Effects of Estradiol-17β on the Feminization of Japanase Eel, Anguilla japonica (극동산 뱀장어, Anguilla japonica 자성화에 미치는 estradiol-17β 유도 효과)

  • The effects of oral administration of estradiol-$17{\beta}$ (E2) on glass eels (Body weight: $0.16{\pm}0.05g$, Total length: $6.2{\pm}0.9cm$) and young eels (Body weight: $2.6{\pm}0.6g$, Total length: $13.2{\pm}0.6cm$) on gonadal sex and growth were examined, respectively. Glass eels were fed a diet containing E2 at a dose of 10 mg/kg or 25 mg/kg, respectively, for five months. The female ratio significantly increased in all E2-treated groups (10 mg/kg diet group: 70%; 25 mg/kg diet group: 90%) when compared to the control group (10%). Young eels were fed a diet containing E2 at a dose of 25 mg/kg for four months. The female ratio also significantly increased in the E2-treated groups (60%) compared to the control group (20%). The highest female ratio was observed in the stage of glass eels rather than young eels. In all experiments, however, the growth of eels treated with E2 was similar to that of controls to the end of the experiment. Thus, oral administration of E2 could be a good approach to controlling sex differentiation.

The Effect of Exercise on the Conversion Rate of Ingested Glueose to Glycogen in the Hindlimb Skeletal Muscles in Rats (흰위에서 운동부하후 경구투여한 Glucose가 특성이 다른 골격근에서 Glycogen으로 합성되는 속도)

  • In the present study the effect of exercise on the conversion rate of ingested glucose to glycogen in the different types of hindlimb skeletal muscles in Sprague-Dawley male rats was studied. The amounts of synthetized glycogen from ingested glucose of fast-twitch white(WV), fast-twitch red(RV), mixed type of fast-twitch white and red(EDL), and slow-twitch(SOL) muscles were determined at 30 and 90 min. after ingestion of 25% glucose solution which contained $^{14}C$-glucose($2m{\ell}(1uCi)$/100gm B. W.)in control and exercise loaded rats. The result was summarized as follows : The about 55% at 30 min. and 70% at 90 min. after glucose ingestion were absorbed from gastrointestinal tract. This result shows no effects of exercise on absorption rate from gastrointestinal tract. The amount of synthetized glycogen of SOL from ingested glucose at 30 and 90 min. after glucose ingestion were highest, whether WV were lowest in hindlimb skeletal muscles in control and exercise loaded rats. In the exercise loaded rats, the amounts of synthetized glycogen of SOL, RV, and EDL at 90 min. after glucose ingestion was much higher than control rats, but not different in WV between exercise-loaded and control rats. At 30 min. after glucose ingestion, only SOL of exercise loaded rats was higher than control rats. In the control rat, the synthesis of glycogen was almost completed during initial 30 minutes. On the other hand, in the exercise loaded rat, except WV was opposite result of control rats, i. e., amounts of synthetized glycogen were major during late period. The amount of synthetized glycogen of liver at 30 and 90 min. after glucose ingestion in exercise loaded rats was higher than control rats. The rate of glycogen synthesis in control and exercise loaded rats were higher between 30-90 minute than initial 30 minute.

Effects of Oyster Shell Extract on Papain-induced Osteoarthritis in C57BL/6J Mice (굴 패각 추출물이 Papain으로 유도된 골관절염 C57BL/6J Mice에 미치는 영향)

