Monitoring on Alcohol and Acetic Acid Fermentation of Potatoes Using Response Surface Methodology (반응표면분석에 의한 감자의 알콜 및 초산발효조건 모니터링)

  • 상품성이 떨어지는 감자의 효율적 활용을 위하여 감자식포 제조공정을 반응표면분석에 의해 최적화하였다. 품질이 일정하고 단기간에 대량생산이 가능한 발효조건을 확립하기 위하여 반응표면분석으로 식초 제조조건 설정을 위해 알콜 발효와 초산발효 2단계로 구분하여 모니터링하였다. 그 결과 1단계 알콜발효에서 알콜함량이 최대치를 나타내는 조건은 감자에 대한 백분율로 첨가한 가수량 241.35%(v/w), 교반속도 8.05 rpm 및 발효시간 34.81 hr으로 나타났다. 알콜발효에서 알콜함량에 대한 최적조건으로 알콜발효 후 2단계로 초산발효를 행하여 초산발효조건을 예측하였다. 그 결과 총산이 최대치를 나타내는 조건은 교반속도 169.89 rpm 및 발효시간 285.12 hr로 나타났다. 최적조건에서 예측된 알콜발효 및 초산발효의 결과는 최적조건에서 실제 발효를 행한 결과와 유사하였다.

  • In this study, it was observed that hydrogen productivity varied with changes of input g1ucose concentration and dilution rate in FBR( Fixed Bed Reactor), and CSTR(Continuous Stirred Tank Reactor). We evaluated and compared reaction rate Parameters and internal external and overall mass transfer resistances of immobilized carrier in both reactors. Apparent $K_m$ decreased with increasing dilution rate in FBR but showed a constant value above $0.4h^{-1}$ of dilution rate in CSTR. The experimental results in FBR showed nearly analogous to those in CSTR, however, the performance of FBR resulted in lower hydrogen productivity and an external effectiveness factor but a higher internal effectiveness factor than in CSTR. The overall effectiveness factor obtained with various input 91ucose concentrations showed similar values in both reactors.

Xylitol Production from D-Xylose by Candida mogii ATCC 18364 (Candida mogii ATCC 18364를 이용한 D-Xylose로부터 Xylitol 생산)

  • Fermentation characteristics of D-xylose into xylitol by Candida mogii ATCC 18364, a potential xylitol producer from rice straw hemicellulose hydrolyzates, were investigated. The influences of the most important operational variables on xylitol production were examined. The best results in xylitol production were obtained in shake-flask fermentations when 3.0 g/L initial cell concentration of 12 hr-old cells grown in D-glucose containing medium were used as inoculum. The oxygen availability is a critical factor in xylose fermentation, therefore, xylose conversion into xylitol was investigated in a 2-L fermenter at different stirring rates. Maximum xylitol production was obtained with an aeration rate of 1 vvm at a stirring rate of 200 rpm.

  • One of the most important factors which affect the impact strength of high impact polystyrene (HIPS) is the rubber-phase particle size and size distribution. In this study, HIPS was prepared from a batch reactor to observe the influence of reaction conditions such as rubber content, agitation speed and prepolymerization time on the particle size and size distribution. Measurements concerning the particle size distribution were conducted using a particle size analyzer. Due to swelling, the particle suspended in toluene increases in size with lower heat-treatment temperature and shorter heat-treatment time, while the particle in methyl ethyl ketone shows quite reasonable size without any effort of heat-treatment. As rubber content increases, the average particle size increases substantially, but the increase in agitation speed at lower rubber contents does not have much influence on the size. However, the polystyrene-phase particles occluded in rubber-phase become more uniform as agitation speed increases. Longer prepolymerization time produces rubber-phase particles with narrower particle size distribution.

Optimization for The Alcoholic Fermentation of Concentrated Grape Juice Using Response Surface Methodology (반응표면분석을 이용한 농축 포도즙의 알코올발효 조건 최적화)

  • Response surface methodology (RSM) was applied to optimize alcohol fermentation of concentrated grape juice. Sugar concentration ($X_1$), agitation rate ($X_2$) and fermentation time ($X_3$) were chosen as the independent variables of the central composite design (CCD). Dependent variables were alcohol content ($Y_1$) and total acidity ($Y_2$). To optimize two dependent variables, desirability function was defined as $Y_1$=10.0% and $Y_2$=minimum. The optimum conditions for alcohol fermentation were $19.98^{\circ}Bx$ (sugar concentration), 104.1 rpm (agitation rate) and 89.67 hr (fermentation time). The predicted responses were 10.0% in alcohol content and 0.86% in total acidity. The coefficients of determination ($R^2$) were 0.948 and 0.958, which indicate that the model fit was highly significant (p<0.001). The experimental values were 10.1% for alcohol content and 0.88% for total acidity. These values were similar to the predicted values from RSM.

