, comparable to regular products.

Changes in Quality Characteristics of Chopped Garlic with Various Storage Method (저장방법에 따른 다진 마늘의 품질특성 변화)

  • This study was carried out to investigate quality characteristics of chopped garlic stored at various temperature. Chopped garlic was stored at room temp.($25^{\circ}C$), refrigerator temp.($5^{\circ}C$) and frozen temp. ($-18^{\circ}C$) for 1, 2, 4, 8, 24, 48, 72 hours(room temp.) and 1, 3, 5, 7, 10, 15, 20, 30 days(refrigerator temp. and frozen temp.), respectively. Brown color, Hunter color value, vitamine C contents and flavor patterns by electronic noes of chopped garlic were measured. Fresh chopped garlic was used as control and garlic powder by hot air drying was used for comparisons. Brown color of chopped garlic increased with increasing storage time, and the intensity of that was reduced as following order such as chopped garlic stored at room temp., refrigerator temp. and frozen temp. Brown color of garlic powder was most dark amomg all the samples. The results of Hunter color value was consistent with then of brown color in chopped garlic, whereas those of garlic powder showed relatively low browning. Above results might be caused by the difference of measurement as only surface color was measured in Hunter color value unlike brown color. Vitamine C contents of chopped garlic decreased with increasing storage time, and in particular those stored at room temp. decreased markedly as retention of those stored at room temp. was 60% of control after 1 day storage. Vitamine C retention of chopped garlic stored at refrigerator temp. was 78% of control after 10 day storage and that stored at frozen temp. was 86% of control after 30 day storage. At room temp., flavor pattern by electronic nose showed a difference between chopped garlic stored for 1hour and control, and showed occurrence of off odor after 1 day storage. Flavor pattern of garlic powder by electronic nose showed a mild flavor. Flavor patterns of chopped garlic stored at refrigerator temp. were similiar until 10 day storage, whereas those showed occurrence of volitile components after 15 day storage. Flavor patterns of chopped garlic stored at frozen temp. were similiar after 30 day storage, and especially those after 3 day storage were almost the same as control. Therefore it is desirable to store chopped garlic within 1 day at room temp. and within 10 days at refrigerator temp. And it is most desirable to store chopped garlic at frozen temp. as retention of quality characteristics was superior than other storage temp.

Development of Freezing Time Prediction Model and Thermo-physical Properties of Frozen Kimchi (김치 동결시의 물리적 특성 및 동결시간 예측 모델 개발)

  • This study was carried out to investigate the thermo-physical properties and design Freezing time prediction model from data of freezing test of Kimchi. Density of Kimchi were measured as 1001.9 ${\pm}$0.03 kg/㎥ at unfrozen state, 987.0 ${\pm}$0.07 kg/㎥ at frozen state and volume of the Kimchi expanded 4.67% at -l5$^{\circ}C$. Initial freezing point of Kimchi and seasoning were -4.0$^{\circ}C$ and -2.5$^{\circ}C$, respectively. Freezing ratio of Kimchi were estimated more than 50% at -5.0$^{\circ}C$, more than 75% at -l0$^{\circ}C$ and approximately 90% at -25$^{\circ}C$. To obtain equation for freezing time prediction of Kimchi, freezing time(Y) was regressed against the reciprocal( $X_3$) of difference of initial freezing point and freezing medium temperature, reciprocal( $X_4$) of surface heat transfer coefficient, the initial temperature( $X_1$) and thickness( $X_2$) of samples. As results of the multiple regression analysis, equations were obtained as follows. Y$_{kimchi}$=3.856 $X_1$+13982.8 $X_2$+8305.166 $X_3$+ 3559.181 $X_4$-639.189( $R^2$=0.9632). These equations shown better results than previous models, and the accuracy of its was very high as average absolute difference of about 10% in the difference between the fitted and experimental results.

Low-temperature aging and drying treatments of restorative rice to improve its microbial safety and texture (복원용 밥의 미생물 안전성 및 식감 향상을 위한 저온 숙성 및 건조 처리)

  • In this study, a commercial production stepwise method for restorative rice with high quality and microbial safety was developed. The stepwise treatment method included steaming, refrigerated aging, and low temperature drying. The soaking rice was steamed twice at $90-100^{\circ}C$, and then, the rice was aged at $0-10^{\circ}C$, frozen at $-20^{\circ}C$, and dried at low temperatures with 5 m/s wind speed at $1-20^{\circ}C$ and 85% relative humidity. Applying the three steps improved sensory qualities compared with the conventional hot air drying and made storage at room temperature for 3 months possible. Specifically, the moisture content of the restorative rice was increased to 30%, which was 4.3 times higher than the 7% of the conventional air dried rice, and the rice grain shape was well maintained. The texture and appearance of the three-step rice were significantly improved (p<0.05) in a sensory evaluation.

