Biological antagonists of Phytophthora capsici were isolated from soil in Gyeongbuk, Korea. Among the isolated bacteria, a Bacillus sp. was identified from l6S rDNA sequence analysis and named Bacillus sp. AM-651. Bacillus sp. AM-65l strain which can strongly a antifungal activity against Phytophthora capsici. Culture conditions for the maximum production of the antagonistic substance were optimized. The production of antibiotic were high on modified Davis mineral medium pH 7 at $30^{\circ}C$. The medium for highest production of the agonistic substance optimized. It is composed the best activity on glucose, $(NH_4)_2SO_4$ and $K_2HPO_4$ at 0.5%, 0.1%, and 0.7%, respectively. By time course of culture solution selected Bacillus sp. AM-65l, the culture solution after 48hrs had strongly growth inhibition rate against P. capsici. And culture solution of Bacillus sp. AM-651 was stable within a pH range $5{\sim}11$ and temperature range $4{\sim}70^{\circ}C$. Bacillus sp. AM-651 cultured broth shown fungal growth inhibitory activity against B. sorokiniana, B. cinerea, R. solani avove and beyond P. capsici and comparatively showed a high activity against C. gloeosporioides, B. dothidea, B. cinerea and F. graminearum by agar diffusion method.
These studies were aimed to find out the representative values of physico-chemical properties in Korean soils and to serve the basic informations to improve the soil properties for increasing the soil productivity. By the results of detailed soil survey, 14 physico-chemical properties (soil texture, moisture characteristics, organic matter content etc) were collected from 315 series of soils except Cheju soils, computerized the simple mean and the distributed area by weighted mean values with grouping of land use, drainage class and soil type. The results were as follows; 1. Simple mean values within the efficient soil depth in whole country soils were clay 20.0%, organic matter 2.03% and CEC 10.3me/100g. But weighted mean values with land area belonged to the same soil series were clay 18.0%, OM 1.85% and CEC 8.6me/100g. 2. Mean values of paddy soils weighted by area were clay 19.7%, OM 2.0% and CEC 9.1me/100g but those of upland soils were 17.9%, 1.8% and 8.1me/100g and forest soils were 16.7%, 1.8% and 8.6me/100g respectively. 3. Weighted mean values of moderately well soils with covered area were clay 18.9%, organic matter 1.7%, CEC 8.4me/100g but those of imperfectly drained soils were 21.3%, 2.2% and 9.5me/100g, and those of poorly drained soils were 15.1%, 1.8% and 9.9me/100g respectivcly. 4. Simple mean and area weighted mean values of clay content, pH, organic matter contents, cation exchange capacity and base saturation were calculated by the types of paddy and upland.
Field experiments were conducted on the cropping system soybean-barley-soybean to investigate the effect of phosphorus and wollastonite application, effective application methods of phosphrus, and their residual effect in a typical Jeju island volcanic ash soil when the fertilizers were applied to the first crop of the three crops sequence. The results are summarized as follows. 1. Response of phosphorus was higher for barley than for soybean. The maximum application rate of phosphate was extimated to be 10-20kg/l0a for cultivation of soybean first crop) and 100kg/l0a for barley (second crop). Barley yield under 100kg/l0a of phosphate was double as compared to the no phosphate plot. 2. With application of 100kg/10a phosphate at the sowing time of soybean, the available phosphate in the soil after harvesting of soybean was increased to 175ppm but decreased to 60 ppm by cultivation of barley (second crop.) On the other hand, the phosphate absorption coefficient was 1,146mg/100g after soybean harvest but increased to 1,764mg/100g after barley harvest. 3. High residual effect was found in the plot to which phosphate applied with compost in the sowing band. 4. Response to wollastonite under heavy application of phosphate was high and residual effect was also great. 5. Response of soybean to wollastonite was higher than that of barley. Application of wollastonite increased the yield of all three crops soybean (first crop) by 30%, barley (second crop) by 12% and soybean (third) by 20%, respectively. Highest yield was obtained from the 1,000kg/10a wollastonite treatment for soybean and 500kg/l0a for barley.

An organic acid tolerance mutant (M-200) was obtained from Leuconostoc mesenteroides KCCM 35471, followed by the screening procedure using a specific organic acid medium (lactic acid: acetic acid, 2:1). The characteristics of the acid tolerance M-200 and the wild type LM-W were examined at various temperature and pH ranges $(l0-30^{\circ}C$ of temp, 3.5-4.5 of pH). The growth of strain M-200 at HCl adjusted medium $(10^{\circ}C\;and\;pH 3.5)$ was observed. In the case of organic acid adjusted medium, the strain showed its growth at the pH range of 3.8. When the strain M-200 was used as a starter for Kimchi fermentation, a constant acid level (0.55) was observed during the whole fermentation period. This result indicates that the strain produces a proper level of acid content for the Kimchi fermentation. This result also indicates that the edible period of Kimchi can be extended to 3.5 fold compare to the result obtained from the LM-W used Kimchi fermentation. However the excess use of the strain M-200 showed the inhibition of growth of Lactobacillus plantarum, low lactic acid level content and low level of organoleptic test. In the case of organic acid content during the Kimchi fermentation, the strain M-200 showed relatively low production rate compare to the wild type (M-200: 3.5 mg/L at 21 days of fermentation, LM-W: 7 mg/L at 21 days of fermentation). Therefore a mixed Kimchi starter containing M-200 and other strains probably maintain a good Kimchi quality during the fermentation.
