Comparative biodegradation studies of liquid and solid alkanes and of two different solid alkanes were conducted by an isolated Acinetobacter sp., which degraded crude oil alkanes simultaneously. for the determination of degradation mechanism of hydrophobic crude oil constituents. Also a model oil experimental system composed of a solid alkane. heneicosane, as a substrate and a non-degradable non-aqueous phase liquid. pristane, as an oil matrix was established and studied. It was proposed that the Acinefobacter sp. utilized hydrophobic substrates directly on the surface of them with no difference in the degradation rates between the liquid and solid alkanes. On the basis of the results from the heneicosane/pristane system which imitates crude oil matrix containing solid constituents. the crude oil matrix was considered to reduce the bioavailability of contained substrates by reducing the specific surface area of substrates to contact with microorganisms.
The formation of soluble microbial products(SMP) and molecular weight distribution on loading rate were observed in batch-type culture medium, which phenol was fed as a substrate. The molecular weight destribution was obtained by using 30K, 100K dalton and $0.45{\mu}$ membrane filters. When the phenol concentrationas a substrate was 120, 230 and 440 mg/L , the specific substrate utilization rate(q) showed 0.639, 1.281 and 1.744 mgTOC/mgMLSS/day, respectively. The endogenous biomass decay rate constant($K_d$) at each substrate concentration was 0.00536, 0.0661 and 0.0749($day^1$), respectively. The $SMP_e$ product rate constant($k_{SMP}_ e$) showed 0.006, 0.0058 and 0.0057($day^1$), respectively. The initial influent substrate during the course of time degraded and produced $SMP_s$. The $SMP_s$ was converted to the $SMP_{nd}$ and endogenous phase converted to the $SMP_e$ ingredients. The molecula weight distribution on loading rate was converted to a higher MW during the course of time.
세계적인 자원고갈과 지구온난화와 같은 환경문제가 발생됨에 따라 대체에너지 개발에 대한 연구가 활발히 이루어지고 있다. 섬유소 기질을 이용한 바이오에탄올 생산은 세계적으로 막대한 자원이 있으며 광합성에 의해 재생산되는 무한한 재원으로서 환경적으로도 대기오염물질을 적게 배출하여 유용한 에너지원으로 각광받고 있다. 하지만 섬유소 기질은 cellulose, hemicellulose, lignin과 같은 고분자 화합물이 유기적으로 결합된 단단한 결정구조로 이루어져 있어 이를 분해하여 원하는 물질을 얻기 위해서는 전처리 과정이 필요하다. 전처리 공정은 바이오에탄올을 생산하는 당화 및 발효공정의 효율 및 반응시간 단축에 기여하며, 특히 섬유소 기질일 경우에는 필수불가결한 공정이다. 전처리 공정은 물리적, 화학적, 생물학적 방법으로 나누어지며, 이러한 방법들 중 기질의 특성과 처리효율에 따라 기술들을 병합하여 사용하기도 한다. 이에 본 연구에서는 산 처리, 암모니아 처리, 과산화수소 처리 및 효소를 이용한 생물학적 처리를 단독 또는 병행하여, 전환된 당 성분 및 함량을 조사함으로서 섬유소계 기질인 채소 음식물류 쓰레기를 대상으로 바이오에탄올을 경제적으로 생산하기 위한 적합한 전처리법을 검토하였다. 전처리 방법별 당화율을 살펴보면, 산 처리와 암모니아-과산화수소-계면활성제 처리가 각각 65.3 % 및 65.7 %로 가장 높았으며, 과산화수소 처리는 16.2 %로 가장 낮았다. 반면 전처리 공정 없이 효소를 이용한 당화만을 실시한 경우에는 4.3 %의 낮은 당화율을 나타내었다. 섬유소계 기질의 전처리 효율을 향상시키기 위해 첨가하는 계면활성제의 효과는 암모니아-과산화수소 및 암모니아-과산화수소-계면활성제 처리의 당화율을 비교한 결과, 뚜렷한 효과를 확인할 수 없었다. 전처리 방법별 당의 성분 및 함량을 비교한 결과 육탄당은 암모니아-과산화수소-계면활성제 전처리에서 가장 많이 검출되었다. 오탄당은 산 처리 후 그 함량이 현저히 높았으며, 오탄당 중 xylose의 함량이 60.49 mg/g로 가장 많이 차지하고 있었다. 이 결과로부터 전처리 방법에 관계없이 당화율은 유사한 수준을 보이지만, 당화된 당의 성분 및 함량에는 큰 차이가 있음을 알 수 있었다. 이당류의 경우 과산화수소 및 암모니아-과산화수소 처리를 제외한 나머지 전처리 방법에서 유사한 수준을 나타내었다. 암모니아 처리 및 과산화수소 처리를 순차적으로 병행한 암모니아-과산화수소 처리에서는 각각의 처리시보다 육탄당의 함량은 증가하였으나 암모니아 처리시보다 이당류의 함량은 감소한 것으로 나타났다.
