Interaction between Transforming Growth Factor $\beta$ and Cumulus Cells during In Vitro Maturation in Porcine Oocytes (돼지난자의 체외성숙시 Transforming Growth Factor$\beta$와 난구세포의 상호작용)

  • This study was undertaken to evaluate the interaction between cumulus cells and TGF $\beta$1 on in vitro maturation in porcine oocytes. No differ ences were found in maturation rates when follicular oocytes were cultured in medium with various concentrations of TGF $\beta$. At 24 h after maturation, the oocytes matured to metaphase-II were found in medium with TGF $\beta$ regardless of cumulus cells. On the other hand, the maturation rates were significantly(P < 0.01 higher cumulus-enclosed(70 and 52%) than cumulus-denuded oocytes(35 and 26%) in medium with or without TGF $\beta$ at 48 h after culture. In a another experiment, the same maturation rates (54-71%) were observed when cumulus-enclosed oocytes were cultured with various addition time of TGF $\beta$. However, the maturation rates in cumulus-denuded oocytes were significantly (P < 0.05) higher in medium added at 0~24 h (59%) or 24-48 h(57%) after culture than in medium with(27%) and without(38%) TGF $\beta$ for 48 h. These results indicated that cumulus cells is essential for in vitro maturation in porcine oocytes but TGF $\beta$ can promote oocytes maturation in cumulus-free oocytes.

Differentially Expressed mRNA Profiles between Immature Germinal Vesicle(GV) and Mature Metaphase II(MII) Mouse Oocytes (미성숙 난자와 성숙 난자에서 서로 다르게 발현하는 유전자에 관한 연구)

  • Oocyte maturation refers to the process that prophase I arrested germinal vesicle(GV) drives the progression of meiosis to metaphase II(MII) to have the capacity for fertilization and embryo development. To better understand the molecular mechanism(s) involved in oocyte maturation, we identified differentially expressed genes(DEGs) between GV and MII mouse oocytes using a new innovative annealing control primer (ACP) technology. Using 20 ACPs, we successfully cloned 32 DEGs between GV and Mll oocytes, and 26 out of these 32 DEGs were functionally known genes. Four genes including Pscd2 were GV-specific, 10 genes including PKD2 and CSN3 were highly expressed in GV oocytes(GV-selective), and 12 genes including Diva were highly expressed in MII oocytes (MII-selective). Ail of the genes identified in this study were first reported in the oocyte expression using ACP system and especially, we could characterize the existence of PKD-CSW signaling pathwayin the mouse oocytes. Results of the present study would provide insight for studying molecular mechanisms regulating oocyte maturation.

Effect of Meiotic Maturation of Canine Oocytes Cultured in Reproductive Tract (개 미성숙난자의 체내이식 배양이 핵성숙에 미치는 영향)

  • This study were carried out to evaluate the possibility of nuclear progression of canine immature oocytes, of which was cultured in a reproductive tract, such as oviduct, ovarian bursa and uterus of estrus bitch for 4, 5 and 6 days following immediately collection. Cumulus intact oocytes(COC) fore collected from domestic dog following ovariohysterectomy at local veterinary clinics. In Exp. 1, COCs $of>110\;{\mu}m$ diameter were selected and cultured in vitro at $39^{\circ}C$, $5\%\;CO\_{2} $ in air atmosphere. The nuclear progression of canine oocytes checked at 24, 48 or 72 h after in vitro maturation. There was not increased the nuclear progression to the M II stage depending on culture periods at 24, 48 and 72h $(1.3\%,\;3.7\%\;and\;4.7\%)$. In Exp. 2, to evaluate of nuclear progression of immature oocytes, collected or in vitro cultured oocytes were transfer into a canine reproductive tract (oviduct, ovarian bursa and uterus). The recovery rates of canine oocytes from a reproductive tract after 4 days $(33.7\%)$ in vivo culture were significantly higher than those 5 $(17.7\%)$ 6 day $(3.4\%)$ (P<0.05). The survival rates of collected oocytes after 4 days $(60.0\%)$ were also significantly higher than those of 5 days $(30.2\%)$ and 6 days $(38.9\%)$ (P<0.05). The meiotic resumption rates of canine oocytes were not significantly difference among the culture periods at 4 days $(5.9\%)$, 5 days $(0.0\%)$ and 6 days $(0.0\%)$. These results show that the nuclear progression of canine immature oocytes from in in vivo culture was not affect the nuclear resumption of oocytes.

