Implications of Streptomyces coelicolor RraAS1 as an activator of ribonuclease activity of Escherichia coli RNase E (Streptomyces coelicolor RraAS1의 Eschechia coli RNase E의 RNA 분해작용에 대한 활성제로서 기능 암시)

  • RNase E (Rne) is an essential enzyme involved in the processing and degradation of a large portion of RNAs in Escherichia coli. The enzymatic activity of RNase E is controlled by regulators of ribonuclease activity, namely, RraA and RraB. Gram-positive bacterium Streptomyces coelicolor also contains homologs of Rne and RraA, designated as RNase ES (Rns), RraAS1, and RraAS2. In the present study, we investigated the effect of S. coelicolor RraAS1 on the ribonucleolytic activity of RNase E in E. coli. Coexpression of RraAS1 with Rne resulted in the decreased levels of rpsO, ftsZ, and rnhB mRNAs, which are RNase E substrates, and augmented the toxic effect of Rne overexpression on cell growth. These in vivo effects appeared to be induced by the binding of RraAS1 to Rne, as indicated by the results of co-immunoprecipitation analysis. These results suggested that RraAS1 induces ribonucleolytic activity of RNase E in E. coli.

Studies on Polymorphism of Transferrin of Serum Proteins in Tilapia (Oreochromis niloticus) (Tilapia(Oreochromis niloticus)의 혈청과 단백질의 transferrin의 다형현상에 관한 연구)

  • This study was taken to isolate transferrin fractions from the sera of tilapia(Oreochromis niloticus) by physico-chemical analyses such as the rivanol precipitation, iron-staining method, SDS-polyacrylamide gel electrophoresis and $^{59}FeCl_3$ autoradiography, and to calculate gene frequencies by using Hardy-Weinberg Law. The results obtained in this experiment were summarized as follow : 1. The transferrin fraction is composed of several components possessing relative lower electrophoretic mobilities and higher molecular sizes than the albumin components. 2. When different staining method was compared with transferrin in band, it was not found difference. 3. It was concluded that the optimun ratio of rivanol to serum was 2 : 1 and this ratio was used in all further fractionation. 4. The molecular weight of transferrin component was about 70,000 $\pm$ 2,000. 5. Tilapia transferrin fractionations were found to be polymorphic. 6. There transferrin variants(A, B and C) have been found in tilapia(Oreochromis niloticus) and Tf types were assumed to be controlled by three codominant alleles Tf A, Tf B and Tf C. Six different phenotypes can be theoretically expected Tf AA, Tf AB, Tf AC, Tf BB, Tf BC, and Tf CC. Only five types of these were observed and Tf CC types(homozygotes) was not found. 7. The frequencies of the three allele Tf A, Tf B and Tf C were 0.795, 0.15 and 0.055 respectively.

HIDE, a Testis Specific Deubiquitinating Enzyme, Interacts with HSP90 (고환 특이적으로 발현되는 탈유비퀴틴효소 HIDE와 HSP90의 상호작용)

  • 연구목적: 본 연구는 아직 그 기능이 파악되지 않은 탈유비퀴틴효소 중 하나인 HIDE에 대한 기본적인 생화학적 특징과 고환에서의 발현 양상을 파악하고 있다. 연구재료 및 방법: 인간의 HIDE 유전자를 클로닝하여 효소활성이 있는지 세포 외 실험을 통해 확인하였고, 아미노산 서열을 분석하여 진화상 보존된 부분을 찾아 그 기능을 파악한 다음 HSP90과의 상호작용을 공동면역침전반응으로 확인하였다. HIDE의 조직별 발현양상을 파악하기 위해서 인간과 쥐의 RNA 블롯과 쥐의 단백질 블롯을 이용하여 각각 노던 블롯팅과 웨스턴 블롯팅을 수행하여 고환에서 많이 발현된다는 것을 알았고 이 사실을 바탕으로 쥐의 고환을 절개하여 면역조직화학반응으로써 고환 내의 HIDE 단백질의 발현양상을 파악하였다. 결 과: HIDE는 세포 외에서 유비퀴틴 잔기를 제거하는 탈유비퀴틴 활성이 있으나 세포 내에서 전체적인 유비퀴틴 복합체를 줄여주는 효과는 없었다. HIDE는 HSP90이라는 분자 샤페론과 상호작용한다. HIDE의 전사체는 고환에서 가장 많이 발현되며 다른 조직에서도 소량 발현된다. HIDE의 단백질은 웨스턴 블롯상에서 고환에서만 확인되었다. 고환 내에서의 HIDE의 발현양상은 왕성한 감수분열을 하는 정모세포에서 높았으며 지지세포나 정조세포에는 발현되지 않았다. 결 론: HIDE는 분자 샤페론 HSP90과 상호작용하며 고환 내의 감수분열 중인 세포에서 많이 발현되는 것으로 보아 감수분열이나 정자형성에 관여하는 것으로 보인다.

