Three-dimensional micro photomachining of polymer using DPSSL (Diode Pumped Solid State Laser) with 355 nm wavelength (355nm 파장의 DPSSL을 이용한 폴리머의 3차원 미세 형상 광가공기술)

  • The basic mechanistic aspects of the interaction and practical considerations related to polymer ablation were briefly reviewed. Photochemical and photothermal effects, which highly depend on laser wavelength have close correlation with each other. In this study, multi-scanning laser ablation processing of polymer with a DPSS (Diode Pumped Solid State) 3rd harmonic Nd:YVO$_4$ laser (355 nm) was developed to fabricate a three-dimensional micro shape. Polymer fabrication using DPSSL has some advantages compared with the conventional polymer ablation process using KrF and ArF laser with 248 nm and 193 nm wavelength. These advantages include pumping efficiency and low maintenance cost. And this method also makes it possible to fabricate 2D patterns or 3D shapes rapidly and cheaply because CAD/CAM software and precision stages are used without complex projection mask techniques. Photomachinability of polymer is highly influenced by laser wavelength and by the polymer's own chemical structure. So the optical characteristics of polymers for a 355 nm laser source is investigated experimentally and theoretically. The photophysical and photochemical parameters such as laser fluence, focusing position, and ambient gas were considered to reduce the plume effect which re-deposits debris on the surface of substrate. These phenomena affect the surface roughness and even induce delamination around the ablation site. Thus, the process parameters were tuned to optimize for gaining precision surface shape and quality. This maskless direct photomachining technology using DPSSL could be expected to manufacture tile prototype of micro devices and molds for the laser-LIGA process.

Evaluation of biological activities of plasma-treated phloridzin (플라즈마 처리 phloridzin 반응물의 생리활성 평가)

  • Phloridzin is a predominant member of the chemical class of dihydrochalcones and mainly found in apple. The biological activity of phloridzin treated with dielectric barrier discharge (DBD) plasma was evaluated to investigate whether exposure to plasma can be used as a tools to enhance the biological activity of natural resources. DBD plasma treatment of phloridzin was carried out for three different exposure times. The antioxidant effects of degraded phloridzin for different reaction time were evaluated via radical scavenging assay using DPPH radical. In addition, the anti-diabetic and anti-obesity properties of the degraded phloridzin were measured based on ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities. Phloridzin treated for 60 min showed significantly higher radical scavenging, ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities compared to the tested positive controls. Degradation of phloridizin induced by DBD plasma might be responsible for enhancing the biological activity of phloridizin.

  • Apoptosis is an important mechanism that regulates cellular populations to maintain homeostasis, and the caspases, a family of cysteine proteases, are key mediators of the apoptosis pathway. Caspase-8 is an initiator caspase of the extrinsic apoptotic pathway, which is initiated by extracellular stimuli. Caspase-8 have two conserved domains, N-terminal tandem death effector domains (DED) and C-terminal two catalytic domain, which are important for this extrinsic apoptosis pathway. In extrinsic apoptosis pathway, death receptors which members of TNF superfamily are activated by binding of death receptor specific ligands from cell outside. After the activated death receptors recruit adaptor protein Fas-associated death domain protein (FADD), death domains (DD) of death receptor and FADD bind to each other and FADD combined with death receptor recruits procaspase-8, a precursor form of caspase-8. The DED of FADD and procaspase-8 bind to one another and FADD-bound procaspase-8 is activated by cleavage of the prodomain. This death receptor-FADD-caspase-8 complex called death inducing signaling complex (DISC). Cellular FLICE-inhibitory proteins (c-FLIPs) regulate caspase-8 activation by acting both anti- and pro-apoptotically, and caspase-8 activation initiates the activation of executioner caspases such as caspase-3. Finally activated executioner caspases complete the apoptosis by acting critically DNA degradation, nuclear condensation, plasma membrane blebbing, and the proteolysis of certain caspase substrates.

