Comparison of Pood Chemical Properties in Olive Flounder (Paralichthys olivaceus), Spotted Flounder (Verasper variegatus) and Their Hybrid Muscle (넙치, 범가자미 및 잡종 (넙치 male $\times$ 범가자미 female) 근육의 식품화학적 특성 비교)

  • The proximate composition, mineral content, fatty acid composition, ATP related compounds, amino acid composition, color and texture were investigated with dorsal and ventral muscle from olive flounder, spotted flounder and hybrid (olive flounder m $\times$spotted flounder f). Spotted flounder and hybrid were higher in moisture content, and lower in crude protein content than those of olive flounder, Potassium content in hybrid was higher than that in olive flounder and spotted flounder. Hybrid was lower in calcium, iron, manganese content, and higher in magnesium content than olive flounder and spotted flounder. The contents of saturated fatty acid, unsaturated fatty acid and docosahexaenoic acid (DHA) in hybrid were intermediate level of spotted flounder and olive flounder. Fatty acid composition of dorsal muscle was slightly similar to ventral muscle. Adenosine triphosphate (ATP) and its related compounds contents and amino acid content in hybrid were intermediate level of spotted flounder and olive flounder, and these compounds of dorsal muscle were slightly similar to those of counterpart. The major amino acids such as aspartic acid, glutamic acid, leucine and Iysine were very similar to all the samples. Total amino acid and essential amino acid contents in dorsal muscle were slightly higher than those in ventral muscle. Free amino acid content and composition in hybrid were similar to spotted flounder, and free amino acid content in dorsal muscle was higher than in ventral muscle, The lightness of hybrid and spotted flounder was stronger than that of olive flounder, and was stronger in dorsal muscle. The breaking strength of hybrid was slightly lower than that of spotted flounder, and was stronger in dorsal muscle.

  • The alkaline protease gene was cloned from a halo-tolerant alkalophilic Bacillus clausii I-52 isolated from the heavily polluted tidal mud flat of West Sea in Inchon Korea, which produced a strong extracellular alkaline protease (BCAP). Based on the full genome sequence of Bacillus subtilis, PCR primers were designed to allow for the amplification and cloning of the intact pro-BCAP gene including promoter region. The full-length gene consists of 1,143 bp and encodes 381 amino acids, which includes 29 residues of a putative signal peptide and an additional 77-amino-acid propeptide at its N-terminus. The mature BCAP deduced from the nucleotide sequence consists of 275 amino acids with a N-terminal amino acid of Ala, and a relative molecular weight and pI value was 27698.7 Da and 6.3, respectively. The amino acid sequence shares the highest similarity (99%) to the nattokinase precursor from B. subtilis and subtilisin E precursor from B. subtilis BSn5. The substrate specificity indicated that the recombinant BCAP could hydrolyze efficiently the synthetic substrate, N-Suc-Ala-Ala-Pro-Phe-pNA,and did not hydrolyze the substrates with basic amino acids at the P1 site. The recombinant BCAP was strongly inhibited by typical serine protease inhibitor, PMSF, indicating that BCAP is a member of the serine proteases.

Structure and mechanical properties of Korean commercial silkworm cocoon (장려품종 누에고치의 구조 및 기계적 특성)

  • Korean commercial Bombyx mori silkworm cocoon made by Kumokjam and Daesungjam was examined to characterize for application. Amino acid analysis showed that the main amino acid of Kumokjam and Daesungjam cocoon was glycine, alanine, serine, tyrosine, and aspartic acid. The content of three major amino acids was about 67% and the content of long chain amino acids was about 37%. Stress-strain curve showed that breaking stress of Kumokjam cocoon, 50 MPa, was higher than that of Daesungjam cocoon, 48 MPa. Breaking strain of Kumokjam cocoon, 37%, was also longer than that of Daesungjam cocoon, 30%. These results might be used as basic information for development of non-textile materials using Bombyx mori silkworm cocoon.

