Efficient Light Treatment for Graft-take and Early Growth of Grafted Tomato Seedlings (토마토 접목시 활착과 순화에 효과적인 광 처리 방법 구명)

  • This research was conducted to elucidate the appropriate light environment right after grafting to produce vigorous cherry tomato seedlings. Tomato plants were grafted and then treated in 4 different ways: to keep in the natural light (Non), to cover the grafted stem part with aluminum foil to make only that part dark (Part), to put the grafted seedlings in the acclimation room for two (Day-2) or four days (Day-4) to make the whole seedlings in the dark condition. Tube grafting method was used for grafting, in which silicon tube of 1.5mm in diameter was used. The survival rate was the maximum in the treatment Day-2. The SPAD value, seedling quality and yield of $1^{st}$ and $2^{nd}$ cluster were the best in the treatment Part. The treatment Part needs cost more than other treatments but is more economic thanks to higher yield. Therefore it was concluded to be economically feasible to make the grafted stem part dark right after grafting in case of cherry tomato.

Mass-production of Eleutherococcus seoulensis Seedlings Through Somatic Embryogenesis (체세포배 형성을 통한 서울오갈피(Eleutherococcus seoulensis) 묘목의 대량생산)

  • This study was conducted to establish the optimal condition for acclimatization from somatic embryos of Eleutherococcus seoulensis. Torpedo-shaped embryos of Eleutherococcus seoulensis were cultured on 1/3 MS and WPM media supplemented with $GA_3$ (3.0, 5.0 mg/L) for 4 weeks. Plentlets were transferred to 1/2 SH solid medium with 1.0 mg/L $GA_3$ and 0.2% activated charcoal for shoot and root elongation and them elongated plantlets further developed on 1/2 SH medium for 4 weeks. Developed plantlets further elongated into well-shaped leaf and root system on 1/3 SH medium under ventilation condition for 4 weeks. Plantlets grew normally on 1/3 SH basal medium, were acclimated on various soil. Survival frequency of plantlets was influenced by soil type(peatmoss+perlite, perlite, soil on Nam mountain). The highest survival rate to soil was more than 70% when plantlets were 1/3 SH medium under ventilation condition in Nam mountain soil. These results indicate that the systematic procedure of plant production in Eleutherococcus seoulensis could be practically applicable for mass propagation.

In vitro Multiple-propagation of Wolly Grass (Imperata cylindrica 'Rubra') (홍띠(Imperata cylindrica 'Rubra') 기관분화에 의한 기내대량증식)

  • 홍띠(Imperata cylindrica 'Rubra') 식물자원의 기내대량증식과 재분화식물체의 순화체계를 구축하고자 기내 재분화에 적합한 식물재료부위, 생장조절물질을 조사하고, 재분화 유식물체로부터 적정 순화조건을 구명하였다. 기본배지로 MS (Murashige and Skoog, 1962) 배지를 사용하였고, 배양은 $26{\times}2^{\circ}C$, $25{\mu}mol/m^2/s$, 14h/10h (day/night) 광조건 하의 배양실에서 수행하였다. 캘러스 형성은 뿌리 끝, 줄기절편, 생장점 부위 중생장점 부위에서 가장 양호하였고, 이 생장점 조직에 0.1 mg/L의 2,4-D와 2 mg/L의 BA를 처리하였을 때 양호하였다. 캘러스 증식은 0.1 mg/L의 2,4-D와 0.05 mg/L의 BA 배지, 0.05 mg/L의 2,4-D와 0.5 mg/L의 BA를 첨가한 배지 중 0.1 mg/L의 2,4-D와 0.05 mg/L의 BA 배지에서 양호하였고, 이들 캘러스로부터 신초 재분화는 0.01 mg/L의 NAA와 2 mg/L의 BA 처리에서 양호하였다. 초기 치상으로부터 실제 경과시간은 캘러스 유도에 19주간(2018. 03. 18~07. 27), 캘러스 증식 9주간(2018. 07. 27~09. 28), 신초 유도 11주간(2018. 09.28~12. 14), 순화에 10주간(2018. 12. 14~2019. 02. 23)에 걸쳐 진행하였으나 확립된 배양계를 적용하면 캘러스 유도 4주, 캘러스 증식 3주, 신초유도 및 증식 4주, 순화 7주 정도가 소요될 것으로 계측되었다. 순화는 다경줄기 형성후 MS배지를 멸균한 상토(버미큘라이트) 또는 종이포트로 교체하여 재분화식물체를 배양병에서 7주간 배양하고, 7주후에 배양병 뚜껑을 1/10 정도 1차 개방하여 1주일 후 3/10 정도 개방하여 2주간 경과한 후 컵포트(직경 6 cm)에 이식하여 성공적으로 활착시켰다.

