A gene encoding the dextransucrase(dsCB) that synthesizes mostly $\alpha-(1\rightarrow6)$ linked dextran with low amount(10%) of $\alpha-(1\rightarrow3)$ branching was cloned and sequenced from Leuconostoc mesenteroides B-742CB. The 6.1 kbp DNA fragment carrying dsCB showed one open reading frame(ORF) composed of 4,536bp. The deduced amino acid sequence shows that it begins from the start codon(ATG) at position 698 of the cloned DNA fragment and extends to the termination condon(TAA) at position 5,223. The enzyme is consisted of 1,508 amino acids and has an calculated molecular mass of 168.6kDa. This calculated Mw was in good agreement with an activity band of 170kDa on non-denaturing SDS-PAGE. A recombinant E. coli DH5 $alpha$ harboring pDSCB produced extracellular dextransucrase in 2% sucrose medium, and synthesized both soluble and insoluble dextran. To compare the properties of enzyme with B-742CB dextransucrase, the acceptor reaction, hydrolysis of dextran and methylation were performed. The expressed enzyme showed the same properties as B-742CB dextransucrease, but its ability to synthesize $\alpha-(1\rightarrow3)$ branching was lower than that of B-742CB dextransucrase. In order to identify the critical amino acid residues known as conserved regions related to catalytic activity, Asp-492 was replaced with Asn. D492N resulted in a 1.6 fold decrease in specific activity.
The American bullfrog, Rana catesbeiana was imported from Japan for farming for the human consumption in 1970's and introduced populations were a great threat to native habitats in the pond and lake ecosystem. However, it is thought that the population of bullfrog has rapidly declined for past years in Korea. In this study, we investigated the intra-genetic diversity of R. catesbeiana habitated in Korea. The nucleotide sequences of 1,215bp mitochondrial ND1/tRNA region in bullfrogs sampled from 5 sites in Jeollanamdo were analyzed and compared to the original sequence of R. catesbeiara reported in Genbank. The nucleotide similarity between Korean and North American bullfrog was ranged from 98.7% to 100% based on kimura-2-parameter distance. In addition, bullfrogs analyzed in this study were clustered into two groups with one including Jangheung and the other including Gwangju populations in the neighbor-joining tree. North American R. catesbeiana was grouped in Jangheung cluster, indicating that there is the very low genetic difference between Korean and North American populations. The maximum parsimony tree in which North American R. catesbeiana was set as an outgroup suggests that Jangheung group represents the introduced population to Korea. Taken together, the results indicate that the population of R. catesbeiana in Korea has not segregated geographically yet, after the introduction.

Although GPS technology for location positioning system has been widely used, it is difficult to be used in intelligent transport systems, due to the large positioning error and limited area for receiving radio signals. Thanks to the rapid development of LED technology, LED lights become popular in many applications. Especially, visible light communications (VLC) has raised a lot of interests because of the simultaneous functioning of LED illumination and communication. Recent studies on positioning system using VLC mainly focused on indoor environments and still difficult to satisfy positioning accuracy and simple implementation simultaneously. In this paper, we propose a positioning system based on VLC using the coordinate information of LEDs installed on the road infrastructure. Extracting the LED signal, obtained through VLC, from the easily accessible camera image, it is possible to estimate the position of the car on the road. Simulation results show that the proposed scheme can achieve a high positioning accuracy of 1 m when large number of pixels is utilized and the distance from the LED light is close.

Researches that apply the acceleration signal due to user's gait measured at the wearable device to the authentication technology are being introduced recently. The gait acceleration signal based authentication technologies introduced so far have assumed that an attacker can obtain a user's gait acceleration signal only by attaching accelerometer directly to user's body. And the practical attack method for gait acceleration signal based authentication technology is mimic attack and it uses a person whose physical condition is similar to the victim or identifies the gait characteristics through the video of the gait of the victim. However, mimic attack is not effective and attack success rate is also very low, so it is not considered a serious threat. In this paper, we propose Video Gait attack as a new attack method for gait acceleration signal based authentication technology. It is possible to know the position of the wearable device from the user's gait video signal and generate a signal that is very similar to the accelerometer's signal using dynamic equation. We compare the user's gait acceleration signal and the signal that is calculated from video of user's gait and dynamic equation with experiment data collected from eight subjects.
Conservation and analysis of wall painting Goguryeo was performed to classify the unknown fragments. The conservation naked eye observation, optical microscopy, and infrared examination were carried out in order to figure out the structure, quality of constituting materials, and damages such as cracks, and discolored fragments of colored areas. Based on such investigation, conservation was proceeded. and it was completed with strengthening the weakened pigment layer of wall blocks. In addition tombs where the wall painting fragments were excavated were investigated by making comparison with gelatin dry plates and copies possessed by National Museum of Korea. According to the result, they were Kaemachong, Gosan-ri Tomb No.1 Gamsinchong, and Wonbong-ri Tomb. The components of colors with which Goguryeo wall painting fragments were painted and the mineral pigments of the wall layer were analyzed. Portable µ-XRF spectrometer and X-ray diffractometer were employed. It showed that lime (CaCO3) used for the wall layer, and the brown color is hematite(Fe2O3) and cerusite (PbCO3) and lead oxide(PbO) were identified. In the red color, cinnabar (HgS) were detected.

