Hepatic Oxygen Free Radical and Alcolhol Metabolizing Enzyme Activities in Rats Fed Diets Supplemented with Lycium chinense Ethanol Extract (흰쥐에 있어서 구기자 추출물 첨가식이가 간조직의 유해산소 및 알콜대사 효소활성에 미치는 영향)

  • To investigate the oxygen free radical and alcohol metabolizing system in liver of rats fed diets with 30% ethanol extract of Lycium chinense (LCEE), Sprague-Dawley male rats weighing 225~235g have been fed a diet supplemented with 2% or 4% LCEE for a month. The rats fed LCEE supplemented diets gained less body weight compared with the control, and had no remarkable changes of liver function. In rats fed 2% LCEE supplemented diet, hepatic cytochrome P450 contents appeared to be increased, but catalase (204.88$\pm$20.06 $H_2O$$_2$nmoles/mg protein/min), and superoxide dismutase (13.18$\pm$0.74 Unit/mg protein) activities were significantly increased compared with control 120.28$\pm$26.99 $H_2O$$_2$nmoles/mg protein/min and 10.49$\pm$0.80 Units/mg protein). There was no difference in hepatic glutathione content, glutathione peroxidase, and glutathione-S-transferase ctivities between the rats fed LCEE suplemented diets and the control diet. On the other hand, hepatic alcohol dehydrogenase activity were not changed by LCEE feeding, but hepatic aldehyde dehydrogenase activities were significantly increased in rats fed both 2 and 4% LCEE diets(5.01$\pm$0.21 and 4.47$\pm$0.06 $\mu$moles NADPH/mg protein/min) compared with control (3.28$\pm$0.21 $\mu$moles NADPH/mg protein/min) and its Vmax value was 1.9 fold increased in rats fed 2% LCEE and 1.5 fold in those fed 4% LCEE compared with control. In conclusion, it is likely that rats receiving a diet supplemented with LCEE may have the oxygen free radicals and alcohol detoxication potential.

Serum cholinesterase activities and serum bromide concentrations in farmers (농업인에서의 체내 잔류농약 검출)

  • The usage or agricultural chemicals is on the increasing tendency. Methyl bromide and organophosphate are the most widely used toxic agricultural chemicals in Korea. We try to set up the methods to detect the accumulation of these chemicals in Korean farmers. Blood samples were collected for 121 farmers of slack's season in February 1998. And survey about arable acreage and usage of the gloves and masks was also performed. Serum cholinesterase activities and bromide concentrations were measured with gold chloride method and colorimetric method. The reference ranges of serum cholinesterase activity and bromide concentration were 1.6~15.9 U/mL and below $72.9{\mu}g/mL$. Serum bromide concentrations of farmers and normal controls showed no differences. Serum cholinesterase activities of farmers were significantly higher than those of normal controls. According to the arable acreages and usage of the gloves and masks, serum bromide concentrations and cholinesterase activities showed no differences. In conclusion, serum cholinesterase activities and bromide concentrations showed no differences between farmers of slack's season and normal controls.

  • Objective : The aim of this study was to rivew systemically literature and clinical trials in the treatment of urinary incontinence or lower urinary tract syndrome(LUTS). Methods : Computerized literature searches were carried out on two electronic database, and computerized searching on some korea oriental medicine journals in library of Kyung-Hee Medical center. Results : 1. Three reports of review study, six reports of experimental study and fourteen reports of clinical trials were collected and reviewed. Three reports of review study were all printed in the korea oriental medicine journal. From 2000, researches and studies have been increased in quantity and improved in quality. 2. Urinary disturbance include variable symptoms of lower urinary tract symptoms, urinary incontinence, in theaspect of Oriental medicine these symptoms are anurin, dysuria, urinary incontinence, nochumal enuresis, uracratia and so on. 3. Roughly physiological procedure of Acupuncture in Treatment of Urianry Disturbance may be that effect of acupuncture stimulation for parasympathetic nerve, sleep-arousal system in cerebrum, pontine/spinal urination center and pudendal/pelvic nerve affect bladder in expansion of bladder capacity, inhibition of urinary contraction and affection in periurethral muscle by continuous excitement of spinal annular circuit and synapse of neuron. 4. Clinical result for acupuncture treatment in urinary disturbance is summarized that acupuncture treatment in urianation disturbance of Neurogenic Bladder, Incontinence, Cycitis, Nocturnal Enuresis, Prostatitis/Pelvic Pain Syndrom and so on is significant clinical trials and technique. Conclusion : Hereafter, in the old age society these variable urinary disturbance patients are increased and desire of treatment may be also increased. So study of various and formal treatment and tecnnique is needed.

