Screening of Microorganisms Secreted High Efficient Enzymes and Properties of Enzymatic Deinking for Old Newsprint(IV) -Culture conditions of fungal extracellular enzyme production for biological deinking system- (고효율 효소를 분비하는 균주의 선발 및 신문고지의 효소탈묵 특성(제4보) -고지탈묵용 Fungal Cellulase와 Xylanase의 생산-)

  • This study was focused on the optimum culture condition in CMCase, FPase and xylanase activities of two fungal strains that secret extracellular enzymes for using enzymatic deinking agent to old newsprint. The results of this study were as follows. When Fusarium pallidoroseum was grown on the medium, containing of rice bran+xylan $2.0\%,\;peptone\;0.6\%,\;KH_2PO_4\;0.075\%\;and\;MnSO_4\;0.06\%\;with\;pH\;9.0,\;at\;29^{\circ}C$ for 6 days, the quantitative degree of extracellular enzyme production was the highest. Optimum culture condition for Aspergillus niger was pH 5.0, $27^{\circ}C$ incubating temperature and 7 days incubation period on liquid medium, containing of CMC+xylan $2.5\%,\;yeast\;extract\;0.4\%,\;K_3PO_4\;0.05\%\;and\;CaCl_2+FeSO_4\;0.08\%$. Aspergillus niger was fairly higher FPase and xylanase activities than Trichoderma reesei ATCC 28217.

Effect of ginseng residue on the growth of Ganoderma lucidum (Ganoderma lucidum의 생육에 미치는 홍삼박의 영향)

  • The ginseng residue, a by-product of ginseng tea manufacture, was used as growth promoting substance in the submerged cultivation of Basidiomycetes for its effective utilization. Ginseng residue contained about 46% of total sugar, 14% of crude protein, 12% of ash, and 0.16% of crude saponin. Among inorganic substances in ginseng residue, amount of Mg, Na, K and Ca were much more than other inorganic substances. All ginsenosides existed in red ginseng residue. Ganoderma lucidum and Pleurotus ostreatus could be excellently cultured in potato dextrose broth. Most effective additional concentration of residue extract for growth of these fungi was shown to be 0.2%, and the contents of crude protein and amino acid in mycelium were increased when 0.5% of residue extract were added to the medium.

The Suitable Condition for Mycelial Growth of Volvariella volvacea strains and Selection of the Superior strain (풀버섯의 균사배양 적합조건 및 우량균주 선발)

  • This study was carried out to obtain basic data on mycelial growth characteristics for an artificial cultivation of Volvariella volvacea. Twelve URP primers were used to assess genetic relationship of V. volvacea and its result was divided into two groups. But there was nothing different in morphological characteristics in V. volvacea. Among five kinds of mushroom media, MCM medium was selected as the favorable culture medium. The optimal range of temperature and pH for mycelial growth on V. volvacea were 35oC and pH 6~8, respectively. Carbon sources had not an effect on 10 strains of V. volvacea, and nitrogen source for the optimum mycelial growth was yeast extract. Also, we selected GMVV 79004 as a superior strain of 10 strains V. volvacea based on the mycelial growth and yield.

  • Clinically isolated methicillin-resistant Staphylococcus aureus strains were exposed to subinhibitory concentration of DW286, DW-224a, gemifloxacin, trovafloxacin, sparfloxacin and ciprofloxacin during 26- to 39-days period. Subculturing led to resistance development, and most of the selected mutants were above susceptible breakpoints. Selected mutants had broad cross resistance to other quinolone antibiotics and only one mutant was completely susceptible to all fluoroquinolones. Twenty five among 42 mutants revealed mutations on DNA gyrase and topoisomerase IV by sequencing. Also 16 mutants had fluoroquinolones MICs that were 4-32 times lower in the presence of reserpine. In conclusion, alterations in DNA gyrase or topoisomerase IV and action of efflux pumping out system are the resistance mechanisms of DW-224a.

