Germinability and Physiological Properties of Maize Seeds Affected by Matriconditioning (Matriconditioning에 의한 옥수수 종자 활력증진과 생리적 특성변화)

  • To observe the effects of seed matriconditioning on the emergence, plumule growth, and seed physiological properties of normal and aged seeds of three corn(Zea mays L.) genotypes (dent, sugary, and shrunken-2), normal and artificially aged seeds were matriconditioned at 50, 75, 100, and 125% moisture contents of vermiculite at $25^{\circ}C$ for 24 hours and the germinability was examined at $25^{\circ}C$ and in cold test (7 days at 1$0^{\circ}C$followed by $25^{\circ}C$). Emergence rate, emergence speed, and plumule growth were smaller in the order of dent>sugary>shrunken-2. Matriconditioning did not affect the emergence rate of normal dent and sugary seeds, while it enhanced emergence speed and plumule growth. In contrast, the emergence rate, emergence speed, and plumule growth of normal shrunken-2 and aged seeds of all genotypes were enhanced as the moisture content of vermiculite increased up to 125% both at $25^{\circ}C$ and in cold test. Matriconditioning promoted $\alpha$-amylase activity and DNA and soluble protein contents, while it reduced the leakage of total sugars and electrolytes from the seeds when soaked in water.

  • The leakage characteristics of electrical conductivity, inorganic constituents-K, Ca, Mg and Na, total sugar and total amino acid from the exudates of some vegetable seeds of viable and non-viable(artificially aged) were quantified to get basic information about the detection of the non-viable seeds. The crops studied were radish, cabbage, broccoli, onion and carrot. The time course electrolyte leakage was different from viable and non-viable seed of cruciferae but not sensitive in onion and carrot seed In inorganic constituents, potassium leakage was the greatest amount and difference between viable and non-viable seeds, but Ca, Mg and Na leakages were not as much differences as potassium. Total sugar as glucose and total amino acid as glycin leaked a lot more in aged radish, cabbage and broccoli seed than non-aged seed and the large differences were appeared after 4 hour imbibition. As a results, in general the leakages from the aged seeds were greater than from the non-aged seeds in most components tested but they were varied depending on species or varieties and components.

A Study on the Interannual Variation of Seedfall and Seed Viability of Acer pictum subsp. mono in Natural Broadleaved Forests at Mt. Jungwang (중왕산 천연활엽수림에서 고로쇠나무 낙하종자량 및 종자활력의 연년변이 연구)

  • To examine the interannual variation of seedfall and seed viability rates of Acer pictum subsp. mono (APSM), we investigated the annual seedfall and seed viability of APSM in the natural broadleaved forest in Mt. Jungwang, Gangwon-do, from 2009 to 2014. The amounts of annual seedfalls from 2009 to 2014 were revealed to be 5,700, 4,060, 60,900, 403,450 and 310 ea/ha respectively. 2013 is presumed to be the masting year since large amounts of seedfall were revealed, compared to that of the other years in the study period. Also, the amount of seeds was significantly different among seed viabilities. Most fallen seeds were damaged or decayed, accounting for 47.88% of the total, followed by empty seeds, undeveloped seeds and sound seeds in that order. The percentage of sound seeds ranged from 6.7 to 31.0%, which marks the lowest average with 16.74%. In the masting year, 2013, a significantly larger number of sound seeds fell at 125,070 ea/ha, on the forest floor than in the other years (320~17,840 ea/ha). Meanwhile, the ratio of damaged or decayed seeds was the lowest at 38%, although the number of those seeds was the largest, 153,310 ea/ha, for the same year. These results imply that the surplus seeds are supplied for natural regeneration in the masting year, supporting the seed predator satiation hypothesis. More long-term researches should be conducted on the seed production of broadleaved trees.

The Change of Peroxidase Activity in Soybean Seed Followed by Infection with Cercospora kikuchii (대두종자의 자반병 감염과 Peroxidase 활성도변화)

  • The present study was carried out to investigate the change of peroxidase activity of soybean seed infected with Cerrospora kikurhii. The protein content, polyphenol oxidase activity and peroxidase isozyme pattern in health and infected soybean seed were also compared. 1. The peroxidase activity was substantially higher in the infected soybean seeds than that in the healthy seeds either cracked or not. No significant differences in protein content were recognized among the seeds tested. 2. No significant differences in peroxidase activities and protein contents were notified between healthy and infected seeds from the measurements on each parts of dissected seeds, cotyledon and seedcoat, however the peroxidase activity in the seed coat of the stained seed was 2.5 times to healthy seed. 3. The activities of polyphenel oxidase were undectable in both healthy and diseased seeds. 4. The electrophoretic Patterns of the Peroridase isozyme were the same between healthy and in footed seed. 5. Therefore, the increase of peroxidase activity in infected soybean seedcoat was mainly due to the biochemical reaction against the pathogen.

