Phylogenetic Relationships of Mushroom Agrocybe spp. Based on rDNA-ITS Analysis (rDNA-ITS분석에 의한 볏짚버섯 (Agrocybe spp.) 수집균주의 계통분류학적 특성구분)

  • Phylogenetic relationships were investigated in the mushroom genus Agrocybe based on sequence data from the internal transcribed spaces(ITS) of nuclear ribosomal DNA(rDNA). Thirty strains including Agrocybe aegerita, A. grocybe cylindracea, A. grocybe praecox, A. grocybe pediades and Unknown Agrocybe species were subjected for the analysis. The Agrocybe spp. were separated into eight distinct groups. Phylogenetic analysis divided these species into eight groups as follows: A. chaxingu within group I, A. salicacola within group II, A. cylindracea and A. aegerita within group III, Agrocybe sp. within group IV and V, and other Agrocybe spp. were grouped within VI, VII, and VIII.

Probiotic Properties of Lactic Acid Bacteria isolated from Feces and Kimchi (베트남인 분변 및 김치로부터 분리된 유산균의 프로바이오틱스 기능성 연구)

  • The purpose of this study was to investigate the probiotic properties and antioxidant capacity of lactic acid bacteria isolated from Vietnamese feces and the Korean traditional food kimchi. Six isolated strains were identified as Lactobacillus sp. by 16S rRNA sequencing. All strains showed good resistance to low pH (1.5, 2.0, and 3.0) and 0.3% oxgall bile acids. Culture filtrates from the six strains showed various antioxidant effects, including DPPH, ABTS, reducing power, and metal chelating ($Fe^{2+}$) activities. Two of the six Lactobacillus strains showed potential probiotic activity. Heat resistance and adhesion assays were conducted by mixing the selected strains, Lactobacillus acidophilus V4, Lactobacillus plantarum V7, and Lactobacillus paracasei DK121 isolated from kimchi. The results showed that the heat resistance of these strains was similar to that of a commercial strain, L. plantarum LP. In addition, a mucin attachment assay using the mixture of selected strains (V4, V7, and DK121) showed high binding activity to the mucous layer. In conclusion, a mixture of V4, V7, and DK121 shows promising probiotic activity and may be useful for the development of health-related products.

Isolation of Fibrinolytic Enzyme and β-Glucosidase Producing Strains from Doenjang and Optimum Conditions of Enzyme Production (된장으로부터 혈전용해능 및 β-Glucosidase 활성을 가진 균주 분리 및 효소생산 배지의 최적화)

  • Bacterial strains showing the firinolytic and $\beta$-glucosidase activity were screened from Doeniang. The strain of KH-15 revealed a high level of fibrinolytic and $\beta$-gluocosidase activity. The isolated bacterium was identified and desingnated as Bacillus sp. KH-15. The carbon, nitrogen and salts sgnificantly influenced te fibrinolytic enzyme and $\beta$-glucosidase production. The optimized composition of medium appeared to be 2% glucose, 0.5% yeast extract and 0.1% calcium chloride. The optimum pH and temperature for fibrinolytic enzyme and $\beta$-glucosidase activities were pH 7∼8, 4$0^{\circ}C$ and pH 6∼8, 30∼4$0^{\circ}C$, respectively.

Pathogenesity of Colletotrichum gloeosporioides from Other Hosts on Strawberry (다른 기주 탄저병균의 딸기에 대한 병원성)

  • The pathogenesity of 25 isolates of Colletotrichum gloeosporioides from apple, 42 isolates from pepper, 5 isolates from jujube, 8 isolates from persimmon was evaluated to know transmission to strawberry from other infected plants. Followings are the results. Colony morphology and spore size on potato dextrose agar was similar. When each isolate was inoculated on leaf and petiole of strawberry, isolates from persimmon was the most pathogenic. Five isolates, one pathogenic isolate per each host, were evaluated in simulated field condition under natural rainfall for their natural infectivity. All isolates infected strawberry in field condition, so C. gloeosporioides from other hosts are potential inoculum source of strawberry anthracnose.

