Real-Time RT-PCR for Validation of Reovirus Type 3 Safety During the Manufacture of Mammalian Cell Culture-Derived Biopharmaceuticals (세포배양 유래 생물의약품 생산 공정에서 Reovirus Type 3 안전성 검증을 위한 Real-Time RT-PCR)

  • Validation of viral safety is essential in ensuring the safety of mammalian cell culture-derived biopharmaceuticals, because numerous adventitious viruses have been contaminated during the manufacture of the products. Mammalian cells are highly susceptible to Reovirus type 3 (Reo-3), and there are several reports of Reo-3 contamination during the manufacture of biopharmaceuticals. In order to establish the validation system for the Reo-3 safety, a real-time RT-PCR method was developed for quantitative detection of Reo-3 in cell lines, raw materials, manufacturing processes, and final products as well as Reo-3 clearance validation. Specific primers for amplification of Reo-3 RNA was selected, and Reo-3 RNA was quantified by use of SYBR Green I. The sensitivity of the assay was calculated to be $3.2{\times}10^0\;TCID_{50}/ml$. The real-time RT-PCR method was proven to be reproducible and very specific to Reo-3. The established real-time RT-PCR assay was successfully applied to the validation of Chinese hamster ovary (CHO) cell artificially infected with Reo-3. Reo-3 RNA could be quantified in CHO cell as well as culture supernatant. When the real-time RT-PCR assay was applied to the validation of virus removal during a virus filtration process, the result was similar to that of virus infectivity assay. Therefore, it was concluded that this rapid, specific, sensitive, and robust assay could replace infectivity assay for detection and clearance validation of Reo-3.

  • A 75-tonf-class LRE(liquid rocket engine) thrust chamber with ablative material for technology demonstration was manufactured on the basis of development technologies of 30-tonf-class LRE. Hydraulic characteristics of the thrust chamber were examined through cold flow test and ignition test of low flow condition. Test result showed that hydraulic function was good. Side ignition method with igniter ring also showed a fine function of ignition in operating ways of static condition. But a close review is required to understand the phenomena of generation and extinction of specific frequencies showed in dynamic characteristics ways. To achieve these, a large combustion test facility which is capable of performing combustion test at design condition of the 75-tonf-class thrust chamber should be constructed as soon as possible.

Design and Construction of Test Field for Low Voltage Under Cable Fault Location Detection (저압 지중케이블 고장 위치 검출 실증 시험장 설계 및 구축)

  • Various reflectometry methods to locate power cable fault have been studied. But, most related studies has been verifying by simulation and laboratory test and study in conditions similar with real cable fault filed was not performed due to the absence of cable fault test field. Therefore, this paper design and construct test field for the standardized performance test and the operating education of cable fault location equipments. In the constructed test, open, short, half open and poor contact fault at 100m, 200m location of cable was produced and 1km cable role was installed for maximum distance measurement test. The test field will be used in the development and standardization of cable fault location technology, and te performance evaluation and certificate test of the related equipments.

  • A new propeller open water testing device is developed and tested in a tow tank. The devised dynamometer consists of a torque measuring part, a thrust measuring part and a driving motor with an RPM counter. Torque is designed to be measured directly from the torsion of the structure holding the motor by using strain gauges and, consequently, conventional slip ring is removed. Also, in order to make the device fit in relatively small model ships, the weight and the size of the whole integrated structure are reduced in various ways. The developed tester is proved to be easily utilized in small circulating water channel experiments and is believed to have provided us with essential information for future design of various types of conventional or object oriented force measuring device.

Development and Verification of a Large Scale Resonant Column Testing System (대형 공진주시험기의 개발 및 검증)

  • In this study, a resonant column testing system which is the largest in Korea has been developed to evaluate the dynamic deformation characteristics of coarse granular geomaterials, and the performance and the applicability of the testing system have been verified. The system has been developed as a typical Stokoe type device whose boundary conditions are fixed bottom and free top with additional mass, and can adopt a large specimen with 200 mm in diameter and 400 mm in height. The driving and measurement instruments are configured as high performance and precision systems, hence the automated testing system is appropriate to drive enough stress and to measure the behavior precisely for the test in practical manner. The dynamic response of the mechanical components and the applicability of the system have been evaluated using metal specimens as well as polyurethane specimens, and its precision was verified by comparing its results with those from other equipment and/or methods. To confirm the applicability of the large system for coarse geomaterials, the resonant column test results from both large and normal scale apparatus for the same material were compared and it was found that the result can be partially affected by scale. Finally, the dynamic deformation characteristics of coarse geomaterial which is used for construction of large dam was evaluated using the large system and its practicality could be confirmed.

