Studies on the Effects of Liver-Fluke Infection on the Reproductive Disorder and Milk and Meat Production in Korean Native Goat (Liver-Fluke의 기생(寄生)이 산양(山羊)의 번식장해(繁殖障害)와 유육생산(乳肉生産)에 미치는 영향(影響)에 관한 연구(硏究))

  • These studies were carried out to investigate the effects of liver-fluke infection on the reproductive disorder and milk and meat production in Korean Native Goat. A survey of infected goat in Chungnam district was conducted with 474 head by interdermal reaction and also a clinical observation was founded. A experiment was carried out to determine the effects of liver fluke extract on the reproductive disorder by subcutaneous injection and milk and meat yield were measured from treat ed goats. The results obtained were summarized as follows. 1. The rate of positive goat was 71.3% among the surveyed goat and the highest rate was appeared at 3~4 years old goats as 81.4%. 2. The treated goats lost condition and failed to thrive, and became progressive weaker. 3. In hemogram, observation, the rate of eosinophil leucocyte was increased significantly by extract treated. 4. It was appeared the pathogenic signs such symptoms poisoning liver function disturbance and reproductive disorder from treated goats with liver fluke extract. 5. In milking goat the milk yield droped significantly and fattening goat did not fatten. 6. The gross income value per capita was lower from infected goat and it was analysed more sensitivly in net income value.

Survey on Soybean Parasitic Nematodes (대두증산을 위한 기생선충 조사)

  • Survey on soybean parasitic nematodes has been undertaken in order to get an ideas about the distribution of nemic fauna. Total 71 soil samples were collected from soybean fields in Gyeongbug and Chungbug provinces. Eighteen species belonging to fourteen different genera were found associated with soybean, Glycine max. The results were as follows: Aphelenchus avenae, Circonemoides informis, Ditylenchus dipsaci, Helicotylenchus pseudorobustus, Heterodera glycines, Hirschmanniella imamuri, Meloidogyne hapla, Meloidogyne incognita, Paratylenchus pandus, Pratylenchus thornei, Pratylenchus vulnus, Psilenchus hilarulus, Rotylenchus sp. Tylenchorhynchus clay toni, Tylenchorhynchus dubius, Tylenchorhynchus nudus, Tylenchus sp. Xiphinema americanum were found in soybean fields. In frequency of occurring nematodes by localities, soybean cyst nematode was higher in Chungbug by $70\%$, while $50\%$ in Gyeongbug. But rook-knot nematode was higher in Gyeongbug by $54.1\%$, while $19.1\%$ in Chungbug. In density of nematode, Heterodera was the most dominant as 200.4 populations per 100ml soil, Meloidogyne as 107.1 populations, Rotylenchus, Xiphinema, Aphelenchus in decreasing order. In survey of loss, density of soybean cyst nematode in heavily infested fields was 155 cysts per 100ml soil while 35 cysts in slightly infested fields. Seventy percent of soybean yields was decreased when comparing with average 64.2kg/10a in heavily infested fields and average 209.5kg/10a in slightly infested. Correlation coefficients among number of soybean cyst and yields, pod weight, number of pod was negative as r- 0.57, 0.47, 0.38 over all localities.

  • The influence of temperature on the rate of egg production and embryonic development of Microcotyle sebastis was investigated to determine the precise time of a second treatment. The survival time of the adults of M. sebastis was inversely proportional to temperature. The number of laid eggs per each replicate during the first 24 h was $39.3{\pm}4.0$ at $10^{\circ}C$, $62.7{\pm}14.2$ at $15^{\circ}C$, $101.0{\pm}5.6$ at $20^{\circ}C$ and $89.0{\pm}11.0$ at $25^{\circ}C$. The time required for egg hatching of M. sebastis was $31.30{\pm}4.88$, $17.52{\pm}3.24$, $11.59{\pm}3.02$ and $10.76{\pm}3.10$ days at 10, 15, 20 and $25^{\circ}C$, respectively. The regression models of the time required for the beginning and 50% point of hatching according to the different temperatures were as follows; Beginning of hatch: D=58.2000-$4.2067{\times}Temp+0.0867{\times}(Temp)^2$ ($P\leq0.01$), 50% of hatch: D=91.3833-$7.5767{\times}Temp+0.1767{\times}(Temp)^2$ ($P\leq0.01$).