Processing and Fermentation of Food Wastes with MS Microorganism Complex for Swine Feeds (MS 미생물복합군에 의한 음식폐기물의 양돈사료화를 위한 적정처리공정)

  • In order to investigate the proper processing of food wastes with miraculous soil-microorganisms (MS) for final use of swine feeds, calory, amino acid and fatty acid in food wastes were determined in relation with fermentation process with MS microorganism complex. Aflatoxin test was also performed to check safety of the fermented food wastes. Calory of food wastes was determined in average $7.60\;Kcal{\cdot}g^{-1}\;D.W.$ In finally processed food wastes, total content of amino acid was $93.0\;mg{\cdot}g^{-1}]\;D.W$, showing 18.5% of increase by the anaerobic fermentation. Essential and non-essential amino acids were measured at respectively 34.43 and $58.56\;mg{\cdot}g^{-1}\;D.W.$ Leucine, phenylalanine, isoleucine and threonine of essential amino acids and proline and glutamic acid of non-essential amino acids were highly composed as compared to others. The composition of fatty acid in food wastes was also increased by anaerobic fermentation for 3 weeks. Palmitic acid, oleic acid and palmitoleic acid were more important in quantity. Present results indicate that food wastes properly processed with MS have enough calory and are safe from aflatoxin, and that anaerobic fermentation with MS microorganism in an efficient process for hydrolyzing protein and lipids in food wastes.

Comparison of Free Amino Acids and Dipeptide Contents, Warner-Bratzler Shear Force and Sensory Property of Beef Jerky Manufactured with Different Drying Methods (건조방법에 따른 우육포의 유리아미노산 및 Dipeptide 함량, 물성 및 관능 특성 비교)

  • This study was conducted in order to investigate the effect of different drying methods on free amino acids and dipeptide contents, Warner-Bratzler shear force and sensory property of beef jerky products. The drying methods used for this study were sun-drying, shade-drying and hot air-drying methods. The sun-dried beef jerky had the highest free amino acids (277.45 mg/100 g) followed by shade-dried (206.43 mg/100 g) and hot air-dried (111.88 mg/100 g) ones, whereas the amount of dipeptides were the highest in the shade-dried beef jerky followed by sun-dried and hot air-dried ones. The fatty acid composition of the beef jerky was not shown any difference among the beef jerkies with 3 different drying methods. The color $L^*$-value of the beef jerky was lower in the hot-air dried, while the $a^*$-value was higher in the sun-dried one. The shade-dried jerky showed a lower $b^*$-value. The shear force of beef jerky was lower when it was either sun-dried or shade-dried than when it was hot air-dried. Sensory evaluation revealed that hot air-dried beef jerky scored lower than the sun-dried and shade-dried ones (p<0.05). Therefore, using either the sun drying or shade drying methods is more desirable than using the hot air drying method when manufacturing beef jerky for superior physicochemical qualities.

STUDIES ON THE UTILIZATION OF ANTARCTIC KRILL 1. Compositional Characteristics of Fresh Frozen and Preboiled Frozen Krill (남대양산 크릴의 이용에 관한 연구 1. 크릴의 식품원료학적인 성상)

  • For the use of antarctic krill as a fond protein source its compositional characteristics were investigated as the first part of the work includes other subjects such as processing of drill paste, concentrates, and fermented or seasoned product. In general composition of fresh frozen and preboiled frozen krill on board, the contents of crude fat and free amino nitrogen were higher in the former than in the latter which contained a high amount of ash. VBN was rather high as much as 37.6 and $26.4\;mg\%$ in both fresh frozen and preboiled krill. The pH of drill homogenates was 7.1 to 7.2 in both cases. Such a low pH might be attributed to a long term storage and temperature fluctuations during frequent transshipping. The amino acid competition of fresh frozen krill meat showed relatively high amount of glutamic acid, aspartic acid, lysine, proline, and leucine while methionine, histidine, serine, tyrosine, and phenylalanine were lower. Among the essential amino acids lysine and leucine were higher and methionine was lower. In tile composition of free amino acid proline, lysing, arginine, and alanine were higher comparatively to the contents of histidine, aspartic acid, serine, and threonine. It is noteworthy for nutritional qualification that tile essential amino acids particularly as lysine were abundant similarly to that of fishes. Heavy metal contents of krill meat 0.039 to 0.048 ppm as Hg, 0.06 to 0.11 ppm as Pb, less than 0.32 ppm as Zn, 0.008 to 0.012 ppm as Cd, 0.61 to 0.68 ppm as Fe, 0.87 to 1.37 ppm as Cu, and nondetective as Cr. A high Cu content seems to be resulted by tile blood pigment of crustacea. The ratio,1 of edible portion to non-edible portion were 37:63 in fresh frozen and 42:58 in preboiled frozen krill respectively. Release of drip after thawing was more in fresh frozen than in preboiled frozen drill marking $36\%$ and $24\%$ of both respectively.

