Biological control of Paraconiothyrium minitans CM2 on Lettuce Sclerotinia Rot Caused by Sclerotinia sclerotiorum (Paraconiothyrium minitans CM2의 상추 균핵병균(Sclerotinia sclerotiorum)에 대한 생물적 방제)

  • A mycoparasite, Paraconiothyrium minitans CM2 was selected for biological control of sclerotinia rot of lettuce caused by Sclerotinia sclerotiorum. The experiment was carried out in a lettuce greenhouse in Yangpyeong from March to April.. When lettuce sclerotinia rot showed in the early stage of occurrence, Conidial suspension of the mycoparasite was weekly treated once to three times onto soil surface around lettuce plants. Incidence of sclerotinia rot in the once-application plot of the mycoparasite ($1{\times}10^7$ spores/$m{\ell}$) and in the benomyl(WP)-treated plot was 11.0% and 2.7%, respectively, whereas that of control was 31.0%. Incidence of twice- and three-application plots of the isolate was 7.9% and 12.8%, respectively. For increasing the effect of the mycoparasite, the experiment for the timing of application of P. minitans CM2 was carried out in a lettuce greenhouse in Yangpyeong and Suwon. Control efficacy against lettuce sclerotinia rot in the soil-drenching plots of P. minitans CM2 ($5{\times}10^6$ spores/$m{\ell}$) in the planting was 75.3~84.7%, and control effect by treatment of the isolate at the pot drenching+the soil-drenching plots in the early stage of disease occurrence was 63.8~58.0%. As the results, P. minitans CM2 could be a prospective biofungicide for biological control of sclerotinia rot of lettuce.

  • This study was conducted in order to find out biological control of sesame wilt and blight caused by Fusarium of oxysporum f. sp. vasinfectum and Phytophthora nicotianae var. parasitica by using Streptomyces spp. Two sesame pathogens, Fusarium oxysporum f. sp. vasinfectum and Phytophthora nicotianae var. parasitica were purely isolated from diseased sesame plants of the field. Streptomyces species were isolated from 72 soil samples collected from red pepper and sesame uplands in Chungbuk and selected as antagonists according to the results of dual culture. The selected Streptomyces isolates such as St-11 and St-20 were confirmed their antagonistic effect through mycelial inhibition zone and inhibitory effects on the mycelial growth of the pathogens by culture filterate of the antagonists. Inhibitory effects on the conidial germination of Fusarium oxysporum vasinfectum and Phytophthora nicotianae parasitica by the antagonists were also tested in addition to mycelial Iysis. The antagonists St-11 and St-20 showed inhibitory effect on growth of sesame seedlings after seeds soaked in the suspension. Effect of soil inoculation with antagonist St-11 showed 40 to 78 percent of control effect for two diseases in comparison with control under greenhouse.

Effect of some antibiotics on control of bacterial brown blotch of oyster mushroom caused by Pseudomonas tolaasii (몇가지 항생제의 느타리버섯 Pseudomonas tolaasii에 의한 세균성갈색무늬병 방제 효과)

  • Antibacterial activities of 33 antibiotics against Pseudomonas tolaasii causing the brown blotch disease on the edible mushroom Pleurotus ostreatus, were tested in vitro for the control of the disease. Tetracyclin, kanamycin, kasugamycin, and streptomycin showed strong antibacterial activity against P. tolaasii, having the minimal inhibitory concentration of 10, 10, 100 and 200ppm, respectively. These antibiotics showed similar control value of 72.9, 71.2, 68.1 and 62.7%, respectively when applied on the artificially infected mushroom beds. Mushroom yields in the tetracycline treated boxes were increased about 31.8% comparing to the control ones. Mycelial growth of P. ostreatus on the PDA supplemented with streptomycin and kanamycin were not affected, but were inhibited 10~20% and 40% with tetracyclin and kasugamycin treatment, respectively.

  • An isolate of Ampelomyces quisqualis 94013(AQ94013) was selected as an effective parasite for biological control of cucumber powdery mildew. In the greenhouse, occurrence of cucumber powdery mildew was significantly suppressed for nine days by pre-treatment with $5.0{\times}10^6/ml$ and $5.0{\times}10^7/ml$ of conidial suspension of AQ94013. The disease was effectively controlled within three to seven days by post-treatment with the $5.0{\times}10^6/ml-conidial$ suspension of AQ94013. When AQ94013 was treated at concentration of $5{\times}10^6/ml$ three times at seven-day interval in the vinylhouse, the control effect was higher than that treated twice at ten-day interval and that treated with fenarimol twice. As the results, Ampelomyces quisqualis 94013 could be a prospective biofungicide for biological control of powdery mildew of cucumber.

  • This study was conducted to investigate the major mycotoxins occurred during storage from apple, peat, citrus and grape. Analyses of the mycotoxins were conducted by TLC and HPLC. Patulin was only detected from apple, pear, citrus and grape infected by penicillium mycotoxins, but citrinin did not detected. The detected amount except grape ranged from $5.68{\sim}46.81\;{\mu}g/g$ in apple, $3.48{\sim}84.7\;{\mu}g/g$ in pear, $0.16{\sim}0.27\;{\mu}g/g$ in citrus, respectively. When compared to the effect of control on penicillium mycotoxins during storage in apple, pear and citrus, sodium hypochloride gas and heat at $37^{\circ}C$ treatment exhibited effective control. Especially, heat at $37^{\circ}C$ treatment exhibited 100% effective control without any injuring by treatment. In pear and citrus sodium hypochloride gas treatment caused injury by treatment.

