Soil washing by surfactants is a technology to enhance mobilization and subsequent degradation of oil pollutants by reducing the surface tension of pollutants which is combined with soil. In this study, biosurfactant, rhamnolipid was produced from Pseudomonas aemginosa ATCC 9027 which had an excellent biodegradable activity in soil without causing secondary pollution. Effects of chemical surfactants on the removal of diesel from diesel-contaminated soil were compared to those of biosurfactants including rhamnolipid. Diesel removal efficiency by rhamnolipid extracted from P. aeruginosa culture broth was over 95% in both batch and column washing test in 5,000ppm diesel-contaminated soil with 1% surfactants after washing for 24 hours. On the contrary, the results of chemical surfactants were below 50∼80%, The chemical surfactants with HLB value(8∼15) showed more then 75% efficiency of diesel removal. But, when the HLB values were below 8 or over 15. their efficiency were observed as less then 60% of diesel removal. Rhamnolipid, biologically produced surfactants, may also be promising agent for enhancing diesel removal from contaminated soil.

유류오염토양의 자연복원 시 생물이용도(bioavailability)를 높이기 위해 사용되는 계면활성제의 영향을 살펴보고자 제지 폐수로부터 분리한 미생물과 5가지 계면활성제를 사용하였다. 계면활성제로는 Brij 35, Brij 30, Triton X-100, Tween 40, Tween 80을 사용하였으며 유류오염물질로는 PAHs를 사용하여 계면활성제 별 미생물의 성장률 및 PAHs 분해율을 살펴보았다. 그 결과 각각의 계면활성제에 따라 서로 다른 미생물 성장 및 PAHs 분해능을 보였다. 계면활성제에 따른 미생물 성장은 Tween 계열에서 뚜렷했으며 이들 중 Tween 40에서는 PAHs의 분해능 또한 가장 활발하였다.
The biodegradation of ($C^{14}$)phenanthrene was studied in water and soil-water systems with nonionic surfactants and biosurfactant : polyoxyethylene alkyl ester($C_{17}$$H_33$COO($C_2$$H_4$O)nH) and sophorolipid. The extents of so1ubilization and biodegradation were monitored by radiotracer technique. Experimental results showed that surfactant concentrations above the critical micelle concentration were toxic to the phenanthrene-degrading bacteria in soil or active sludge and the presence of surfactant micelles inhibited mineralization of PAHs. Solubility and bioavailibility of phenanthrene in water and soil-water system were enhanced by mixed surfactants system. The optimum water content and hydrogen concentration were 30% (w/v) , pH 7, respectively.
Productivity of biosurfactant (rhamnolipid) by Pseudomonas aeuginosa F722 was investigated in the several culture conditions and culture composition. Biosurfactant production by P. aeuginosa F722 was amounted to 0.78 g/l as the result of the nitrogen sources and carbon sources without investing of optimum conditions. As for that one was investigated, biosurfactant production by P. aeruginosa F722 was amounted to 1.66 g/l. Biosurfactant production increased twofold because the composition of a modified C-medium was investigated efficiently. $NE_4$Cl or $NaNO_2$ inorganic nitrogens and yeast extract or trypton organic nitrogens were effective, but others inorganic nitrogens and organic nitrogens tested were not efficient far biosurfactant production by P. aeruginosa F722. The optimum concentration of $NH_4$Cl; inorganic nitrogen and yeast extract; organic nitrogen were 0.05% and 0.1%, respectively. In various carbon sources, others with the exception of hydrophobic property substrate (n-alkane) and hydrophilic property substrate (glucose, glycol) were not found to be effective fur biosurfactant production, and 3.0% was better in yield than other concentration of glucose. This yielded C-to-N ratios between 17 and 20. In our experiment, the highest biosurfactant production by P. aeruginosa F722 were observed in 5 days cultivation, containing glucose 3.0%, $NH_4$Cl 0.05%, and yeast extract 0.1% and C-to-N ratio was 20. Optimal pH and temperature for biosurfactant production were 7.0 and $35^{\circ}C$, respectively. Under the optimal culture conditions with glucose, biosurfactant production was amounted to 1.66 g/l. Velocity of biosurfactant production and strain growth increased after nitrogen depletion. The average surface tension of 30 mN/m after the 3 days of incubation under optimal culture condition was measured by ring tensionmeter.
In this study, the effect of the addition of fluorinated surfactant FS-606 on the solubilization of n-hexane by hydrocarbon surfactant CDP-W was investigated. Oil drop contacting experiments revealed that solubilization rate is independent of initial oil drop size and proportional to the initial surfactant concentration, suggesting that solubilization of n-hexane oil by the surfactant mixture of FS-606 and CDP-W is controlled by an interface-controlled mechanism. In addition, the solubilization rate has been shown to increase with an increase in FS-606 composition, reach a maximum, and then decrease with a further increase in FS-606 composition. On the other hand, the interfacial tension between micellar solution and n-hexane oil has been found to decrease with an increase in FS-606 composition, reach a minimum, and then increase with a further increase in FS-606 composition.
