나일틸라피아 치어에 미치는 염화나트륨, 포르말린 및 과망간산칼륨의 급성독성

  • 본 연구에서는 나일틸라피아, Oreochomis niloticus를 대상으로 염화나트륨, 포르말린 및 과망간산칼륨에 의한 24시간 급성독성 조사를 실시하였다. 부화 후 14일의 양식산 나일틸라피아 치어(평균 전장 14.8±1.1 mm, 평균 체중 0.05±0.01 g)를 공시어로 사용하였다. 3반복 실험 후의 각 농도에 따른 24시 간 LC/sub 50/ 값과 95% 유의수준에서의 상한치와 하한치는 Litchfield and Wilcoxon 법(US EPA, 1978)에 따른 'probit analysis' software로 계산하였다. 각 처리군의 24시간 LC/sub 50/은 매우 좁은 농도 범위 내에서 급성독성 값에 대한 신뢰범위가 결정되었으며, 처리 농도 증가에 따라 매우 유의한 농도 의존성을 나타내었다. 대조군 나일틸라피아 치어는 희석수에 24시간 처리시 모두 생존하였다. 염화나트륨의 24시간 LC/sub 50/ 값은 18.6‰ 이었으며 95% 신뢰한계의 상한치는 18.7‰, 하한치는 18.5‰ 이었다. 포르말린의 24시간 LC/sub 50/ 값은 152 ppm 이었으며 95% 신뢰한계의 상한치는 156 ppm, 하한치는 148 ppm 이었다. 과망간산칼륨의 24시간 LC/sub 50/ 값은 2.09 ppm 이었으며 95% 신뢰한계 상한치는 2.11 ppm, 하한치는 2.07 ppm 이었다. 포르말린과 과망간산칼륨의 24시간 처리시 죽은 나일틸라피아 치어 개체들은 공통적으로 호흡 장애로 기인된다고 판단되는, 아가미덮개 열림과 아울러 입이 열렸고, 표피 점액에 손상을 입는 독성 증후를 보였다. 본 연구 결과와 더불어 앞으로, 1시간 혹은 2시간의 영향농도(Effect concentration) 파악, stage 민감도 조사, 처리 시간 및 처리 환경을 달리한 급성독성에 관한 연구가 필요할 것이다.}C$에서 전반적으로 증가하였으며, 저 수온인 15$^{\circ}C$는 4 시간까지 낮아지다가 이후 증가 되었다. Hb농도는 3$0^{\circ}C$$25^{\circ}C$에서 전반적으로 증가하는 경향을 보였으나 4 시간째 일시적으로 감소하는 경향을 보였다. 15$^{\circ}C$에서는 4시간째까지는 감소되었다가 6 시간째에는 높아지는 경향을 보였다. 이러한 증가된 경향은 온도가 높아질수록 용존 산소의 포화도가 낮아져 산소부족 요인에 의해 적혈구수의 증가와 함께 Ht수치도 높아지며 아가미운동을 통한 혈액으로의 산소운반이 능동적으로 이루어지고 있음을 나타내고 있다. 혈장 코티졸의 농도는 $25^{\circ}C$에서는 증가하는 경향을 보였으며, 1$0^{\circ}C$및 3$0^{\circ}C$에서는 1 시간까지 증가되었다가 회복되는 경향을 나타내었다. 신장에서의 SOD활성은 $25^{\circ}C$인 경우 대조구보다 높은 값을 유지하였다. 하지만, 30도에서는 4 시간째까지 증가하는 경향을 보였으나 이후 감소하는 경향을 보였으며, 저온인 1$0^{\circ}C$에서는 감소하였다. 간장에서의 CAT의 활성은 3$0^{\circ}C$인 경우 4시간까지 $25^{\circ}C$ 및 15$^{\circ}C$에서는 2 시간까지 증가하였다가 이후 낮아지는 경향을 보였다. 이상의 결과로 고ㆍ저 수온변화에 따른 스트레스가 장기화될 경우 어체의 건강상에 문제점의 발생하여 대량 폐사 및 질병발생의 야기될 것으로 사료된다.은 먹이 공급 6시간 후 가장 높은 값인 44.19$\pm$