In vitro CaCO3 Crystallization at Room Temperature and Atmospheric Pressure Using Recombinant Proteins GRP_BA and GG1234 (재조합단백질 GRP_BA 및 GG1234를 이용한, 상온상압조건에서의 In vitro 탄산칼슘 결정화)

  • The exquisite structure and attractive biological properties of biominerals have great potential and increased interest for use in a wide range of medical and industrial applications. Calcium carbonate biomineralization, mainly controlled by shell matrix proteins, has been used as a representative model to understand the biomineralization mechanism. In this study, in vitro calcium carbonate crystallization was carried out under room temperature and atmospheric pressure using recombinant shell matrix protein GRP_BA and artificial shell matrix protein GG1234. Both proteins inhibited the growth of typical rhombohedral calcite crystals in the calcium carbonate crystallization using $CaCl_2$ solution and $(NH_4)_2CO_3$ vapor, and spherulitic calcite crystals with rosette-like structures were synthesized in both the presence of GRP_BA and GG1234. These results might be caused by the properties of block-like domain structure and intrinsically disordered proteins. We expect that this study can contribute to enhance understanding of the calcium carbonate biomineralization controlled by shell matrix proteins.

Expression of H-ras, RLIP76 mRNA and Protein, and Angiogenic Receptors in Corpus Luteum Tissues during Estrous Cycles (난소 내 황체조직에서 발정주기별 H-Ras, RLIP76, Angiogenic Receptors mRNA와 Protein의 발현)

  • Corpus luteum (CL) is a transient endocrinal tissue that undergoes repeated formation and regression during the estrous cycle. In this study, we hypothesized that the functional and structural mechanism of angiogenesis is similar between CL and tumor formation. First, we measured mRNA and protein expression of angiogenic receptors (vascular endothelial growth factor receptor-2, VEGFR2; Tie 2) in the early, middle, and late phase CL tissues during the estrous cycle. Ral-interacting protein of 76 kDa (RLIP76) and H-ras mRNA and protein were also expressed in the CL tissues. VEGFR2 and Tie 2 mRNA and protein were expressed in the early and middle phase CLs and decreased in the late phase. H-ras mRNA and protein were expressed in the early and middle phase CLs, but not in the late phase. RLIP76 mRNA was expressed in all phase CLs, and the protein expression was highest in early phase CLs. We suggest that RLIP76 and H-ras, an oncogenic gene, regulate the function of the CL during the estrous cycle, and the proteins will play an important role in the angiogenic mechanism of the CL.

Precise Deterioration Diagnosis and Restoration Stone Suggestion of Jungdong and Banjukdong Stone Aquariums in Gongju, Korea (공주 중동 및 반죽동 석조의 정밀 손상도 진단과 복원석재 제안)

  • This study focus on the restoration stone selection of break-out part based on material characteristics analysis and the conservational safety diagnosis using various nondestructive techniques for Jungdong and Banjukdong Stone Auariums. As a result, the original rocks of the stone aquariums body are porphyritic granodiorite with magnetite-series having igneous lineation, microcline phenocryst, veinlet and basic xenolith. As a result of the provenance presumption of the host rock, a rock around Gamgokri area in Nonsan City was identified the genetically same rock. Therefore, the rock is appropriate for restoration materials of the break-out part. The deterioration assessment showed that the stone aquariums were highly serious scaling, scale off and blackening. Particularly, the front face of Banjukdong stone aquarium needs reinforcement of structural crack (760mm) caused from igneous lineation of biotite. Blackening contaminants on the stone aquariums surface occurred by combining iron oxide, manganese oxide and clay mineral. Also, major factors of efflorescence contaminants were identified as calcite (Jungdong stone aquariums) and gypsum (Banjukdong stone aquariums). The physical characteristics of stone aquariums appeared that the original and new stone is third (moderately weathered) and second grade (slightly weathered), respectively. This study sets up an integrated conservation system from material analysis to restoration stone selection and conservational safety diagnosis of Jungdong and Banjukdong stone aquariums.

Raman Spectroscopic Study on Corrosion Layers of Archaeological Bronzes (라만분광분석을 통한 출토 청동유물의 부식층 연구)

  • Buried bronze objects are naturally corroded by their surrounding environment, which results in producing corrosion layers containing a number of constituents. Corrosion layers in stable condition protect the objects from the environment and also could provide information in terms of the objects. Characteristic and mechanism of the corrosion layers is likely to be valuable information for the conservation treatment. Many research have been conducted to figure out the formation and characteristic of the corrosion layers, but the more research should be conducted with various approach and analytical methods. Raman spectroscopy is one of the analytical methods to identify microcrystal as a compound while other analytical methods are used to identify element. Therefore, the aim of this research is to identify the characteristic of corrosion layers of both excavated bronze objects through the raman spectroscopy. Two analytical methods, which are raman spectroscopy and SEM~EDS, were used to analyse four excavated bronze bowls. In the case of bronze bowls, malachite was found from the exterior corrosion layer and albite, quartz, and microcline, which are minerals, were also found. Cuprite was detected from the interior corrosion layers illustrating slightly different spectrum due to the combined compound. Lead segregation shows the form of PbO, $PbSO_4$ and $PbCO_3$ or it replaced as cuprite. In this study, small number of samples were analysed. This research is likely to be useful information to figure out not only the characteristic of the corrosion layers but also the authenticity of the artifacts if relevant research will be conducted. Therefore, further comprehensive researches on the various archaeological objects and corrosion environment condition are required in the future.