  • Hand deboned and mechanically deboned chicken meat were produced from domestic broilers and spent layers. Meat yield, chemical composition, functional characteristics, storage stability and microbiogical properties were investigated. The results obtained were as follows: 1. 35% of carcass freight was recovered primarily as hand deboned chicken meat (HDM) and 45% secondarily as mechanically deboned chicken meat(MDM), total meat yield reaching 80% of carcass weight. 2. Moisture, protein, fat. ash and calcium content of MDM were 65, 12, 20, 1.7 and $0.2{\sim}0.4%$, respectively MDM was higher than HDM in fat, ash and calcium, but significantly lower in moisture and protein Total pigment content of MDM was 2.5 times higher than that of HDM, such high content being attributed to the increased inclusion of hemoglobin during the mechanical masceration of carcass in the deboning process. 3. The emulsifying capacity (ES) of MDM per g meat was only 70% that of HDM, but when ES was expressed on unit g of protein basis MDM showed even higher ES than HDM primarily due to the higher proportion of salt soluble protein fraction of MDM. 4. Since the TBA value of MDM increased rapidly after 4 weeks of frozen storage at $-20^{\circ}C$, the maximum possible storage period of MDM is estimated to be about 4 weeks. 5. Total microbial counts of MDM was approximately $1.8{\times}10\;cells/g$ showing no great difference from HDM or red meat.

Effects of Gibberellin Treatment on the Dormancy-breaking and Germination Promotion of Carpinus laxiflora Seeds Stored for 16 Years (지베렐린 처리에 따른 서어나무 16년 저장 종자의 휴면타파 및 발아촉진 효과)

  • This study assessed the vitality of Carpinus laxiflora seeds stored for more than 15 years in order to discover optimal pre-treatment conditions for removingthe causes of seed dormancy and improving revitalization rate. Seeds were collected in October 2000 and stored at -18℃ for 16 years. Experiments to assess the revitalization ability of the seeds were performed under the following conditions: a controlled environment; in warm stratification (30 days at 23℃); in cold stratification (30 days at 4℃ andfor 30, 45, 60, and 120 days); gibberellin (GA3) treatment (24 hours per day in GA3 solutions of 100, 500, 1000, 2000, and 3000 mg·L-1); and both cold stratification and GA3 treatment (30 days at 4℃, then in a GA3 solution of 100, 500, and 1000 mg·L-1 for 2 hours). The average germination percentage(GP) of untreated seeds was 2%, and the average GP of warm-stratification seeds was 10%. Cold-stratification seeds had the highest GP at 81% for the 45-day process, while the 120-day cold-stratification seeds had the lowest GP at 67.3%. The average GP of seeds treated with GA3 ranged from 77% (100 mg·L-1) to 99% (1000 mg·L-1), indicatingsignificant differences between the treatment concentrations. The treatment effect of GA3 was highest at 500 mg·L-1 and 1000 mg·L-1, and lowest at 100 mg·L-1. The average GP of seeds treated with GA3 following cold stratification was 68%, which was lower than the cold stratification-only (73.2%) and GA3-only (88.4%) treatments. A comprehensive comparison of the seed germination characteristics according to the four treatments determined that a GA3 500 mg·L-1 pre-treatment, with the highest average GP, was ideally suited to the revitalization of long-term stored C. laxiflora seeds. Consequently, C. laxiflora stored at -18℃ for 16 years indicated strong vitality and could be regenerated by proper pre-treatment.

Storage and Acceptability of a Smoked Sebastes schlegeli Product (훈연처리에 의한 조피볼락의 저장성 및 기호도)

  • This study was conducted to develope a new type of ready-to-eat smoked Sebastes schlegeli product with high acceptability and extended shelf-life. A Sebastes schlegeli was salted at 4% salt concentration for 6 hr at $4^{\circ}C$. The cold smoking conditions for the salted Sebastes schlegeli consisted of drying for 2 hr at $22{\sim}24^{\circ}C$ followed by smoking for 2 hr at $22{\sim}24^{\circ}C$. The warm smoking conditions for the salted Sebastes schlegeli consisted of drying for 2 hr at $22{\sim}24^{\circ}C$, smoking for 2 hr at $22{\sim}24^{\circ}C$, and smoking again for 30 min at $47{\sim}50^{\circ}C$. The rancidity of the smoked Sebastes schlegeli did not change after 1 year storage at $-20^{\circ}C$ by monitoring the iodine value, peroxide value, and acid value. The number of viable cells in the cold and warm smoked samples were counted as $7.4{\times}10^5$ and $6.2{times}10^5$ CFU/g, respectively. Viable cells were not detected after 1 year of storage at $-20^{\circ}C$. The sensory evaluations of the processed Sebastes schlegeli showed that elastic texture increased with smoking as compared to with salting. There were no significant differences between cold and warm smoking in terms of sweetness, elastic texture, color, and smoke flavor. However, for overall acceptability, preference were in the oder of cold smoked, warm smoked, and salted.