The approval of use of certain food-grade phosphates as food additives in a wide variety of meat products greatly stimulated research on the applications of phosphates in foods. Phytic acid is a natural plant inositol hexaphosphate constituting 1-5% of most cereals, nuts, legumes, oil seeds, pollen, and spores. In this study, we investigated antibacterial activity of sodium phytate (SPT) against Salmonella typhimurium in tryptic soy broth with different pHs and in chicken, pork and beef. In tryptic soy broth, SPT at the concentrations of 0.1, 0.5, and 1.0% effectively inhibited the growth of Salmonella typhymurium in a concentration-dependent manner. At pH 5.5-7.0 similar to meat pHs, 1% SPT almost completely inhibited the bacterial growth. The inhibitory effect of SPT was stronger at pH 7.0 than pH 5.5. In chicken, pork, and beef, SPT at the concentrations of 0.1, 0.5, and 1% significantly inhibited the growth of Salmonella typhimurium in a dose-dependant manner (p<0.01). The addition of 1% SPT in the meats significantly increased the meat pHs. These results indicate that SPT is very effective for inhibition of bacterial growth as a muscle food additive for increasing food safety and functions.
Development of expression vectors is important for the basic and applied researches on kimchi LAB (lactic acid bacteria). An expression vector, pSJE6c was constructed by inserting P6C promoter sequence from Lactococcus lactis into pSJE, a shuttle vector for E. coli and Leuconostoc species. To test the efficiency of pSJE6c, aga ($\alpha$-galactosidase) and lacZ ($\beta$-galactosidase) genes were expressed in Lactobacillus brevis 2.14. Compared to the pSJE, expression levels of both genes were increased, indicating P6C promoter was better than indigenous promoters. Enzyme activities of L. brevis cells harboring pSJE6caga (pSJE6c with aga) or pSJE6Z (pSJE6c with lacZ) were 1.5-2 fold higher than those with pSJEaga (pSJE with aga) or pSJEZ (pSJE with lacZ). More RNA transcripts were detected in cells harboring pSJE6c based recombinant plasmid. The results indicated that heterologous gene expressions in kimchi LAB could be improved significantly by use of efficient expression vectors.
It is generally agreed that the country's population will grow up to the level of 52 million by the year of 2000 and that due the active growth of industry, urbanization and road constructions, sizable portion of existing arable lands will be utilized for other purposes than agriculture in near future. From 1966 to 1977, it was estimated that, the average annual conversion of arable lands to other uses, was 12,909 ha. If this trend persists, it is predicted that from 1978 to 1991 when the 6th Five Years Economic Development Plan will terminate, approximately 181,000 ha of arable lands will be converted for other uses again. On the other hand, it is certain that the increased population (39 million in 1981, 45 million in 1991, 52 million in 2001) and the changes in food pattern along with the enhancement of living standards will bring about the phenomenal increase in demands for not only the staple food but also the livestock products such as meat, milk and eggs, vegetables and fruits. These future increased demands for various foods, naturally mean the increased needs for the expansion of arable lands at the same time. It is predicted that, if more activities than present scale are not taken for the expansion of arable lands, the national food self sufficiency level will drop from 79% in 1977 down to 62% in 1991. To meet the increased food demands in future, there are several ways and means. These will include the increased land use intensity, elevation of unit area yield levels, minimization of conversion of arable lands to other uses and expansion of arable lands through the reclamations of idle hillside lands and tidal lands. Among these, the expansion of arable lands through reclamations of idle hillside lands and tidal lands are more positive measures to cope with the increased production of foods in future. The reclamation of hillside lands demands more attention because it needs more advanced technologies in agronomical and engineering aspects, larger scale fundings and integrated socioeconomic considerations. In agromical aspects, the thechniques for early improvement of chemical and physical properties of soils, proper soil conservation measures and rational cropping systems are of particular importance. As to the financial supports to encourage the farmings in hillside land, much bold fund inputs are essential for the construction of roads, installation of irrigation and drainage facilities, soil conservation mechanisms, which will ensure the stabilized farming with reasonable incomes in the newly reclaimed lands.