Molecular Characterization and Expression Analysis of Nucleoporin 210 (Nup210) in Chicken

  • Nucleoporin 210 (Nup210) is associated with several physiological processes including muscle and neural cell differentiation, autoimmune diseases, and peripheral T cell homeostasis. Chicken Nup210 (chNup210) gene was originally identified as one of the differentially expressed genes (DEGs) in the kidney tissues of chicken. To elucidate the role of Nup210 in metabolic disease of chicken, we studied the molecular characteristics of chNup210 and analyzed its gene expression under the stimulation of Toll-like receptor 3 (TLR3) ligands. The Nup210 genomic DNA and amino acid sequences of various species including fowls, fishes, and mammals were retrieved from the Ensemble database and subjected to bioinformatics analyses. The expression of Nup210 from several chicken tissues was probed through qRT-PCR, and chicken fibroblast DF-1 cell line was used to determine the change in expression of chNup210 after stimulation with TLR3 ligand, polyinosinic-polycytidylic acid (poly (I:C)). The chNup210 gene was highly expressed in chicken lung and spleen tissues. Although highly conserved among the species, chNup210 was evolutionary clustered in the same clade as that of duck compared to other mammals. Furthermore, this study revealed that chNup210 is expressed in TLR3 signaling pathway and provides fundamental information on Nup210 expression in chicken. Future studies that offer insight into the involvement of chNup210 in the chicken innate immune response against viral infection are recommended.

An Exploratory Study on Sales and Operations Planning as SCM Supporting Tool (공급망 관리 지원도구로서의 S&OP 운영에 관한 탐색적 연구)

  • S&OP(Sales and Operations Planning) is an ongoing process of periodic planning, reviewing, and evaluation through the involvement of all key stakeholders. Within this process, performance is regularly reviewed and early warning signals are generated, so that the company can react quickly to changing market and operational environment. This paper presents a framework for effective S&OP for fair alignment, accountability, teamwork, visibility, and risk management. This framework focuses on supply chain information governance, level of information sharing through S&OP, role of S&OP as coordination mechanism, APS effectivesness as a planning tool and SCM performance. In addition, a brief case study on the operating characteristics of S&OP at three Korean firms is presented. Implications of the study finding are also provided. It will also make companies that are considering the introduction of S&OP aware of the importance of S&OP, which will provide practical guidelines for the introduction of S&OP.

The Analysis of Stakeholders' Conflict Surrounding Water use Charges: Targeting the Han River region (한강수계 물이용부담금을 둘러싼 이해당사자 간 갈등분석)

  • This research purposes to design a methodological framework to suggest the optimal method to resolve the conflicts of stakeholders surrounding the water use charge of the Han River region, and to use the analysis results to provide the direction of policy. For this, it was preceded that the process of understanding the mechanisms of the multifaceted conflict between decision makers taking different positions over water use charge of the Han River region, and an optimal method to resolve the conflict of water use charge of the Han River region was derived by using a graph model for conflict resolution(GMCR). According to the analysis results, the optimal state to find a solution to the water use charge of the Han River is that the Seoul-Incheon-Gyeonggi region pays the charge according to the original rate while Seoul-Incheon requesting discount the water use rate. In addition, the Han River management committee should establish policies desired by Seoul-Incheon-Gyeonggi region including rationalization of the decision-making structure to determine the rate of water use charge, making the basis to support the Han River management fund system for the Seoul-Incheon region, and transparent management of the Han River management fund system considering the characteristics of beneficiary regions and residents. This study is expected to provide objective decision-making information in establishing environmental policy directions related to conflict resolution in the water use charge of the Han River region and to offer a methodological basis for similar follow-up studies related to conflicts derived from sharing nature environment.