  • Infrequent voiding is defiled as two or less micturitions in a day without organic causes. It can 1ead to bladder capacity enlargement and increase in the volume of residual urine and as a consequence it may produce recurrent urinary tract infections(UTI) and or renal damages. We report a case of acute pyelonephritis due to infrequent voiding in a 13 year old girl. The imaging studies revealed floating debris in the bladder on VCUG and dilated ureter on ultrasonography, and parenchymal defects on 99mTc DMSA scan. (J.   • A protective effect of activated charcoal against the acute lead poisoning of kidney was studied in mile. Mice approximately 30 gm in weight were grouped into the control, lead acetate-treated, and activated charcoal-treated after lead acetate groups. Lead acetate (60 mg/kg) and activated charcoal (40mg/kg) were delivered orally. Serum AST, ALT and glucose were measured and the ultrastructural alteration of liver was examined by electron microscopy. Activated charcoal decreased the increase of serum AST, ALT and glucose levels induced by lead. Lead acetate-treated hepatic cells characterized by irregular nuclei, enlarged and reduced number of mitochodria, enlarged rough endoplasmic reticulum, loss of riboscomes. Cells treated with activated charcoal were similar to those of the control group. In conclusion, activated charcoal may protect the lead-induced toxicity on liver.

Effects of Ethanol Extract from Lathyrus palustris on Anti-inflammation Response of RAW 264.7 Cell (RAW 264.7 대식세포 염증반응에 대한 털연리초 에탄올 추출물의 항염증 효과)

  • Lathyrus palustris often used as a treatment for inflammation of the kidneys in Korean traditional medication. Generally, drugs for arthritis have anti-inflammatory and antinociceptive properties. However, the validity of the anti-inflammatory effect has not been scientifically investigated so far. Therefore, the purpose of the research was to investigate the latent anti-inflammatory ability of L. palustris using the ethanol extract. To evaluate the anti-inflammatory activities, we examined the inflammatory arbitrators such as a nitric oxide (NO) and prostaglandin E2 (PGE2) on RAW 264.7 cells. Our results indicated that ethanol extract significantly inhibited the lipopolysaccharide E (LPS) derived PGE2 production in RAW 264.7 cell. The inhibitory activity of ethanol extract for PGE2 tests with inhibition ratio showed in 40 ㎍/mL. Overall, PGE2 tests had a higher inhibitory effect on inflammation than NO tests. This result anticipated that the ethanol extract from L. palustris is a good candidate for developing the origin of anti-inflammatory agents.

Urinary Tract Infection in Febrile Infants with Pyuria (발열과 농뇨가 있는 영아에서 요로감염에 관한 연구)

  • Objective : Urinary tract infection(UTI) is a frequent serious bacterial infection in young infants. The clinical presentation may be non-specific and variable, depends on factors such as the age and the level of infection. Children with renal involvement may be at risk of permanent renal damage. Experimental studies have shown that renal lesions caused by acute febrile UTI may be prevented or diminished by early diagnosis and treatment. Therefore, it is important to find a method that can permit early diagnosis and identification of patients who are at risk for progressive renal damage. We designed this study to identify related factors in culture positive UTI infants, and also to identify related factors in culture negative UTI infants, who are febrile with pyuria, by using renal imaging and functional studies including renal sonography, DMSA scan and VCUG. Methods : Retrospectively analyzed the medical records of 136 febrile infants with pyuria over 2 years(from January 2001 to February 2003). Urine culture was done in all cases, and regardless of urine culture findings, renal imaging study was done if symptomatic UTI suspected. Results : Total 57 organisms were isolated in 53 patients. E. coli was the most common organism(86%), followed by E. faecalis, M. morganii, Proteus species, P. aeruginosa, S. aureus and E. fergusonii. Most of the isolates had high sensitivity on cephalosporins or amikacin and had low sensitivities on aminopenicillins. Abnormal acute phase DMSA scan or VCUG findings were seen in both urine culture positive and negative group without statistical differences(P>0.05). In febrile infants with pyuria, fever over 48 hours, older age and high CRP related to abnormal acute phase DMSA scan findings regardless urine culture results. Conclusion : 1st or 3rd generation cephalosporins with amikacin could be the first choice of treatment for UTI. Febrile infants with positive urine culture dose mean urinary tract infection but not acute pyelonephritis which directly relates to cortical damage which could be confirmed by acute phase DMSA scan. Even cases with negative urine culture findings, acute pyelonephritis should be concerned in febrile infants with pyuria who are older than 3 months of age, has fever over 48 hours or high CRP level. And in such cases, acute phase DMSA scan and VCUG should be evaluated for early treatment and long term prognosis.