Screening Method for Non-viable Seeds in Brassicaceae Vegetable Crops by Sinapine Leakage (Sinapine누출을 이용한 십자화과 채소의 퇴화종자 선별법)

  • A seed coating system was developed to screen non-viable seeds in the Brassicaceae. The crops studied were radish, chinese cabbage, broccoli, cauliflower and brussel sprout. Sinapine leaked more from artificially deteriorated seeds than non-deteriorated seeds. Seed coating with cellulose was to trap the sinapine leakage in the non-viable Brassicaceae vegetable seeds. The seeds were first hydrated, then coated with cellulose powder to capture the leakage. Coated seeds were dried, then sorted two fractions-fluorescent seeds and non-fluorescent seeds under the UV light. The ratio of the fluorescent seeds were higher in bad seedlot than good one. The germination rate were increased 3∼35% by eliminating the fluorescent seeds in tested Brassica vegetable seeds. Sowing non-fluorescent seeds resulted in a greater percent normal seedling than non-coated seeds in all seedlots. The fluorescent seeds contained a high percentage of the dead and abnormal seedlings.

Leakage of Sugars, Amino Acids and Protein from Differently - Aged Seeds of Sesame, Welsh Onion and Lettuce (퇴화처리에 따른 참깨, 파, 상추 종자의 당, 아미노산, 단백질 누출)

  • Experiments were conducted to obtain information on separation of nonviable seeds from seed lots by the nondestructive ways. Seeds of sesame, welsh onion and lettuce were artificially aged at 90% relative humidity and 45$^{\circ}C$ to get different seed qualities. The relationships between seed quality and leakage of total sugars, amino acids, and proteins into soaking water were determined to know a possibility of grading seeds. Dead seeds of lettuce leaked significant amounts of total sugars, amino acids, and proteins, while high quality seeds leaked negligeable amounts of total sugars and some of amino acids and proteins. Dead seeds of welsh onion leaked significant amounts of amino acids and some total sugars and proteins, while high quality seeds leaked negligeable amounts of these compounds. Sesame seeds leaked little total sugars, amino acids, and proteins regardless of seed quality.

Nondestructive Vigor Test of Seed (비파괴적 방법에 의한 종자의 활력 검정)

  • Experiments were conducted to obtain information on separation of nonviable seeds from seed lots of rape, Chinese cabbage and radish, by nondestrutive ways. Seeds were artificially aged at 90% relative humidity(RH) and 45$^{\circ}C$ to get different seed qualities. Large amounts of amino acids, proteins and sinapine were leaked from the dead seeds into water in the course of 4~8 hours soaking, while high quality seeds did not leak in all crops. Percentage of normal seedlings from nonfluorescent cellulose coated seeds of rape, Chinese cabbage and radish under ultraviolet light was 96, 96 and 74%, while that of fluorescent seeds was 8, 9 and 1%, respectively. Cellulose coating and storage of Chinese cabbage and rape seeds at 60 and 75% RH for 3 months and 90% RH for 2 months did not deteriorate seed quality. But percentage of normal seedlings from nonfluorescent radish seeds stored at 60, 75 and 90% RHs for 3 months was 63, 64 and 2%, respectively.

Sugar Leakage from Differently-aged Seeds of Rape, Chinese Cabbage and Radish (퇴화처리에 따리 유채, 배추, 무 종자의 당 누출)

  • Experiments were conducted to know the relationships between seed quality and leakage of sugars from differently-aged seeds of rape, Chinese cabbage and radish. Seeds were artificially aged at 90% relative humidity and 45$^{\circ}C$ to get different seed qualities. Significant amounts of total sugars were leaked from the dead seeds of all crops during the 8~24 hours soaking period, while high quality seeds leaked negligible amounts of total sugars. High quality seeds of all crops contained lots of sucrose and glucose and some stachyose, raffinose and fructose. During the artificial aging of seeds stachyose, raffinose, and sucrose decreased, while glucose and fructose increased.

  • This study were conducted to investigate the viability and germination of dehisced Panax ginseng seeds stored for long period in the storage chamber with $5^{\circ}C\;and\;30{\%}$ humudity. The staining reaction times for viability test were 150 min, 90 in, 60 min at $0.1{\%},\;0.5{\%}\;and\;1{\%}$ triphenyltetrazolim chloride(TTC), respectively. the more the storage period, the less the healthy seeds ratio. It was $96.6{\%},\;89.2{\%},\;63.4{\%}$ for 1-year storage, 7-years storage, 9-years storage, respectively. Germination ratio were $84.0{\%},\;80.5{\%},\;73.5{\%},\;2.5{\%}$ for 1-year stroage, 6-years storage, 7-years storage and 9-years storage, respectively. Therefore it was confirmed that ginseng seed can be stored up to 6-7 years.
