Antibacterial and Antibiofilm Activities of Leaf Extracts of Stewartia koreana against Porphyromonas gingivalis (Porphyromonas gingivalis에 대한 노각나무 잎 추출물의 항균활성 및 생물막 형성 억제 효과)

  • This study was conducted to investigate the potential of Stewartia koreana as oral healthcare materials. The antibacterial activity of ethanol extracts from leaves and branches of S. koreana against oral bacteria was confirmed. The leaf and branch extracts (1 mg/disc) showed antibacterial activity against P. gingivalis only among several tested oral bacteria. The leaf extracts showed higher antibacterial activity, with values similar to those of chlorhexidine, which was used as a positive control. The MIC of the leaf extract against P. gingivalis was 0.4 mg/ml and showed bacteriostatic action. The inhibitory effects of the extract on biofilm formation and on gene expression related to biofilm formation by P. gingivalis were determined by biofilm biomass staining, scanning electron microscopy (SEM), and qRT-PCR analysis. The biofilm production rate and cell growth of P. gingivalis in the cultures treated with 0.2-2.0 mg/ml of S. koreana leaf extracts were significantly decreased in a concentration-dependent manner. The inhibitory effect on the formation of P. gingivalis biofilms at concentrations of 1 mg/ml was confirmed by SEM. The qRT-PCR analysis showed concentration-dependent suppression of the fimA and fimB gene expression associated with fimbriae formation in the cultures treated with 0.2-2.0 mg/ml S. koreana leaf extract. These results support the conclusion that S. koreana leaf extracts can be used as oral healthcare materials derived from natural materials, as demonstrated by the antibacterial action and inhibition of biofilm formation of P. gingivalis.

Antibacterial, Antioxidative and Antiaging Effects of Allium cepa Peel Extracts (양파껍질 추출물의 항균, 항산화 및 항노화 효과에 관한 연구)

  • In this study, the antibacterial, antioxidative and inhibitory effects of Allium cepa peel extracts on tyrosinase and elastase were investigated. MIC values of the ethyl acetate fraction of Allium cepa peel on especially, S. aureus among the skin resident flora (Staphylococcus aureus, S. aureus; Propionibacterium acnes, P. acnes; Pityrosporum ovale, P. ovale; Escherichia coli, E. coli) were 0.06%. The aglycone fraction showed more excellent free radical (1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activity (FSC50=5.05μg/mL). Reactive oxygen species (ROS) scavenging activities (OSC50) of the ethyl acetate fraction and aglycone fraction in the luminol-dependent Fe3+EDTA/H2O2 system were 0.05 and 0.03μg/mL, respectively. The cellular protective effect of the aglycone fraction on the rose-bengal sensitized photohemolysis of human erythrocytes exhibited more prominent (τ50, 480 min at 25μg/mL). The inhibitory effects (IC50) of the ethyl acetate fraction and aglycone fraction on tyrosinase were 9.16 and 8.68μg/mL, the inhibitory effect (IC50) of the aglycone fraction on elastase was 14.12μg/mL The transepidermal water loss of the cream containing 0.1% ethyl acetate fraction was decreased from 8.3g/m2h in control to 6.8g/m2h in the subjects applied with cream containing the ethyl acetate fraction. These results indicate that extract/fractions of Allium cepa peel can function as antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging 1O2 and other ROS, and protect cellular membranes against ROS, and possibly as antiaging agents. Allium cepa peel extract could be used as a new cosmeceutical for whitening and anti-wrinkle products.

Comparison between the biomass and habitat suitability index(HSI) of marine forest forming seaweeds (바다숲 조성 해조류의 생물량과 서식지적합지수 비교)