  • Mercury, one of the most diffused and hazardous organ-specific environmental contaminants, exists in a wide variety of physical and chemical states. The murcury with the nature which evaporates easily can cause an acute or chronic mercury poisoning to workers at mercury-handling workplaces. Although many studies indicate that mercury induces a deleterious damage, little has been reported from the investigations of mercury effects at surrounding levels in living things. The purpose of this study was to evaluate the biological effects of mercury chloride and ionizing radiation. Prepubertal male F344 rats were administered mercury chloride in drinking water throughout the experimental period or were given wholebody irradiation with a dose of 6.5 Gy. The amount changed of body weight during the experimental period showed a 4.9% rise in the mercury-treated group and 14.4% decline in the irradiated group compared with the level of the control group. The results of hematological analysis (red blood cells, white blood cells, hemoglobin, and hematocrit) indicated the differential effects of mercury chloride and ionizing radiation. However the concentration of cortisol as assessed by radioimmunoassay increased in both of the groups. Relative expressions of mRNA related to mitochondrion-mediated apoptosis were investigated using semiquantitative reverse transcription polymerase chain reaction on gonad and urinary organs of the experimental groups. While the expression of Bcl-2 mRNA exhibited different patterns depending on the organs or the experimental groups, both of the experimental groups showed a conspicuous expressions of Bax mRNA. In conclusion, the target organ of mercury chloride seems to be a urinary organ and the pattern of damage induced by mercury chloride differs from that by ionizing radiation.

Low T3 syndrome in Kawasaki disease: Relation to serum levels of tumor necrosis factor-α, interleukin-6 and NT-proBNP (가와사끼병에서의 저 T3 증후군 : 혈청 tumor necrosis factor-α, interleukin-6 및 NT-proBNP 농도와의 관계)

  • Purpose : We investigated the relationship between thyroid hormone and serum tumor necrosis factor (TNF-${\alpha}$), interleukin (IL-6) and N-terminal fragment of pro-brain natriuretic peptide (NT-proBNP) in patients with Kawasaki disease (KD). Methods : Serum levels of thyroid hormone, TNF-${\alpha}$, IL-6, and NT-proBNP were measured in 52 KD patients in the acute and subacute phase and 10 patients with acute febrile illness (control group). TNF-${\alpha}$ and IL-6 were determined by sandwich enzyme-linked immunosorbent assay (ELISA). Echocardiography was performed to detect coronary artery lesions (CAL) in KD patients. Results : Low $T_3$ syndrome occurred in 63.5% of KD patients. $T_3$ in the acute phase of KD was lower than that in the control. In KD patients, $T_3$ was lowered in the acute phase and elevated in the subacute phase, whereas TNF-${\alpha}$, IL-6 and NT-proBNP were elevated in the acute phase and decreased in the subacute phase. NT-proBNP, and IL-6 were higher in patients with low $T_3$ than in those with normal $T_3$. In addition, $T_3$ inversely correlated with IL-6 and NT-proBNP. Of the 4 patients with CAL, 3 had very low $T_3$. Compared with intravenous immunoglobulin (IVIG)-responsive patients, IVIG-resistant patients had lower $T_3$ and higher IL-6 and NT-proBNP. Conclusion : $T_3$ decreases in the acute phase of KD and normalizes in the subacute phase without thyroid hormone replacement. Low $T_3$ may be partially induced by IL-6 rather than TNF-${\alpha}$, and is strongly associated with high NT-proBNP. $T_3$ in KD may be used for the differential diagnosis, monitoring the activity of the disease, and predicting the severity of inflammation.