Effects of Omija (Schizandra chinensis Baillon) Concentrate on Blood Alcohol Clearance and Hepatoprotective Function in Rats Induced by Acute Ethanol Intoxication and Chronic Ethanol Treatment (오미자농축액이 급성주정중독량 및 장기간 주정을 투여한 흰쥐의 혈중 알코올 분해율과 간 기능 보호에 미치는 영향)

  • We investigated whether or not Schisandra chinensis (SC), a traditional herbal medicine, has protective effects against alcohol-induced fatty liver and blood alcohol clearance. Two tests focused on acute intoxication and chronic ethanol treatment were carried out. For the chronic ethanol treatment test, rats were fed ethanol by intragastric administration everyday for 8 weeks to induce alcoholic fatty liver. Ethanol treatment significantly increased blood alcohol concentration at 90 min after acute ethanol intoxication. Compared with the two ethanol-treated groups, rats administered ethanol along with SC extracts showed an approximately 13% increased blood alcohol clearance rate at 360 min. Chronic ethanol treatment significantly increased serum and hepatic triglyceride (TG) levels, and caused fatty degeneration of liver. Ethanol treatment also elevated the serum total-cholesterol (TC) level. However, after feeding of ethanol plus SC extracts, ethanol-induced elevation of hepatic TG levels reversed, whereas elevation of serum TG and TC levels was not observed after treatment with SC extracts. Ethanol treatment significantly increased ${\gamma}$-GT, aspartate aminotransferase, and alanine aminotransferase activities after 8 weeks. Compared with the ethanol-fed group, rats administered ethanol plus SC extracts for 4 weeks showed attenuated fatty degeneration as well as decreased hepatic function test values. SC administration also significantly increased intracellular lipid accumulation in hepatocytes and reduced steatosis score and hepatic TG levels, as measured by biochemical and histolopathological analyses. Our results indicate that the protective effects of SC are accompanied by a significant decrease in hepatic TG levels, thereby suggesting SC has the ability to prevent ethanol-induced fatty liver, by reducing hepatic TG and enzyme levels in alcoholic rats.

Efficacy of deferoxamine on paraquat poisoning (파라쿼트 중독 환자에서 deferoxamine의 치료 효과)

  • Background: Paraquat is known to induce oxidant injury that results in multiorgan failure and lung fibrosis. Iron has been considered to play a key role in paraquat-induced oxidant lung injury. This study examined the effect of deferoxamine, an iron-chelating agent, in the treatment of paraquat poisoning. Methods: From September, 2001 to April, 2005, 28 patients with paraquat poisoning who were admitted at a medical intensive care unit of a University-affiliated hospital, were enrolled in this study. Sixteen patients were treated according to the paraquat poisoning treatment guidelines and 12 received an intravenous infusion of deferoxamine in addition to the treatment guidelines. Results: There were no differences between the two groups in terms of age, gender, severity of paraquat poisoning, and the time elapsed from ingestion to presentation at hospital. There was no difference in overall mortality between the two groups but the incidence of respiratory failure in the deferoxamine group was higher than in the conventional group(4/7 versus 0/9, p=0.019). Conclusion: Deferoxamine seems to have no clinical benefit compared with the conventional treatment.

Effects of Aged Deodeok Extract on Serum Lipid Content in Rats Fed a High-Fat Diet (고지방식이를 섭취한 흰쥐에서 숙성더덕추출물이 혈청지질농도개선 효과에 미치는 영향)

  • This research was performed to investigate the effect of aged Deodeok (Codonopsis lanceolata) water extracts on the levels of lipids in the serum of rats fed a high-fat diet for 10 weeks. Experimental groups were divided into basal diet only (BDG), high fat diet control (HFDCG), high-fat diet and 10% aged deodeok extract powder (HF10S), and high-fat diet and 20% aged deodeok extract powder (HF20S) groups. The levels of hematological variables were not significantly different among the four groups. Compared with the control group's serum total cholesterol level of $339.38{\pm}4.06mg/dL$, the levels of the HF10S and HF20S groups were significantly lowered to $225.38{\pm}5.44$ and $215.02{\pm}4.77mg/dL$, respectively. Compared with the control group's LDL-cholesterol leve of $64.91{\pm}3.67mg/dL$, the LDL-cholesterol levels of the HF10S and HF20S groups were significantly lowered to $54.16{\pm}3.46$ and $46.14{\pm}1.79mg/dL$, respectively. Also, compared to the control group's serum triglyceride level of $103.07{\pm}13.2mg/dL$, the level of the HF20S group was significantly lowered to $48.25{\pm}11.52mg/dL$. These results suggested that dietary supplementation of aged deodeok extract does not have any adverse effect on the hematological variables, while improving the lipid content and reducing hepatic damage of the high-fat fed rats.