  • The seasonal and vertical biomass of marine forest seaweeds were examined to select a suitable species at 12 sites of the South Sea in Korea between 2018 and 2019. The Habitat Suitability Index (HSI) was also calculated in terms of biomass for six species (three kelp and three Sargassum species). A total of 16 marine forest-forming species including four kelp and 12 Sargassum species were observed at the 12 sites. The average annual seaweed biomass by season and depth ranged from 843.73-2,925.85 g wet wt. m-2 at the eastern South Sea and from 343.87-4,580.10 g wet wt. m-2 at the western South Sea. In the kelp species, the Ecklonia cava biomass was predominant, followed by E. stolonifera. The macroalgal species with the greatest biomass was Sargassum macrocarpum, followed by S. horneri. The HSI values of E. stolonifera were between 0.76-1.0 at eight sites and those of E. cava were 0.58-0.92 at four sites, indicating that E. stolonifera was more suitable than E. cava. In the HSI values of the Sargassum species, S. horneri ranged between 0.84-1.0 at all 12 sites and the S. macrocarpum values were between 0.68-0.99. The results indicate that E. cava and S. macrocarpum were the most suitable for the marine forest construction in terms of the seaweed biomass, and E. stolonifera and S. horneri in terms of the HSI values. Thus, we suggest that seaweed biomass and HSI values should be considered when choosing suitable forest-forming species.

Quality Properties and Antioxidant Activities of Korean Traditional Rice-Based Wine, Makgeolli Added with Sweet Pumpkin (단호박을 첨가하여 제조한 막걸리의 품질특성 및 항산화 활성)

  • Sweet pumpkin is rich in minerals such as calcium, phosphorus and fiber, and also contains a large amount of β-carotene, which has antioxidant effects. In this study, Makgeolli containing steaming sweet pumpkin (SP) was manufactured to enhance the antioxidant activity. To prepare the Makgeolli, SP was added in amounts of 5, 10, 20, and 30% (w/w), and the chemical (pH, total acidity, alcohol), microbiological (lactic acid bacteria, yeast) properties, and antioxidant activities (DPPH, ABTS) were examined during fermentation and storage for 9 days. The pH of SP Makgeolli was 4.00-4.23 at day 1 of fermentation, and then the pH gradually decreased as fermentation progressed, showing the lowest results at day 9 (3.28-3.52). At day 1, the total acidity was significantly increased (P<0.05) as the amount of SP (0-30%; 0.09-0.55%) and the total acidity in SP Makgeolli (1.01-1.20) was also rapidly increased by 5 days of fermentation. Alcohol content was significantly increased (P<0.05) as the amount of SP (0-30%; 4.59-5.77%) increased at day 9. The counts of lactic acid bacteria and yeast in SP Makgeolli were 8.0-8.1 and 7.9-7.8.0 CFU/mL, respectively, which was higher than SP 0% at day 9. DPPH and ABTS radical scavenging activities were significantly increased (P<0.05) as the amount of SP (0-30%; 48.29-78.97% for DPPH, 62.12-86.68% for ABTS) increased at day 9. This study suggests that Makgeolli added with SP could be potentially and commercially developed due to its superior microbiological and chemical properties, including high antioxidant activities.

Effect of Nuruk protease activity on the quality of anchovy sauce (누룩의 protease 활성이 멸치액젓의 품질에 미치는 영향)

  • This study investigated the quality characteristics of anchovy sauce fermented using Nuruk to maintain a unique flavor, reduce fishy smell, and improve the fermentation rate. Six kinds of fermented fish sauces, including the control, fermentation using traditional Nuruk; SH Koji (Fs-A), JJ Koji (Fs-B), GJ Koji (Fs-C); and fermentation with improved Nuruk; Aspergillus luchuensis (Fs-D) and Aspergillus oryzae (Fs-E), were prepared. Samples were collected at 15 days intervals with 10% Nuruk added to raw anchovy and fermented at 25o C for 60 days. The free amino acids, especially glutamic acid content and amino nitrogen, were the highest in Fs-C, reflecting the high protease activity of Nuruk C (GJ). Regarding overall sensory evaluation, the control was the lowest, whereas Fs-C was highly evaluated among the sample groups. The addition of Nuruk not only shortened the fermentation period, but also increased the overall sensory level by adding umami and reducing fishy odor.