The Therapeutic Effect of Hovenia dulcis Thunberg Extracts and 6 Types of Herbal Extracts on $CCl_4-Induced$ Acute Liver and Kidney Damages in Rats (헛개나무추출액 및 생약성분의 혼용이 사염화탄소 투여로 유발된 흰쥐의 급성 간 및 신장손상의 회복에 미치는 영향)

  • This study was performed to investigate the therapeutic effects of Hovenia dulcis Thunberg (HDT) and HDTmix extracts on the biochemical analysis, histopathology and histomorphometry of liver and kidney in carbon tetrachloride $(CCl_4)$ administrated rats. Extract was prepared by autoclave ($121^{\circ}C$, 15 psi, 3 hours) and filtered with nylon cloth and filter paper then freezing dried. In blood chemistry analysis, HDTmix group, aspartate aminotransferase and alanine aminotransferase were significantly (p<0.01) decreased compared to the $CCl_4$ group, on 3rd day, respectively. In histologic and histomorphometry analysis, the $CCl_4-related$ hepatopathies and nephropathies were dramatically decreased (3rd, 5th day), and well corresponded to the histopathological changes significantly (p<0.01) decreases of degenerative regions, degenerative cells and glomeruli were detected in liver and kidney with significantly decreases of $CCl_4$ group. HDTmix group, quite similar effects on the liver and kidney were observed compared to that of HDT extracts group but more favorable efficacies were detected especially HDTmix also inhibit the hepatopathies (1 day), in which HDT extract does not showed any inhibit effects.

A Child of Severe Mycoplasma pneumoniae pneumonia with Multiple Organ Failure Treated with ECMO and CRRT

  • Mycoplasma pneumoniae (MP) is the most common causative agent of community-acquired pneumonia in school-aged children. An 8-year-old boy who had been diagnosed with autism looked severely ill when he presented to our hospital due to dyspnea and lethargy. He had fever and cough 7 days prior to hospitalization. He had signs and symptoms of severe respiratory distress. The percutaneous oxygen saturation was 88% at high oxygen supply. Chest radiography showed diffusely increased opacity with moderate pleural effusion. He was intubated immediately and admitted to the intensive care unit. Under the clinical impression of mycoplasmal pneumonia, intravenous clarithromycin was started. Laboratory findings showed leukocytosis, hepatitis, decreased renal function, and presence of serum MP immunoglobulin (Ig) M (+) IgG (+) and sputum MP polymerase chain reaction (+). On hospital day 2, the patient developed multiple organ failure with acute respiratory distress syndrome (ARDS). Veno-venous extracorporeal membrane oxygenation (ECMO) was performed with continuous renal replacement therapy (CRRT) and was weaned successfully. This is the first reported case of an ARDS due to MP infection complicated by multiple organ failure that was successfully treated with ECMO and CRRT in South Korea.

Antihepatotoxic effect of ethanol extracts from steam-dried ginseng berry on ᴅ-galactosamine/lipopolysaccharide-sensitized mice (ᴅ-galactosamine/lipopolysaccharide로 감작된 급성간독성 마우스 모델에서 인삼열매추출물의 간독성 개선 효과)

  • The present study aimed to examine the hepatoprotective effects of ethanol extracts from steam-dried ginseng berry (SGBE) in both $\text\tiny{D}$-Galactosamine/Lipopolysaccharide ($\text\tiny{D}$-GalN/LPS)-sensitized mice and in vitro models. Our results clearly demonstrated that SGBE significantly reduced the level of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, and lactate dehydrogenase in blood, and $TNF{\alpha}$ was normalized in 8 h after the treatment with $\text\tiny{D}$-GalN/LPS. Coincidently, major organs remained unimpaired when compared to $\text\tiny{D}$-GalN/LPS control group. Moreover, p38, which stimulates expression of NAFLD-associated cytokines, was markedly inhibited when treated with SGBE. In vitro analysis revealed that the main components of SGBE, linoleic acid and ginsenoside Re/Rd, may play a role in protecting liver from $\text\tiny{D}$-GalN/LPS-induced toxicity. Finally, we concluded that SGBE may be a promising therapeutic agent for preventing damage to the liver.
