Convenient Nucleic Acid Detection for Tomato spotted wilt virus: Virion Captured/RT-PCR (VC/RT-PCR) (Tomato spotted wilt virus를 위한 간편한 식물바이러스 핵산진단법: Virion Captured/RT-PCR (VC/RT-PCR))

  • Virion captured reverse transcription polymerase chain reaction (VC/RT-PCR) could detect plant virus quickly and accurately. In the VC/RT-PCR, no antibody is needed unlike immuno-captured RT-PCR (IC/RT-PCR) which had been improved method of RT-PCR for plant viruses, and virus nucleic acids can be obtained easily within 30minutes by property of polypropylene PCR tube which is hold and immobilized viral particles on its surface. For the virion capture of Tomato spotted wilt virus (TSWV), the extraction buffer was tested. The optimum macerating buffer for TSWV was 0.01M potassium phosphate buffer, pH 7.0, containing 0.5% sodium sulfite. The viral crude sap was incubated for 30 min at $4^{\circ}C$. The virions in the PCR tubes were washed two times with 0.01M PBS containing 0.05% Tween-20. The washed virions were treated at $95^{\circ}C$ immediately for 1 min containing RNase free water and chilled quickly in the ice. Disclosed virions' RNAs by heat treatment were used for RT-PCR. Dilution end point of $10^{-5}$ from plant's crude sap infected with TSWV showed relatively higher detection sensitivity for VC/RT-PCR. During multiple detection using two or more primers, interference was arisen by interactions between primer-primer and plant species. The result of multiplex RT-PCR was influenced by combinations of primers and the kind of plant, and the optimum extraction buffer for the multiplex detection by VC/RT-PCR should be developed.

No-Tillage Agriculture of Korean-Style on Recycled Ridge III. Changes in Pepper Growth and Biodiversity at Plastic Film Greenhouse Soil in Organic Cultivation of No-tillage Systems (두둑을 재활용한 한국형 무경운 농업 III. 시설 무경운 유기재배 고추의 생육 및 생물다양성의 변화)

  • Hot pepper growth in no-tillage cultivation on recycled ridge was increased by 22% compared with tillage cultivation. At 3 years after no-tillage cultivation, hot pepper growth was increased by 12% compared with tillage cultivation. Dry weight of unripe hot pepper at 2 years of no-tillage cultivation was 348.4 kg/10a increasing 16% compared with tillage cultivation while dry weight of unripe hot pepper was decreased at 3 years of no-tillage cultivation. Bacteria flora at 2 years of no-tillage cultivation was significantly increased compared with tillage cultivation. Bacteria flora was not significantly different at 3 years of no-tillage cultivation. Actinomyces flora at 2 years of no-tillage cultivation was significantly increased compared with tillage cultivation. Actinomyces flora was decreased at 3 years of no-tillage cultivation. Fungi flora at 2 and 3 years of no-tillage cultivation was increased by 1.3 and 1.7 times respectively, compared with tillage cultivation. Generation amount of carbon dioxide at no-tillage cultivation soil was remarkably decreased by 41% compared with tillage cultivation. Population of animalcule in early stage of hot pepper soil was 2 species and 6 individuals on Collembola and Acari at tillage cultivation. Population of animalcule in hot pepper soil was 5 species and 11 individuals including Chilopode at one year of no-tillage cultivation. Population of animalcule in hot pepper soil was 3 species and 5 individuals including Coleoptera and Chilopode at 2 years of no-tillage cultivation. Population of animalcule was 4 species and 40 individuals including Hypogastrurigae and 8 species and 97 individuals including Earwig (Labidura japornica) at 46 days after transplanting on tillage cultivation. Population of animalcule was 9~10 species and 101~107 individuals on no-tillage cultivation. Nature status for environmental change as index organism was 19 points and 33 points, at tillage and no-tillage cultivation, respectively. These results indicate that no-tillage agriculture of korean-style on recycled ridge plays a very important roles on pepper growth, biodiversity of animalcule, and greenhouse gases at plastic film greenhouse soil in no-tillage systems.