The glycosylated human MINPP (multiple inositol polyphosphate phosphatase), which was recombinantly over-expressed by using industrial host, Pichia pastoris, showed the phytase activity against phytate ($InsP_6$) and the enzyme activity of the unglycosylated counterpart was decreased to 30%. The optimal phytase activity occurred at pH 7.4. The human MINPP showed high substrate specificity for $InsP_6$ with little activity on other organic phosphate conjugates such as para-nitrophenylphosphate (pNPP), ATP, and ribose-1-phosphate (R-1-P). The phosphatase activity against 2,3-bisphosphoglycerate (2,3-BPG) by human MINPP was increased to 1.2-fold in the presence of stimulator, 1 mM 2-phosphoglycolate (2-PG) but the phytase activity against $InsP_6$ was not affected by addition of 1 mM 2-PG. The phosphatase activity against 2,3-BPG by human MINPP was not increased in the presence of 2 mM $Mg^{2+}$ or 100 mM $Cl^-$.
In the present study, two kinds of D-xylanases (1, 4-$\beta$-D-xylan xylanohydrolase (EC 3.2.1.8) were purified and characterized from crude extract of Aspergillus niger KG79. Xylanase I was most active at pH 5.0, whereas xylanse II at pH 4.0 Both enzymes demonstrated their maximum activity at 45$^{\circ}C$. They were relatively stable between pH 4.0 and 6.0 at 3$0^{\circ}C$ for 6 hours. Molecular weight of xylanse I and II were 12, 500 and 11, 500, respectively. Michaelis-Menten constants of xylanse I and II were 0.28% and 0.26% of xylan, respectively. Both enzymes could degrade commercial D-xylan to xylose, xylobiose, and xylotriose to the degree of about 10% of total reducing power. Xylanse I could, however, liberate arabinose from barley straw xylan in addition to xylose and xylooligasaccharides more rapidly than xylanase II. The degree of hydrolysis was about 25%. The reconstituted D-xylanase system with purified xylanases and $\beta$-xylosidase degraded commercial xylan and barley straw xylan to the degree of 28% and 54% respectively. The limit of hydrolysis by the enzymes was suggested to be resulted from the physical structure of the substrate.
Bedrock is inhomogeneous for its genetically diverse origins and geological conditions when it forms, and especially, conglomerates and core-stones are one of these typical composite geo-materials composed of weak matrixes and strong pebbles. Mechanical properties of these composite bedrocks, like a conglomerate, generally vary depending on the mechanical properties and distributions of pebbles and the matrix. Therefore, regarding the consequence of understanding mechanical property of bedrocks in the designing slopes, tunnels, and other engineering facilities, empirical rock classification methods generally applied in the mechanical property modeling may not be suitable and rather, we may need some other classification methods, or tests more specific for these inhomogeneous composite bedrocks. This study includes a series of analyses to see elastic behaviors and modulus of composite geo-materials using homogenization theory. Forty nine case models were made for the elastic analysis with considering 5 factors such as gravel content, gravel size, strength of matrix, sorting and dip angle. The results analyzed are applicable to calculate elastic modulus of composite geo-materials as conglomerates and core-stones.

