식품중의 발암성 물질 -니트로소 화합물-

  • 니트로소디메틸아민이 발암성 물질이라는 것이 1954년 Barnes와 Magee에 의해 발견된 이래 이것이 지금까지 학자들에게 관심의 대상이 되고 있는 이유는 발암력이 강하여 극미량으로도 생체내에 암을 유발시킬 수 있을 뿐만 아니라, 대부분의 발암성 물질이 신체의 특정기관에 발암작용을 나타내는데 비해 이 물질은 신체의 여러 부위에 암을 유발시킬 수 있다는 점 그리고 이 물질이 햄, 소시지, 베이컨, 알코올음료, 김치 및 어류 가공품 등 어려 가지 식품에 널리 분포되어 있다는 사실 등을 들 수 있다. 니트로소 화합물은 구조로 보아 티트로소아민과 니트로소아미드로 구분되는데 전자는 제 2급 아연이 산화질소 유도체와 반응하여 생성된 니트로소 유도체이과 후자는 오소, 아미드 등이 치환된 니트로소 유도체로서 화학적인 성질이나 생물학적인 작용이 상이하다. 즉 니트로소아민은 식품에서 안정한 화합물인 반면에 대부분의 니트로소아미드는 불안정하다. 지금까지 연구된 바에 의하면 3백 여종의 니트로소 화합물에 대하여 동물실험을 행한 결과 발암성이 90% 이상 인정되었다. 식품 중 니트로소 화합물의 전구물질 중 질산염과 아질산염은 식품의 가공 저장 및 조리과정 중 니트로화의 된 전구물질인데 이는 육가공품의 색소고정, Cl. Botulinum에 의한 식중독 방지 및 풍미의 향상을 위하여 수 세기 동안 식품첨가제를 사용되어 왔으며, 유럽이나 미국 등지에서는 아직도 육가공품에 아질산염의 첨가가 논란의 대상이 되고 있다. 식품 중 니트로소 화합물에 대한 북유럽 식품 3천여 점을 분석한 결과 검출된 니트로소 화함물은 니트로소디메틸아민이 대부분이며 맥주에서 66%로 검출률이 가장 높았고 다음으로 염지육 및 치즈의 순 이었다. 조리한 일본산 해산 식품 중에 니트로소디메틸아민이 최고 313$\mu$g/kg, 캐나다산 해산 건조 식품에서는 67$\mu$g/kg, 홍콩산 염건어에는 1,400 $\mu$g/kg , 훈연어류에는 N-nitrosothiazolidine이 13,700 $\mu$g/kg , 우리 나라 해산 식품 중 니트로소디메틸아민은 건조가오리, 동결건조명태, 건조오징어, 굴비 및 소건새우 등에서 2.8~86.0 $\mu$g/kg 으로서 비교적 높은 양이 검출되었을 뿐 아니라 이들 식품을 조리할 경우 3.6~13배 증가하였다. 또한 김치와 젓갈류 중에서도 니트로소디메틸아민이 검출된다는 연구가 있다. 식품 중 니트로소 화합물의 생성율 억제시키기 위하여 최근 20여년간 연구된 바를 요약하면 아스코르브산과 같은 억제제의 첨가, 가공방법 및 조리방법의 개선이 비교적 바람직한 방법으로 인정되고 있다. 위의 방법을 적용하여 베이컨을 조리한 결과 낮은 온도에서 오랜 시간 가열하는 것이 높은 온도에서 짧은 시간 가열한 것보다 니트로화 반응이 훨씬 낮았고 또 마이크로웨이브로 조리하는 것이 니트로소아민을 최소화 시키는 방법이었다. 염지육은 아스코르브산이나 토코페롤 등의 니트로화 억제제를 첨가할 경우 니트로소 화합물이 현저히 감소 하였는데 이는 산화질소의 소거 능력이 우수하기 때문인 것으로 밝혀졌다. 가공방법의 개선으로서는 가공시 식품을 공기에 노출시킬 경우 특히 직화로 가열된 공기에 노출되면 니트로소아민의 생성이 매우 높은 것으로 보고되어 있는데 그 대표적인 예로 맥아를 직화로 건조할 경우 맥주 중에 니트로소 화합물이 훨씬 높은 양이 검출된다고 보고되어 있다. 대체로 식품의 가공 조리 및 저장 중 니트로소화합물에 대한 메커니즘은 상세히 밝혀져 있으나 생체내에서의 생성이나 억제 등에 대한 연구는 아직도 미흡한 실정이라이 분야에 대한 연구가 절실히 요구된다.