Method Validation for Monitoring of Agricultural Worker Exposure to Insecticide Fenthion (살충제 Fenthion에 대한 농작업자 노출 측정을 위한 분석/시험방법 검증)

  • Exposure measurement of agricultural worker to pesticide is one of important part of health risk assessment of pesticide. Therefore exposure matrices, apparatus, instruments and methods must be validated in advance to field experiment. In this study, method validation with an organophosphorus insecticide fenthion was carried out for exposure monitoring of agricultural worker. LOD and LOQ were 0.01 and 0.05 ng, respectively. Calibration curve linearity ($R^2$ > 0.999) and reproducibility (C.V. < 3%) were also excellent. Recovery at LOQ, 10LOQ and 100LOQ levels from gloves, socks, mask, patch, solid sorbent, glass fiber filter was 76~113% (C.V. < 3%). Trapping efficiency was 95~105% while no breakthrough was observed. Method validation for the exposure monitoring was established successfully through several experiments. Such method validation can be usually performed in laboratory and not much different for each pesticide so that, this techniques will be applied widely in research for pesticide exposure monitoring by combination with body surface area and respiration rates.

High Temperature Application of Iron Removal Chemical Cleaning Solvent in the Secondary Side of Nuclear Steam Generators (증기발생기 2차측 제철화학세정액의 고온적용)

  • A qualification test was performed for the iron removal chemical cleaning of the secondary side of nuclear steam generators at the selected temperature, 1$25^{\circ}C$, higher than the standard application temperature, 93$^{\circ}C$. The field cleaning condition for a nuclear unit was tested in a bench scale test loop including a SUS 316 stainless steel autoclave with one gallon capacity as a test vessel. The kinetics of sludge dissolution, corrosion of the secondary side materials and change of solvent chemistry were monitored. Test results indicated that more thorough cleaning was accomplished in less than half of the cleaning time required at 93$^{\circ}C$. And the total corrosions of the secondary side materials were found to be less than the values at 93$^{\circ}C$. While the solvent is recirculated and heated by an external chemical cleaning equipment for the conventional 93$^{\circ}C$ process, the secondary side is heated by the lateral heat of the primary coolant without the recirculation of the cleaning solution, and the solvent is mixed by vigorous boiling induced by periodic ventilation for the high temperature process. The requirement that the reactor coolant pumps should be running during the cleaning operation is the major disadvantage of the high temperature process which also should be considered when chemical cleaning is planned for steam generators under operation.

KAUSAT-5 Development and Verification based on 3U Cubesat Standard Platform (3U 큐브위성 표준 플랫폼에 기반한 한누리 5호 개발 및 검증)

  • The major objective of this study is to develop and verify the KAUSAT-5 based on the modular 3U CubeSat standard platform. In the mechanical system design of a 3U standard platform, subsystem and micro equipment functions/performance should be integrated and miniaturized on micro-sized PCBs and electrical capability was maximized to accommodate multiple payloads. KAUSAT-5 is 3U-sized Cubesat which will be operated in Low Earth Orbit(LEO), which implements mainly two scientific missions; one is to observe the Earth through infrared camera and the other is to measure space radiation with a Geiger Muller tube. An additional mission is to verify the equipment(device) such as VSCMG and fuzzy logic-based MPPT internally developed. The results of ETB, qualification and acceptance level environmental tests were shown to verify standard platform and KAUSAT-5 Cubesat.

  • In this paper, a case of a high-rise building supported by a large-diameter drilled shaft socketed in a weathered ground has been studied. The design case of pile foundation presented in this paper could be a representative example of execution of pile design through three(3) design stages of "preliminary design-proof test-supplementary design". As proof test, two(2) BDH PLT (Bi-directional High Pressure Pile Load Test) were performed at the early construction stage. By following the design process of this way, the economic pile design could be carried out and a large amount of construction cost saving could be realized.

  • A Fuel Feeding Unit of electric propulsion system has been developed for the small-satellite applications. The fuel feeding unit stores the xenon gas with high pressure and density as a fuel. Xenon can affect to system stability since xenon has the transient condition under the critical point which is in ambient temperature on the launch environment. The functional and structural stability on the launch environment needs verification through the ground tests. The design points and verification tests of the system were discussed and test results were described on this text. The system stability on the launch environment was verified through these verification tests.