  • Use of disposer at the kitchens of Korean apartments is inevitable in treating their foodwaste having a water content of more than 80%. Also we have to ensure that this extra disposer-foodwaste BOD loadings be treated properly by installing/operating a pre-treatment system before this wastewater enters public sewer system. However, the degree of BOD removal should not be excessive since a BOD/N ratio higher than 5 is required for removing N/P at a municipal wastewater treatment plant. The removal of BOD/N in the pretreatment system rather than BOD alone can be an alternative solution in solving this problem. The particles separated by sedimentation, screen or packed-bed can be anaerobically digested at apartment sites to generate biogas that can be used for simple digester heating and to generate volatile fatty acids (VFAs) for nitrogen removal. We suggest that Korean government grants a temporary license (say for 5 years) to foodwaste treatment companies in collaboration with apartment construction companies which may do business and develop various kinds of disposer-foodwaste treatment systems in diverse wastewater discharge systems of Korean apartments.

Comparison of Blood Test Results and Symptoms of Patients with COVID-19 Monoinfection and with COVID-19 and Influenza Virus Co-Infection (COVID-19 단일 감염 환자와 COVID-19 및 인플루엔자 바이러스 동시 감염 환자의 혈액 검사 결과 및 증상 비교)

  • In December 2019, the coronavirus disease 2019 (COVID-19) caused by the virus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) emerged in China and spread rapidly around the world, infecting millions of people. Cases of COVID-19 infection were observed to lead to viral pneumonia. Thirty-five patients admitted to the Gyeonggi Medical Center, South Korea, between November 2020 to January 2021, were found to have been infected with the influenza virus A and B, which cause symptoms similar to COVID-19. The records of these patients and those of COVID-19 patients who visited the hospital for medical examination were compared. The study patients included thirty patients with COVID-19 and/or influenza, five of those with influenza alone. A group of 121 patients without infection was used as control. Patients with COVID-19 and influenza had significantly higher lactate dehydrogenase levels than the patients with COVID-19 alone. The erythrocyte sedimentation rate (ESR) was higher in patients with COVID-19 alone than in other groups. Significant clinical outliers were observed in the COVID-19 and influenza infection group compared with the COVID-19 alone group. These results are expected to play an important role in the analysis of the hematological data of infected patients and the comparison of simultaneous and single infection data to determine clinical symptoms and other signs. These results may also assist in the development of vaccines and treatments for COVID-19.

Deterioration Diagnosis and Source Area of Rock Properties at the West Stone Pagoda, Gameunsaji Temple Site, Korea (감은사지 서탑의 풍화훼손도 진단 및 석재의 산지추정)

  • The rock properties of the West pagoda in the Gameunsaji temple site are composed mainly of dark grey porphyritic granodiorite with medium grained equigranular texture and developed with small numerous dioritic xenoliths. These xenoliths occurred with small holes due to different weathering processes. As a weathering results, the rock properties of this pagoda occur wholly softened to physical hardness because of a complex result of petrological, meteorological and biological causes. Southeastern part of the pagoda deteriorated seriously that the surface of rock blocks showed partially exfoliations, fractures, open cavities in course of granular decomposition of minerals, sea water spray and crystallization of salt from the eastern coast. The Joint between blocks has small or large fracture cross each other, contaminated and corrupted for inserting with concrete, cement mortar, rock fragments and iron plates, and partially accelerated coloration and fractures. There are serious contamination materials of algae, fungus, lichen and bryophytes on the margin and the surface on the roof stone of the pagoda, so it'll require conservation treatment biochemically for releasing vegetation inhabiting on the surface and the discontinuous plane of the blocks because of adding the weathering activity of stones and growing weeds naturally by soil processing on the fissure zone. Consisting rock for the conservation and restoration of the pagoda would be careful choice of new rock properties and epoxy to reinforce for the deterioration surfaces. For the attenuation of secondary contamination and surface humidity, the possible conservation treatments are needed.

