Pathophysiology of Chronic Obstructive Pulmonary Disease (만성폐쇄성폐질환의 병태생리)

  • Chronic obstructive pulmonary disease (COPD) is a chronic progressive disease, characterized by irreversible airflow limitation, with a partially reversible component. The pathological abnormalities of COPD are associated with lung inflammation, imbalances of proteinase and antiproteinase, and oxidative stress, which are induced by noxious particles and gases in susceptible individuals. The physiological changes of COPD are mucus hypersecretion, ciliary dysfunction, airflow limitation, pulmonary hyperinflation, gas exchange abnormalities, pulmonary hypertension, cor pulmonale and systemic effects. The airflow limitation principally results from an increase in the resistance of the small conducting airways and a decrease in pulmonary elastic recoil due to emphysematous lung destruction. This article provides a general overview of the pathophysiology of COPD.

Changes of membrane resistance and membrane potentials by hyperpolarizing current stimulation in mouse unfertilized eggs (과분극 자극에 대한 mouse 미수정란의 세포막저항과 세포막전압의 변화)

  • For the observations of both the membrane properties and the excitability on the unfertilized eggs of female mice, changes of the membrane resistance and the membrane potential by hyerpolarizing current stimulation were recorded. As current-voltage relation was linear over the entire range (-180mV~+60mV), membrane resistance($R_m$) was calculated from the amplitude of electrotonic potential to a given stimulus current. Also the presence of anode-break excitation was confirmed. The results were as follows; 1. There was a linear relation between the membrane resistance and resting membrane potential, the expected input resistance was 61. 4M$\Omega$(resting membrane potential was $-18.9{\pm}8.7mV$, mean${\pm}$SD, n=30). 2. Transient depolarization with overshoot was generated just after hyperpolarizing current stimulus and showed the dependency of stimulus duration. 3. Transient depolarization lasted over 30ms, amplitude of these depolarization was increased by high $Ca^{{+}{+}}$(20mM) and inhibited by $Ca^{{+}{+}}$-antagonist, $Mn^{{+}{+}}$. 4. From the above results, it was suggested that the unfertilized mouse egg showed the characteristics of the excitable cell.

Fitting Distribution of Accident Frequency of Freeway Horizontal Curve Sections & Development of Negative Binomial Regression Models (고속도로 평면선형상 사고빈도분포 추정을 통한 음이항회귀모형 개발 (기하구조요인을 중심으로))

  • 교통사고예측 및 예방을 위해서는 실제적으로 도로설계과정에서 제어가 가능한 도로 기하구조요소에 대한 사고관계를 파악함이 타당하다. 즉, 도로의 설계자는 도로건설에 앞서 기하구조요소와 사고와의 관계를 현장자료를 통해 정확히 밝혀 도로설계에 반영해야 한다. 이를 위해, 교통사고의 빈도분포를 박히는 것은 가장 기본이 되는 일이며, 교통사고 예측모형개발에 선행되어야 한다. 일반적으로 교통사고건수의 경우 분산이 평균보다 큰 과분산(overdispersion)의 특징을 가지고 있어 음이항 분포를 따른다고 알려져 있다. 따라서 본 논문은 사고모형의 개발에 앞서, 사고발생지점에 대한 도로설계요소와 기타 잠재적인 사고발생 관련요인이 비교적 잘 파악되어있는 호남고속도로를 중심으로 평면 선형상 곡선부에 대하여 교통사고의 분포를 적합도 검정을 통해 알아보고자 하였다. 사고자료는 한국도로송사의 호남고속도로 5년(1996∼2000)간 자료를 분석에 맞게 정리하였으며, 강민욱과 송봉수(2002)에서 제시한 평면선형에 있어서의 구간분할법을 이용하여 배향곡선구간과 단일곡선구간에 대한 사고분석을 하였다. 적합도 분석결과, 예상대로 음이항분포가 사고건수를 설명하기에 가장 적합한 확률분포로 제시되었으며, 이를 통해 최우추정법을 이용한 음이항회귀모형을 개발하였다. 구간분할법을 적용한 음이항회귀모형의 경우, 기존의 확률회귀토형에 비하여 높은 결정계수를 갖았으며, 모형에서 적용된 기하구조요소로는 차량 노출계수, 곡선반경, 단위거리 당 편경사변화값 등이다.

