To investigate whether genistein regulates menopause-induced obesity, it was studied the effects of genistein on anti-obesity effects in female ovariectomized (OVX) mice, an animal model of postmenopausal women. 7-week-old female mice (C57BL/6J) were randomly divided into three groups. All the animals received a high fat diet or a high fat diet supplemented with genistein for 8 weeks and variables and determinants of obesity were measured. The OVX mice had significantly higher body weight and adipose tissue mass than sham mice. However, genistein supplementation reduced body weight, adipose tissue mass, and adipocyte size of OVX mice. The OVX mice treated with genistein had significantly lower levels of serum triglycerides and total cholesterol than the vehicle-treated OVX mice. Lipid accumulation in liver was also markedly decreased by genistein in OVX mice. The results suggest that genistein can effectively prevent adiposity, adipocyte phertrophy, and llipid disorders caused by ovariectomy. Moreover, this study may contribute to the alleviation of metabolic syndrome, including obesity and hyerlipidemia in postmenopausal women.

Osteoporosis that is associated with estrogen deficiency in menopause is by far the most common cause of age-related bone loss. Since isoflavone had been reported as a natural substance that minimizes bone loss, we have begun this study to examine the effect of the substance on bone metabolism in ovariectomized rats. Five week-old (n=22) and 25 week-old (n=22) Sprage-Dawley female rats were classified into young (Y) and adult (A) groups. Each group consisted of three subgroups : sham operated group (SH), ovariectomized group (OVX), and isoflavone supplemented group (OVX+ISO 80 mg/kg B.W.). They were fed chow for 9 weeks. The result showed that body weight gain was increased in YOVX in comparison to YSH group, (p<0.05) serum osteocalcin concentration and urinal deoxypyridinoline (DPD) excretion had significantly increased in YOVX more than in YSH group, and significantly decrease in OVX+ISO than in YOVX group (p<0.05). We concluded that soy isoflavones may decrease bone turnover in young rats. However, isoflavone supplement didn't show significant influence on bone metabolism of adult rats.
The ovarian hormone-deficiency induced ovariectomy rat is widely used as an aging model due to its practicality, convenience, and cost effectiveness. The surgically ovariectomized rat induces reactive oxygen species (ROS) generation like aging phenomena. Free oxygen radicals have been proposed as important causative agents of aging. The purpose of this study was to investigate the effect of chondroitin sulfate (CS) to prevent ovariectomy (OVX)-induced oxidative stress. The OVX rats were given intraperitoneally CS at doses of 100 mg/kg and 200 mg/kg daily for fifteen weeks. Malondialdehyde (MDA) levels were determined as well as the activities of superoxide dismutase (SOD), catalase (CAT), reduced-glutathione (GSH), oxidized-glutathione (GSSG), glutathione peroxidase (GPx) in the liver. The liver antioxidative enzyme activity was elevated while MDA concentration decreased in all CS treated animals. The results demonstrated that CS reduced oxidative stress in a dose dependent manner. These results suggest that CS might be a useful candidate for antioxidative reagent.
이 논문은 대두 이소플라본 섭취가 난소를 절제한 쥐의 골격 손실에 미치는 영향을 투여량에 따라 조사한 것이다. 실험 초기에 암컷의 Wistar rats(7개월령; n=55)을 절제 후 봉합시킨 군(SH; n=14)과 난소절제군(OVX; n=41)으로 각각 구분하여 사육하고 80일 후에 각 5마리씩을 해부하여 난소절제로 인한 골손실 진행여부를 확인하였다. 그 후 난소절제 쥐(OVX)를 9마리씩 4그룹으로 나누고 대두 이소플라본(IF, 대두단백질이 없는 실험식이에 혼합하여 공급)을 각각 0, 20, 40, 80mg/ (kg body.d)씩 84일간 공급하였다. 동시에 SH rats에게도 이소플라본이 없는 실험식이를 섭취시켰으며 164일 후에 해부하였다. 예상했던 대로 골 무기질 밀도(대퇴골, 골간부위와 골간단부위) 및 골 표면적에 대한 다공성 골 면적의 비(대퇴골 원위부 골간단에서)가 OVX군이 SH군보다 낮았다(P<0.05). OVX군은 SH군보다 혈장 osteocalcine 농도가 높았으며 소변으로의 deoxypyridinoline 배설량이 많았다(P<0.05). 164일째 osteocalcine 과 deoxypyridinoline 농도는 OVX군에서보다 IF40 및 IF80군에서 더 낮았다(P<0.05). 그럼에도 불구하고 골 무기질 밀도와 다공성 골 면적이 OVX군에서보다 IF군들에서 더 낮아지지는 않았다. 그러므로 난소를 절제한 성숙한 쥐에게 대두 이소플라본을 매일 섭취시키는 것은 골격조직의 재흡수를 감소시킬 수 있으나 이미 진행된 골감소증(osteophenia)을 회복시킬 수는 없는 것으로 나타났다.