Free Amino Acid and Polysaccharide Content of Submerged Mycelial Culture of Fomitopsis pinicola (귤피 및 녹차추출물에서 배양한 소나무잔나비버섯(Fomitopsis pinicola) 균사체의 유리아미노산 및 다당류 함량)

  • Free amino acids and polysaccharide content of submerged mycelial culture of Fomitopsis pinicola using $2\%$ citrus peel water extracts(CP), $2\%$ green tea water extracts(GT) and yeast-malt broth(YM) were investigated. Cultivations were conducted at $30^{\circ}C$ and 150 rpm for 10 days. Yields of the mycelium as fresh weight basis in GT-, CP- and YM-broth were $42.3\%,\;34.2\%\;and\;9.89\%$, and their turbidity(OD at 660 nm) of the broth without mycelium were 0.14, 0.16 and 0.22, respectively. Total free amino acid content in the mycelium were 928.19 $mg\%$ in YM, 1060.53 $mg\%$ in CP, 764.83 $mg\%$ in GT, and the major free amino acid was lysine in YM, glutamic acid in CP and GT. Total free exo-amino acid contents were 659.75 $mg\%$ in YM, 954.55 $mg\%$ in CP, 838.69 $mg\%$ in GT, and the major in the all broths was glutamic acid Total amino acid derivatives content of the mycelium were in order of CP>GT>YM, and the major was cystathionine in YM, hydroxy proline in CP and GT. The major among exo-amino derivatives was hydroxy proline in the all broth. Exo-AIS content was in order of CP>GT>YM. Acid soluble polysaccharide content of the mycelium was GT($0.69\%$)>YM($0.39\%$)>CP($0.18\%$). The exo-polysaccharide content was in order of GT($0.87\%$)>CP($0.69\%$)>YM($0.09\%$). Alkali soluble polysaccharide content of the mycelium was in order of CP($5.21\%$)>GT($5.18\%$)>YM($4.56\%$), and exo-polysaccharide was in order of GT($6.79\%$)>YM($3.57\%$)>CP($3.01\%$). The alkali soluble polysaccharide eluted from mycelium cultivated in CP broth was supposed to polysaccharide(about 500,000 daltons) composed of hexose and uronic acid bounded with protein(below 10,000 daltons).

Effects of Feeding Citrus by Products on Nutritional Components of Korean Native Chickens (토종닭 고기의 영양 성분에 미치는 감귤부산물 급여의 영향)

  • In this study, the effects of feeding citrus byproducts on nutritional components of Korean native chickens were investigated. Two samples of Korean native chickens were used for this study: T0 (chickens fed with only feed for laying hen until they became 39 weeks old, not with citrus byproducts) and T1 (chickens fed with citrus byproducts). The feed for T1 was the same as T0 for the first 16 weeks, and then was made by adding 4% of citrus byproducts to the feed for T0 between $17{\sim}39$ weeks. The chicken used for the experiment was obtained by chilling them for 2 days after slaughter. There was no significant difference between T0 and T1 regardless of feeding citrus byproducts, in terms of their breast/thigh's calorie, contents of moisture, protein, fat and ash, total structural amino acid, total free amino acid and composition of saturated/unsaturated fatty acids. The breast's cholesterol amount of T0 and T1 were 48.4 mg/100 g and 47.6 mg/100 g, respectively, while that of thigh for T0 and T1 were 75.7 mg/100 g and 72.8 mg/100 g, respectively, which implies that T1 showed lower amount of cholesterol than T0. T1 showed significantly higher amount of phosphorus, potassium, magnesium and vitamin B2 for thigh than T0 (p<0.05).