Plant Regeneration via Secondary Somatic Embryogenesis and Acclimatization in Panax ginseng (장뇌삼의 2차 체세포배 발생을 통한 식물체 유도 및 순화)

  • This study was conducted to establish the optimal condition for plant regeneration and acclimatization from somatic embryos of Panax ginseng. Cotyledon segments of Panax ginseng produced primary and secondary somatic embryos when cultured on MS and WPM media supplemented with 7% sucrose. To induce plantlet conversion, cotyledonary somatic embryos were cultured on WPM solid medium with $GA_3$ at various concentrations (1~30 mg/L) for 4 weeks. Plantlets were transferred to 1/2 WPM solid medium with $GA_3$ at various concentrations (0~5 mg/L) and 0.5% activated charcoal for shoot and root elongations. Elongated plantlets further developed into well-developed leaf and root system on 1/3 SH medium with 0.5% activated charcoal under ventilation condition for 5 months. The highest survival rate to soil was 75% when plantlets were regenerated on 1/3 SH medium without sucrose under ventilation condition.

  • 참전복은 그동안의 연구에서 그 적수온이 2$0^{\circ}C$에서 23$^{\circ}C$ 사이로 알려져왔다. 그러나, 이 종이 무척추 연체동물로서 개체의 순화 수온과 서식 환경에 따라서 그 적수온이 다소 차이가 있을 것으로 예상된다 실제 전 실험의 결과 참전복 치패가 온도에 대한 높은 내성을 가지는 것으로 미루어 보아, 알려진 적수온보다 다소 높은 온도에서도 성장이 원활할 것으로 기대된다. (중략)

  • Growth of M.9 (Malus domestica Bark. cv) and M.26 (Malus domestica Bark. cv) of dwarf apple rootstock, cultured on MS agar medium in a vessel with ventilating stopper (VS) and then in vivo rooting and acclimatization under combined-treatment by some materials with IBA, were investigated. Concentration of $CO_2$ and ethylene in the vessel with VS was lower then in the vessel with non-VS. Change of temperature and humidity in the vessel with VS was repeated by light condition. Stomatal pares of tissue in the vessel with VS were immediately closed after plantlets were exposed to room humidity but those in the vessel with non-VS were opened after 20 minutes exposure to room humidity. Leaf area and chloroplast index of tissue in the vessel with VS was higher then in the vessel with non-VS. In vivo rooting ratio and acclimatization ratio of M.9 and M.26 was highest in 300mg/L IBA+3% sucrose dip-treatment among other combined- treatments.

The Photoautotrophic Culture System Promotes Photosynthesis and Growth of Somatic Embryo-derived Plantlets of Kalopanax septemlobus (독립영양방식 액체대량배양 시스템하에서 배양한 체세포배 유래 음나무 기내묘의 생장과 광합성)

  • A photoautotrophic micropropagation methodology in liquid culture medium has a number of advantages for large-scale propagation of plants. This paper describes an improved system for the mass propagation via somatic embryogenesis of the medicinal plant Kalopanax septemlobus Nakai. Somatic embryo-derived young plantlets of K. septemlobus were cultured either under heterotrophic conditions with sucrose on half-strength MS medium with $30gL^{-1}$ sucrose, under heterotrophic conditions without sucrose, or under photoautotrophic conditions (MS liquid medium without sucrose under forced aeration) for four weeks before transferring the plantlets for acclimatization. Plantlets grown under photoautotrophic conditions had more leaves, higher chlorophyll content, a higher net photosynthetic rate (NPR), and a higher survival rate. The results indicate that the photoautotrophic conditions with a forced ventilation system are effective in enhancing the growth of plantlets and the rate of net photosynthesis. The plantlets grown under photoautotrophic conditions had a high survival rate (92%) upon ex vitro transplantation. Our study shows that autotrophically produced plantlets acclimatize better and sooner upon ex vitro transplantation than conventionally cultured plants.