  • In the course of screening for antifungal agents, a bacterial strain, DM3 was isolated from a mud sample collected at Daechon in Chungnam province and identified as Bacillus licheniformis based on API 50CHB test. It has antifungal activity against 12 plant pathogenic fungi in paper disc assay. At the 95% lethal dose of gamma radiation ($^{60}Co$, 10 kGy, $D_{10}=2.32\;kGy$), the antifungal activity deficient mutant (mDM3) against Colletotrichum gloeosporioides was induced From 2-D electrophoresis analysis, serine hydroxymethyltransferase (45.0 kDa), hypothetical protein(40.7 kDa), NifU protein homolog(15.4 kDa), and resolvase(12.5 kDa) homologous proteins were detected only in B. licheniformis DM3. Lysozyme(18.1 kDa) and alkyl hydroperoxide reductase(15.6 kDa) homologous proteins were expressed uniquely in B. licheniformis mDM3. Further studies are needed to reveal that these proteins from B. licheniformis DM3 could be closely related to the antifungal activity against plant pathogenic fungi.

Effects of Rhodobacter sp. SA16 on Lettuce(Lactuca sativa L.) in Plastic Film House (시설 상추에 대한 Rhodobacter sp. SA16 처리 효과)

  • This experiment was conducted to determine the development of mixed organic fertilizer using photosynthetic bacteria and mass production of mixed microbial compound for the environment-friendly agriculture. Photosynthetic bacteria, Rhodobacter sp. SA16 was isolated from soil collected by plastic film house. The SA16 strain was identified based on 16S rDNA sequence analysis and it is closely related to Rhodobacter sp.(100% similarity). The mixed organic fertilizer using SA16 was made of $N-P_2O_5-K_2O=60-10-20\;g\;kg^{-1}$ with combined soybean cake, sesame cake, powdered blood, fish meal, powdered bones and red-yellow soil. The mixed organic fertilizer 0.45, 0.90 and 1.35 Mg $ha^{-1}$ application in Ihyeon series was treated based on soil testing for lettuce cultivation in plastic film house. These results showed that the yield was increased the 18 and 19%over control by the mixed organic fertilizer application 0.45 and 0.90 Mg $ha^{-1}$, respectively. In the physical properties of the soil, the porosity of mixed organic fertilizer 1.35 Mg $ha^{-1}$ treatment was highest at 58.8%. Our results clearly revealed that the organic fertilizer using Rhodobacter sp. SA16 and mass production of mixed strains could be a useful technology in pursuing environment-friendly agriculture.

Establishment of the Optimum Culture Conditions for Mozzarella Cheese manufacturing by Streptococcus macedonicus LC743 with Immunomodulating Activity (면역 활성능을 가진 Streptococcus macedonicus LC743을 이용한 모짜렐라 치즈 제조의 최적 배양조건 확립)

  • This study aimed to establish the optimum culture conditions for Mozzarella cheese by using Streptococcus macedonicus LC743, a strain selected for its immunomodulatory activity. For process optimization, 1.0% and 2.0% strain was inoculated and incubated at $32^{\circ}C$ and $35^{\circ}C$, respectively. The general components, bacterial count, total solids, yields, and immunomodulatory activity of the Mozzarella cheese were investigated. When the strain was inoculated at 2.0% and incubated at $32^{\circ}C$, the product quality and immunomodulatory activity was optimal and required minimal processing time. Therefore, 2.0% of S. macedonicus LC743 starter culture was added to milk at $32^{\circ}C$, after pasteurization at $63^{\circ}C$ for 30 min, and agitated for 4~5 min after addition of $230{\mu}L/kg$ of rennet. Curd was made by setting the milk 25~35 min after addition of 0.01~0.02% calcium chloride. The curd was cut at 0.1~0.12% acidity (81 min later) and after heating the cheese to up to $43^{\circ}C$. Whey was removed at an acidity of 0.17~0.18% by agitation for 53 min. Next, cheddaring for 210 min up to an acidity of 0.6~0.65%, stretching at $72{\sim}75^{\circ}C$, and molding at $65{\sim}70^{\circ}C$ were performed, and the product was allowed to cool down to $5^{\circ}C$. Salting was done with a solution of $18{\sim}20^{\circ}B{\acute{e}}$ at $12^{\circ}C$ for 20 min and drying occurred at 80~90% relative humidity at $10^{\circ}C$ for 2~3 days.