Manufacture of Koji Using fungi Isolation from Nuruk and Identification of Koji Molds (전통누룩 진균류를 이용한 입국의 제조 및 입국곰팡이의 동정)

  • Various koji were prepared by fungi isolated from traditional nuruk and their quality characteristics were investigated. Acidity and saccharification power of their koji were ranged in 5.0~6.8 and 128sp~241sp. Nine fungi which were showed good quality and sensory evaluation were identified by analysis of their nucleotide sequences with PCR-amplified 18S rDNA internal transcribed spacer-1(ITS-1) and ITS-4 genes. Among them, six strains were identified as Aspergillus oryzae and the other strains were identified as Mycocladus corymbiferus, Rhizo-pus oryzae, Lichtheimia corymbifera.

Phylogenetic relationships of genera Polyporus on the basis of ITS region sequences (rDNA의 ITS 부위 염기서열 분석에 의한 겨울우산버섯(Polyporus)속 균주의 유전적인 유연관계 분석)

  • This study was carried to identify a correct species and asses genetic diversity within the same species of Polyporus spp. preserved in Division of applied Microbiology. Contaminated isolates showed different growth rates, morphology and color of hyphae. We have reconstructed the phylogenetic tree of a select group of Polyporus spp. using nucleotide sequences of the internal transcribed spacer region(ITS) region. The phylogenetic tree was constructed by using the neighbor-joining method. PELF primers of 20-mer were used to assess genetic diversity of preserved isolates. Sequence analysis showed that three strains were different species and four strains were identified completely different nomenclature. According to the analysis of ITS sequences, the genus Polyporus clustered into five distinct group, most of which correlated with species-groups identified by RAPD method. Four isolates included strain PM02 showed high similarity with P. arcularius, four isolates included strain PM03 high similarity with P. alveolaris, three isolates included strain PM01 high similarity with P. tuberaster, and PM 06 and PM04 high similarity with P. brumalis and P. squamossus. Isolates were collected in the United States(PM10, PM11) was identified as P. alveolarius and P. arcularius. RAPD analysis of genetic polymorphisms of genus Polyporus showed a very different band patterns. As the result of RAPD and ITS region sequences analysis for preserved isolates, it seems likely that 6 isolates of Polyporus spp. may be need to reclassify or eliminate from preserved catalogue.

  • On the basis of the specific interrelationship between the species or variety of leguminous crops and the species or strain of nodule bacteria, Rhizobia, the rhizobial species and strain must be effectively chosen for the successful inoculation. The present paper describes on some results of the isolation and taxonomic study on the native rhizobial strains isolated from the nodules of five species of leguminous crops such as numerous varieties of soy bean, lespedeza, birdfoot trefoil, ladino and red clovers. The isolated strains of soy bean nodule bacterium, Rhizobium japonicum were grouped through the inoculation test on variety Changdanbaikmock into the effective, noneffective and toxic strain for the nodule formation. In the study of the effect of some inorganic and organic nitrogenous compounds on the growth of Rhizobium japonicum strain Ac 20, a promotive response was showed by asparagine, and glutamine, but hydroxylamine, nitrite, hydrazine and azide was inhibitory at the concentration of $10^{-2}M/l$ in mannitol-yeast extract basal medium. In the physiological characteristics each strain showed somewhat different activities of the indole-3-actic acid formation and hydrogenase and discussed with these characters in relation to nodule forming ability.