Accumulation of oxyresveratrol in Ramulus mori upon postharvest storage (숙성에 의한 뽕나무 상지 내 옥시레스베라트롤 축적)

  • Oxyresveratrol (trans-2, 3', 4, 5'-tetrahydroxystilbene), found in many plants including grape, peanut and mulberry, is a phytoalexin, an antimicrobial and antioxidative substance that rapidly accumulates in areas infected by the pathogen. We examined the accumulation of oxyresveratrol in nine Morus alba L. cultivars with respect to storage time and temperature postharvest and infection with GRAS microorganisms. Among the nine cultivars, the Suwon cultivar showed the highest oxyresveratrol content (9.6-fold increase) postharvest, when stored at $30^{\circ}C$ for 7 days. The optimal temperature and postharvest storage time for oxyresveratrol accumulation was $30^{\circ}C$ and 6 days. When Ramulus mori was infected with five microorganisms, the accumulation of oxyresveratrol increased over 4-fold in response to B. coagulans infection. These results suggest that oxyresveratrol accumulation is influenced by storage temperature, storage time, Ramulus mori cultivars, and microbial attack. Therefore, postharvest storage for an appropriate time period at a suitable temperature might be a useful way to industrially produce Ramulus mori cultivars with high oxyresveratrol content.

The Effect of Rain on Traffic Flows in Urban Freeway Basic Segments (기상조건에 따른 도시고속도로 교통류변화 분석)

  • An earlier study of the effect of rain found that the capacity of freeway systems was reduced, but did not address the effects of rain on the nature of traffic flows. Indeed, the substantial variation due to the intensity of adverse weather conditions is entirely rational so that its effects must be considered in freeway facility design. However, all of the data in Highway Capacity Manual(HCM) have come from ideal conditions. The primary objective of this study is to investigate the effect of rain on urban freeway traffic flows in Seoul. To do so, the relations between three key traffic variables(flow rates, speed, occupancy), their threshold values between congested and uncontested traffic flow regimes, and speed distribution were investigated. The traffic data from Olympic Expressway in Seoul were obtained from Imagine Detection System (Autoscope) with 30 seconds and 1 minute time periods. The slope of the regression line relating flow to occupancy in the uncongested regime decreases when it is raining. In essence, this result indicates that the average service flow rate (it may be interpreted as a capacity of freeway) is reduced as weather conditions deteriorate. The reduction is in the range between 10 and 20%, which agrees with the range proposed by 1994 US HCM. It is noteworthy that the service flow rates of inner lanes are relatively higher than those of other lanes. The average speed is also reduced in rainy day, but the flow-speed relationship and the threshold values of speed and occupancy (these are called critical speed and critical occupancy) are not very sensitive to the weather conditions.

Some Instances of Manchurian Naturalization and Settlement in Choson Dynasty (향화인의 조선 정착 사례 연구 - 여진 향화인을 중심으로 -)

  • In the late Koryo period, until 14th century, there had been at least two groups of Manchurians who were conferred citizenships; one group was living as an original inhabitant in the coastal area of north­eastern part of Korean peninsular, long time ago, and they were over one thousand households. The other was coming down from inland, eastern part of Yoha River, to the area of Tuman River to settle down and they were at least around one hundred and sixty households, including such tribes as Al-tha-ry, Ol-lyang-hap, Ol-jok-hap and others. They were treated courteously, from the early days of Choson dynasty, with governmental policies in an economic, political, and social ways. They were given, for instance, a house, a land, household furniture, and clothes. They were allowed to get marry with a native Korean to settle down. They were educated how to cultivate their lands. It was also possible for them to be given an official position politically or allowed to take a National Civil Official Examination. The fact they could take such an Examination, in particular, means they were treated fairly and equally, because they also had a privilege to improve their social positions through the formal system as much as common people. Two typical families were scrutinized, in this paper, family Chong-hae Lee and family Chon-ju Ju. All of them were successful to settle down with different backgrounds each other. The former were from a headman, Lee Jee-ran, who controlled his tribe, over five hundred households. He was given three titles of a meritorious retainer at the founding of Chosun dynasty, at the retrieval of armies, and an enshrined retainer. His son, Lee Wha-yong, was also given a vassal of merit who kept a close tie successfully with the king's family through a marriage. Upon the foundation of their ancestors, their grandsons, family Lee Hyo-yang and family Lee Hyo-gang, each, had taken solid root as an aristocratic Yang-ban class. The former became a high officer family, generation by generation, while the latter changed into a civil official family through Civil Official Examinations. They lived mainly around Seoul, Kyong-gi Province and some lived in their original places, Ham-kyong Province. Chu-man, the first ancestor, was given a meritorious retainer at the founding of the dynasty and Chu-in was also given a high officer position from the government. They kept living at the original place, Ham-heung, Ham-kyong Province, and then became an outstanding local family there. They began to pass the Civil Official Examinations. After 17th century on the passers were 17 in Civil Official Examinations and 40 were passed in lower civil examinations. The positions in government they attained usually were remonstrance which position was prohibited particularly to North­Western people at that time. The Chosun dynasty was open to Machurians widely through the system of envoy, convoy, and naturalization. It was intended to build up an enclosure policy through a friendly diplomatic relation with them against any possible invasion from outside. This is one reason why they were supported fully that much in a various way.