  • The rice blast fungus is a representative model phytopathogenic fungus in which Gene-for-Gene interaction with host rice is applicable. After 1980, eight differential varieties have been constructed and classified to analyze the race of rice blast isolates in Korea. However, since there is limited information about the genetic background of rice blast resistance genes within the Korean differentials, scientific analysis on the emergence of new race or resistance break down was difficult. Recently, a differential system has been developed using monogenic resistance lines to understand the interactions of pathogen race and rice resistance genes. In this study, a total of 50 isolates were selected from four different races isolated in Korea, and they were inoculated into monogenic lines. As a result, the isolates in the same race classified by the Korean differential system reacted differently in single monogenic lines. This suggests that the isolates categorized as the same race group contains different avirulence genes and furthermore, it is presumed that the Korean differential system is difficult to provide useful information for breeding program. For this reason, introduction of differential system using monogenic resistance lines is required in addition to the current system.

Screening of Anti-inflammatory Compound-producing Wild Yeasts and Their Microbiological Characteristics (항염증 물질 생산 능력이 우수한 야생효모의 선별 및 이들의 균학적 특성)

  • To screen for potent anti-inflammatory compound-producing yeasts, we evaluated nitric oxide production inhibitory activities in RAW 264.7 macrophage cells using cell-free extracts from 182 non-pathogenic yeasts. Rhodotorula graminis YJ36-1 and Meyerozyma guilliermondii YJ34-2 showed high inhibitory activities of 57.4% and 47.0%, respectively. The microbiological characteristics of these yeasts were investigated. Rhodotorula graminis YJ36-1 formed ascospores and pseudomycelium. This species grew well at $25^{\circ}C$ in yeast extract-peptone-dextrose (YPD) medium, vitamin-free medium, and 5% NaCl-containing YPD medium. Meyerozyma guilliermondii YJ34-2 was an asporogenous yeast and did not form pseudomycelium. This strain also grew well at $30^{\circ}C$ in YPD medium, vitamin-free medium, and 5% NaCl-containing YPD medium.

Antifungal Activity of Bacterial Strains isolated from Tidal Mudflat and Salted Seafood (traditional Jeotgal) Against Six Major Plant Pathogens (갯벌 및 젓갈에서 분리한 세균의 작물 주요 병원균에 대한 항균활성 효과 검정)

  • Many bacterial strains inhabit strong saline condition, such as tidal mudflat and salted seafoods, were identified and reported for the proposed protease activities and salt resistance; however antifungal activities against plant fungal pathogen have not well been studied until now. In this study, primary screening was performed for the isolation of promising strains against major plant pathogens like Sclerotinia sclerotiorum, Fusarium oxysporum, Phytophthora capsici, Botrytis cineria, Collectotrichum acutatum and Pythium ultimum. Totally 423 bacterial strain were isolated from laboratory media which was based on different morphological characteristics and all the strains were dual cultured against major fungal pathogens on PDA, finally 40 strains were selected as antifungal bacterial strain and identified by fatty acid phylogenic difference analysis from MIDI shorlock gas chromatography system. As a result, antifungal strains from tidal mudflat were 10 species of 6 genus. Paenibacillus macerans was dominant species; 5 strains among the 17 isolates from tidal mudflat. Antifungal strains from salted seafoods were 7 species of 3 genus and Bacillus atrophaeus was dominant species; 12 strains among the 23 isolates from salted fishes.