Rice yield and grain guality were investigated with treatments of silicate fertilizer, straw compost and NPK fertilizer in the paddy soil of silty clay loam from 1975 to 2001. Rice yield of the treatments of silicate fertilizer. $1,500kg\;ha^{-1}$ and straw compost $10,00kg\;ha^{-1}$ were increased by 15%, 10%, respectively, for 27years average yield in comparison with NPK plot. In the plots of NPK+silicate fertilizer $1,500kg\;ha^{-1}$, NPK+silicate fertilizer $2,500kg\;ha^{-1}$, taste value(Toyo instrument) of cooked rice and quality assessment were higher than those in NPK control plot and NPK+straw compost $10,000kg\;ha^{-1}$ plot, but the contents of crude protein, amylose of milled rice grain were lower than those in other treated plots. Nitrogen and phosphorous contents of rice plant were lower, but $SiO_2$ content were higher in the treatment of silicate ferti lizer 1,500, $2,500kg\;ha^{-1}$ than straw compost $10,000kg\;ha^{-1}$ plot. Rice quality assessment was positively correlated with taste value, but it was negatively correlated with crude protein and amylose content of milled rice grain. Ratios of Mg/N, $Mg/N{\cdot}K_2O$, $SiO_2/N$, $SiO_2/P_2O_5$ and $SiO_2/K_2O$ were positively correlated with taste value and quality assessment of rice grain, but negatively correlated with crude protein, amylose and fatty acid content. In soil chemical characteristics related with rice quality after experiment in 2001, soil pH and $SiO_2$ content were positively correlated with taste value and quality assessment of rice grain, but contents of soil $P_2O_5$, ex-K were negatively correlated with them.
Silkworm is known to be damaged under the unfavorable environmental conditions. Recently, it has been often argued that nicotine is a main factor to affect the silkworm nutritional balance and to result in decrease of quantitative characters, and unbalance of larval nutrition. Especially in spring season, there is tendency to produce nicotine damage to silkworm fed with the mulberry leaf near to tobacco field early cultivated in mulching system. In these connections, the trial was made in the different pots placing by the different distance 10rn, 20m, 40m, and 60m in the direction of east-north from the tobacco field. Location of pot near to the tobacco fold is 40m below the tobacco Held and gradually higher than the tobacco field by 10 degree of slope. In addition, when the mulberry field is divided into two areas from the east-south to west-north, southern east zone is lowering down below 10 degree of slope and has 36% of east-south wind, 21% of south-east wind and 16% of south wind, etc. Pots are left on the mulberry field from the completion of lower part leaf harvest to just before upper part leaf harvest is completed for 32 days. Then silkworm at 4th-5th stage was fed with the leaf grown during the period and larval characters are statistically analyzed. The results are summarized as follows: 1. 10-40m distant pot statistically at the same level has the longest larval duration and 60m distant pot is. not significant at 5% level. Whole larval duration has same tendency as in the 5th larval duration. 2. Percentage of delayed molting larvae at the 5th stage is increased in 10m distant pot and there is no significance statistically in other pots, as compared with that of control. 3. It is not showed that there is no damage to mortality of the old larvae in 60m distant pot. 4. 10-20m distant pot produces a low pupation and others are at the same level statistically at 5% significance, 5. It is showed that nicotine damage is occurred in single cocoon freight and cocoon shell weight up to 60m distant pot, rather higher damage in cocoon shell weight than in single cocoon weight. 6. It is showed that cocoon yield from 10, 000 molted larvae including double cocoon is decreased up to 40m distant pot far from the tobacco field in which 40% of cocoon yield in 10m distant pot by 7kg, 26% of cocoon yield in 20m distant pot by 3.9kg, 24% of cocoon yield in 40m distant pot by 3.6kg, and 12% of cocoon yield in 60m distant pot by 1.8kg, as compared with control. However, there is no significance statistically at 5% level in 60m distant pot. With these above results, it is concluded that nicotine da mage could be occurred in cocoon yield and cocoon quality up to 60m distant far from the tobacco field.
To design more efficient agricultural machinery, the accurately measuring system among many other factors is essential. A light-beam oscillographic recorder is generally used in measuring dynamic strain but it is not compatible with the extremely high speed measuring system such as 1,000 m/s, also is susceptable to damage due to vibration while using the system in field. The recorder used light sensitive paper for strip chart recording. The reading and analysis of data from the strip charts is very cumbersome, errorneous and time consuming. A microcomputer was interfaced with A/D converter, microcomputer program was developed for measuring, system calibration was done and the strain generated from a cantilever beam vibrator was measured. The results are summarized as follows. 1. Microcomputer program was developed to perform strain measuring of agricultural machine elements and could be controled freely the measuring intervals, no. of channels and no. of data. The maximum measuring speed was $62{\mu}s$. 2. Calibration the system was performed with triangle wave generated from a function generator and checked by an oscilloscope. The sampled data were processed using HP 3000 minicomputer of Chungnam National University computer center the graphical results were triangle same as input wave and so the system have been out of phase distorsion and amplitude distorsion. 3. The strain generated from a cantilever beam vibrator which has free vibration period of 0.019 second were measured by the system controlled to have l.0 ms of time interval and its computer output showing vibration curve which is well filted to theoretical value. 4. Using microcomputer on measuring the strain of agricultural machine elements could not only save analyzing time and recording papers but also get excellent adaptation to field experiment, especially in measurement requiring high speed and good precision.