CUEDC2, CUE Domain Containing Protein 2, Associates with Kinesin-1 by Binding to the C-Terminus of KIF5A (CUE 도메인 포함 단백질인 CUEDC2는 KIF5A의 C-말단과 결합을 통하여 Kinesin-1와 결합)

  • Kinesin-1 is a motor protein identified as the first member of the kinesin superfamily (KIF), which plays a role in intracellular cargo transport by acting as microtubule-dependent motor proteins within cells. Kinesin-1 consists of two heavy chains (KHCs, also known as KIF5s) and two light chains (KLCs). The 93 amino acids in the carboxyl (C)-terminal tail region of KIF5A are not homologous to the C-terminal tail region of KIF5B or the C-terminal tail region of KIF5C. In this study, we used a yeast two-hybrid screen to identify the binding proteins that interacted with the C-terminal region of KIF5A. We found an association between KIF5A and CUE domain containing 2 (CUEDC2), which is proposed to function as an adaptor protein involved in ubiquitination pathways and protein trafficking. CUEDC2 bound to the C-terminal region of KIF5A and did not interact with KIF5B (the motor of kinesin-1), KIF3A (the motor of kinesin-2), or kinesin light chain 1 (KLC1). KIF5A specifically bound to the C-terminal region of CUEDC2. Furthermore, KIF5A did not interact with another isoform: CUEDC1. In addition, glutathione S-transferase (GST) pull-downs showed that KIF5A directly bound GST-CUEDC2 but did not interact with GST-CUEDC1 and GST alone. When myc-KIF5A and EGFP-CUEDC2 were co-expressed in HEK-293T cells, CUEDC2 co-immunoprecipitated with kinesin-1, and myc-KIF5A and FLAG-CUEDC2 colocalized in the cells. These results suggest that in intracellular cargo transport by kinesin-1, CUEDC2 serves as an adaptor protein connecting kinesin-1 and cargo by binding to KIF5A.

Cyanobacterial Development and Succession and Affecting Factors in a Eutrophic Reservoir (부영양 저수지에서 남조류의 발달과 천이 및 영향 요인)

  • This study was conducted to evaluate the causes and effects of cyanobacterial development and succession in a shallow eutrophic reservoir from March 2003 to February 2004. Phytoplankton succession, sedimentation rate, and sediment composition were analyzed. Algal bioassay also was conducted with the consideration of light, water temperature and nutrients. Cyanobacteria dominated throughout the year, except for spring season (March${\sim}$April) in which diatoms and flagellates dominated. Total cell density increased in July and November when P loading through inflows was high. Oscillatoria spp. and Aphanizomenon sp. were dominant in May and June, respectively, but replaced with Microcystis spp. in July. Thereafter, Microcystis spp. sustained until December, and again shifted to Oscillatoria spp. and Aphanizomenon sp. The dominance of Oscillatoria spp. in May was accompanied with high TN/TP ratio and the increase of water temperature and light intensity. While the dominance of Microcystis spp. was related with relatively low TN/TP ratio, ranging from 46 to 13 (average: 27). The sedimentation rate was highest in March (0.6 m $day^{-1}$) when diatoms dominated. During the period of cyanobacterial dominance, relatively high sedimentation rate was observed in May (0.4 m $day^{-1}$) and October (0.36m $day^{-1}$). C/N ratio of the sediment ranged $6{\sim}8$. Inorganic P concentration in the pore water was low when DO concentration was < 2 mg $O_2$ $L^{-1}$ in the hypolimnion, reflecting the P release from the sediment. Cyanobacterial growth rate depended on phosphorus concentration and water temperature, and high P concentration compensated for the low temperature in the growth rate. Our results suggest that the potential of cyanobacterial development and substantiality in eutrophic reservoirs be high throughout the year, as being supplied with enough P, and emphasize the consideration of sediment man. agement for the water quality improvement and algal bloom control.