Media Scholars and Power: The politicized intellectuals hanging on the dangerous rope (언론학자와 권력: 정치화된 지성의 위험한 줄타기)

  • Media scholars take a lion stake in power circle. Not only do they take a part in media policies but seize prestigious positions like board members in Korea Communication Commission(KCC). Unfortunately, though, little has been known about who they are, what qualifications they have, and whether they meet public interests. This paper attempts to unveil the mechanism of those politicized intellectuals who are specialized on the media. Two categories divided into 'representative' and 'expertise' are employed for this purpose. On the one hand, the representative means the degree of committment into such public services as participation in conferences or non-profit organizations. On the other hand, the number of research articles, books and projects belong to the expertise. Evaluation levels consist of 'excellence, good and average' were allocated to those scholars who are(were) in 'Power Hole,' where decision makings come into being. Some interesting observations were made though this study. First of all, such criteria as representative and expertise vaguely suggested by the laws were hardly fit into those intellectuals, Rarely did they commit into public service let alone showing vigilance in academic activities. Secondly, both ideological loyalty and political activities in line with the government had much to do with taking such positions. Thirdly, not surprisingly, it showed that to graduate from Seoul National University and have Ph.D. degree from U.S.A. was one of the most essential factors. In final, most of them were very good at taking advantage of the press in way of boosting their publicity. To attend at policy making processes either in form of board members or advisers is inevitable for media experts. However, as shown in this study, such qualification of public service and academic eagerness shouldn't be underestimated. Academic integrity not selling intelligence solely for private interests needs to be protected as well. The authors hope this study to provide a valuable opportunity to establish a kind of ethical standards in participating into politics.

A Comparative Analysis of Masan's Democratic Movement : The 3·15 Uprising in 1960 and the 10·18 Buma Uprising in 1979. (마산의 민주화운동 비교 분석: 1960년 3·15의거와 1979년 10·18부마항쟁)

  • The purpose of this article is to bring about 10.26 accident by providing a fuse for the overthrow of the Park Chung-hee administration in the 10.18 Buma uprising. So, first of all, this article has a main purpose in analyzing the comparison between Masan's 3.15 uprising and 10.18 Buma uprising. The purpose of the study is to compare the 3.15 democratic movement in Masan with the 10.18 Buma uprising, given that the incident laid the foundation for the dictator to be overthrown in the event of an anti-dictatorial movement in Masan. The research method of this article is intended to be used as a research method in the 3.15 and 10.18 protests, given that if a person in power conducts election fraud or suppresses anti-government movements in order to maintain the system, it could lead to the destruction of those in power. In the end, the Masan 3.15 uprising and 10.18 uprising failed to reach a direct attempt to overthrow the regime due to unfinished democratic movements, but with the revolution of 4.19 and the massive political transformation of 10.26 Accident, he achieved the leading role theory that allowed the Rhee Syngman and Park Chung-hee administrations. In eradicating authoritarian regimes, however, the historic significance of the democratization movement was that the authoritarian regime eventually brought about the collapse of the regime by making a hard-line stance on election schemes or popular protests over the trap of power boomerang, which causes the regime to collapse.

Understanding of the Duplex Thrust System - Application to the Yeongwol Thrust System, Taebaeksan Zone, Okcheon Belt (듀플렉스트러스트시스템의이해 - 옥천대태백산지역영월트러스트시스템에의 적용)

