Formation, cellular distribution and structural changes of storage lipid, and active site and cellular localization of lipase in endosperms and cotyledons of lipid-rich seeds such as Helianthus annuus, Ricinus communis and Pinus koraiensis, and in those of starch-rich seeds such as Pisum sativum and Zea mays were investigated in relation to the seed development by cytochemical methods. In endosperms and storage cotyledons of lipid- and starch-rich seeds after seed-gathering, there were widely distributed storage material which was composed of spherical protein bodies, spherosomes, and starch granules. But cellular organelles were hardly observed in the cytoplasm. Staining pattern of vesicles released from SER, and of low electron dense membraneous granules, which were perhaps at an early stage of spherosomes, were the same as in the spherosome. Electrondense granules released from RER were observed in the vicinity of plasma membrane. As a result of lipid staining, the spherosomes were more electron dense and were uniform as compared with the protein matrix within the protein body and cytoplasmic proteinaceous granules. The major component of the spherosome was determinated to be lipid. Spherosomes and vesicles containing SER-released materials showed the same as in the electron density. Lipase activity was especially strong in the inner region and on the surface of decomposed spherosomes and near the plasma membrane.mbrane.

Ultrstructural studies of germ cell differentiation and vitellogenesis in the oocytes of the female Rapana venosa in the brackish water area of Seomjin River were investigated by transmission electron microscope observations. In the early vitellogenic oocytes, the Golgi complex and mitochondria were involved in the formation of glycogen particle, lipid droplets, and yolk granules. In the late vitellogenic oocytes, the rough endoplasmic reticulum and multivesicular bodies were involved in the formation of proteid yolk granules in the cytoplasm. However, heterosynthetic vitellogenesis in this species were not observed in vitellogenic oocytes during oogenesis. A mature yolk granule was composed of three components: crystalline core, electron lucent cortex and the limiting membrane. As shown in some large gastropods, vitellogenesis in R. venosa occurred by way of endogenous autosynthesis without heterosythetic vitellogenesis (exogeneous endocytosis), which are found in the oocytes in bivalves. The mating period and spawning activity were related with the increases of seawater temperatures and salinities.

Tungsten alloys are used widely in general industrial fields, but they are difficult to cast, so products are manufactured using powder metallurgy. In this study, a mixed powder of tungsten, nickel, and iron homogenized using a ball mill was added to pure water as a solvent, and PVA as a binder was added to prepare a spray drying mixture. The mixed liquid was prepared using a spraying machine. A study was carried out to produce a granular powder that can reduce the variations between products during the molding and sintering process of the powder metallurgy method. A preliminary experiment was conducted to confirm the influence of the variables in the granulation process. Through the preliminary test results, this experiment was performed with the volume of solvent of the spray drying the mixture as an independent variable, and granular powder having a mean particle size similar to that of the existing mass-production conditions and an increased apparent density was prepared. In addition, a pilot test was conducted for the molding and sintering process. The improved granular powder reduced the characteristic variation (weight variation) of the mass-produced product.
Granular gland regeneration in the toad after dorsal skin wound histologically was examined using scanning and transmission electron microscopy. After cutaneous wounds were induced by excision, animals were maintained in special cages for up to 20 days. In transmission electron microscopy (TEM), newly formed granular gland, though poorly developed, was seen on 4 day after injury. Epithelial cells moved toward apical region of newly formed gland. The cells had smooth surface and were not connected to other cells by desmosomes. Mitochondria rich cell (MRC) possessing long cytoplasmic processes formed a gland cavity and hemidesmosomes were found under the cell processes. Basal cavity of newly formed gland consisted of MRC, pro-granular producing cells (pGPC), and granular producing cell (GPC). Moreover it was observed that xanthophores moved to the base of the epithelial tissue on 10 day after the injury. These cells contained numerous pterinosomes and carotenoid vesicles. Immature pterinosomes were large and carotenoid vesicles were moderately electron dense. On 13 day after the injury, xanthophores contained abundant carotinoid vesicles and lammelated pterinosomes. Iridophores were also observed adjacent the developing xanthophores on 16 day post-injury. These observations indicated that regeneration of granular gland from glandular precursor cells during wound healing and subsequent expansion of the glandular cells might be dependent on maturation and proliferation of these newly formed cells.