A Study on Efficiency of Water Purification of Korean Village Bangjuk[dike] as a Means of Ecological Watershed Management (생태적 유역관리 도구로써 마을방죽의 수질정화 효율성 고찰)

  • This study centering on 10 village - Bangjuks analyzed multifunctionality value of village Bangjuks which have been main water treatment system in Korean traditional villages. On the basis of understanding the structure and character of components such as the well, common spring, village waterway and others which making water-flow and consisting of aquatic system in Korean traditional village Bangjuk, the conclusion as the instrumental device of social and ecological role and ecological watershed management, securing the ecosystem soundness of the damaged or deteriated aquatic ecosystem due to the industrialization and urbanization is as below; 1. The traditional village Bangjuk was environmentally friendly hydraulic system which gathers waterways of village into a point including sewage water, retains and flows out to village through agricultural waterway. Through this Bangjuk, they have managed sewage and rainfall runoff flowed out village efficiently. It is not only a detention system of water but a kind of eco-friendly system that flow out water into the rivers after reusing and filtering it. 2. Around five traditional villages and five villages after modernization, this study classified the types of village Bangjuk as three types considering geographic location, size, etc; marsh type of low swamp, high water -low rice field type of natural flow stucture, low water - high rice field type requiring artificial irrigation facility. All the five traditional villages were turned out to be marsh type of low swamp. Geoji, Sanjeri, Ma-am, Yangchon of the agricultural villages were high water-low rice filed type, and Sangchoenri village was classified low water-high rice field type. 3. This study checked up the function of water purification of village Bangjuk. In Wonteo and Geji villages affected by discharge of village sewer and domestic sewage, the efficiency of ammonia nitrogen($NH_3-N$) and total phosphorus(T-P) was 56~95%, which was high. In Sangcheonri and Sanjeri villages strongly affected by stall and farmland, the efficiency of suspended solids(SS) was 70~85%, and that of total nitrogen(T-N) and total phosphorus(T-P) was 5.3~65%. 4. A water purification system can be found out in the system of village Bangjuk that filter out village sewage and rainfall runoff flowed through the settle and filter of pollution source and denitrification of plants. Through this system of village Bangjuk, it must be used as the basic facilities for the ecological watershed management. The sewage management system of village Bangjuk as a eco-filter must be used and studied as an eco-friendly facility for the ecological watershed management around the subwatershed and catchment.

Zeolitization of the Dacitic Tuff in the Miocene Janggi Basin, SE Korea (장기분지 데사이트질 응회암의 불석화작용)

  • Dacitic tuffs, 97 to 118 m thick, were recovered from the lower part of the subsurface Seongdongri Formation, Janggi Basin, which was drilled to assess the potential for underground storage of carbon dioxide. The tuffs are divided into four depositional units(Unit 1 to 4) based on internal structures and particle componentry. Unit 1 and Units 3/4 are ignimbrites that accumulated in subaerial and subaqueous settings, respectively, whereas Unit 2 is braided-stream deposits that accumulated during a volcanic quiescence, and no dacitic tuff is observed. A series of analysis shows that mordenite and clinoptilolite mainly fill the vesicles of glass shards, suggesting their formation by replacement and dissolution of volcanic glass and precipitation from interstitial water during burial and diagenesis. Glass-replaced clinoptilolite has higher Si/Al ratios and Na contents than the vesicle-filling clinoptilolite in Units 3. However, the composition of clinoptilolite becomes identical in Unit 4, irrespective of the occurrence and location. This suggests that the Si/Al ratio and pH in the interstitial water increased with time because of the replacement and leaching of volcanic glass, and that the composition of interstitial water was different between the eastern and western parts of the basin during the formation of the clinoptilolite in Units 1 and 3. It is also inferred that the formation of the two zeolite minerals was sequential according to the depositional units, i.e., the clinoptilolite formed after the growth of mordenite. To summarize, during a volcanic quiescence after the deposition of Unit 1, pH was higher in the western part of the basin because of eastward tilting of the basin floor, and the zeolite ceased to grow because of the closure of the pore space as a result of the growth of smectite. On the other hand, clinoptilolite could grow in the eastern part of the basin in an open system affected by groundwater, where braided stream was developed. Afterwards, Units 3 and 4 were submerged under water because of the basin subsidence, and the alkali content of the interstitial water increased gradually, eventually becoming identical in the eastern and western parts of the basin. This study thus shows that volcanic deposits of similar composition can have variable distribution of zeolite mineral depending on the drainage and depositional environment of basins.