  • The duplex system has been considered as an important slip-transfer mechanism to evaluate the evolution of orogenic belts. Duplexes are generally found in the hinterland portion of fold-thrust belts and accommodate large amounts of total shortening. Thus, understanding its geometric and kinematic evolution can give information to evaluate the evolution of the entire orogenic belt. Duplexes are recognized as closed-loop thrust traces on map view, indicating higher connectivity than imbricate fans. As originally defined, a duplex is an array of thrust horses which are surrounded by thrust faults including the floor and roof thrusts, and imbricate faults between them. Duplexes can accommodate regional layer-parallel shortening and transfer slip from a floor thrust to a roof thrust. However, an imbricate fault is not the only mean for layer-parallel shortening (LPS) and displacement transfer within duplexes. LPS cleavages and detachment folds can also play the same role. From this aspect, a duplex can be divided into three types; 1) fault duplex, 2) cleavage duplex and 3) fold duplex. Fault duplex can further be subdivided into the Boyer-type duplex, which was firstly designed duplex system in the 1980s that widely applied most of the major fold-thrust belts in the world, and connecting splay duplex, which has different time order in the emplacement of horses from those of the Boyer-type. On the contrary, the cleavage and fold duplexes are newly defined types based on some selected examples. In the Korean Peninsula, the Yeongwol area, the western part of the Taebaeksan Zone of the Okcheon Belt, gives an excellent natural laboratory to study the structural geometry and kinematics of the closed-loops by thrust fault traces in terms of a duplex system. In the previous study, the Yeongwol thrust system was interpreted by alternative duplex models; a Boyer-type hinterland-dipping duplex vs. a combination of major imbricate thrusts and their connecting splays. Although the high angled beds and thrusts as well as different stratigraphic packages within the horses of the Yeongwol duplex system may prefer the later complicate model, currently, we cannot choose one simple answer between the models because of the lack of direct field evidence and time information. Therefore, further researches on the structural field investigations and geochronological analyses in the Yeongwol and adjacent areas should be carried out to test the possibility of applying the fold and cleavage duplex models to the Yeongwol thrust system, and it will eventually provide clues to solve the enigma of formation and its evolution of the Okcheon Belt.

Anti-inflammatory Activity of Antimicrobial Peptide Protaetiamycine 2 Derived from the Protaetia brevitarsis seulensis (흰점박이꽃무지 유래 항균 펩타이드 프로테티아마이신 2의 항염증활성)

  • The white-spotted flower chafer Protaetia brevitarsis seulensis is a medicinally beneficial and important edible insect species. We previously performed an in silico analysis of the Protaetia brevitarsis seulensis transcriptome to identify putative antimicrobial peptides and then tested their antimicrobial and hemolytic activities. These peptides had potent antimicrobial activities against bacteria and yeast without inducing hemolysis. In the present study, the cationic antimicrobial peptide, protaetiamycine 2, was selected for further assessment of its anti-inflammatory properties in mouse macrophage Raw264.7 cells. Protaetiamycine 2 treatment of Raw264.7 cells suppressed LPS-induced nitric oxide production and reduced the expression of inducible nitric oxide synthase and cyclooxygenase-2, as determined by real-time PCR and western blotting. The expression of proinflammatory cytokines ($TNF-{\alpha}$, IL-6, and $IL-1{\beta}$) was also attenuated through the MAPKs and $NF-{\kappa}B$ signaling. We also confirmed that protaetiamycine 2 bound to bacterial cell membranes by a specific interaction with LPS. Collectively, these data obtained from LPS-induced Raw264.7 cells indicated that protaetiamycine 2 could have both antimicrobial and anti-inflammatory properties.

National brand development research strategy using traditional Korean patterns (한국 전통 문양을 활용한 국가 브랜드 연구 개발 전략 - 금문(錦紋)을 중심으로 -)

  • The present study is about a brand development strategy that utilizes Korean traditional patterns. The global culturenomics phenomenon confirms the value of the cultural aspects of design when a national brand is established. People succeed with their unique aesthetic sense and reinterpret it in a modern view, and this design acts as today's national brands. In this way, people used traditional patterns and original designs, along with regional characteristics and formative style, in developing their designs. However, due to recent changes in the global environment, modern people live in an "untact" world, and consumption culture shifted toward online marketing. In this environment, where one is isolated from social activities, there needs to be a strong image that can dramatically change the mood of one's home. I would like to re-examine the Dan-Chung pattern, whose painting depends on the aesthetic characteristics of architecture to protect the wooden members of the framework in traditional architecture. The pattern and color of Dan-Chung, coated in traditional architecture, differs by the type of construction used, which includes a palace, a Buddhist temple, and a Confucianism Dan Chung. The Geummoon pattern contains aesthetic factors to add solemnity to the Main Buddhist Halls, which contain Buddha. This is a new medium that continues the current traditions instead of remaining in the past. Among different Dan Chung patterns, Geummoon has magnificent decoration consisting of the highest grade materials and unique composition; therefore, it is suitable to be reinterpreted in modern terms. The same pattern can be interpreted in different ways with different colors, so there is a great aesthetic impression in the Geummoon pattern. The value of preservation for exploration and theoretical study of the traditional pattern is important, but recreating the pattern into modern formative art can present a new angle of view and national brand, bearing pride in our cultural assets. The study used multidimensional molding methods for realistic presentation after going through the two-dimensional design process. The significant value of Korean molding beauty which hangs onto the past will play a crucial role in establishing our national brand.