  • We investigated the effects of oyster shell extract (OSE) on papain-induced osteoarthritis in C57BL/6J mice. Osteoarthritis was induced in mice by a papain injection into the knee joint. The mice were divided into a total of five groups (n=8). The normal group was untreated, whereas the papain group (negative control) was induced with osteoarthritis and treated with water daily. The papain+DS group (positive control) was treated with diclofenac sodium. Papain+OSE groups were treated with OSE concentrations of 100 and 200 mg/kg/bw for 20 days. Proteoglycan content in articular cartilage was analyzed through safranine-O fast green staining and H&E staining. The histopathological changes in cartilage were measured by the Rudolphi score approach. The contents of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin (IL)-$1{\beta}$, and IL-6 in plasma were analyzed by the ELISA method. After experiments, body weights of the treated groups were not significantly different compared with the normal group. Cartilage loss and joint instability significantly improved in a dose-dependent manner in the OSE-treated group compared with the papain group (P<0.05). Proteoglycan content was significantly higher in the OSE-treated group than the papain group (P<0.05). Osteoarthritis scores of the OSE-treated group were significantly decreased compared with the papain group (P<0.05). TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 content in the plasma of the papain+OSE treated groups significantly decreased in a dose-dependent manner compared with the papain group (P<0.05). These results suggest that OSE treatment might have anti-arthritic effects on papain-induced osteoarthritis in C57BL/6J mice.

Pharmacokinetics of oxolinic acid in cultured olive flounder Paralichthys olivaceus by oral administration, injection and dipping (Oxolinic acid의 경구투여, 주사 및 약욕에 따른 넙치, Paralichthys olivaceus 체내 약물동태학적 특성)

  • The pharmacokinetic properties of oxolinic acid (OA) were studied after oral administration, intraperitoneal injection and dipping to cultured olive flounder, Paralichthys olivaceus (average 90 g, $23{\pm}1{^{\circ}C}$). Plasma samples were taken at 3, 5, 10, 15, 24, 30, 48, 96 and 144 h post-dose. In oral dosage at 15, 30 and 60 ㎎/㎏, the peak plasma concentrations of OA, which attained at 10~15 h post-dose, were 1.92, 2.45 and 3.72 $\mu{g}/m\ell$, respectively. In intraperitoneal injection with 10 and 20 ㎎/㎏, the peak plasma concentrations of OA, which attained at 10 h post-dose, were 4.1 and 4.8 $\mu{g}/m\ell$, respectively. In dipping in 30 and 50 ppm for 1 h, peak concentrations were observed at 5 h and 30 h post-dose, were 0.22 and 0.38 $\mu{g}/m\ell$, respectively. The kinetic profile of absorption, distribution and elimination of OA in plasma were analyzed fitting to a one-compartment model by WinNonlin program. Calculated parameters for a single oral dosage of 15, 30 and 60 ㎎/㎏, respectively, were: AUC (the area under the concentration-time curve)=70.93, 120.0 and 141.86 $\mu{g}$ $h/m\ell$ $T_{max}$ (time for maximum concentration)=16.22, 20.39 and 17.33 h; $C_{max}$ (maximum concentration)=���D1.61, 2.40 and 3.01 $\mu{g}/m\ell$. Following intraperitoneal injection of 10 and 20 ㎎/㎏, these parameters were AUC=184.7 and 315.92 $\mu{g}$ $h/m\ell$ $T_{max}$=5.91 and 6.26 h; $C_{max}$=4.19 and 4.45 $\mu{g}/m\ell$. Following dipping at 30 and 50 ppm, these parameters were AUC=17.58 and 21.69 $\mu{g}$ $h/m\ell$ $T_{max}$=19.08 and 31.43 h; $C_{max}$x=0.22 and 0.25 $\mu{g}/m\ell$.

Effects of Lespedeza Caneata ethanol extract on the Liver, Kidneys of Lead administered Mice (야관문(Lespedeza Caneata) Ethanol 추출물이 납 투여한 생쥐의 간장과 콩팥에 미치는 영향)