The effects of gamma irradiation on the survival and development of Clonorchis sinensis metacercariae (감마선 조사가 흰쥐에서의 간흡충 생존 및 발육에 미치는 영향)

  • The effects of gamma irradiation on the survival and development of C. sinensis metacercariae were studied to evaluate the feasibility of irradiation as a control measure for clonorchiasis. Pseudorasbora parve were collected at an endemic river of clonorchiasis and were used for irradiation of the fluke in three schemes. The first (Scheme 1) was irradiation of the isolated metacercariae from the fish followed by infection to experimental rats. The second (Scheme 2) was irradiation of the fish, and then the metacercariae were isolated and in(ected to rats. The third (Scheme 3) was irradiation on the rat livers after infection with normal metacercariae. Irradiation doses varied from 5 to 100 Gy for Schemes 1 and 2, and 10 to 25 Gy for Scheme 3. The rats were sacrificed 2 to 6 weeks after infection. In Scheme 1, the metacercariae irradiated at 50 Gy failed to survive in the rats after 2 or 6 weeks. However, 1 to 44% of the metacercariae irradiated at 5∼30 Gy survived. The estimated LD50 of Scheme 1 was 16.5 Gy. The flukes irradiated in Scheme 2 survived better than those in Scheme 1. The average worm recovery rate in 50 Gy was 28%(7∼39% individually). Increasing the dose up to 100 Gy brought a remarkably low survival rate of an average 1%(0∼3% individually). The LD50 of Scheme 2 was 47.5 Gy. Worm recovery rates in the 10 Gy group of Scheme 3 were 21∼39%, and those in the 25 Gy group were 2% and 34%. Although the metacercariae were irradiated, all of the recovered worms were morphologically normal. Only the worms irradiated with 10 Gy or 25 Gy after 9 days from infection in Scheme 3 showed underdeveloped testes and seminal receptacle. The present results suggest that irradiation of the fish by 100 Gy could be adopted as a control measure for clonorchiasis.

Susceptibility of the Alfalfa Weevil, Hypera postica (Coleoptera: Curculionidae) to Korean Entomopathogenic Nematodes in Laboratory Assays (한국산 곤충병원성 선충에 대한 알팔파바구미의 감수성 실내 검정)

  • Laboratory tests were conducted to determine the susceptibility of the alfalfa weevil, Hypera postica to Korean entomopathogenic nematodes, Steinernema carpocapsae GSN1 strain (ScG), S. glaseri Dongrae strain (SgD), Heterorhabditis bacteriophora Hamyang strain (HbH), and Heterorhabditis sp. Gyeongsan strain (HeG) at the petri dish assay. The larval mortality of H. postica was significantly different depending on nematode species and treatment concentration. SgD and HeG strain were more effective against H. postica larva than ScG and HbH strain. When SgD and HeG strain were treated with the rate of >20 infective juveniles (ijs) per alfalfa weevil larva, mortality was ca. 77.5-100% at the late instars of H. postica in 3 days. The number of established nematode was significantly different depending on nematode species, whereas number of progeny was not significantly different. The mean number of established us of SgD strain in a host was the highest at 80 ijs by 30.2. The highest progeny number of HeG strain was 2,671.5 with 80 ijs. All nematode strains were not parasitic entirely to H. postica adults. These results show that alfalfa weevil late larva is highly susceptible to Korean entomopathogenic nematodes in the laboratory.

Effect of Ultrasound on the Growth and Short-term Behaviour of the Carp, Cyprinus carpio (초음파가 잉어 Cyprinus carpio의 성장 및 단기적 행동에 미치는 영향)

  • In this study, mortality, body wet weight, health assessment, and short-term behavioral mode were observed to determine the effects of ultrasound on the ecological response of the carp Cyprinus carpio. Mortality in the treatments was less than 5% during ultrasound exposure (31 dB re 1 ${\mu}Pa$, $14{\sim}15$ kHz) and there were no significant differences among the replicates (P>0.05). The treatments, based on mean wet weight, exhibited greater effect than the controls, but the magnitude of the differences was not large. Skins and tail fins of some test animals exposed to the controls and intensive treatment groups had light injury such as hemorrhaging, whereas, damages in other organs such as eyes, other fins, parasites, thymus, and gills were not observed (P>0.05). Sudden stimuli in low intensity at a short distance caused a directional avoidance of the fish from the sound. However, the carp exposed to ultrasound at the large scale field was not observed clear behavior changes. Overall results suggest that fish exposed to the low-intensity ultrasound had no serious stress during the tests.

Isolation of Cryptosporidium parvum oocysts from fecal samples - The combination of ether extraction and discontinuous sucrose gradients (분변에 함유된 Cryptosporidium parvum 오오시스트의 순수분리 -Ether extraction과 discontinuous sucrose gradients의 병용)