Confirmation of Enzymatic Synthesis of 1, 2-Octanediol Galactoside using Mass Spectrometry and NMR Spectroscopy (Mass spectrometry와 NMR Spectroscopy를 이용한 1, 2-Octanediol Galactoside의 효소합성 확인)

  • 1, 2-Octanediol galactoside (OD-gal) has been synthesized from 1, 2-octanediol (OD), as a safer cosmetic preservative, using recombinant Escherichia coli β-galactosidase (β-gal). To confirm the molecular structure of synthesized OD-gal, mass spectrometry and NMR (1H- and 13C-) spectroscopy of OD-gal were carried out. In the reaction mixture, a sodium adduct ion of OD-gal (m/z=331.1732) was identified using mass spectrometry analysis. In addition, 1H NMR spectrum of OD-gal showed multiple peaks corresponding to the galactosyl group, which is evidence of galactosylation on OD. Downfield proton peaks at δH 4.39 ppm and multiple peaks from δH 3.98~3.55 ppm were indicative of galactosylation on OD. Up field proton peaks at δH 1.52~1.26 ppm and 0.89 ppm showed the presence of CH2 and CH3 protons of OD. 13C NMR spectrum revealed the presence of 24 carbons suggestive of α- and β-anomers of OD-gal. Among 14 carbon peaks from each anomer, the 4 peaks at δC 31.4, 29.0, 22.3, and 13.7 ppm were assigned to be overlapped showing only 24 peaks out of a total of 28 peaks. The mass value from mass spectrometry analysis of OD-gal, and 1H and 13C NMR spectral data were in good agreement with the expecting structure of OD-gal. Finally, we identified a galactose molecule from the hydrolysate of OD-gal using β-gal. We are expecting that through future study it will eventually be able to develop a safe cosmetic preservative.

Development of integrated microbubble and microfilter system for liquid fertilizer production by removing total coliform and improving reduction of suspended solid in livestock manure (가축분뇨 내 대장균 제거와 부유물질 저감 효율 향상을 통한 추비 생산용 미세기포 부상분리와 마이크로 필터 연계 시스템 개발)

  • Livestock manure is used as an organic fertilizer to replace chemical fertilizers after sufficient fermentation in an aerobic bioreactor. On the other hand, liquid manure disposal problems occur repeatedly because soil spraying is restricted during the summer when the crops are growing. To use liquid fertilizer (LF) as an additional nutrient source for crops, it is necessary to reduce the amount of suspended solids (SS) in the liquid fertilizer and secure stability problems against pathogenic microorganisms. This study examined the effects of the simultaneous SS removal and E.coli sterilization in the LF using the microbubble (MB) generator (FeMgO catalyst insertion). The remaining SS were further removed using the integrated microbubble and microfilter system. During the floating process in the MB device, the SS were removed by 57.9%, and the coliform group was not detected (16,200→0 MPN/100 mL). By optimizing the HRT of the integrated system, the removal efficiency of the SS was improved by 92.9% under the 0.1h of HRT condition. After checking the properties of the treated LF, 64.5%, 70.1%, 54.9%, and 51.5% of the TCOD, SCOD, PO4-P, and TN, respectively, were removed. The treated effluent from such an integrated system has a lower SS content than that of the existing LF and does not contain coliforms; therefore, it can be used directly as an additional fertilizer.

Characterization of Exolytic GH50A β-Agarase and GH117A α-NABH Involved in Agarose Saccharification of Cellvibrio sp. KY-GH-1 and Possible Application to Mass Production of NA2 and L-AHG (Cellvibrio sp. KY-GH-1의 아가로오스 당화 관련 엑소형 GH50A β-아가레이즈와 GH117A α-NABH의 특성 및 NA2와 L-AHG 양산에의 적용 가능성)