Difference of Gray Mold Severity at Roses Caused by Botrytis cinerea Strains (잿빛곰팡이병원균 Botrytis cinerea 균주 종류별 장미 발병 정도의 차이)

  • Botrytis cinerea is the pathogen for a gray mold generating problems during the cultivation and transportation of roses. But there is little information about the difference of the symptom severity caused by gray mold on rose varieties and pathogen strains. 16 strains were collected from the rose cultivation area to confirm the degree of disease occurrence against strains and each variety. Collected 16 strains were identified based on the sequences analysis of ITS region of ribosomal DNA by using specific primers. The sequence analysis was performed by comparing the sequences to find a difference. To confirm the difference in disease occurrence for each strains, the difference was classified from 0 to 5 stages using charmant variety as a control. The data was confirmed through Kruskal-Wallis ANOVA. The result showed the significant difference in the pathogenicity caused by strains. WNG6_5 showed the lowest pathogenicity with 0.24 and WNG6_3 showed the highest with 3.20. The difference between two strains were almost 3.0. In addition, nine varieties of roses were more investigated with three strains such as the strains of WNG6_5, Hwa_1, and WNG6_3. The result showed that the Love Letter variety showed resistance and the Ice Bear variety was sensitive to three strains. Taken together, this study showed the significant difference by the interactions of rose varieties and gray mold strains.

Selection and Characterization of Antagonistic Microorganisms for Biological Control of Acidovorax citrulli Causing Fruit Rot in Watermelon (수박에 과실썩음병을 유발하는 Acidovorax citrulli의 생물학적 방제를 위한 길항 미생물 선발과 특성 검정)

  • This study was performed to screen the efficacy of antagonistic bacterial isolates from various sources against the bacterial fruit blotch (BFB) causing pathogen (Acidovorax citrulli) in cucurbit crops. In addition, plant growth promoting traits of these antagonistic bacterial isolates were characterized. Two thousand seven hundred ninety-four microorganisms were isolated from the collected samples. Molecular identification revealed two A. citrulli out of 2,794 isolates. In vitro antagonistic results showed that, among the 28 antagonistic bacterial isolates, 24 and 14 bacterial isolates exhibited antagonism against HPP-3-3B and HPP-9-4B, respectively. Antagonistic and growth promotion characterization of the antagonistic bacterial isolates were further studied. Results suggested that, 4 antagonistic bacteria commonly showed both antagonism and growth promotion phenotypes. Moreover, 3 isolates possessed growth promoting activities. Overall results from this study suggests that BFB causing bacterial pathogen (A. citrulli) was suppressed in in vitro antagonism assay by antagonistic bacterial isolates. Furthermore, these antagonistic bacterial isolates possessed growth promotion and antagonistic enzyme production ability. Therefore, data from this study can provide useful basic data for the in vivo experiments which ultimately helps to develop the eco-friendly agricultural materials to control fruit rot disease in cucurbit crops in near future.

Studies on the Formation of L-Threonine by auxotrophic mutants of Brevibacterium flavum (Brevibacterium Flavum의 Auxotrophic Mutants에 의한 L-Threonine 생성(生成)에 관한 연구(硏究))

  • This study was attempted to increase the production of L-Threonine by Brevibacterium Flavum ATCC 14067, To select the strain which produce the highest threonine, mutants ere induced by N-methyl-N'-nitro-N-nitrosoguanidine treatment. The composition of media and cultural condition for its overproduction of threonine were also studied. In a threonine producer, strain B-13(Met-) was the strain producing the highest amount of threonige among methionine, lysine and isoleucine auxotrophs. The following results were obtained. 1. The wild strain and B-13(Met-) produced threonine 1.4mg/ml and 4.86mg/ml , respectively. 2. The optimum composition of medium for producing threonine by Brevibacterium Flavum B-13 was glucose 10%, ammonium sulfate 4%, potassium phosphate monobasic 0.2%, magnesium sulfate 0.05%, biotin $200{\mu}l$, thiamine $300{\mu}l$. Addition of nicotinic acid also led to increase L-threonine production. 3. In addition of organic nutrients to the fermentation medium, peptone n'ere effective and addition of methionine $100{\mu}g/ml$ produced the highest amount of L-Threonine. Aspartic acid and homoserine were also effective when these amino acid were added to the fermentstion medium. 4. Cultural conditon on threonine production by B-16 were investigated. The optimum pH was 7.0-8.0. The highest amount of threnine was produced after 4 days of cultural period.
