  • 최근의 연구에 의하면 자연계 또는 인류가 합성한 수많은 화합물질들이 내분비계를 교란하여 야생 도물, 조류, 어류 및 인체에 부작용을 일으킨다고 보고되고 있다. 이들 내분비계를 교란시키는 물질들을 과학자들은 "내분비계 장애물질 (endocrine disruptors, 언론에서는 환경호르몬으로 소개됨)"이라고 부른다. 이들 내분비계 장애물질은 일상 생활에서 우리 주변에서 흔히 볼 수 있는 것으로서 플라스틱, 캔, 세제, 내연제, 음식, 완구, 화장품, 살충제, 약품 등에 광범위하게 분포되어 있다. 우리는 음식물, 음료수, 약품, 화장품 등에 포함된 내분비계장애물질을 구강 또는 피부로 이들 물질을 섭취하고 있다. 아직까지 이들 화합물질에 대한 정확한 부작용이 규명되지 않고 있는 문제점이 있음에도 불구하고 동물과 사람은 다양한 이들 화합물에 노출되고 있다.

  • Endocrine disrupting chemicals (EDCs) have been attracting significant attention in modern society, owing to the increased incidence rate of various diseases along with population growth. EDCs are found in many commercial products, including some plastic bottles and containers, detergents, liners of metal food cans, flame retardants, food, toys, cosmetics, and pesticides. EDCs have a hormonal effect on the human body, which disrupts the endocrine system, notably affecting sexual differentiation and normal reproduction, and can trigger cancer as well. Recently, the association between neurological diseases and EDCs has become a hot topic of research in the field of neuroscience. Considering that EDCs negatively affect not only neuronal proliferation and neurotransmission but also the formation of the neuronal networks, EDCs may induce neurodevelopmental disorders, such as autism spectrum disorders and attention-deficit/hyperactivity disorder as well as neurodegenerative diseases, including Parkinson's disease and Alzheimer's disease. In light of these potentially deleterious outcomes, important efforts have been underway to minimize the exposure to EDCs through appropriate regulations and policies around the world, but chemicals that have not yet been associated with endocrine disrupting properties are still in wide use. Therefore, more epidemiological investigations and research are needed to fully understand the effects of EDCs on the nervous system.

Safty of Alternatives for Endocirne Disrupting Substances (내분비계장애물질 대체소재의 안전성)

  • Endocirne disruptors (EDs) can cause fertility decrease, developmental disorder, and even cancer in animals. Until 90's, EDs were used in various synthetic products including paints, coatings, detergents, plastics, and plasticizers. Currently, in several countries, the production, trade and use of EDs or EDs-suspected chemicals have been regulated while activity to screen the alternatives for EDs including bisphenol-A, phthalate and nonylphenol is active. Although various toxicity test method was developed and applied for screening of alternatives, however, the safety of alternatives has been not fully demonstrated. Some alternatives have high structural similarity with existing EDs, raising the possible risk of endocrine disruption by alternatives. In an effort to develop the safe alternatives, we reviewed the effects of EDs such as bisphenol-A, phthalates, nonylphenol and their substituents. In addition, in-silico analysis for endocrine disrupting activities of some alternatives was presented.

Effects on Biomarkers and Endocrine in Muddy Loach (Misgurnus anguillicaudatus) under 21 day Exposure to Methomyl (21일간 methomyl에 노출한 미꾸리의 생물지표 및 내분비계 영향)