Minerals (Calcium, Iron, Zinc) Analysis and Interaction of Emphasized Nutrition Indication on Products (영양강조표시제품의 무기질 (칼슘, 철, 아연) 함량 분석과 상호작용에 대한 연구)

  • The present study aimed to calcium (Ca), iron (Fe) and zinc (Zn) contents were compared with reference values indicated on the nutrition label of nutrition emphasized food. Also, this paper will briefly review and evaluate what is currently known about Fe-Zn and Ca-Fe interaction. Ca, Fe or Zn-emphasized product, including cereal, snack, soy milk, chocolate product, other cocoa product, sugars product, beverage and tea were analyzed. The contents of Ca, Fe, and Zn in samples after dry-ashing were examined by ICP. The measured Ca (n = 42), Fe (n = 13), Zn (n = 6) values were ranged 87~176%, 98~167%, 98~275% of reference value indicated product, respectively. All samples were ranged more than 80% of the reference value, which were complied with food regulation. High intake of Ca has been shown to adversely affect Fe absorption and the negative effect of Fe on Zn absorption is well known. Therefore, it is important to consider Ca-Fe and Fe-Zn absorption interaction, when Ca, Fe or Zn-emphasized product produce and nutrition labeling policy should be managed regularly with continuous monitoring.

Determination of Isoflavone, Total Saponin, Dietary Fiber, Soy Oligosaccharides and Lecithins from Commercial Soy Products Based on the One Serving Size - Some bioactive compounds from commercialized soy products - (대두 가공품 1회분량 내 이소플라본, 사포닌, 식이섬유, 대두 올리고당 및 레시틴의 함량 - 상업용 대두 가공품 1회 분량 당의 생리활성 물질 함량 분석 -)

  • The levels of biologically active compounds, such as isoflavone, total saponin, dietary fiber, soy oligosaccharides, and lecithin from each serving size of commercial soy products, were quantitatively determined from the raw soybean, soymilk, tofu, isolated soy protein (ISP), soybean paste(toenjang), natto, and tempeh from local and foreign market. Soy flour, natto, and soymilk contained 489.1 mg, 308.3 mg, and 138.1 mg of isoflavone in each 100 g of dry matter, respectively. The ratios of aglycone to glucoside of soybean paste and tempeh showed relatively high level compared with other tested soy products. Commercial soymilk showed the highest ratio of soluble fiber to total dietary(59%). The higher levels of dietary fiber (20.1 g) and lecithin (1.13 g) were also found in tofu. The lecithin and saponin content of isolated soy protein(ISP) were highest (0.63 g and 0.65 g/ 100 g of dry matter) among the tested samples. In conclusion, soy flour showed the highest level of biologically active compounds, such as saponin, isoflavone, dietary fiber, and soy oligosaccharides. But when the evaluation was based on the serving size, soymilk containing 31.5 mg of isoflavone, 2.59 g of dietary fiber, 0.57 g of oligosaccharides, 0.10 g of lecithin, and 0.11 g of saponin showed similarity to those of the tested soybeans(20 g).

