  • A cytochrome c-552 from a thermophilic hydrogen-oxidizing bacterium, Hydrogenobacter thermophilus, was amplified using PCR. The cytochrome c-552 gene was cloned into E. coli vector pAlter-1 and transferred to JM109. Glutamine of cytochrome c-552 protein was changed to cysteine through point mutation.

Purification and Properties of Protease from Thermophilic Actinomyces (고온성 방선균이 생산하는 단백질 분해효소의 정제와 특성)

  • Microbial protease has been interesting due to the biological roles in the producing microorganism. A thermophilic Actinomyces produing protease was isolated from soil. The optimal medium composition and culture conditions for maximum protease production was as follows 0.5% soluble starch, 0.5% yeast extract. 0.1% K2HPO4, 0.05% CaCl2, initial pH 8.0 at 50$^{\circ}C$ for 48hours. The protease was purified by the procedure of ammonium sulfate precipitation, anion exchange chromatography(LC), DEAE high performance liquid chromatography and GPC HPLC. The purification fold of the purified enzyme was increased about 22.6. The optimal pH and temperature for reaction of the purified enzyme were 7.5 and 60$^{\circ}C$. The purified enzyme was stable for the pH range from 6.0 to 8.5, but was unstable when treated at 80$^{\circ}C$ for 10 minutes. The activity of the enzyme was inhibited by Ag+ and Cu2+.

  • 고순도의 수소를 정제 및 분리하기 위한 팔라듐계 합금 수소분리막은 높은 수소 투과-선택 특성이 요구된다. 그러나 종래의 스퍼터 공정 조건으로 팔라듐계 분리막을 제조하면 주상정 구조에 기인하여 표면에 기공이 형성되게 된다. 본 연구에서는 개선된 스퍼터 시스템에서 고진공, 고온 및 높은 직류 전원 공정조건 하에 치밀하고 균일한 팔라듐/금 수소분리층을 제조하였다. 이와 같은 공정 조건에 의해 종래의 제조 공정 조건 보다 얇은 분리막을 제조하여 공정의 경제성을 향상 시켰으며, 기공이 포함되지 않은 수소분리층을 형성하여 수소 선택 특성이 무한대의 값을 가지는 것으로 관찰되었다.

  • 합성가스는 C1화학을 시작하는 반응원료 물질로 최근 DME(dimethyl-ether), 메탄올, GTL(gas to liquid), CTL(coal to liquid), 암모니아 생성 공정 등 많은 화학공정에 사용되고 있다. 합성가스를 생산하는 방법은 천연가스 개질반응과 석탄의 가스화반응, 그리고 원유의 정제 등을 통해 얻을 수 있다. 삼중개질반응은 천연가스와 산소, 수증기, 이산화탄소를 원료로 $1000^{\circ}C$ 이상의 고온에서 반응시켜 합성가스를 생산하며, 균일반응계와 불균일반응계로 이루어져 있다. 균일반응계에서는 천연가스와 산소가 주로 반응하며, 원료로 투입된 대부분의 산소는 균일반응계에서 소모되어 일산화탄소와 이산화탄소를 생성한다. 삼중개질반응의 균일반응계에서는 산소와 천연가스와의 반응으로 많은 발열이 발생하여 전체 반응계의 온도를 유지할 수 있도록 해준다. 본 연구에서는 산소로 인한 삼중개질반응의 온도 조절과 균일반응계의 온도 분포를 위치에 따라 관찰해 보았으며, 실험과 모사를 통해 비교해 보았다.

Reason and Prevention of Color Reversion of Corn Oil in Summer (옥수수기름의 하절기 변색현상 원인규명 및 방지대책)

  • Crude corn oil (CCO) was obtained through the expression-extraction process from corn germ. The CCOs of final process at $90^{\circ}C,\;70^{\circ}C\;and\;50^{\circ}C$ were stored in outdoor storage tanks. From the samples, refined CCO (RCCO) were prepared with $0\%,\;10\%\;and\;20\%$ excessive of phosphoric acid, caustic solution and acidic clay were used in degumming-alkali refining-bleaching process. RCCOs were stored at room temperature in dark places. The color change was not effected by the amount of phosphoric acid, caustic solution and acidic clay, but temperature of process affects the color change. Finally, the prevention for color reversion of RCCO could be obtained by lowering the temperature of final process and optimal temperature of RCCO in summer was found about less than $50^{\circ}C$.

  • ZnO-CuO mixed sorbents for desulfurization in hot gas cleaning process Were prepared and investigation on their characteristics was performed in this study. The rate of sulfidation increased as the amount of copper oxide in the composite sorbent was raised. TPO experiments were carried out to investigate the characteristics of the regeneration of the sorbents with severa1 different ratios of Zno to CuO. Copper sulfate was formed at temperatures above $400^{\circ}C$, while it was decomposed by pyrolysis above $700^{\circ}C$. $SO_2$ slippage due to $CuSO_4$ was observed in the sorbent regenerated at temperatures above $600^{\circ}C$. However, it was not observed when regenerated above $700^{\circ}C$. It was confirmed in the ZnO-CuO mixed sorbent system that CuO suppressed the vaporization of ZnO on the one hand and Zno minimized the $SO_2$ slippage due to CuO on the other hand.