This study investigated whether swimming exercise suppresses(: inhibites) the inflammatory response in adipose tissue in obese mice induced by a high fat diet after ovariectomy in female mice. Experimental group and control group were divided into sham-opereated group (Sham), ovariectomized group (OVX), and ovariectomized group with swimming exercise (OVX/Ex). All female mice fed high fat diet (45% fat) for 8 weeks. Compared with the Sham, the OVX increased the weight of visceral adipose tissue including parametrial, retroperitoneal, and mesenteric adipose tissue. Compared to the OVX, the OVX/Ex demonstrated reductions the weight of visceral adipose tissue. Compared to the Sham, the OVX decreased the mRNA expression of IκBα and increased the mRNA expression of inflammatory cytokines (L-6, MCP-1, TNF-α and leptin) in adipose tissue. But the OVX/Ex appeared to increase the mRNA expression of IκBα and decrease the mRNA expression of inflammatory cytokines in adipose tissue, compared with the OVX. This findings suggested that swimming exercise is useful for preventing adipose tissue inflammation, which will be effective in the prevention and treatment of obesity in obese mice induced obesity by a high fat diet after ovariectomy.

This study investigated whether swimming exercises improves obesity through regulation of angiogenesis in white adipose tissue. Female mice with high-fat diets were divided into sham-operated group (Sham), ovariectomized group (OVX), and swim-trained ovariectomized group (OVX + Swim). Compared to the Sham, OVX increased body weight, adipose tissue mass and size of adipocyte. However, these factors (: such as body weight, adipose tissue mass and size of adipocyte) of OVX + Swim decreased compared with OVX. Compared with the Sham, OVX increased the mRNA expression of angiogenic activator and MMPs and decreased the mRNA expression of angiogenic inhibitors in white adipose tissue. But OVX + Swim decreased the mRNA expression of angiogenic activator and MMPs and increased the mRNA expression of angiogenic inhibitors in white adipose tissue, compared with the OVX. Theses results suggested that swimming exercises the angiogenesis in white adipose tissue, resulting to improve obesity in high-fat diet-fed female OVX mice.

This study investigated the effects of exercise on genistein-induced adipose tissue reduction and anti-inflammation of adipose tissue in ovariectomized mice fed a high-fat diet. Compared to mice fed a high-fat diet (C), both genistein (G) or exercise only (Ex) not only decreased adipose tissue weight and adipocyte size, but also glucose and free fatty acid levels in serum, which were more effectively reduced when genistein and exercise were treated simultaneously (G/Ex). The expression of lipogenesis and inflammatory cytokine genes of both G and Ex in adipose tissue was decreased compared to C. In particular, the decrease of these genes expression in G/Ex was more effective than each treatment alone. Thus, this study revealed that simultaneous treatment of genistein and exercise resulted in more effective improvement of obesity and adipose tissue inflammation than genistein single treatment in ovariectomized mice fed a high-fat diet, and it is a result of positive regulation of lipogenesis genes. This study suggests that the exercise has a beneficial effect on anti-obesity and adipose tissue anti-inflammation of genistein.

This study was carried out to investigate the effects of dietary calcium-salt, estrogen-treatment, and estrogen/calcium treatment on calcium, phosphorus and nitrogen metabolism. Female Sprague-Dawley rats with a body weight of 250~280g were underwent ovariectomy or sham-operation. The ovariectomized rats were divided into 9 different experimental groups including the saline-treated group, the estrogen-treated group, the high calcium salt-treated group, and the estrogen/calcium treated groups and fed for 6 weeks. Each group daily intake and excretion of calcium, phosphorus and nitrogen were measured and apparent digestibility and balance were also studied. The results were as follows: The excretion level of winn calcium was significantly higher the ovariectomized rats than the sham-operation group(p<0.01) and reduced with estrogen treatment but this difference was not statistically significant. Fecal loss of calcium was higher the ovariectomized rats than the sham-operation group(p<0.001). When the estrogen was treated, fecal loss was decreased and then apparent digestibility of calcium was increased. Calcium balance was significantly higher the high calcium treated groups than the control diet groups. The excretion level of urinary Phosphorus was higher the ovariectomized rats than sham-operation group, while these showed to be decreased in the calcium salt, the estrogen and the estrogen/calcium treated groups(p<0.01). Fecal loss of phosphrous was higher in the ovariectomized rats. When the estrogen was treated, the fecal loss was decreased in the avariectomized rat than that of other groups. The excretion level of urinary nitrogen was higher the ovariectomized rats than the sham-operation, while these showed to be decreased in the estrogen, the estrogen/calcium, and the estrogen gradually reduction/calcium intensification. Fecal loss of nitrogen was decreased in tile estrogen treated group. The results in this study showed that high calcium, estrogen/calcium and estrogen gradually reduction/calcium intensification in the ovariectomized rats enhanced calcium balance compared to the ovariectomized rats without calcium treatment, but little effects on the phosphorus and nitrogen balance.
The present experiment was performed to study whether or not ovariectomized female rats can be used as an animal model to investigate the effects of estrogen on body compositions. Six-month-old, female rats were ovariectomized and the changes in body compositions were evaluated 6 months after surgery. As summarized in Fig. 2, body fat accretion was double in ovariectomized rats compared to shamoperated rats. Muscle weights were not d!fferent, while bone weights were lower in ovx rats than in sham rats when the values were expressed as a per\ulcornercentage of body weight. Results of the present study suggest that these ovariectomized rats can be utilized as an animal model to study the body compositions of postmenopausal obese women and related health problems.