Component Analysis and Antioxidant Activity of Kalopanax pictus Leaf (개두릅의 함유성분 분석과 항산화 활성)

  • The purpose of this study is to determine the possibility of using Kalopanax pictus leaf as natural health food source. The contents of proximate and antioxidative nutrients of Kalopanax pictus leaf were measured. The contents of carbohydrate, crude protein, crude lipid and ash were 41.42%, 45.23%, 3.29% and 10.07%, respectively. The calories of Kalopanax pictus leaf was 401.52 kcal/100 g and total dietary fiber was 30.37%. The percentages of water soluble dietary fiber to insoluble dietary fiber were 9.16% and 21.21%, respectively. The protein contained a total of 18 different kinds of amino acids. The contents of essential and non-essential amino acids were 10.51 g/100 g and 17.69 g/100 g. The K was the largest mineral followed by P, Ca, and Mg, which means Kalopanax pictus leaf is alkali material. The contents of saturated fatty acids, monounsaturated fatty acids and polyunsaturated fatty acids were 1.56 g/100 g, 0.11 g/100 g and 1.84 g/100 g, respectively. The antioxidant activity of 70% ethanol extract and fractions of the Kalopanax pictus leaf has been determined by the scavenging of the stable radical DPPH; the result showed that the ethyl acetate fraction was the most active, as the amount required for 50% reduction of DPPH after 30 mins ($RC_{50}$) was $105.7\;{\mu}g$, followed by 70% ethanol extract ($247.3\;{\mu}g$), hexane fraction ($120.7\;{\mu}g$), chloroform ($107.3\;{\mu}g$), butanol fraction ($110.1\;{\mu}g$) and aqueous fraction ($491.9\;{\mu}g$).

Varietal Analysis and Quantification of Amino Acid in Mulberry Fruits (뽕나무 계통별 오디의 아미노산 함량 분석)

  • To reveal the possibility as a source of functional food and to increase the usability of mulberry fruits, amino acid analysis with mulberry fruits was carried out with L-8800 Amino acid auto analyzer. We analyzed 16 amino acid components including 6 essential amino acids. According to the results, the mean content of total amino acid was 4,640.8 mg/100 g DW. 'Sabangso(II)' showed the highest total amino acid content (6,919.3 mg/100 g DW) among the fifty accessions tested, whereas 'Gukkwang' showed the lowest content (3,389.1 mg/100 g DW). The distribution rate of amino acid components decreased in the order of Glu ($20.1\%$) > Gly ($12.7\%$) > Asp ($12.5\%$) > Thr ($8.2\%$) > Ala ($7.9\%$) > Arg ($7.5\%$) > Pro ($6.1\%$) > Ser ($6.1\%$) > Val ($5.7\%$) > Leu ($5.3\%$) > Ile ($2.7\%$) > His ($2.5\%$) > Met ($0.9\%$) > Tyr ($0.8\%$) > Lys ($0.7\%$) > Cys ($0.3\%$). 'Sabangso(II)' showed the highest content in 11 amino acid components, which were Glu (1,293.2 mg/100 g), Ser (390.2 mg/100 g), Thr (564.4 mg/100 g), His (175.3 mg/100 g), Arg (590.4 mg/100 g), Lys (53.5 mg/100 g), Gly (589.4 mg/100 g), Ala (499.3 mg/100 g), Leu (481.9 mg/100 g), Ile (240.9 mg/100 g), and Val (407.6 mg/100 g). Tyr and Cys were the highest in 'Jangsosang' (70.8 mg/100 g, 19.3 mg/100 g, respectively), and Asp, Pro, and Met were the highest in 'Jasan(I)', 'Hwansipchosaeng' and 'Palcheongsipyung(I)' (1,098.8 mg/100 g, 364.4 mg/100 g, 68.8 mg/100 g, respectively).