  • To evaluate the effect of endocrine disruption chemicals (EDCs) to aquatic organisms, muddy loach (Misgurnus anguillicaudatus) was exposed to low concentration methomyl for 21 days in order to identify the effect of biomarkers and endocrine. Vitellogenin (VTG) in blood plasma, which used widely as validated biomarker for endocrine disruption, was significantly greater in male fish exposed to 0.4 mg/L and 2 mg/L methomyl, and in female fish exposed to 0.08 mg/L, 0.4 mg/L, and 2 mg/L methomyl for 21 days (p<0.05). This results suggest that methomyl have probability of endocrine disruption to organism on aquatic system. While inhibition of Acetylcholinesterase (AChE) activity and increase of DNA damage in comet assay were verified by fish exposed to methomyl, change of ethoxyresorufin-O-deethylase (EROD) activity was not occurred, comparing the control group (p<0.05). Indicators at the level of organism such as condition factor (CF), hepato-somatic index (HSI), and gonado-somatic index (GSI) were not influenced by exposure of methomyl. In conclusion, these results showed the possibility of methomyl in regard to not only endocrine disruption but also impacts on biochemical biomarkers to aquatic organisms.

연안의 유해물질 오염과 평가에 관한 사례연구

  • 현대의 우리들의 생활환경 중에는 약 10만 종류 이상의 아주 많은 화학물질이 존재하고 있으며 매년 약 1000종류의 새로운 화합물이 상품화되고 있다고 알려져 있다. 이들 중 일부는 우리의 일상생활에 편리한 기능을 가져다 주는 역할을 하고 있으나 최근 생태계 및 인체에 유해한 영향을 미치는 물질로 판명되는 사례가 증가하고 있다. 그들 중에는, 성호르몬과 유사한 구조를 가지기 때문에 호르몬 수용체에 잘못 결합하여 호르몬 활성을 異常히 높이는 등의 영향을 미치는 것이 알려져 있으며, 또, 약물대사 효소의 유도나 효소활성의 저해 등에 의한 물질대사의 교란, 면역계통의 저해, 발암작용 등의 영향에도 미치는 것이 알려져 있다. 이러한 일군의 물질은 내분비계 장애물질(Endocrine disruptors; 환경 호르몬)로 불리며, 최근의 여성에 있어서 유방암의 발생율의 상승이나 남성의 정자수의 감소, 혹은 어떤 종의 야생 생물에 있어서 생식기능 장해(악어 수컷 생식기의 퇴화, 수컷 잉어등의 암컷화, 일부 권패류의 임포섹스 등)는 이러한 내분비계 장애물질에 의하여 유발되고 있다고 하는 가능성이 지적되고 있다. 그렇지만, 이러한 내분비계 장애물질의 환경 중에서의 분포나 다양한 생물에의 영향에 관한 연구는 세계적으로도 아직 부족하고 내분비계 장애물질의 실태나 그 메카니즘 및 그것이 가지는 의미에 대하여는 아직 명확하게 규명되어 있지 않고 있어, 그것에 의한 생체영향의 평가를 총합적으로 검토하기까지에는 이르지 않고 있다. 이러한 물질에 관한 연구는 최근 선진국을 중심으로 활발한 연구가 수행되고 있으나, 우리 나라에 있어서의 연구는 지금까지는 아직 미미한 수준에 머물고 있다. 따라서, 이러한 내분비계 장애물질을 빠른 시기에 발견하여 그것들에 의한 영향을 평가하고, 그 영향을 미연에 방지하는 데에 필요한 대책을 강구하기 위한 많은 연구가 필요 불가결하다. 본고에서는 이러한 환경호르몬 물질 중 본 연구실에서 수행한 내용을 중심으로 유해화학물질 오염 및 대책에 대하여 간단히 발표하고자 한다. 발표내용은, 남해안을 중심으로 조사한 유기주석화합물 오염, 광양만에서의 PAHs(다환방향족탄화수소류) 및 Bisphenol A 오염에 관한 연구결과와 이러한 유해화학물질 오염을 평가하고 관리하는 하나의 유용한 방법으로서 위해성 평가에 대하여 실례를 중심으로 간단히 소개하고자 한다.