Two-step Acid Hydrolysis Method for Producing Fermentable Sugar from Lignocellulosic Biomass (발효당 생산을 위한 목질계 바이오매스의 2단 산당화)

  • For obtain fermentable sugar, we conducted acid hydrolysis with lignocellulosic biomass without enzyme. The lignocellulosic biomass used pinus rigida and Palm residues (EFB; empty fruit bunches). In the acid hydrolysis, we consider the hydrolysis condition to reduce a denatured sugar. So this study was conducted 2-step acid hydrolysis. First-step hydrolysis used high concentration (72 wt%) sulfuric acid at $80^{\circ}C$. At the condition, we obtained 11.49 wt%, 32 wt% glucose conversion for pinus rigida and EFB. After the step, the liquor was dilute until 9~15 wt% acid concentration and conducted second hydrolysis at $50{\sim}120^{\circ}C$. In the second hydrolysis, we obtained maximum glucose conversion (pinus rigida 86.8 wt% (39 g/L) and EFB 95.3 wt% (32.4 g/L)) at 9 wt% acid concentration and $120^{\circ}C$ for 80 min. All samples through the process are analyzed on the basis of mass balance.

Effects of Cyclic-GMP on Hyperpolarization-activated inward Current $(I_f)$ in Sino-atrial Node Cells of Rabbit (동방결절에서 과분극에 의해 활성화되는 내향전류에 대한 Cyclic-GMP의 영향)

  • The aim of present study is to investigate the effects of cGMP on hyperpolarization activated inward current ($I_f$), pacemaker current of the heart, in rabbit sino-atrial node cells using the whole-cell patch clamp technique. When sodium nitroprusside (SNP, $80{\mu}M$), which is known to activate guanylyl cyclase, was added, $I_f$ amplitude was increased and its activation was accelerated. However, when $I_f$ was prestimulated by isopreterenol (ISO, $1{\mu}M$), SNP reversed the effect of ISO. In the absence of ISO, SNP shifted activation curve rightward. On the contrary in the presence of ISO, SNP shifted activation curve in opposite direction. $8Br-cGMP(100\;{\mu}M)$, more potent PKG activator and worse PDE activator than cGMP, also increased basal $I_f$ but did not reverse stimulatory effect of ISO. It was probable that PKG activation seemed to be involved in SNP-induced basal $I_f$ increase. The fact that SNP inhibited ISO-stimulated $I_f$ suggested cGMP antagonize cAMP action via the activation of PDE. This possibility was supported by experiment using 3-isobutyl-1-methylxanthine (IBMX), non-specific PDE inhibitor. SNP did not affect $I_f$ when $I_f$ was stimulated by $20{\mu}M$ IBMX. Therefore, cGMP reversed the stimulatory effect of cAMP via cAMP breakdown by activating cGMP-stimulated PDE. These results suggest that PKG and PDE are involved in the modulation of $I_f$ by cGMP: PKG may facilitate $I_f$ and cGMP-stimulated PDE can counteract the stimulatory action of cAMP.

Breeding of a Seedless Table Grape Cultivar 'Heukisul' (Vitis sp.) with High Quality (고품질 무핵 포도 품종 '흑이슬' 육성)

  • 'Heukisul' (3x) a cross made in 2000 between 'Kyoho' (4x, Vitis sp.) and 'Thompson Seedless' (2x, Vitis vinifera L.) in a seedless grape breeding program, was preliminarily selected in 2004. After regional adaptation testing under the name of 'KTS014' at two sites during 2004-2005, it was finally selected in 2006. 'Heukisul' showed low incidence of berry shattering, resulting in a good berry set compared to 'King Dela'. 'Heukisul' had budburst on April 23, flowering on in June 9, and fruit maturation in September 29 (almost the same as 'King Dela' at Chuncheon), and it was considered a midseason cultivar. The mean berry weight was 4.0 g, about 0.7 g heavier than 'King Dela', and mean soluble solids were $21.9\;^{\circ}Brix$, about $2.7\;^{\circ}Brix$ higher than 'King Dela'. The skin color was dark violet with abundant bloom and the flesh was very firm. Although the cluster was compact, it required no cluster thinning. Also the incidence of berry cracking was very low.