  • This study determined the effect of Lespedeza Caneata extract on the livers and kidneys of lead-administered mice. The experimental groups were divided into a normal group, Pb 4W group, Pb-LC 4W group, Pb 8W group, and Pb-LC 8W group. The normal group was supplied single distilled water, and the pb group was provided distilled water in which lead acetate was dissolved at 1,000 ppm. The Pb-LC group was provided with lead as drinking water, and the Lespedeza Caneata was orally medicated at a concentration of 500 mg/kg daily. AST, ALT, and BUN enzyme activities and histological experiments on the livers and kidneys resulted in the following conclusions. AST, ALT, and BUN activities increased in the experimental group compared to the normal group and decreased more in the Pb-LC group than the pb group during the same period. The histological results reveal that portions of the livers and kidneys were deformed in the Pb 4W group, and most of the Pb 8W group experienced necrosis and deformation. pb-LC4W and Pb-LC 8W groups experienced less deformation than the Pb 4W and Pb 8W groups. During the same period, the Pb-LC group experienced less histological changes than the Pb group. These results suggest that Lespedeza Caneata extract may have some protective effect on hepatic tissue and renal tissue damage with lead-administered in mice.

Effects of BCL oral administation and herbal acupuncture at BL18, BL19 on Liver function changes induced by Alcohol in the mice (죽력(竹瀝)(죽작액(竹昨液)) 경구투여(經口投與)와 간유(肝兪)·담유(膽兪) 약침(藥鍼)이 Alcohol 대사(代謝) 및 간기능(肝機能)에 미치는 영향(影響))

  • Objective : This dissertation was designed to evaluate the effect of BCL(refinded Bambusae Caulis in Liqua-men) oral administration and herbal acupuncture on alcohol metabolism and liver function. Methods : Mice were damaged by a large quantity of alcohol and received treatment of either BCL $1mg/kg$ in oral or BCL $250{\mu}g/kg$ in herbal acupuncture-BL18 BL19 bilateral. and then such parameters as GOT, GPT, catalase and superoxide dismustase(CuZn-SOD, Mn-SOD) were measured. Results : 1. Compared with control group, the activity of GOT, GPT in serum was significantly reduced and the proper degree of alcohol in serum was not significantly differ from oral administration group and herbal acupuncture group. 2. The activity of catalase in liver cell tissue and the activity of CuZn-SOD in liver cell tissue, compared with control group, was not sigificantly affected either by oral administration and herbal acupuncture group. But the activity of Mn-SOD was significantly increased in oral administration group, while it was not the case in acupuncture group. Conclusion : we consider that BCL oral administration and herbal acupuncture is highly effective in recovering alcohol metabolism and liver disfunction induced by alcohol.

The activity of Mn-SOD was significantly increased in oral administration group. while it was not the case in acupuncture group. In conclusion. we consider that BCL oral administration and herbal acupuncture is highly effetive in recovering alcohol metabolism and liver disfunction induced by alcohol.

Effect of Extract Agrimonsa Pilosa L. on biological Activity in Rats (선학초(仙鶴草) 추출액이 흰쥐의 생리활성(生理活性)에 미치는 효과)

  • Agromonia pilosa L. has been used as a medicinal plant in traditional folk remedy, and it shows increasing tendency at various sections such as medicine-making material, functional food, and agricultural chemicals using plant or extract. Dosage effect of extract from Agrimonia pilosa L. plant on rat performance experiments were summarized as follows : Body weight was increased with 0.02% dosage treated-rat by 5% as compared to non-treated one, however, decreasing tendency was observed with 0.04% extract of Agrimonia pilosa L. plant fed rat to control Considering feed efficiency was similar result between extract dosage with 0.02% and non-treated rat. The number of leucocyte was increased by dosage-treated level except 0.02% dosage-treated rat. Number of erythrocyte was increased with 0.06% extract dosage-treated rat by 20% as compared to non-treated one.