  • Recently, we isolated the mutant Kluyveromyces marxianus 36907-FMEL1, which demonstrated improved xylose reductase activity as compared to the parental strain, K. marxianus ATCC 36907. Effects of agitation conditions on xylitol production were verified using a bioreactor system. Under an agitation speed of 400 rpm, K. marxianus 36907-FMEL1 exhibited the highest xylitol yield (0.57 g/g) and productivity ($0.64g{\cdot}l^{-1}{\cdot}h^{-1}$) at $30^{\circ}C$. When the fermentation temperature was increased to $40^{\circ}C$, interestingly, xylitol yield and productivity were found to be increased to 21% (0.64 g/g) and 58% ($0.90g{\cdot}l^{-1}{\cdot}h^{-1}$), respectively, under the optimized agitation conditions.

The Recovery of Silver from Thiourea Leaching Solution by Cementation Technique (침전법을 이용한 Thiourea 용출용액으로부터 Silver 회수)

  • In order to apply the silver cementation method using Fe powder from pregnant thiourea leaching solution. Parameters such as the amount of Fe powder addition, agitation speed, and temperature were investigated. The silver cementation rate was increased by the increasing of Fe powder addition, agitation speed, and temperature. The highest silver cementation rate was found when the addition of Fe powder was 50 g/L at the agitation speed of 500 rpm. The silver cementation rate increase with increasing temperature according to the Arrhenius equation and obeys $1^{st}$ order kinetics. The activation energy from the kinetics data was found to be between 13.73 KJ/mol and 17.02 KJ/mol. In the XRD analysis, goethite was detected in the precipitate of the thiourea leach solution. This indicates that an oxidation-reduction reaction had occurred in the thiourea solution due to the addition of the Fe powder.

  • Metal emulsions have been studied for several decades as a method of increasing the efficiency of the steelmaking process. This study was performed using a water model, observable at room temperature, to compensate for the disadvantages of the high-temperature experiment, the results of which are difficult to observe visually. As a substitute for metal-in-slag emulsions, experiments were conducted by dropping distilled water into silicone oil and comparing the results with the results of a calculation by momentum balance equations. The descending velocity of the water droplet decreased as the diameter of the droplet and viscosity of the fluid (silicon oil) increased. To simulate the descending velocity of a water droplet in silicon oil under stirring conditions, the flow rate of the fluid (silicon oil) was measured by particle image velocimetry (PIV) methods. The calculation of the descending velocity of the water droplet was in good agreement with the measured values, with and without stirring a viscous silicone oil.

Enhanced Production of Gellan by Sphingomonas paucibilis NK-2000 with Shifts in Agitation Speed and Aeration Rate after Glucose Feeding into the Medium (Sphingomonas paucibilis NK-2000 균주가 생산하는 젤란의 생산 농도 향상을 위한 포도당 첨가 및 교반속도와 통기량 변화 방법의 최적화)

  • Optimal agitation speed and aeration rate for the production of gellan by Sphingomnas paucibilis NK2000 in a 7 l bioreactor were found to be 400 rpm and 1.0 vvm. The best time for glucose feeding into the medium for enhanced production of gellan by S. paucibilis NK2000 was 36 hr after cultivation. The concentrations of gellan produced by S. paucibilis NK2000 from 1) 20.0 g/l glucose without additional feeding, 2) 20.0 g/l glucose with feeding of 200.0 g/l glucose at 36 hr, in which the final concentration in the medium was 10.0 g/l, 3) 20 g/l glucose with feeding of 200.0 g/l glucose and a shift in an agitation speed from 400 to 600 rpm, 4) 20.0 g/l glucose with feeding of 200.0 g/l glucose at 36 hr and shifts in an agitation speed from 400 to 600 rpm and an aeration rate from 1.0 to 1.5 vvm, 5) and 20.0 g/l glucose with feeding of 200.0 g/l glucose at 36 hr and shifts in an agitation speed from 400 to 600 rpm and an aeration rate from 1.0 to 2.0 vvm, were 5.19, 5.74, 6.73, 7.93, and 9.40 g/l, respectively, and their conversion rates from glucose were 26.0, 19.1, 22.4, 26.4, and 31.3%, respectively. Compared to those developed using a normal process, production of gellan by S. paucibilis NK2000 from 20.0 g/l glucose was 1.81 times higher, and and its conversion rate was 1.20 times higher when the optimized process developed in this study was used.

The Effects of Iron Powder Agglomeration on the Copper Removal Efficiency during Cementation Process for Treating Mine Drainages (광산배수 처리를 위한 세멘테이션 공정 중 구리제거효율에 대한 철분 응집의 영향)

  • The effects of equivalent of iron powder, particle size, agitation speed on the removal efficiency of copper ion were investigated by adding iron powder as cementation agents to simulated mine drainage solution with 117.15 mg/L Cu ion. The 50 % of Cu was removed at 90 min with 2 equivalent of iron powder while more than 99 % of Cu was removed at 60 min with 16 equivalent at 200 rpm and $20^{\circ}C$. The removal efficiencies of Cu ion were not different using 2 equivalent of $48{\mu}m$ and $150{\mu}m$ iron powder, and the removal efficiency increased rapidly with increasing the agitation speed to more than 400 rpm. This lower removal efficiency resulted from agglomeration of iron powder observed by SEM, which could reduce the effective specific surface area. More than 99 % of copper ion was removed using 2 equivalent of $48{\mu}m$ iron powder at 60 min, 600 rpm and $20^{\circ}C$.