$K^{+}$ 통로 개방제의 혈관근 이완작용에 대한 연구 : sarcoplasmic Reticulum에서의 $Ca^{++}$ 유리에 대한 효과

  • 1.목적 $K^{+}$ 통로 개방제인 cromakalim과 levcromakalim이 원형질막의 $K^{+}$ 통로를 개방시켜 막의 과분극을 일으킴으로서 강력한 혈관 이완작용을 일으킨다는 것은 주지하는 바다. 본 연구진들은 지난 2년간의 신약개발 연구사업을 통하여 cromakalim과 pinacidil 등 여러종의 K+통로개방제가 건전한 평활근 세포에서 phenylephrine뿐만 아니라 saponin처리에 의한 투과성 근세포 (permeabilized cells)에서 inositol 1,4,5-triphosphate (IP$_3$)에 의한 수축도 억제함을 보고 하였다. 본 연구에서는 이러한 수축작용에 대한 SPEX fluolog-2 spectrophotometer를 사용하여 돼지의 관상동맥 혈관근의 세포질내 $Ca^{++}$의 농도 ([$Ca^{++}$])의 변동을 관찰하였다. 정상 관상동맥 혈관근 조직에서 acetylcholine (0.1-1$\mu$M)에 의한 [Ca$^{++}$]농도의 증가와 b-escin 처리에 의한 skinned strip에서의 IP3 (1-5$\mu$M)에 의한 [Ca$^{2+}$]의 증가는 cromakalim과 levcromakalim의 전처치에 의하여 현저히 억제되었다. Skinned strip에서 이러한 K+ 통로 개방제에 의한 $IP_3$-요도 ($Ca^{2+}$)i 증가의 억제가 apamin (1-5 mM)과 glibenclamide (1$\mu$M)에 의하여 봉쇄되었으나, chrybdotoxin (0.1$\mu$M)에 의하여는 영향을 받지 아니하였다 한편 skinned strip에서 cromakalim은 GTP${\gamma}$s에 의한 [$Ca^{2+}$]i의 증가는 봉쇄하였으나 caffeine에 의한 [$Ca^{2+}$]i의 증가는 영향을 미치지 아니하였다. 이러한 연구결과로 보아 cromakalim과 levcromakalim과 같은 $K^{+}$ 통로 개방제가 세포막의 $K^{+}$ 통로를 개방하는 작용 뿐만 아니라 적어도 sarcoplasmic membrane에서 $Ca^{2+}$의 유리를 봉쇄함으로써 강력한 혈관 평활근 이완 작용을 나타내는 것으로 시사된다.

Load balancing method of overload prediction for guaranteeing the data completeness in data stream (데이터 스트림 환경에서 데이터 완전도 보장을 위한 과부하 예측 부하 분산 기법)

  • A DSMS(Data Stream Management System) in ubiquitous environment processes huge data that input from a number of sensor. The existed system is used with a load shedding method that is eliminated with a part of huge data stream when it doesn't process the huge data stream. The Load shedding method has to filter a part of input data. This is because, data completeness or reliability is decreased. In this paper, we proposed the overload prediction load balancing to maintain data completeness when the system has an overload. The proposed method predicts the overload time. and than it is decreased with data loss when achieves the prediction overload time. The performance evaluation shows that the proposed method performs better than the existed method.

Production of Biosugar from Red Macro-algae Eucheuma cottonii using Acid-hydrolysis (Eucheuma cottonii로부터 산 가수분해를 통한 biosugar 생산)

  • In this study, biosugar production by the red-algae Eucheuma cottonii was investigated using dilute sulfuric acid-catalyzed hydrolysis and data analysis by response surface methodology. This approach yielded 25.8 g/l total reducing sugar under the conditions of $160.1^{\circ}C$, 1% (v/v) sulfuric acid, and 13.1 min. The sugar concentration showed a linear inverse correlation with the combined severity factor (CSF) of the reaction conditions. CSF was calculated as $log(t{\cdot}e{xp}[(T_H-T_R)/14.75])-pH$, where t is the coupling reaction time, $T_H$ is the target temperature, and $T_R$ is the reference temperature ($100^{\circ}C$). In addition, levulinic acid production showed a linear positive correlation with CSF. E. cottonii may represent a useful feedstock for sugar production in the field of bioenergy.

  • The Port Logistics Information System (Port-MIS) is the system that processes all of port management such as entry and departure of ships, using facilities within ports, port traffic control, cargo entering and carrying and tax collection in 31 trade ports over the whole country. Lately, The Port Logistics Information System (Port-MIS) has been reformed as a WEB-based system and established basis to provide real time information support structure, diversification of civil complaint system, and optimized civil complaint service by using wire and wireless internet. The typical study on the Port Logistics Information System was C/S program and EDI-centered. But it has significance to study on service quality measurement of WEB Port-MIS that has been served as a new web-based platform since April 2010, for there was no such a study until now.