To improve the type error of osmotic tablet which is one of the drug delivery system, osmotic granule could be manufactured by fluidized bed coating. It has drug layer containing different amount of osmogant and is coated with membrane including different types of binder. We confirmed that the morphology of osmotic granule was different at each coating step. The more mont of osmotic agent, the faster drug release was observed due to increasing the driving force for drug release from osmotic granule. And drug release from osmotic granule coated with membrane using different types of binder was differed by solubility of binders to water. The formation of pore in membrane was confirmed by SEM and DSC Membrane using water soluble binder released more amount of drug. From these results, we assured that difference of osmotic pressure between the inside and the outside of granule and porosity of membrane have an effect on drug release from osmotic granule.
The aim of this study was to identify the chronological growth characteristics of Gaeryangmeoru grapes, by observation of the developmental process of the axillary bud, fruit quality, and anatomical structure of the pericarp. No necrosis of axillary buds was observed, irrespective of node position of the shoot, from shoot occurrence to leaf fall. Berry weight, cluster weight, soluble solid content and titratable acidity were analyzed to be 1.2 g, 128.8 g, $16.9^{\circ}Brix$, and 1.57%, respectively, at maturity. The sugar was composed of fructose and glucose, with a ratio of 1:1 (fructose: glucose) at maturity. Malic acid was predominant among the organic acids, and conformed to the changes of chronological titratable acidity. Sugar and organic acid content changed dramatically starting a week before veraison. The degraded ovules were observed before and after fertilization, and the pericarp was composed of one epidermis, and seven to ten hypodermis layers at maturity.

The pars distalis of the Korean frogs (Rana dybowskii Guenther) during hibernating and active periods was observed with the electron microscope. Seven cell types were classified according to the size and shape of secretory granules and to the ultrastructural characteristics. There were many differences between hibernating and active frogs in type 5 cells. Therefore the following results were observed. Cell type 1; This type cell contains spherical secretory granules, $375{\sim}687m{\mu}$ in diameter. Cell type 2; This type cell contains various secretory granules, $250{\sim}437m{\mu}$ in diameter Cell type 3; Spherical and rod-shaped granules, $l25{\sim}187m{\mu}$ in diameter were observed. Cell type 4; In this type cell, the electron density is the lowest and the density of granules is the highest of all type cells. This type cell contains various secretory granules and large secretory granules, $2l0{\sim}420m{\mu}$ in diameter, were also observed. Cell type 5; The electron density of this cells is similar to that of type 4 cells. The density of granules is lower than that of type 4 cells. And the shapes of the secretory granules are similar to those of type 4 cells. But many rod shaped granules, $200{\sim}863m{\mu}$ in diameter, were also observed. Cell type 6; This type was similar to type 2. The electron density of cytoplasm is very low. Spherical granules, $232{\sim}316m{\mu}$ in diameter, were observed. Cell type 7; This type of cell has no secretory granules. This cell is not developed very well. The type 5 cells in hibernating frogs are different from cells in active frogs. In type 5 cells, many secretory granules were observed during active period. But the number of secretory granules were greatly declined and there were many vacuoles in cytoplasm during hibernating period.