This study aims to think about the problem of dolmens: the reasons for the presence of dolmens and how to construct it, at the point of view of geographical and geomorphological. The subjects of this study is the dolmens which locate between at Dogok Hyosanri and Chunyang Daesinri Hwasoon-gun, Jeonlanamdo. The study areas in Hyosanri, Daesinri has been observed as follows. First, the long axis direction of Dolmen upper stone and the slope one are the same. Second, tor, block stream, hockey stick, etc. Third, Composition of the soil silt> sand> clay is distributed in the order. Forth, The soil of high quality silt and the roundness of angular, sub-angular-level and the high frequency, peaks of quarts and illite clay minerals show. Fifth, in the result of $SiO_2/Al_2O_3$, $SiO_2/R_2O_3$, and CIA(Chemical Index of Alteration), Hyosanri, Daesinri areas show mechanical weathering was dominant and chemical weathering environment was not being progressed. The blocks used in construction of dolmens had moved to the bottom of slopes by mass movement such as solifluction then them which had been placed in the position seem to be used by people of Bronze. Based on the above results, the process of construction of dolmens can be estimated as follows. They would dig up the ground under the upper stone of dolmens, put the supporting stone in the place, then dig up earth, place into remains, close the obturating stones, then heal up earth.
Since now, the study of wooden pagodas in ancient Korea have been proceeded to the site plan, the devices and the goods of sarari containers and the reconstruction. They are based on the result of archaeological excavations and the historical material documents. But the study of the foundations of wooden pagodas have been unsatisfactory. In this paper, the investigations of the wooden pagodas in the Three Kingdoms are proceeded. Through this action, we have checked the method of the foundations, the type and installation of Simchoseok(central base stone) and the erection of Simju(central base pillar). In the result of this study, three types were in the method of the foundations-the construction of the upper on the surface of the earth, the construction of the upper and lower on the surface of the earth. And we know the types of the installation of Simchoseok were the underground-type central base stone and the ground-type central base stoned. The factors of the central base pillars were changed with the rising of central base stones from the lower on the surface of the earth to the surface of the foundation. This change was needed to construct the larger wooden pagodas than the ones of the past. In especially, the large wooden pagodas with the ground-type central base stoned were appeared in the king of Baekje, Muwang(A.D. 600~640). We assume that the method of the construction of larger wooden pagodas like that was forwarded to Silla and Japan.
Reinforced concrete bridge decks are the first to be damaged by vehicle loads and rain infiltration. Concrete deterioration primarily occurs owing to the corrosion of rebars and other metal components by chlorides used for snow and ice melting. The structural condition and concrete deterioration of the bridge decks within the pavement were evaluated using ground-penetrating radar (GPR) survey data. To evaluate concrete deterioration in bridges, it is necessary to develop GPR data analysis techniques to accurately identify deteriorated locations and rebar positions. GPR exploration involves the acquisition of reflection and diffraction wave signals due to differences in radar wave propagation velocity in geotechnical media. Therefore, a full-waveform inversion (FWI) method was developed to evaluate the deterioration of reinforced concrete bridge decks by estimating the radar wave propagation velocity in geotechnical media using GPR data. Numerical experiments using a GPR velocity model confirmed the deterioration phenomena of bridge decks, such as concrete delamination and rebar corrosion, verifying the applicability of the developed technology. Moreover, using the synthetic GPR data, FWI facilitates the determination of rebar positions and concrete deterioration locations using inverted velocity images.

To investigate the exchange rates of mercury(Hg) across soil-air boundary, we undertook the measurements of Hg flux using gradient technique from a major waste reclamation site, Nan-Ji-Do. Based on these measurement data, we attempted to provide insights into various aspects of Hg exchange in a strongly polluted soil environment. According to our analysis, the study site turned out to be not only a major emission source area but also a major sink area. When these data were compared on hourly basis over a full day scale, large fluxes of emission and deposition centered on daytime periods relative to nighttime periods. However, when comparison of frequency with which emission or deposition occurs was made, there emerged a very contrasting pattern. While emission was dominant during nighttime periods, deposition was most favored during daytime periods. When similar comparison was made as a function of wind direction, it was noticed that there may be a major Hg source at easterly direction to bring out significant deposition of Hg in the study area. To account for the environmental conditions controlling the vertical direction of Hg exchange, we compared environmental conditions for both the whole data group and those observed from the wind direction of strong deposition events. Results of this analysis indicated that the concentrations of pollutant species varied sensitively enough to reflect the environmental conditions for each direction of exchange. When correlation analysis was applied to our data, results indicated that windspeed and ozone concentrations best reflected changes in the magnitudes of emission/deposition fluxes. The results of factor analysis also indicated the possibility that Hg emission of study area is temperature-driven process, while that of deposition is affected by a mixed effects of various factors including temperature, ozone, and non-methane HCs. If the computed emission rate is extrapolated to the whole study area we estimate that annual emission of Hg from the study area can amount to approximately 6kg.
In order to determine the origin and genetic diversity among chicken breeds, mitochondrial (mt) DNA D-loop sequences have been widely used. In this study, 41 individuals from four breeds (Korean native chicken (Black and Brown) and two imported breeds, Rhode Island Red and Cornish) were used for identifying genetic relationships with other chicken breeds. We obtained ten haplotypes and the highest number of haplotype was represented by eight individuals each from haplotype 1 and haplotype 2. Neighbor-joining phylogenetic tree indicates that the black and brown Korean native chicken breeds were mixed in haplotype 2 and they were closely related with the red jungle fowl (Gallus gallus). We also investigated whether the D-loop hypervariable region in chicken mtDNA can be used for the breed identification marker. The results indicated that the combination of the SNPs in the D-loop region can be possibly used for the breed discriminating markers. The results obtained in this study can be used for designing proper breeding and conservation strategies for Korean native chicken, as well as development of breed identification markers.