Drugs Most Frequently Used in OPD of Yeungnam University Hospital: March to August, 1985 (영남대학교 의과대학 부속병원 외래환자에 대한 약물처방 동향의 분석)

  • This report offers descriptive data about the drugs utilized in out patient department (OPD) of Yeungnam University Hospital (YUH) in the period of march to august in 1985. The data in this report were produced by the computerized totalization of the number of mentions of individual drugs included in the prescriptions. The 100 drug entries that were most frequently recorded are listed in rank order. The listing is arbiturarily restricted to the drugs that were prescribed as single preparations, the drugs of basis of compound preparations and the drugs of adjuvent or corrective of compound preparations that have significant therapeutic effects either by generic names. And in addition, the listing also involves the compound preparations used in relatively large frequency, and the individual components of which have the unique pharmacological actions each other by proprietary names. And all routes of administrations were allowed. The 10 drugs most frequently named are diazepam, aluminum compounds, acetaminophen, isoniazid, metoclopramide, $polaramine^{(R)}$, carboxymethylcystem, ephedrine, codeine and caroverine in order. The 521,855 drug mentions listed as above are described by the chief therapeutic usage that each is intended to apply generally. The drugs which account the largest proportion of total mentions were those acting on the central nervous system (20.57%), including tranquilhzers and sedative hypnotics (11.71%), analgesic antipyretics (5.55%), antidepressants (2.15%) etc. Gastrointestinal drugs and smooth muscle preparations (18.64%) included antacids and anti-ulcer drugs (9.24%), antiemetics (3.57%), spasmolytics (3.14%) and others. Respiratory drugs (16.11%) included expectorants and cough preparations (10.99%) and bronchodilators (5.12%). Chemotherapeutic agents (15.12%) included the antiTbc drugs (7.09%) most frequently, and the penicillins (3.33%) accounted the largest proportion among the antibiotics. Cardiovascular drugs (5.64%) included cardiac drugs and coronary vasodilator (4.12%) and antihypertensives and vasodilators (1.06%). And anti-inflammatory drugs (4.33%), vitamins of single preparations (3.76%), hormones and their antagonists (3.29%), common cold preparations (3.12%), diuretics (2.81%), drugs supporting liver function (2.02%), drugs affecting autonomic nervous system(1.89%) including anti-glaucomas, atropine and cerebral vasodilators, antihistamine drugs (1.02%) and disinfectants (0.74%) were following in order. The data in this report were compared to those reported by H. Koch, et al. in United States (US), 1981 as "Drugs Most Frequently Used in Office Practice:National Ambulatory Medical Case Survey, 1981." Cardiovascular drugs prescribed in YUH were much less in proportion than in US (10.56%), but gastrointestinal drugs accounted the larger proportion than in US (3.72%). Expectorants and cough preparations in YUH also accounted the larger proportion than in US (2.74%). In conclusion, in the period of march to august, 1985, OPD of YUH prescribed the CNS drugs including diazepam most frequently, and gastrointestinal, repiratory and chemotherapeutic drugs in next orders. It is supposed that the eating habits of Koreans and a unique atmospheric condition in Taegu as a basin were some important factors that affected the proportions of drugs acting on gastrointestinal and respiratory tracts.