This study was performed to investigate the removal characteristics of volatile organic compounds (VOCs) in the gasphase biofilters, and to propose a stoichiometric analysis approach to characterize biological reaction through carbon mass balance. The VOCs studied were toluene, styrene, methyl ethyl ketone (MEK), and methyl isobutyl ketone (MIBK) as a single substrate for each biofilter. The critical loading rate was determined to be $46.9\;g/m^3{\cdot}hr$, $25.8\;g/m^3{\cdot}hr$, $96.3\;g/m^3{\cdot}hr$, and $66.5\;g/m^3{\cdot}hr$ for toluene, styrene, MEK, and MIBK, respectively. The obtained results indicated that the critical loading rate was well correlated the octanol-water partition coefficient. In the analysis of carbon mass balance, carbon recovery to $CO_2$ became relatively lower as substrate loadings increased, but higher for carbon recovery to biomass. Stoichiometric analysis revealed that biomass yield increased as substrate loadings increased, and its coefficient (g biomass/g substrate) varied from 0.31 to 0.57 for toluene, 0.29 to 0.57 for styrene, 0.08 to 0.56 for MEK, and 0.14 to 0.53 for MIBK.
Batch kinetics during the exo-polysaccharide (EPS) fermentation of Ganoderma lucidum was investigated as a function of different substrates (glucose and starch), substrate concentration $(1{\sim}7%,\;w/v)$ and subculture (3 times). Logistic model for mycelial growth fitted the experimental data better than Monod and two thirds power model. The Luedeking-Pirt equation was adequate to fit the kinetic data of product formation and substrate consumption. The EPS production was strongly non-growth associated, although it was mixed type. The product formation and sustrate consumption by growth associated mechanism decreased as the concentration of glucose increased, while those of the non-growth associated mechanism increased. However, starch medium increased the growth associated and non-growth associated substrate consumption indicating higher availability of substrate. Also, batch culture in starch medium showed the higher specific growth rate and stability during subculture than those in glucose medium. In conclusion, the enhanced EPS production and stability in the subculture was found to be remarkably improved by use of starch as sole carbon source in medium. The maximum mycelium dry weight and EPS production of 9.463 and 10.410 g/l, respectively, were obtained after shake culture of 7 days at $30^{\circ}C$ from the media containing 7% starch.
In this study, to explore the psychological and biosocial characteristics of the temperament and character's latent profile group, first, the latent group was identified with the seven variables of the Temperament and Character Inventory(TCI), and second, the difference between the psychological and biosocial characteristics of three identified latent groups. A total of 287 university students participated, and the latent groups was identified through latent profile analysis, a human-centeted statistical method, using Cloninger's TCI, Cognitive Emotion Regulation Questionnaire(CERQ), Positive Affect and Negative Affect Schedule(PANAS), Composite Scale of Moriningness(CSM), Pittsburgh Sleep Qulity Index(PSQI), and Satisfaction With Life Scale(SWLS). As result, first, three latent groups were identified through latent profile analysis using the seven variables of TCI. second, significant differences were identified in CERQ, PANAS, which are psychological variables, CSM, PSQI, and SWLS, which are biosocial variables among the latent groups. In conclusion, the importance of Self-Directedness(SD), a character factor that can be developed rather than Harm-Avoidance(HA), a temperament factor from nature, was confirmed. And the necessity of follow-up studies on psychological and biosocial variables for adaptive and mature personality was discussed.
We prepared non-porous membrane on the $Al_2O_3$ substrate with the different pore by the size by the plasma polymerization of $CHF_3$. We studied the permeability characteristics of membrane by Ar treatment and the effect of substrate pore size on gas permeation mechanism. The selectivity was increased with Ar plasma treatment time and rf-power near the substrate to the cathode while the permeability was decreased. It was observed that the solution-diffusion model would be applied to non-porous layers while it is applied Knudsen diffusion model to the substrate. From the experimental observation, it could be concluded that the pore size of $Al_2O_3$ membrane influenced on the permeability and the selectivity.