Growth Inhibition of Yeast Isolated from Processed Rice Cake with Ethanol and Organic Acids (에탄올과 유기산에 의한 가공쌀 부패효모의 생육저해 효과)

  • To defend putrifaction of the processed rice cake from gas-forming yeast during storage and distribution it needed to reduce and remove them. The sanitizers of ethanol and organic acids were applied on Pichia anomala, Candida tropicalis, and isolated yeasts from the putrified cut rice cake. Although growth inhibition effect by the sanitizer of 20% ethanol, 1% acetic acid, or 1% lactic acid respectively were very low, the combined sanitizer of 20% ethanol and 1% acetic acid, or 1% lactic acid showed very high sterilizing effect toward the yeasts. Six log cfu/ml of the yeast was reduced with this combined sanitizers for 30 minutes. In addition, the combined sanitizer heated from 20 to $50^{\circ}C$ had more the increased sterility. Therefore, the sanitizer of the combined ethanol with the acetic acid or the lactic acid for 30 minutes at $50^{\circ}C$ might reduce or sterilize the putrifying yeast at the processed rice cake. The result might be also applied to the effective pre-treatment of many agricultural food stuffs, against yeast, especially unsterilized stuffs, without any hazards from the special sanitizers and nutritional loss from harsh sterilization.

Exposure Assessment of N-nitrosamines in Foods (식품을 통한 니트로사민 노출량 평가)

  • In this study, contamination levels of 7 N-nitrosamine were investigated in processed meat products (n=51), processed fish products (n=62), salted fish pickles (n=20), and beer and malt beverages (n=21) using a GC/PCI-MS/MS method. The limits of detection (LOD) of the N-nitrosamines ranged from 0.2 to 0.5 ${\mu}g$/kg. In addition, methods were used to estimate the recovery of 7 N-nitrosamines, which ranged from 84 to 112%. N-nitrosamines were detected in 89 (58%) out of 154 samples. The exposure of an entire population group to N-nitrosamines through food intake was estimated using the average body weight of the total population and average daily food consumption, to perform risk assessment based on reports of a national health and nutrition survey. The results indicated that the daily intake of N-nitrosamines over a life time was $4.92{\times}10^{-7}$ mg/kg b.w./day. The margin of exposure (MOE) for the general population, estimated using the benchmark dose lower confidence limit 10 ($BMDL_{10}$) of N-nitrosodimethylamine, was 208,939, which was found to be safe.

Determination of polycyclic aromatic hydrocarbons in processed foods (가공식품 중 다환방향족탄화수소 분석)

  • In this study, the following concentrations of some PAHs (Polycyclic Aromatic Hydrocarbons) were investigated; [benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene, indeno(1,2,3-c,d)pyrene] in processed foods (n=165) and cooked meats (n=45) and established the analytical method by characteristics of processed foods. The methodology involved extraction (alkali digestion, liquid-liquid extraction, microwave extraction), clean-up on Sep-Pak Florisil Cartridges and determination by HPLC/FLD (High Performance Liquid Chromatography/Fluorescence Detector). The recovery of overall method for 8 PAHs spiked into these products ranged from 92 to 103%. The mean level of detected foods was found to be benzo(b)fluoranthene $0.9\;{\mu}g/kg$ in smoked salmon, benzo(b)fluoranthene $1.0\;{\mu}g/kg$, benzo(k)fluoranthene $0.3\;{\mu}g/kg$, benzo(a)pyrene $0.9\;{\mu}g/kg$ in dried banana, benzo(b)fluoranthene $0.2\;{\mu}g/kg$, benzo(k)fluoranthene $0.1\;{\mu}g/kg$, benzo(a)pyrene $0.2\;{\mu}g/kg$ in smoked chicken, benzo(b)fluoranthene $1.3\;{\mu}g/kg$, benzo(k)fluoranthene $0.3\;{\mu}g/kg$, benzo(a)pyrene $0.9\;{\mu}g/kg$ in charcoal grilled pork, respectively.