  • A calf and 50 mice were infected with Cryptosporidium parvum and their fecal materials were collected and treated 10 ether extraction (EE), followed by discontinuous sucrose gradients (DSG) method. EE method was to remove some of fat or lipid from feces. Sediments were washed by centrifugation ($1,500{\;}{\times}{\;}g$ for 10 min., 3 times) In phosphate-buffered saline and then these washed sediments were sleeved sequentially through stainless steel screens with a final mesh of 250 ($61{\;}{\mu\textrm{m}}$ porosity) to remove other debris. After sieving, the materials were suspended in 2.5% potassium dlchromate solution. Oocysts were counted by using a hemocytometer and the recovery rate of pure oocysts was calculated on the basis of the count. Following centrifugation ($1,500{\;}{\times}{\;}g$ for 30 min.) by DSG method, most oocysts were recovered at the interface between a gravity of 1.103 and 1.064. The recovery rates of pure oocysts from the fecal suspension of the calf ($3.8{\;}{\times}{\;}10^7/ml$) and the mouse ($3.2{\;}{\times}{\;}10^6/ml$) treated with EE method were 81.6% and 51.6%, respectively. It is suggested that the recovery rate was dependent on the number of oocysts In each suspension treated with EE method. To get the 50% recovery rate, there must be more than $2{\;}{\times}{\;}10^6$ oocysts per ml of the fecal suspension treated with EE method. By the combination of the two methods it was possible to isolate C. parvum oocysts from normal feces of the calf and mouse as well as from dlarrhelc feces.

Infectivity of the sparganum treated by praziquantel, gamma-irradiation and mechanical cutting (프라지콴텔, 감마선 조사 및 기계적 절단으로 처리한 스파르가눔의 마우스에 대한 감염력)

  • An experimental study was performed to observe the infectivlty of sparganum (plerocercold of Spirometra erinncel) treated by prazlquantel, gamma-irradiation and mechanical cutting. The spargana were obtained from the naturally Infected European grass snake, Rhabdophis tigrina, or from the expenmentally infected mice. A total of 83 mice (ICR straine were divided Into 3 experimental groups by the source of the damage, fed each UH 5 spargana, and sacnaced 1 month later for worm recovery. In the praziquantel group, the worms were incubated In the concentration of 10㎍/ml (control: Tyrode for 4 hours) for 0.5. 1.2 and 4 hours at 36℃. and fed to mice. The recovery rate from mice in prazlquantel group was not different from that (80%) of control poop and in the range of 76-100%. In the gamma-Irradiation group, the worms were irradiated by 10-1000 Gy with Csl37. The average recovery rates of 69-100% were not different from that of control up to 100 Gy. The rate was 56% under 150 Gy, and 5% by 1000 Gy. In the mechanical cutting group, the worms were cut at 0.5, 1, 2 and 3 m from the anterior end of the scolex. The average recovery rates In each group were 70-90% and that of control was 90%. The present folding suggests that the sparganum be highly resistant to prazlquantel, gamma- irradiation and mechanical cutting. The vitality center of the sparganum must be at the anterior end of Its scolex.

  • Enterocytozoon bieneusi, a microsporidian species, has emerged as an opportunistic pathogen in AIDS patients. This organism has also been identified in a wide range of animals, and the zoonotic potential of human infections is of particular interest. This study revealed that this organism was found with relatively high prevalence in feces of asymptomatic cattle in Korea. Fecal specimens were obtained from a total of 1,720 cattle in a slaughterhouse located in Chungnam province, Daejeon city and Chonbuk province. After removal of fecal debris by sieving and density gradient centrifugation, samples were examined by microscopic examination and then nested polymerase chain reaction (PCR). Microscopic examination with the modified trichrome staining for the fecal specimens revealed 194 (11.28%) positive calves for microsporidia spore. PCR using the specific primer for E. bieneusi revealed 79 (4.59%) positive calves. The infection ratio of microsporidia was higher in March than other season.

Effects of nutrient and salinity in egg and larval development of Aedes togoi (영양과 염도가 토고숲모기 (Aenes togoi)의 난 및 유충발육에 미치는 영향)

  • The study made an observation on periodicity of oviposition, and the effects of nutrient and salinity in egg un larval development of Aedes togoi, and the results are summarized as follows: The 53.9% mosquitoes of one feeding laid eggs once, 26.9% laid hice and 19.2% laid three times. Autogenous rate of Aedes togoi reared in three different nutrient groups in larval stage was 6.9% in 0.8 mg/larva, 22.5% in 1.6 mg/larva and 44.4% in 2.4 mg/larva. The oviposition rate according to different salinity of the oviposition sites (0%, 0.5%, 1.0%, 2.0% and 4.0%) was 25.2% in distilled water, 36.2% in 0.5% salinity 23.5% in 1.0% salinity and 14.6% in 2.0% salinity. Developing period of the aquatic stage of male Aedes toBoi in $25^{\circ}C$ were shorter (10.73 days) than females (11.85 days). The most effective concentration of salinity for the developing period was 1.0% which took 9.25 days in males and 10.44 days in females. In the developmental status of the follicles according to nutrition in the larval stage, the numbers of follicles of groups fed 0.8 mg, 1.6 mg and 2.4 mg per larva were 180.7, 197.% and 202 respectively. The result of ovary dissection on the 1 with day after emergence, three different nutrition groups were in Christopher's stage IIa mostly; each 71.0%, 61.1% and 39.9% of the total follicles and autogenous females observed.