This study was carried out 1) to investigate the pH effect on solubilization of phenanthrene by biosurfactant in aqueous system and 2) to evaluate the pH effect on the biodegradation rate of phenanthrene in the presence and the absence of the biosurfactant by phenanthrene degraders. Tween 80, which is a chemically synthesized surfactant, showed greater solubilizing capacity than rhamnolipid. The solubilization capacity can be expressed as a MSR(molar solubilization ratio=moles of organic compounds solubilized per mole of surfactant). The calculated MSR of Tween 80 and rhamnolipid were 0.1449 and 0.0425 respectively. The kinetic study of phenanthrene solubilization by rhamnolipid showed that solubilization mechanism could reach equilibrium within 24 hours. Addition of 240 ppm rhamnolipid solution, which concentration is 4.3 times of Critical Micelle Concentration(CMC), caused 9 times solubility enhancement compared to water solubility. The highest solubilities were detected around a pH range of 4.5-5.5. Changes in apparent solubility with the changes in pH are possibly related to the fact that the rhamnolipid, an anionic surfactant, can form different structures depending on the pH. Two biodegradation experiments were performed in the absence and the presence of rhamnolipid, with the cell growth investigated using a spread plate method. The specific growth rates at pH 6 and 7 were higher than at the other pH, and the HPLC analysis data, for the total phenanthrene loss, confirmed the trends in the $\mu$(specific growth rate) values. In presence of rhamnolipid, maximum $\mu$ values shifted from around pH 5 which showed maximum enhancement of solubility in the abiotic experiment, compared to the $\mu$ values obtained without the biosurfactant. In this study, the increase in the observed specific grow rate(1.44 times) was not as high as the increase in solubilization(5 times). This was supported by the fact all the solubilized phenanthrene is not bioavailable to microorganisms.
Bacillus subtilis JK-1 showed degradation activity against crude oil, gasoline, kerosene, and light oil, and this strain was used as a crude biosurfactant producing microorganism in this study. To optimize the culture medium for production of crude biosurfactant, the influences of various carbon, nitrogen and mineral sources were assessed. The highest biosurfactant production by B. subtilis JK-1 was observed after 96 h cultivation, containing 1.0% (w/v) soluble starch as a carbon source and 0.5% (w/v) skim milk as a nitrogen source, and carbon to nitrogen concentraion (C/N) ratio was 2.0. For the biosurfactant production 0.1% (w/v) of $KNO_3$ was the most effective mineral source. Comparison of biosurfactant production indicates that B. subtilis JK-1 produces more biosurfactant in the optimum medium established in this study than LB and TSB. Under the optimum medium, the surface tension of culture broth of B. subtilis JK-1 was decreased from 47.3 dyne/cm to 24.0 dyne/cm after cultivation of 48 h.
A kinetic model for evaluating effects of surfactant on the biodegradation of HOC(hazardous organic chemicals) in soil-slurry systems was developed. The model includes the partition of HOC and surfactant, the dissolved-, micellar-, and sorbed-phase biodegradation, the enhanced solubilization of HOC by surfactant addition, and the mass transfer of HOC. Phenanthrene as HOC and Trition X-100, Tergitol NP-10, Igepal CA-720, and Brij 30 were used in the model simulations. The biodegradation rate was increased even with a small micellera-phase bioavailability. The biodegradation was not greatly enhanced due to decreased aqueous HOC concentration by increasing surfactant dose in both cases with and without micellar-phase bioavailability. The effect of sorbed-phase biodegradation on total biodegradation rate was not highly important compared to aqueous- and micellar-phase biodegradation. The model can be applied for surfactant screening and optimal design of surfactant-based soil bioremediation process.
The purpose of this paper is to analyze the characteristics and chemical components of biosurfactant produced by Pseudomonas sp. G314. Pseudomonas sp. G314 was isolated from soil samples which were contaminated with oil in Daejon area. As such, it produced quality biosurfactant [23]. One type of biosurfactant was kept in a refrigerator, whereas another type of biosurfactant was kept in room temperature. The surface tension activities were then compared. As a result, the biosurfactant from Pseudomonas sp. G314 that was kept at room temperature was stable for 10 days, showing 26.2 dyne/cm of surface tension activity. This result was found to be similar to that of the refrigerator storage. The surface tension of batch culture was 25 dyne/cm, but the culture in the 5 l fermentor was 27 dyne/cm. Therefore, it can be suggested that the large-scale culture is feasible via the fermentor. Biosurfactant from Pseudomonas sp. G314 was estimated to be a kind of glycolipid because it dissolved in acetone and methanol much better than in benzene and toluene [23]. A spot was detected through the elution of silica gel column and the spread of TLC, and the Rf value was 0.58. This spot has a positive reaction with Bail's reagent and rhodamine 6G. Hence, we can conclude that biosurfactant from Pseudomons sp. G314 was a glycolipid containing carbohydrate and lipid.
The purpose of this work was to investigate the characteristics of a biosurfactant-producing bacterium isolated from crude-oil contaminated soils. During the incubation of strain HK-3 with 1% crude-oil, bacterial growth pattern, the amount of biosurfactant production, and pH changes were monitored. In order to examine the effect of supplemented carbons on the production of biosurfactant, cultivation of HK-3 cells in BH media with different carbons (e.g. glucose, dextrose, mannitol, citrate, or acetate) revealed that the production of biosurfactant reached the maximal level at the 72 h incubation with mannitol, which the area of clear zone was measured to approximately 7.64 $cm^2$. Identification test using the BIOLOG system, morphology study based on scanning electron microscopy and the 16S rRNA sequence-based phylogenetic analysis assigned strain HK-3 to a Pseudoalteromonas species, designated as Pseudoalteromonas sp. HK-3 which was registered in GenBank as [FJ477041].