Five kinds of neurosecretory cells (type-A, B, C, D and E) and neuropiles surrounding them were observed in the visceral ganglion and the right parietal ganglion of the African giant snail, Achatina fulica, by transmission electron microscopy. Type-A cells (diameter, $35{\mu}m$) are the most popular cells in the cortex of the two ganglions, which are of triangular or irregular forms. In their cytoplasm, there are found large granules of 1 fm in diameters and small round granules of about $0.1{\mu}m$ in diameters. Small granules are classified into the ones of high electron density and the others of middle electron density. Type-B cells (diameter, $19\times12{\mu}m$) are evenly distributed over various portions of cortex and medulla of the two ganglions. They are similar to type-A cells in shapes. The cytoplasm of type-B cells is crowded with high electron dense granules of about $0.1{\mu}m$. Round granules of about $0.7{\mu}m$ in diameters are also found but rarely. Type-C cells are the smallest cells whose sizes are about $8\times6{\mu}m$. Each of them contains a large nucleus of about $6\times5{\mu}m$. Its cytoplasm is full of electron dense granules of about $0.23{\mu}m$, each of which is artually an assembly of tiny granules of about $0.03{\mu}m$. Type-D cells are middle-size cells of about $28\times20{\mu}m$, which take ellipsoidal or irregular forms. They are found in the cortex more than in the medulla. Their cytoplasm looks dark due to the high electron density and, in it, two kinds of round granules whose sizes are $1.6{\mu}m$fm and $0.6{\mu}m$, respectively, are observed. Type-E cells are large cells of about $100\times50{\mu}m$, which are rarely found in the upper and middle portions of the two ganglions. The nucleus of the cell, which is very large $(70\times30{\mu}m)$ for the cytoplasm, contains electron dense round granules of diverse sizes (diameters, $1\sim0.2{\mu}m$). The surface of the cell protrudes filopodia of various forms and phagocytizes decrepit cells. Neuropiles are surrounding the neurosecretory cells. In nerve fibers, synaptic vesicles are observed, which are classified into six classes according to their electron densities , sizes and shapes.

This research focused on determining the effectiveness of cluster and/or flower thinning in highbush 'Jersey' blueberry on the yield and fruit quality. The total yield and quality of fruit were compared between thinning methods of control (no thinning), 1/3 cluster thinning, 1/3 cluster thinning + 1/2 flower thinning, 2/3 cluster thinning, and 2/3 cluster thinning + 1/2 flower thinning per each bearing shoot, respectively. Thinning strength significantly affected the yield of bearing shoot and, thus, total yield of control and 1/3 cluster thinning was higher than others. Both cluster and flower thinning, however, significantly affected the fruit growth resulting in the increase of fruit weight, length and width. Even total yield was similar between control and 1/3 cluster thinning, more larger fruit (> 1.6 g) were produced by 1/3 cluster thinning treatment, indicating that flower and/or cluster thinning contributed to the increase of individual fruit growth. Unlike fruit growth, fruit quality was less affected except total soluble solid contents. Total soluble solid level significantly increased in treatments (2/3 cluster thinning, 2/3 cluster thinning + 1/2 flower thinning) with yield decrease. No significant difference in levels of acidity and firmness of fruit was found. Results indicated that fruit growth and total yield of highbush blueberry was more influenced by the thinning strength regardless of cluster or flower than internal quality of fruit such as solid and acid levels.
In mammalian ovary, major portion(>99%) of ovarian follicles undergo atresia. Recent studies have shown that this phenomenon is mediated via GC apoptosis. Ceramide, a product of sphingomyelin hydrolysis, has been proposed as a novel lipid second messenger with specific roles in mediating antiproliferative responses including apoptosis and cell cycle arrest. In the present study, we have examined the effect of ceramide on apoptotic cell death of GC in vitro. GCs were harvested by squeezing the antral follicles from the immature mice (3-4 weeks) and cultured in MEM medium with 10% fetal bovine serum. The cells were treated with various concentrations of ceramide (0 to 50 \mu M)and cultured up to 24 h.Cell death was determined by MTT cell viability assay and apoptosis was examined by acridine orange staining, in situ 3'-end labeling(TUNEL), and flow cytometry. Ceramid treatment induced apoptotic cell death of GC in a time- and a dose-dependent manner. Results of flow cytometric analysis showed that creamide-induced cell death was mostly confined to the $G_{0}$/$G_{1}$ cells. these results provide an evidence for ceramide as a lipid second messenger of apoptosis in mouse GC.