The Effects of Controlling the Aphis gossypii Glover (Homoptera, Aphididae) on Cucumber of Entomopahtogenic Fungus, Beauveria bassiana

  • Beauveria bassiana can be used as a biological insecticide to control a number of pests. It has been known as that B. bassiana TBI-1 have a insecticidal effect on the twospotted spider mite (Teranychus urticae) and the greenhouse whitefly (Trialeurodes vaporariorum) these days. The biological pest control agent, B. bassiana, showed somewhat insecticidal activities against the cotton aphid, Aphis gossypii, too. After treatment with biological insecticide, the mean of increasing rate of aphids was at around recorded at around 0.21 while at around 0.24 in non-treated TBI-1 biological insecticide condition during 1 week. Consequently, it was found that treatment of Beauveria bassiana reduced an increasing rate of aphid population. Also, we could visually observe dead aphids changed as a reddish mold due to this insecticidal fungus. B. bassiana TBI-1 can significantly reduce an increasing rate of aphid population, so effective biological control against the cotton aphid are possible.

  • A gram-negative bacterium was isolated from spent substrates of Agaricus bisporus and showed significant antagonistic activity against Pseudomonas agarici. The bacterium was identified as Alcaligenes sp. based on cultural, biochemical, physiological characteristics and a 16S rRNA sequence analysis. The isolate is saprophytic, but not parasitic or pathogenic on cultivated mushroom, whereas it showed strong inhibitory effects against P. agarici cells in vitro. The control efficacy of Alcaligenes sp. HC12 against brown blotch of P. agarici was up to 63% on Agaricus bisporus. The suppressive bacterium may be useful for the development of biocontrol systems.

Biocontrol of Ginseng Damping-off by Bacillus velezensis CC112 (Bacillus velezensis CC112 균주의 인삼 잘록병에 대한 생물적 방제)

  • Bacillus velezensis CC112 inhibited the mycelial growth of several plant pathogens, including Rhizoctonia solani, causing damping-off on ginseng. The control efficacies of B. velezensis CC112 against R. solani by seed dipping in LB and BSM broth diluted 10 times, soil dipping, and soil drenching with LB broth diluted 10 times were 65.8%, 67.1%, and 64.2%, respectively. Treatment of soil drenching with the 100 times diluted prototype of B. velezensis CC112 against R. solani and Pythium sp. by soil revealed control efficacies of 77.3% and 65.7%, respectively. These results indicate that B. velezensis CC112 is a prospective biofungicide for the biological control of ginseng damping off.

  • Earthworm cast is a harmful factor which reduces the uniformity of management of turfgrass and play quality in golf courses and playground. This research was conducted to carry out the efficacy of some pesticide (bifenthrin GR + clothianidin GR, ethoprophos GR, imidacloprid SC, picoxystrobin WG, thiophanate-methyl WP) and tee tree (Camellia oliefera) extracts against reducing earthworm cast in golf courses. All tested materials were acted reducing earthworm cast, thiophanate-methyl among them showed the highest effect reducing earthworm casts in golf courses. Tea tree extracts was occurred similar efficacy among tested chemical pesticides. However, pellet type byproduct of tea seed extract and liquid formulation of tea tree extract were better than granule formulation of them. Byproduct of tea seed and tea tree extract can be used for alternative biological management material for reducing earthworm cast in heavily casting occurrence area.

Nematicidal Effect of Root-knot Nematode (Meloidogyne incognita) by Biological Nematicide (생물학적 선충 방제제를 이용한 고구마 뿌리혹선충 (Meloidogyne incognita)의 방제효과)

  • An nematophagous fungi Arthrobotrys thaumasia Nema-1 and Pseudomonas putida C-5, which degrade the collagen and gelatin, was isolated from controlled horticultural soils in Seonnam-myun, Sungju-gun, Kyungpook and Woosung-myun, Gongju-shi, Chungnam to develop biological nematode pesticide. When $5,000mL\;L^{-1}$ of A. thaumasia Nema-1 culture was treated to Meloidogyne incognita, the nematicidal activity resulted in 55% at 72 hours after treatment. While the nematicidal activity increased to 65% by treating the culture mixture of $5,000mL\;L^{-1}$ Nema-1 and P. putida C-5 after 72 hours. Furthermore, the nematicidal activity of the mixture containing cinnamon extract $50mg\;L^{-1}$, each $5,000mL\;L^{-1}$ of Nema-1 and C-5 culture was elevated to 89% at 72 hours after treatment, comparing to the result showed 17% and 57% of the nematicidal activity, respectively by the treatment of chemical nemato pesticide Fosthiazate $50mg\;L^{-1}$ and neem oil $2,000mL\;L^{-1}$. These results suggested that the mixture of microorganisms and plant extract were more effective biological nematicide than the case of only microorganism or plant extract for nematode control.