Effects of EGF, $\beta-ME$, Glucose on the In Vitro Maturation and Development of Porcine NT Embryos (EGF, $\beta-ME$와 Glucose가 돼지 난자의 체외성숙에 미치는 영향과 핵 이식 배의 발생에 대한 영향에 관한 연구)

  • 본 연구는 NCSU-23과 PZM-3 배양액에 EGF, $\beta-ME$와 glucose의 첨가가 돼지 난자의 체외성숙에 미치는 영향과, 배양조건을 다르게 하여 계대배양한 섬유아세포를 이용한 핵이식 배를 다른 배양액과 산소조건에서 배양하였을 때 체외발생율에 미치는 영향을 조사하였다. 난자를 20ng/ml EGF를 첨가 또는 첨가하지 않은 NCSU-23 및 PZM-3 배양액에서 44시간 배양하였을 때 난자의 체외성숙율은 각각 $85.7\pm3.1\%,\;75.2\pm2.8\%,\;87.1\pm2.4\%$$78.2\pm2.6\%$였으며 EGF를 첨가한 배양액에서 배양한 난자의 체외성숙율은 EGF를 첨가하지 않은 배양액에서 배양했을 때의 난자보다 높은 체외성숙율을 나타냈다(p<0.05). 난자를 $25{\mu}M\;\beta-ME$를 첨가 또는 첨가하지 않은 NCSU-23 및 PZM-3 배양액에서 44시간 배양하였을 때 난자의 체외성숙율은 각각 $79.5\pm2.6\%,\;74.7\pm2.5\%,\;80.2\pm2.3\%,\;78.6\pm2.7\%$였고 $\beta-ME$를 첨 가한 PZM-3 배양액에서 배양한 난자의 체외성숙율이 가장 높게 나타났다. 난자를 1.5mM glucose를 첨가 또는 첨가하지 않은 NCSU-23 및 PZM-3 배양액에서 44시간 배양하였을 때 난자의 체외성숙율은 각각 $79.2\pm2.3\%,\;75.0\pm2.6\%,\;85.5\pm2.5\%$$78.9\pm2.7\%$였고, glucose를 첨가한 PZM-3 배양액에서 배양한 난자의 체외성숙율은 glucose를 첨가하지 않은 PZM-3 배양액에서 배양한 난자보다 높은 체외성숙율을 나타냈다(p<0.05). 핵이식 배를 20ng/m1 EGF, $25{\mu}M\;\beta-ME$ 및 1.5mM glucose를 첨가한 NCSU-23 및 PZM-3 배양액에서 48시간, 144시간 배양하였을 때 2세포기 및 배반포로의 체외발생율은 각각 $56.4\pm2.7\%,\;54.3\pm2.9\%,\;70.5\pm2.1\%,\; 69.6\pm1.5\%$$12.0\pm1.3\%,\;9.6\pml.7\%,\;10.9\pm2.1\%,\;11.9\pm1.8\%$였다.

Effects of Catalase and Cumulus Cells during In Vitro Maturation in Porcine Oocytes (돼지난자의 체외성숙시 Catalase와 난구세포의 영향)

  • The effect of catalase on in-vitro maturation in porcine oocytes with or without cumulus cells were studied. The maturation rates were not significantly different in medium with and without catalase during in vitro maturation of oocytes. However, the maturation rates of oocytes with cumulus cells were significantly higher (P<0.05) than in oocytes without cumulus cells regardless of the presence of catalase. On the other hand, the maturation rate in oocytes cultured with cumulus cells for 48 h (57%) was significantly (P<0.05) higher than in oocytes with cumulus cells for first 24 h period (42%) only. In another experiment, the maturation rate was significantly (P<0.05) higher in medium containing catalase for last 24 h period only than in medium containing catalase for first 24 h period during in vitro maturation of ooytes with or without cumulus cells. But the oocytes matured to M-II stage were observed at 24 h of culture of oocytes without cumulus cells only. When oocytes with cumulus cells were cultured for 72 h, the maturation rates was significantly (P<0.05) higher in medium with (79%) than without (65%) catalase during in vitro maturation. These results indicate that cumulus cells are necessary for in vitro maturation of porcine oocytes, catalase have effect according to the addition periods and can prevent aging of porcine oocytes during maturation in vitro.