Effects of Citrus Peel Ethanol Extract on the Serum Lipid and Body Fat of High-fat-diet-fed Rats (고지방식이를 섭취한 흰쥐에서 진피 에탄올추출물이 혈청지질 및 체지방에 미치는 영향)

  • In this work, the major flavonoid content of citrus peel ethanol extract and the changes in the serum lipid and body fat of high-fat-diet-fed rats were examined. The subjects were divided into four groups: the normal, control, O1 (0.1% citrus peel ethanol extract), and O5 (0.5% citrus peel ethanol extract) groups. The experimental groups (O1 and O5) were fed a high-fat diet mixed with 0.1 and 0.5% citrus peel ethanol extract, respectively, for 10 weeks. The results of the analysis showed that the main ingredients of the flavonoid in the citrus peel ethanol extract (60 brix) were $38.21{\pm}1.37$ mg/g hesperidin and $15.02{\pm}0.48$ mg/g narirutin. With regard to the changes in the serum total-cholesterol level, compared with the control group's $79.1{\pm}4.6$ mg/dL, the serum total-cholesterol levels of the O1 and O5 groups were significantly lowered to $64.6{\pm}5.4$ and $59.5{\pm}4.7$ mg/dL, respectively. As for the changes in the LDL-cholesterol level, compared with the control group's $18.4{\pm}3.7$ mg/dL, the LDL-cholesterol levels of the O1 and O5 groups were significantly lowered to $13.0{\pm}3.6$ and $13.0{\pm}1.9$ mg/dL, respectively. For the changes in the serum triglyceride level, compared with the control group's $74.5{\pm}13.2$ mg/dL, the serum triglyceride level of the O5 group was significantly lowered to $48.6{\pm}11.2$ mg/dL. Finally, with regard to the changes in body fat, compared with the control group, the 05 group showed a significant decrease. In conclusion, it was observed that citrus peel ethanol extract was effective in lowering the total-cholesterol, LDL-cholesterol, and triglyceride levels of high-fat-diet-fed rats.

Collection, Identification and Hepatic Effect of Native Cordyceps militaris (새로운 번데기 동충하초의 수집, 동정 및 간기능에 미치는 효과)

  • Entomopathogenic fungus Cordyceps militaris is famous for its medicinal efficacies. It has been reported to have various pharmacological activities such as anti-tumour, insecticidal, antibacterial, immunomodulatory and antioxidant. In this study, we investigated the effect of the extract of C. militaris (MPUN8501), which was identified by the analysis of the nucleotide sequences of 5.8S ribosomal RNA, on the function of liver. C. militaris powder was extracted using hot water extracts method as time, volume and temperature and using method as differential polarity of organic solvent. Each fraction was tested for the improvement of hepatic enzyme alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity. The BuOH extracts (CME) had highest activity which was used for the test of toxicity and efficacy of C. militaris. The enhancing effect of CME on the activity of ADH and ALDH was much more than medicine, drink, natural tea etc. Thus CME promoted the resolution of alcohol and acetaldehyde in rats, inducing recovery to normal condition rapidly. Furthermore, oral administration of CME effectively protected the carbon tetrachloride-induced acute hepatic injury as revealed by the hematological parameters (levels of sGOT and sGPT) and histological observation. CME was ascertained to be safe by regulatory toxicity studies of single dose toxicity and genotoxicity. These results suggest that CME would be useful for the maintaining normal hepatic activity as a functional health food.

Gas Permeation Properties of Sulfonated 6FDA-Based Polyimide Membranes Exchanged with Metal Ions (금속이온이 치환된 설폰화된 6FDA계 폴리이미드 막의 기체 투과 특성)

  • Sulfonic acid of the sulfonated 6FDA-based polyimides were exchanged with the monovalent ($Li^+$, $Na^+$, $K^+$) and divalent ($Mg^{2+}$, $Ca^{2+}$, $Ba^{2+}$) ions. The effect of metal cations exchanged sulfonated polyimides was investigated in terms of gas permeability and selectivity for $CO_2$, $O_2$ and $N_2$ gases. Thermogravimetric analysis showed that thermal stability of sulfonated polyimide was improved by exchanged metal cations. The permeabilities of monovalent cation-exchanged, sulfonated polyimide were reduced as the ion radius reduced [$Li^+$(0.059 nm)>$Na^+$(0.102 nm)>$K^+$(0.138 nm)], and those of divalent cations exchanged were determined by the ionic radii and electrostatic crosslinking between the polymer and metal cations, whereas the selectivities of all the metal cation-exchanged, sulfonated polyimides for $CO_2/N_2$ and $O_2/N_2$, were higher than those of sulfonated polyimide membranes. The sulfonated polyimide exchanged with the potassium cation showed the $O_2$ permeability of 89.98 Barrer [$1\times10^{-10}\;cm^3$(STP) $cm/cm^2{\cdot}s{\cdot}cmHg$] and the sulfonated polyimide exchanged with the lithium cation showed the $O_2/N_2$ selectivity of 12.9.