Effects of the Gamidokwalgisaeng-tang(GDS) Oral Administration and Electro-acupuncture at Chishil$(BL_{52})$, Hyonjong$(GB_{39})$ on Osteoporotic Rats Induced by Ovariectomy (가미독활기생탕(加味獨活寄生湯)의 경구투여(經口投與)와 지실(志室).현종(懸鍾) 전침(電鍼) 병용(倂用) 시술(施術)이 흰 쥐의 골다공증(骨多孔症)에 미치는 영향(影響))

  • Objectives : This study was carried out to determine the effects of Gamidokwalgisaeng-tang(GDS) oral administration and electro-acupuncture at $BL_{52}$ & $GB_{39}$ on the postmenopausal osteoporosis. Methods: In order to study the effects of the Gamidokwalgisaeng-tang(GDS) oral administration and electro-acupuncture at $BL_{52}$, $GB_{39}$ on osteoporotic rats induced by ovariectomy, body weight, femur weight, uterine weight, serum level of osteocalcin, alkaline phosphatase(ALP), cholesterol, triglyceride, estradiol, androstenedione, testosterone and urinary excretions of calcium & phosphorus were measured. Results : 1. Body weight in all experimental group decreased significantly in comparison with ovariectomized group(OVX). 2. The weight of femur compared with body increased significantly in GDS oral administration and combination treatment group in comparison with ovariectomized group(OVX). 3. Uterine endometrial morphology is caused by ovariectomy and the weight of uterus decreased significantly in control and all experimental group in comparison with sham. 4. The serum level of osteocalcin decreased in all experimental group in comparison with OVX, but significantly in electro-acupuncture and combination treatment group. 5. The serum alkaline phosphatase(ALP) activity decreased in electro-acupuncture and combination treatment group in comparison with OVX, but only significantly in electro-acupuncture group. 6. The serum level of cholesterol increased significantly in OVX in comparison with sham and decreased in all experimental group in comparison with OVX, not insignificantly. The serum level of triglyceride increased in all experimental group in comparison with OVX, not insignificantly. 7. Urinary excretions of calcium and phosphorus decreased significantly in all experimental group in comparison with OVX. Conclusion : We suggest that GDS oral administration and electro-acupuncture at $BL_{52}$ & $GB_{39}$ are available for prevention and curing about the postmenopausal osteoporosis.

The Blood Chemical Study of the Oral administration of Guirokyisun-Hwan on the Osteoporosis Induced by Ovariectomy in the Rats (구록이선환(龜鹿二仙丸)의 경구투여(經口投與)가 난소적출(卵巢摘出)로 유발(誘發)된 백서(白鼠)의 골다공증(骨多孔症)에 미치는 혈액화학적(血液化學的) 연구(硏究))

  • In order to investigate the effect of the oral administration of Guirokyisun-Hwan on the estrogen-deficiency osteoporosis induced by ovariectomy in the rats, serum osteocalcin, estradiol, progesterone, follicular-stimulating hormone (FSH), calcium (Ca) and phosphorous (P) levels were monitored. And body weight changes were also monitored throughout whole experimental periods. The results were as follows : 1. Body weight of Control and Treatment group was significantly increased from 6 weeks after ovariectomy but after 2 days of drug administration, body weight of Treatment group were significantly (p<0.01 or 0.05) decreased compared with those of Control group. 2. Serum osteocalcin levels were significantly (p<0.01) increased in control groups compared with those of Sham group but significantly (p<0.01) decreased in the Treatment group. 3. Serum estradiol levels were significantly (p<0.01) decreased in control groups compared with those of Sham group but significantly (p<0.01) increased in the Treatment group. 4. Serum progesterone levels were significantly (p<0.01) decreased in control groups compared with those of Sham group but significantly (p<0.01) decreased in the Treatment groups. 5. Serum FSH levels were significantly (p<0.01) increased in control groups compared with those of Sham group but significantly (p<0.05) decreased in the Treatment group. 6. Serum Ca levels were significantly (p<0.01) decreased in control groups compared with those of Sham group but significantly (p<0.01) increased in the Treatment group. 7. Serum P levels were significantly (p<0.01) increased in control groups compared with those of Sham group and Treatment group. No significant was detected between Control group and Treatment group. According to these results, it is considered that the oral administration of Guirokyisun-Hwan have favorable effect on treatment and prevention of osteoporosis induced by ovariectomy in rats.
