Continuous Ethanol Fermentation by Immobilized Kluyveromyces marxianus F043 Using Jerusalem Arichoke Powder (돼지감자 분말을 이용한 고정화 Kluyveromyces marxianus sp.의 에탄올 연속발효)

  • To produce ethanol from Jerusalem artichoke powder efficiently, Kluyveromyces marxianus F043 cells were encapsulated in 2% sodium alginate and were cultured in a countinuous reactor to investigate the fermentation properties. Immobilized K. marxianus F043 cells were activated for 48 hours in a fermentor for continuous ethanol production. The culture in a CSTR using a Jerusalem artichoke substrate treated with 2% cellulase showed a decrease in ethanol concentration and an increase in residual saccharide concentration with a increasing dilution rate. Optimum conditions for high ethanol productivity and low residual saccharide output were clarified to be given at a dilution rate of 0.2 h$^{-1}$ and a Jerusalem artichoke medium concentration of 75 g/l. Ethanol productivity of 3.1 g/l-h and saccharide utilization of 62.6% were obtained under the optimum condition. When the fermentation was performed for 3 weeks under these conditions, the effluent medium showed stable ethanol concentrations of 16.3 - 17.9 g/l and viable cells of 6.60-7.16 log cells/ml without contamination. Trace amounts of methyl, n-propyl, iso-butyl, isoamyl alcohols besides ethanol were detected.

Production of Itaconic Acid by Various Immobilization Methods (다양한 고정화 방법에 의한 이타콘산 생산)

  • Aspergillus terreus NRRL 1960 was immobilized on various alginate gel beads, Celites, and polyurethane foam cubes, and the comparisons were made for the production of itaconic acid according to the types and sizes of each carrier. The levels of itaconic acid produced from Ca- alginate and Sr-alginate were similar, and the addition of bentonite to Ca- and Sr-alginate resulted in an increase of itaconic acid. The addition of 1.67% bentonite and 0.33% starch to Sr-alginate (SABS bead) produced higher level of itaconic acid (11.59 g/1) than other gel beads. A decrease in the size of Celite increased the itaconic acid production, and the smallest size of Celites, R- 634, produced 6.37 g/l of itaconic acid. Among various types of polyurethane foam cubes, HR 08 (2X2X2 cm) produced about 19 g/l of itaconic acid, which was more efficient than other carriers. In a repeated batch culture using immobilized cells on polyurethane foam cubes (HR 08, 2X2X2 cm), the stability of itaconic acid production was maintained up to 4 batches. Also, the possibility of itaconic acid production by continuous culture was shown in a packed-bed reactor.

  • The effects of methanol. used as a solvent for the hydrophobic substrate progesterone. on the morphology of Rhizopus nigricans and 11$\alpha$-hydroxylation of progesterone was investigated. The methanol tolerance of the 11$\alpha$-hydroxylase system in polyacrylamide immobilized R. nigricans mycelia as well as in free mycelia has been induced by adding various unsaturated fatty acids. biotin and ions into the cultivation medium. Immobilization of the cell seemed to protect the cells from denaturation by methanol. It gave higher reaction rate of progesterone than the free mycelia in the presence of methanol.500 $\mu$g/l of biotin was found to be the most effective induction agent for the methanol tolerance among tested chemicals. R. nixricans cells sustained its enzymatic activity at higher methanol concentrations as a result of accumulation of unsaturated fatty acids. especially oleic acid. in the membrane phospholipid.

Toluene Removal and Microbial Growth of Candida tropicalis Immobilized with Polymer Media in Airlift Bioreactors (효모 Candida tropicalis 고정화 담체를 이용한 Airlift 미생물반응기의 톨루엔 제거 및 미생물 성장)

  • This study was conducted to improve biological degradation efficiency of toluene as a model volatile organic compound (VOC) using yeast Candida tropicalis and to suggest an effective method for bioreactor operation. The yeast strain was immobilized with polyethylene glycol (PEG), alginate, and powdered activated carbon (PAC). The yeast-immobilized polymer media were used as fluidized materials in an airlift bioreactor. Polymer media without PAC were also made and operated in another airlift bioreactor. The two bioreactors showed toluene removal efficiencies ranging 80-96% at loading rates of $10-35 g/m^3-hr$, and the bioreactor containing the polymer media with PAC achieved higher removal efficiency. Protein contents in the liquid phase showed that the bioreactor using the yeast-immobilized polymer media with PAC had a higher rate of microbial growth initially than that without PAC. In addition, the microbial growth rate inside of the polymer media with PAC was five times higher than that without PAC. Consequently, the polymer media containing the yeast strain and PAC could enhance removal efficiencies for VOCs, and the immobilization method improve microbial activity and stability for a long-term operation of biological systems.

  • Hydrogen($H_2$) as a clean, and renewable energy carrier will be served an important role in the future energy economy. Several biological $H_2$ production processes are known and currently under development, ranging from direct bio-photolysis of water by green algae, indirect bio-photolysis by cyanobacteria including the separated two stage photolysis using the combination of green algae and photosynthetic microorganisms or green algae alone, dark anaerobic fermentation by fermentative bacteria, photo-fermentation by purple bacteria, and water gas shift reaction by photosynthetic or fermentative bacteria. In this paper, biological $H_2$ production processes, that are being explored in fundamental and applied research, are reviewed.