  • Recently, we sequenced the entire genome of a freshwater agar-degrading bacterium Cellvibrio sp. KY-GH-1 (KCTC13629BP) to explore genetic information encoding agarases that hydrolyze agarose into monomers 3,6-anhydro-L-galactose (L-AHG) and D-galactose. The KY-GH-1 strain appeared to possess nine β-agarase genes and two α-neoagarobiose hydrolase (α-NABH) genes in a 77-kb agarase gene cluster. Based on these genetic information, the KY-GH-1 strain-caused agarose degradation into L-AHG and D-galactose was predicted to be initiated by both endolytic GH16 and GH86 β-agarases to generate NAOS (NA4/NA6/NA8), and further processed by exolytic GH50 β-agarases to generate NA2, and then terminated by GH117 α-NABHs which degrade NA2 into L-AHG and D-galactose. More recently, by employing E. coli expression system with pET-30a vector we obtained three recombinant His-tagged GH50 family β-agarases (GH50A, GH50B, and GH50C) derived from Cellvibrio sp. KY-GH-1 to compare their enzymatic properties. GH50A β-agarase turned out to have the highest exolytic β-agarase activity among the three GH50 isozymes, catalyzing efficient NA2 production from the substrate (agarose, NAOS or AOS). Additionally, we determined that GH117A α-NABH, but not GH117B α-NABH, could potently degrade NA2 into L-AHG and D-galactose. Sequentially, we examined the enzymatic characteristics of GH50A β-agarase and GH117A α-NABH, and assessed their efficiency for NA2 production from agarose and for production of L-AHG and D-galactose from NA2, respectively. In this review, we describe the benefits of recombinant GH50A β-agarase and GH117A α-NABH originated from Cellvibrio sp. KY-GH-1, which may be useful for the enzymatic hydrolysis of agarose for mass production of L-AHG and D-galactose.

Evaluation of Regional Flowering Phenological Models in Niitaka Pear by Temperature Patterns (경과기온 양상에 따른 신고 배의 지역별 개화예측모델 평가)

  • Flowering time has been put forward due to the recent abnormally warm winter, which often caused damages of flower buds by late frosts persistently. In the present study, cumulative chill unit and cumulative heat unit of Niitaka pear, which are required for releasing the endogenous dormancy and for flowering after breaking dormancy, respectively, were compared between flowering time prediction models used in South K orea. Observation weather data were collected at eight locations for the recent three years from 2018-2020. The dates of full bloom were also collected to determine the confidence level of models including DVR, mDVR and CD models. It was found that mDVR model tended to have smaller values (8.4%) of the coefficient of variation (cv) of chill units than any other models. The CD model tended to have a low value of cv (17.5%) for calculation of heat unit required to reach flowering after breaking dormancy. The mDVR model had the most accurate prediction of full bloom during the study period compared with the other models. The DVR model usually had poor skills in prediction of full bloom dates. In particular, the error of the DVR model was large especially in southern coastal areas (e.g., Ulju and Sacheon) where the temperature was warm. Our results indicated that the mDVR model had relatively consistent accuracy in prediction of full bloom dates over region and years of interest. When observation data for full bloom date are compiled for an extended period, the full bloom date can be predicted with greater accuracy improving the mDVR model further.

Effects of Halogen and Light-Shielding Curtains on Acquisition of Hyperspectral Images in Greenhouses (온실 내 초분광 영상 취득 시 할로겐과 차광 커튼이 미치는 영향)

  • This study analyzed the effects of light-shielding curtains and halogens on spectrum when acquiring hyperspectral images in a greenhouse. The image data of tarp (1.4*1.4 m, 12%) with 30 degrees of angles was achieved three times with four conditions depending on 14 heights using the automatic image acquisition system installed in the greenhouse at the department of Southern Area of National Institute of Crop Science. When the image was acquired without both a light-shielding curtain and halogen lamp, there was a difference in spectral tendencies between direct light and shadow parts on the base of 550 nm. The average coefficient of variation (CV) for direct light and shadow parts was 1.8% and 4.2%, respective. The average CV value was increased to 12.5% regardless of shadows. When the image was acquired only used a halogen lamp, the average CV of the direct light and shadow parts were 2 .6% and 10.6%, and the width of change on the spectrum was increased because the amount of halogen light was changed depending on the height. In the case of shading curtains only used, the average CV was 1.6%, and the distinction between direct light and shadows disappeared. When the image was acquired using a shading curtain and halogen lamp, the average CV was increased to 10.2% because the amount of halogen light differed depending on the height. When the average CV depending on the height was calculated using halogen and light-shielding curtains, it was 1.4% at 0.1m and 1.9% at 0.2 m, 2 .6% at 0.3m, and 3.3% at 0.4m of height, respectively. When hyperspectral imagery is acquired, it is necessary to use a shading curtain to minimize the effect of shadows. Moreover, in case of supplementary lighting by using a halogen lamp, it is judged to be effective when the size of the object is less than 0.2 m and the distance between the object and the housing is kept constant.