Survey of Contaminants of Bound 3-MCPD in Food (식품 중 결합형 3-MCPD 오염실태조사)

  • 3-MCPD created in manufacture process was regulated in our country about soy sauce and HVP. The latest paper reported that Bound 3-MCPD is created as intermediate. Germany common risk assesment reported that Bound 3-MCPD must be reduced because Bound 3-MCPD can be created in estimation circle when this is hydrolyzed in human body, but the data about the toxity of Bound 3-MCPD is lack. Therefore, We analysis about 209 items food such as soy sauce, seasoning food and meat-eating manufactured goods using bound 3-MCPD analysis method developed recently. As result of survey, bound 3-MCPD detected in 8 items among 44 traditional sauce (0.02~0.28ppm), 8 of soup 12 items (0.01~0.96ppm), in 22 items of sauce 60 items (0.01~0.55ppm), in 16 items of meat-eating manufactured foods 30 items (0.04~0.18ppm), in 20 items of snack cookies 28 items (0.09~1.43ppm), in 8 in roasted oil foods 10 items (0.04~1.22ppm), in 6 items of peanut processed food 10 items (0.06~0.25ppm), in 1 of vegetable cream 15 items (0.05ppm). Detected level was lower than the result of monitored by other countries.

Development of an Indirect Enzyme-Linked Immunosorbent assay for Rapid Detection of Adulteration of Food Allergen Mackerel in Processed Marine Foods (수산가공식품 중 알레르겐 고등어 혼입여부 신속 검출을 위한 간접효소면역분석법의 개발)

  • The purpose of this study was to develop an indirect enzyme-linked immunosorbent assay (indirect ELISA) based on a monoclonal antibody (MAb) that is specific to mackerel thermal stable-soluble protein (TSSP), that can be used for the rapid detection of mackerel in processed marine foods. Among the four MAbs (3A5-1, 2, 9, and 12) developed in previous studies, the 3A5-2 MAb that showed high specificity and sensitivity were selected and used to develop the indirect ELISA method. The detection range of the indirect ELISA was 0.02%-0.001% and the detection limit of 0.001% was shown. No cross-reaction to other marine products and food ingredients was observed by the indirect ELISA. Processed marine foods containing mackerel with ${\geq}0.3$ O.D. value at 405 nm were estimated as positive samples by the indirect ELISA. Therefore, the indirect ELISA can be used as a rapid and sensitive method to identify mackerel authenticity and adulteration in processed marine foods.

Characterization of Lipoxygenase in AOT/Isooctane Reversed Micelles (AOT/isooctane 역미셀계내에서의 lipoxygenase 반응 특성)

  • An investigation was carried out to study the characteristics of lipoxygenase in dioctyl sulfosuccinate (aerosol-OT, AOT)/isooctane revered micelles of microaqueous system containing infinitesimal water. ${\alpha}-Linoleic$ acid as a substrate could be analyzed by the colorimetric methodology using 5%(w/v) cupric acetate-pyridine solution and the activity of lipoxygenase was able to be assayed by the degree of ${\alpha}-linoleic$ acid consumption per minute. Optimal pH, temperature, and R-value ([water]/[AOT]) were determined as the value of 5.0, $25^{\circ}C$, and 10.0, respectively. Kinetic analysis of the enzyme reaction under the optimal conditions showed that the values of $K_m$ and $V_{max}$ were 0.31 mM of ${\alpha}-linoleic$ acid and $384.16{\mu}mol$ of ${\alpha}-linoleic$ acid decomposed/min, respectively. The results indicate the reaction to be lipoxygenase-catalyzed oxidation of ${\alpha}-linoleic$ acid in AOT/isooctane reversed micellar system. The inhibitory effect of natural antioxidants on lipoxygenase showed little inhibitory effect of L-ascrobic acid while ${\alpha}-tocopherol$ showed 72% of inhibitory effect.

Determination of Hydroxyproline Contents of Pork Meat for Quality Evaluation of Meat Products (돈육 등급별 Hydroxyproline 함량 분석법을 이용한 육가공품의 품질 평가)

  • This study was conducted to evaluate the feasibility of determination of hydroxyproline (Hyp) contents in pork meat for the quality evaluation of meat products. The relationship between the Hyp contents and pork grade was not high in porcine semitendinosus muscle. Whereas, Hyp contents of pork fresh ham and hamburger patties prepared with them were significantly increased as the decrease of pork grade. These results showed that the determination of Hyp contents might be a useful method for the quality evaluation of meat products.