  • Ammonia is a useful substance which is widely used in various industries. It is generally released by the decomposition of agricultural wastes and known to have toxic effects on human beings. Due to the common usage, it is possible to cause water pollution through either direct or indirect leakage. Such cases, it is preferable to use the adsorption capacity of zeolite to rapidly remove ammonium ions, but it is not sufficiently removed by the adsorption only. In this paper, the removal efficiency of ammonium ion through both the adsorption capacities of commercial synthetic zeolites and the biological mechanism of microorganisms were compared. In addition, microorganisms were immobilized on the zeolite in order to enhance the removal efficiency by applying a chemo-biological process. As a result, the standard commercial zeolite showed 67~81% of the removal efficiency in 2~4 hours at a 100 ppm concentration of ammonium, whereas the selected microorganism Klebsiella pneumoniae subsp. Pneumoniae showed up to 97% within 8 hours. When the microorganism was immobilized on the zeolite, the highest removal efficiency of approximately 98.5% were observed within 8 hours.

Removal Characteristics of Toluene in Biofilters Packed with Reticulated-PU-Foams of Different Porosities (서로 다른 공극률의 망상형 폴리우레탄들이 충전된 바이오필터에서 톨루엔 제거 특성)

  • We studied on the removal of toluene vapors in a lab-scale biofilter. There are three biofilters packed with reticulated polyurethane foams of different porosities of 15, 25, 45 PPI (Pore Per Inch) as media. A toluene-degrading strain (Pseudomonas Putida KCCM 11348, ATCC 12633) was naturally immobilized on the filter media by circulating the culture media. Three biofilters were operated under different sets of continuous experiments, varying both the design and operation parameters such as the inlet toluene concentration and the flow rate. Maximum elimination capacity of 115.5g/㎥hr of biofilter packed with foams of 25 PPI was obtained for toluene degradation. The effect of operating conditions such as flow rate, inlet toluene concentration and porosity on the performance of the biofilter was investigated.

Characteristics of Wastewater Treatment by the Continuous-Flow Fixed Biofilm Process Using Porous Fly Ash Carrier (다공성 석탄회 담체를 이용한 연속류식 고정 생물막 공정의 폐수 처리 특성)

  • We have prepared the porous carrier that has high capacity for immobilization of microorganisms and adsorption capacity for cation using fly ash in the previous work. In this study, we investigated the characteristics of piggery wastewater treatment in comparison with commercial carrier and conventional activated sludge process by continuous-flow fixed biofilm process in laboratory scale at the same experimental conditions to develop the biofilm process using porous fly ash carrier for wastewater treatment. As a result, the prepared fly ash carrier showed that removal efficiency of COD, TN and $NH_4{^+}-N$ items were 80%, 77% and 65%, respectively, which were higher efficiency than the commercial carrier and conventional activated sludge process. And the result of measurement for immobilized microorganisms after treatment showed higher capacity than the commercial carrier, and it was confirmed by SEM observation on fly ash carrier that the colony of microorganisms was stably formed.

  • Nitrogen oxides (NOx) in combustion flue gas are currently mitigated by chemical processes such as catalytic reduction, absorption and adsorption. However, development of environmentally sustainable biological processes is necessary in the near future. In this paper, the up-to-dated R&D trend of biological methodologies regarding NOx removal was reviewed, and their advantages and disadvantages were discussed. The principles and applications of bacterial system including nitrification and denitrification and photosynthetic microalgae system were compared. In order to enhance biological treatment rate and performance, the insoluble nitric oxide (NO) should be first absorbed using a proper solubilization agent, and then microbial degradation or fixation is to be followed. The use of microalgal system has a good prospect because it can fix $CO_2$ and NOx simultaneously and requires no additional carbon for energy source.

Rapid Detection of Salmonella spp. by Antibody Immobilization with Gold-protein A Complex (Gold-protein A Complex 항체 고정화법을 이용한 Salmonella spp.의 신속 검출)

  • A piezoelectric (PZ) biosensor system detecting Salmonella spp. was developed. The system consisted of an oscillator, a frequency counter and an antibody-immobilized quartz crystal. An anti-Salmonella antibody was immobilized on one gold. surface of the quartz crystal with protein A. Salmonella detection was made by measuring resonant frequency shift owing to a mass change by specific binding of microbial cells to the gold surface of the PZ crystal. The PZ antibody sensor was operated optimally at 0.1M phosphate buffer, pH 7.2 and $35^{\circ}C$. The sensor was quite specific to Salmonella spp. The obtained frequency shift was correlated with the Salmonella concentration in the range of $10^5{\sim}10^6\;CFU/mL$. The frequency shift increased further by addition of polystyrene beads. The Salmonella detection which is indicated by a steady-state microbial adsorption to the quartz crystal was accomplished within 50min.