  • Gaseous sulfur compound such as $H_2S$ or COS in coal- or biomass-derived hot syngas can be purified by solid sorbents at high temperatures. In this study, we investigated the physical properties and reactivity of solid regenerable desulfurization sorbents with 37.2, 41.9, and 46.5wt% ZnO to look into the ZnO content effect. The sorbents were produced by spray-drying method to apply to a fluidized-bed process. Sulfidation and regeneration reaction were carried out using a thermogravimetric analyzer. Sorbent prepared with 46.5wt% ZnO had physical properties suitable for a fluidized-bed process applications such as spherical shape, sufficient mechanical strength and density, high porosity and surface area. It showed high sulfur sorption capacity of 10.4wt% (ZnO utilization of 57%) at reaction temperatures of 500 and $650^{\circ}C$ for sulfidation and regeneration, respectively. However, the sulfur sorption capacity and ZnO utilization were significantly reduced and dimple shape appeared when the ZnO content decreased to 37.2 and 41.9wt%. Sulfur sorption capacity and regenerability were improved as reaction temperature increased within the experimental temperatures used in this work. The reaction temperature zones of $1500{\sim}550^{\circ}C$ and $650{\sim}700^{\circ}C$ are recommended for sulfidation and regeneration, respectively, to lead best reaction performances of the ZnO-based spray-dried sorbents developed in this work.

  • 분류층 가스화기는 석탄과 산소(공기) 및 수증기가 반응하여 $1200{\sim}1600^{\circ}C$의 고온, 20~60기압의 고압에서 작동되어 합성가스를 생성하며 합성가스에 포함된 입자 및 황화합물 등을 정제설비를 통하여 정제 후 발전 및 화학원료로 사용한다. 석탄가스화 중 석탄에 포함된 대부분의 회분은 용융슬래그 형태로 가스화기 벽면을 따라 흘러 내려 가스화기 하부의 냉각수조에서 급랭되어 배출된다. 이때 용융슬래그의 원활한 배출을 위해서는 일정범위의 점도를 유지하는 것이 필요하다. 슬래그의 점도는 가스화기 온도 및 Ash의 조성에 따라 크게 변하며 가스화기 설계 및 운전 시 매우 중요한 변수이다. 따라서 최적의 설계 및 운전을 위해서는 Ash의 점도예측이 중요하며, 분류층 가스화기내부에서 Ash 점도 예측을 위한 DooVisco 프로그램을 개발하였다. DooVisco는 가스화기 내부에서 슬래그 용융온도 및 온도별 점도, 가스화기 최소 운전온도 및 석회석 투입 효과 분석뿐만 아니라 석탄의 혼합 사용 시의 특성 예측도 가능하도록 개발되었다. DooVisco는 슬래그 주요 4성분인 SiO2, Al2O3, CaO, FeO 성분에 대한 Phase Diagram을 이용하여 1차적으로 슬래그용융온도(Liquidus Temperature)를 예측하고, 주요 4 성분 외에 Na2O, MgO, K2O, TiO2 등을 고려한 Kalmanovich Model을 이용하여 점도를 예측한다. 최종적으로 슬래그 용융온도와 점도를 활용하여 분류층 가스화기 운전가능 온도범위를 예측한다. 개발된 DooVisco를 활용하여 300MW급 실증 IGCC 플랜트에 사용가능성이 있는 석탄을 대상으로 슬래그의 용융온도 및 점도 등을 예측하였으며 최적 운전을 위한 슬form점도 조절용 Flux인 석회석 투입량 등을 평가하였다. 평가 결과 슬래그 용융온도가 $1700^{\circ}C$ 이상으로 석회석 투입이 필요하다고 판단되었다. 약 가스화기 내부 온도를 $1500^{\circ}C$ 정도에서 원활한 운전을 위해서는 석탄 대비 약 10% 내외의 석회석 투입이 필요할 것으로 평가되었다. DooVisco는 분류층 가스화기 설 계시 가스화기 최적 운전 온도 설정 및 Flux 투입필요성, 종류, 투입량 선정에 활용될 수 있을 뿐만 아니라 플랜트 운전시 석탄의 탄종 적합성 등을 판단하는데 활용될 수 있을 것이라 판단된다.

  • The purpose of this study is to process a seaweed resources that is easy to find in Korea and has outstanding phlogistone and high yield seaweed vinegar solution through mass-production and facility development for utilization. The compound uses wasted seaweeds, such as brown seaweed, kelp, seaweed fusiform, and laver, and carbonizes them in high temperature by drying in poly step trap to extract them. In also involves the development of a process that separates and refines the extracts to create a high value-added anti-bacterial bio solution that can replace chemicals in agriculture and industries and even chemical food additives. so, studied separation and refining of seaweed extracts to analyze the compounds. and processed and compared to seaweed vinegar extracts to prove superiority.

Purification of Waste Acid and Manufacture of Complex Oxide and Mn-Ferrite Powder by Co-Roasting Process (폐산의 정제 기술 및 분무 배소법에 의한 복합 산화물과 Mn-Ferrite 분말의 제조)

  • The purpose of this study is to produce high putity composite powder composed of Fe-oxide, Mn-oxide and Mn-ferrite having superior homogencity in composition and particle size distribution by co-roasting process. Binary component metal (Fe, Mn) chloride solutions were produced by dissolving mill scale and ferro-mangancse alloy in hydrochloric acid. These chloride solutions contained the impurities such as SiO$_{2}$, P, Al, Ca and Na, which were originated from the Fe/Mn source materials. The neutralization and polymeric coagulant method were adoped to refine the hydrochloric liquor. When pH is far below the isoelectric point (pH 2-3), the SiO$_{2}$ was the most effectively reduced element, while other impurities remained unchanged. By increasing pH above 3, most of the impurities could be reduced effectively due to the coprecipitation reaction. The polymeric coagulants such as poly vinyl alcohol, resin amine and ammonium molybdate were found to have no effect on the spray roaster designed by the authors. The produced oxide powders were confirmed to be mixtures of Fe-oxide, Mn-oxide and mn-ferrite. the powders were homogeneously mixed and the particle size increased sleeply with increasing co-roasting temperature.