Effects of Bisphenol A and BPA Alternatives on the Nervous System (Bisphenol A와 대체물질들이 신경계에 미치는 영향)

  • Endocrine disrupting chemicals (EDCs), used in a variety of products in modern society, are hormone-like substances that cause various diseases. Humans are exposed to EDCs through their inclusion in pesticides, plastics, cosmetics, detergents, and drugs. Bisphenol A (BPA), one of the representative endocrine disruptors, is an estrogen-like substance that has been widely used commercially in plastic and epoxy resins. BPA is a chemical that can disrupt the endocrine system, leading to reduced reproductive function, obesity, cancer, and neurodevelopmental disorders. Since the adverse health effects of BPA began to be reported the use of BPA has been regulated worldwide. Various alternatives to BPA have been widely used worldwide; representatively, bisphenol S (BPS) and bisphenol F (BPF) are the most commonly used in commercial contexts. BPS and BPF may cause endocrine-disrupting effects like those of BPA due to their similar chemical structures. Recent studies have reported that BPS and BPF disrupt the neurodevelopmental process and cause neurodevelopmental disorders. Therefore, future studies will be required for safety verification of BPA alternatives and the development of new alternatives to BPA for brain health. In this review, we reviewed the effects of BPA and the alternatives, BPS and BPF, on the nervous system.

Vitellogenin as a Biomarker of Endocrine Disrupter in the Aquatic Environment

  • A number of chemical released into the environment eliciting their effects by disrupting normal hormonal pathways. Endocrine disrupting compounds are present in the aquatic environment and pose potential health consequences to wildlife and humans. This review are designing fur xenobiotic estrogens based on induction of the egg-yolk precursor protein vitelloge-nin. In fish of aquatic environment, it may result in decrease fertility and egg production in females or lead to reduced gonad size or feminization of genetic male fish. It has been known that male fish exposed to estrogenic compounds show induced production of vitellogenin. Vitello-genin production is normally restricted to adult females, which have elevated estrogen levels during egg production. However, vitellogenin can be induced in males by Pollution of environmental endocrine disruptors. Consequently, the presence of vitellogenin in male fish can serve as an indicator of exposure of environmental endocrine disrupting compounds. In immature fish pol-luted at low levels of environmental endocrine disrupter, vitellogenin can serve as a reliable biomarker for exposure to endocrine disrupter. This review demonstrates the utility of vitellogenin as a biomarker fur exposure to estrogenin agents in auqatic environment.

Toxicity Monitoring and Classification of Endocrine Disruptors using Bioluminescent Bacteria.

  • For detecting toxicity of endocrine disruptors (EDs), rapid, sensitive, and simple methods are needed. Therefore, in this study, a new method in which the different toxic effect of EDs can be monitored using 4 different recombinant bacteria was designed and evaluated. It was found that the recombinant bacteria could monitor the toxic effect, not estrogenic effect, due to EDCs through the measurement of bioluminescence and cell growth rate, which were shown to depend upon a form of cellular toxicity, such as DNA damage, protein damage, oxidative damage, and membrane damage. In addition, it was found that the damage done by EDCs can be divided into several groups based upon the toxic mechanisms of the EDCs

Maleficent Effects of Phthalates and Current States of Their Alternatives: A Review (프탈레이트의 유해성과 대체재 현황: 소고)

  • Phthalates, known as typical endocrine disruptors, are plasticizers used to soften plastics such as polyvinyl chloride (PVC). Because of their material properties, phthalates are used extensively in the production of toys, flooring, wood processing, detergents, and even cosmetics as lubricants and perfume solvents. Due to their endocrine disrupting effect and other adverse health effects published, recently, phthalates have been regulated in many countries. Besides, in an effort to replace phthalates, several chemical plasticizers such as trioctyltrimellitate (TOTM) and dioctylterephthalate (DIOP) have been used instead of the existing harmful phthalates, and novel alternatives are continuously being developed. Nonetheless, phthalates are still being detected in several plastic products, and the safety of alternatives that are considered safe is being questioned. In this review, we describe the adverse health effects of phthalates, their regulation and the current status of their alternatives.
