Studies on the Photosynthesis of Korean Ginseng III. Effects of the Light Transparent Rate of Shading on the Photosynthesis Ability of Korean Ginseng Plant (Panax ginseng C. A. Meyer) (고려인삼엽의 광합성능력에 관한 연구 III. 투광율이 광합성 능력에 미치는 영향)

  • This study was conducted to define the effects of light transparent rate of the shading on the photosynthesis ability of the ginseng leaves and their seasonal changes. Regardless the effects of light transparent rate of shading and age of ginseng plant, 10,000 lux was the most adequate light intensity for the maximum photosynthesis of ginseng leaves and seasonal difference was not significant. The ginseng plants which were grown under 10 to 15 percent light transparent shading showed the highest photosynthesis ability. The photosynthesis ability of ginseng leaves was significantly decreased in September than June and the decreasing rate was higher at the ginseng plants planted on back rows than front rows. In June, the ginseng plants grown under 10 to 15 percent light transparent shading showed high respiration amount but in September, those grown under 20 to 25% light transparent shading showed the highest respiration. The amount of chlorophyll of ginseng leaf was significantly decreased by increasing light transparent rate of shading.

Isolation of Photosynthetic Bacterium, Rhodopseudomonas palustris JK-1 and Researches on IAA and Carotenoid Production (광합성세균 Rhodopseudomonas palustis 분리 및 IAA와 Carotenoid 생성에 관한 연구)

  • The JK-1 isolate which was the best producer of indole-3-acetic acid and carotenoid among the 388 strains isolated from 28 wetlands in Jeju, was identified to be Rhodopseudomonas palustirs belongs to a typical group of non sulfur purple bacteria based on 16S sRNA sequencing. This study investigated the effect of different cultural conditions of pH, temperature, agitation, light and aeration on growth, IAA and carotenoid production of photosynthetic bacterium JK-1 for optimization of IAA and carotenoid production. It was found that growth, IAA, carotenoid, and bacteriochlorophyll production with light (3,000~3,500 Lux) and agitation (100 rpm) showed better results than those with dark/static or dark/agitation (100 rpm) in anaerobic conditions. The optimal pH, temperature and agitation speed for cell growth were 7, $30^{\circ}C$, 150 rpm, for IAA production were 9, $30^{\circ}C$, 150rpm and for carotenoid production were 6, $25^{\circ}C$, 50 rpm, cultured for 72 h under anaerobic light, respectively. The growth and IAA production were high in aerobic culture compared with anaerocic culture, whereas carotenoid and bacteriochlorophyll content were decreased extremely in aerobic condition (0.5~1 vvm). Subsequently, the optimal culture conditions for JK-1 were selected with pH 7, $30^{\circ}C$ and 100 rpm under anaerobic light and the effect on plant growth was tested by pot assay. Inoculation of JK-1 with 3% (v/v) level caused increase in shoot and root dry weigh that varied from 20%~58% to 40%~28% in young radish in camparison to uninoculated treatment at 50 days of growth. The study suggests that the JK-1 isolate may serve as efficient biofertilizer inoculants to promote plant growth.

  • To investigate the effect of low light intensities and the inoculation of Frankia and/or Alpova diplophloeus on the symbioses development and their host growth, red alder(Alnus rubra Bong.) seedlings were grown in an air - filtered walk - in growth chamber with either $N_2$ - fixing Frankia inoculation or N - fertilization and live or dead spore inoculation of the ectomycorrhizal fungus A. diplophloeus(Zeller & Dodge) Trappe & Smith. When they were 20 weeks old, the seedlings were grown under three levels of light intensities of 680, 320 and $220{\mu}mol/m^2/s$ PPFD(photosynthetic photon flux density) for three weeks. PPFD of 220 significantly decreased the development of A. diplophloeus mycorrhizae and nodules, the rates of $N_2$ - fixation and $CO_2$ exchange, and the growth of tile seedlings. PPFD 320 significantly decreased the $CO_2$ exchange rate only. Frankia inoculation significantly increased mycorrhiza formation and seedling growth. Alpova inoculation significantly increased seedling growth but not nodule development and $N_2$ - fixation. None of the symbionts affected $CO_2$ exchange rates. Frankia was more critical for seedling growth and mycorrhizal development than the mycorrhizal fungus for seedling growth and nodule development.

  • Effect of effective microorganisms on the growth of plant (salvia sp.) was investigated. Microorganisms used were photosynthetic bacteria, lactic acid bacteria and yeasts. When photosynthetic bacteria were inoculated to soil by 100 dilution, treated plants showed 160% growth by length compared to control. When photosynthetic bacteria, lactic acid bacteria and yeasts were mixed, diluted by 10 and inoculated to soil, the plants showed 212% growth compared to control. Microbial populations were increased in the treated soil.

  • The photochemical reaction of harmaline and N-methylmaleimide has been investigated by spectroscopic methods. A photoproduct was isolated from the irradiation mixture of harmaline and N-methylmaleimide. Spectroscopic results suggested that the amine group of harmaline was added photochemically to the double C=C bond of N-methylmaleimide. This synthesized harmaline derivative has a biological toxicity, because it inhibits the growth of some yeasts and photosynthetic bacteria.