  • In vitro cultrue systems for the growth of sma-II oocytes and for meiotic maturation are expected to provide a new source of a large population of oocytes as well as assistance in basic physiological studies of oogenesis. Mouse oocytes mid-growth phase can complete grovvth and acquire full developmental capacity in vitro. On the other hand, growing pig oocytes need some other factors. FSH at a low concentration maintains the viability of both oocytes and granulosa cells, and hypoxanthine promotes the meiotic competence of the oocytes during culture period. Considerable improvement in the culture systems for growth of pig oocytes, suggested from mouse studies, and for oocyte maturation could help to develop this technology in larger species.

The Effect of Low Concentrated Hypoxanthine and FSH in 10% FBS Supplemented Medium on Immature Oocyte in vitro Maturatio (낮은 농도의 Hypoxanthine과 FSH가 미성숙난자의 체외성숙에 미치는 영향)

  • Objective: We examined the effect of different culture media on oocyte maturation. Methods: Four groups of media, (1) 0.3% BSA mBASAL-XI-HTF, (2) 0.3% BSA mBASAL-XI-HTF with FSH, (3) 10% FBS mBASAL-XI-HTF and (4) 10% FBS mBASAL-XI-HTF with FSH were prepared. Mouse cumulus enclosed oocytes (CEOs) were incubated in each group of medium. Hypoxanthine (Hx) was mixed to each group of medium in increasing concentrations of 1 mM, 2 mM and 4 mM. CEOs were incubated and assessed for GVBD and MII development at 3, 6, 18 hours. Results: CEOs maturation to GVBD was seen in all four groups during 3 hours of culture, however MII stage of oocytes was seen after 6 hours. Complete arrest of GV stage in 4 mM Hx media without FSH and partial arrest in 2 mM Hx media without FSH were seen during 18 hours of culture but development was not suppressed in 1 mM Hx media without FSH. More prominent GVBD suppression was noted at early 3, 6 hours culture in 1 mM, 2 mM Hx media with FSH compared to media without FSH. But the suppression was recovered at 18 hours. This result suggests that low concentrated Hx and FSH supplemented media can suppress CEOs maturation during early culture period but recovery is resumed or even stimulated at late period. 1 mM, 2 mM Hx 10% FBS medium with FSH had significantly higher rates of MII development (71.7%, 66.7%) at 18 hours compared to other media. Conclusion: Our results show that low concentrated Hx and FSH supplemented 10% FBS media may stimulate MII development after an initial inhibitory effect.

고양이에서 난자의 체외성숙과 체세포 핵이식 수정란 생산

  • 고양이 체세포 복제산자의 생산은 Shin등(2002, Nature 415: 859) 1마리의 "Cc"를 성공한 이래 재현 생산한 보도가 없다. 그리하여 본 실험에서는 체세포 복제산자 생산을 위한 기초 실험으로서 난자의 체외성숙, 체세포핵이식 수정란의 체외발달율을 검토하고자 수행하였다. 성성숙한 고양이에서 난소적출수술로 난소를 적출 한 후 4시간 내에 실험실로 운반하여 면도칼로 난소를 절재한 후 미성숙난자를 채취하여 TCM199 + 0.4% BSA + 1000 IU/ml bST에서 24시간 체외성숙하였다. 체외성숙한 난자는 난구세포를 제거 후 핵이식에 공시하였다.   • 소난자 체외성숙에 있어 소 태아혈청 (fetal bovine serum: FBS) 첨가시 높은 성숙율을 보이며 핵 이식 후 배 발달율에 있어서도 높다. 하지만 배양액 내에 포함된 소 태아혈청은 수정란 이식 후 태아의 발육 중 및 태어난 직후에 발생되는 송아지에서 나타나는 몇몇 비정상적인 현상들의 원인인 것으로 보고되고 있다 (Walker 등, 1998). 본 실험은 성숙배양시 소 태아혈청과 이의 대체물질로서 BSA 또는 PVA 와 같은 macromolecules의 첨가가 소 난자의 성숙과 핵 이식 후 배 발달에 미치는 효과를 알아보기 위하여 실시하였다.