  • This study was conducted to define the seasonal differences in the morphological characteristics, the photosynthetic abilities and the dark respirations of the 4-year old ginseng leaves. Chlorophyll-a content in the ginseng leaf was significantly decreased at September than at June but content of chlorophyll-b was not showed seasonal difference. At June, the amounts of chlorophyll a and b in the ginseng leaves grown in the back row were rather abundant than those grown in the front row, but no significant differencies were detected between rows at September. The estimated optimum light intensity for the photosynthesis of ginseng leaves was higher at June than at September and higher in the front row than the back row but was significantly decreased by air temperature above 25$^{\circ}C$. The light compensation point was elevated in higher temperature and at September than June. The amount of photosynthesis was significantly increased in the ginseng plant grown in the front row than the back row at June but the reverse was significant at September. The highest photosynthesis was observed in temperature range of 20 - 25$^{\circ}C$ at June and range of 15 -20$^{\circ}C$ at September. The optimum temperature range of photosynthesis was 21$^{\circ}C$ to 25$^{\circ}C$ at June and 14$^{\circ}C$ to 21$^{\circ}C$ at September, and that was higher in the back row than the front row. High temperature significantly stimulated the dark respiration of ginseng leaves and the respiratory quotients(Q$\_$10/) of the ginseng leaves showed a significant seasonal variation.

Effects Microbial Addition and Incubation Temperatures on Odor of Pig Manure as Fertilizer on Grass and Crop Fields (초지 및 농경지에 살포되는 돼지 분뇨의 냄새에 미생물 첨가 및 분뇨 배양온도가 미치는 효과)

  • Odor in pig manure affects the distribution of the manure over grass and crop fields as fertilizer. The objective of this study was to investigate the effect of different types of microbes (Saccharomyces cerevisiae, Bacillus subtilis and Rodobacter capsulata) and incubation temperatures ($20^{\circ}C$ and $35^{\circ}C$) on the levels of odorous compounds in pig manure. Pig manure was incubated with 0.03% microbes (v/v) at temperatures of $20^{\circ}C$ or $35^{\circ}C$. At incubation temperature of $20^{\circ}C$, the addition of Rodobacter capsulata significantly (p<0.05) decreased the levels of indoles and volatile fatty acid (VFA). At incubation temperature of $35^{\circ}C$, the addition of any microbes of the three used in this study did not significantly (p>0.05) affect the levels of odorous compounds. When incubation temperature was increased from $20^{\circ}C$ to $35^{\circ}C$, levels of odorous compounds were significantly (p<0.05) increased. Taken together, these results suggest that Rodobacter capsulata could be utilized to reduce odor from pig manure in the spring and fall when the average temperature is around $20^{\circ}C$. However, alternative odor-reducing technology is needed to be developed to apply onto pig manure during the hot summer season ($35^{\circ}C$).

Optimal Conditions for Treatment of Swine Wastewater using Rhodopseudomonas palustris KK14 (Rhodopseudomonas palustris KK14를 이용한 돈분폐수처리의 최적조건 검토)

  • For the development of biological wastewater treatment process using photosynthetic bacteria (PSB), photosynthetic sludge process consisted of anaerobic digestion and PSB reactor were designed for the treatment of swine wastewater and the optimal operating conditions in flask-scale were examined. Photosynthetic bacteria from soil, pond, rice field, ditch etc. were isolated in synthetic medium containing high amount of organic acids and finally isolated one strain KK14 which showed the most degradating ability of organic acids was selected for the treatment of swine wastewater. It was identified as Rhodopseudomonas palustris. In the anaerobic digestion stage, the maximum organic acid productivity was obtained at pH 5.0, $37^{\circ}C$, HRT 2 day and under anaerobic standing condition. The optimal operating conditions of PSB reactor for the treatment of swine wastewater were pH 7.0, $30^{\circ}C$ under 4,000 lux illumination, and optimal initial COD loading (kg COD/kg D. C. W of PSB) was 2 (20% v/v seeding) in the main purification stage. Maximum removal rate of COD reached 92% under the above optimal conditions for 5 days.

  • 한국 서해안과 남해안 47개소의 해수와 mud 시료로부터 광합성세균으로 유추되는 13개의 균주를 분리하였고, agar-shake tube method와 RAPD-PCR을 이용하여 4개 서로 다른 균주를 순수분리 하였다. 4개의 균주 중 폐수분해능이 가장 뛰어난 균을 선정하여, 형태학적, 배양적, 생화학적 특성 및 16S rRNA sequencing에 의한 동정결과 purple, sulfur bacteria 쪽에 가까웠으나 형태학적, 배양학적, 생화학적 특성이 purple, non-sulfur bacteria의 Rhodobacter 속에 가장 근접하여, Rhodobacter sp. EGH-24로 명명하였다.

  • To investigate effect of soil conditioners on salt reducing effect, saline soil were treated with photomicrobe, soil plus, pyroligneous liquor and active charcoal and cucumber and tomato were grown. Yield and root development were enhanced in active charcoal treatment overall and tomato blossom end rot frequency was lowest in active charcoal treatment. Total sugar content was highest in soil plus treatment and nitrate and phosphate content were not significantly different within 4 treatments.
