Expression of Inhibin in the Whole-body γ-irradiated Mouse Ovary (감마선이 조사된 미성숙 생쥐 난소 내 인히빈의 발현 변화)

  • Objective: The purposes of the present study were to investigate the effect of ${\gamma}$-radiation on the expression of inhibin-${\alpha}$ proteins and genes for inhibin ${\alpha}$, ${\beta}A$, and ${\beta}B$ in the ovary. Methods: Immature mice were whole-body ${\gamma}$-irradiated with 25% of a lethal dose. At time 0, 3, 6, 12, and 24 hours after the irradiation, the ovaries were collected and used for immunohistochemistry for inhibin-${\alpha}$, and RT_PCR for inhibin-${\alpha}$, ${\beta}A$, and ${\beta}B$. Results: The expression of the immunoreactive inhibins-${\alpha}$ was maintained at 12 hours post-irradiation and reduced thereafter. The expression of inhibin-${\alpha}$ mRNA was significantly increased with the time after the irradiation. However there were no significant changes in the expression of ${\beta}A$ and ${\beta}B$ mRNAs. Conclusion: It might be thought that inhibin acts as one of the regulatory factors in the ${\gamma}$-radiation-induced follicular atresia in mice

Studies on the Preparation of Polished ${\alpha}-Barley$ -I. The Yield and Degree of Gelatinization- (${\alpha}$화 보리쌀의 제조에 관한 연구 -제 1 보 : ${\alpha}$화 보리쌀의 수율과 호화도-)

  • Alpha-barley were prepared by means of soaking, heating, and drying treatment. The degree of gelatinization ${\alpha}-barley$ were determined. The yield of ${\alpha}-barley$ prepared by microwave heating, steaming, and autoclaving were in the range of 95-98%, whereas by boiling, only 74-89% of ${\alpha}-barley$ were obtained. The degree of gelatinization of ${\alpha}-barley$ prepared by 5 min microwave heating and 10 min and 20 min steaming were around 90%. Alpha-barley prepared by boiling, autoclaving and 30 min steaming showed above 95% of degree of gelatinization.

  • As the fuzzy control is applied to servo system, the hardware implementation of the fuzzy information systems requires the high speed operations, short real time control and the small size systems. The aims of this study is to develop hardware of the fuzzy information systems to be apply to servo system. In this paper, we propose a calculation method of approximate reasoning for fuzzy control based on $\alpha$ -level set decomposition of fuzzy sets by quantize $\alpha$ -cuts. This method can be easily implemented with analog hardware. The influence of quantization Bevels of $\alpha$-cuts on output from fuzzy inference engine is investigated. It is concluded that 4 quantization levels give sufficient result for fuzzy control performance of dc servo system. The hardware implementation of proposed operation method and of the defuzzification by gravity center method which is directly converted to PWM actuating signal is also presented. It is verified useful with experiment for dc servo system.

In Vitro Steroidogenesis on Oocyte Development in the Starry Flounder, Platichthys stellatus

  • In this study, oocyte steroidogenesis are investigated in relation to oocyte development in the starry flounder, Platichthys stellatus, a marine multiple spawner. Vitellogenic (0.52 and 0.55 mm oocyte diameter) and mature oocytes (0.63, 0.66 and 0.71 mm oocyte diameter) were incubated in vitro in the presence of $[^3H]17{\alpha}$-hydroxyprogesterone ($[^3H]17{\alpha}$-OHP) as a precursor. Steroid metabolites were extracted from the incubated media and oocytes, the extracts were separated and identified by thin-layer chromatography (TLC), high performance liquid chromatography (HPLC) and gas chromatographymass spectrometry (GC-MS). The major metabolites produced from $[^3H]17{\alpha}$-OHP were androgens [androstenedione ($A_4$) and testosterone (T)] and estrogens [$17{\beta}$-estradiol ($E_2$) and estrone ($E_1$)] and progestins [$17{\alpha},20{\alpha}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\alpha}P$) and $17{\alpha},20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}P$)] in vitellogenic and mature oocytes. The results from this study suggest the potential roles of $E_1$ in the oocytes with diameter 0.52-0.71 mm, $17{\alpha}20{\alpha}P$ and $17{\alpha}20{\beta}P$ at the oocytes of 0.63, 0.66 and 0.71 mm.

A study on the consumer preference according to consumer's need for uniqueness, ALPHA-NUMERIC brand name and types of fashion goods (소비자 독특성 욕구에 따른 알파누메릭 브랜드 네임 및 패션제품 유형의 선호도 연구)

  • The purpose of this study is to analyze the effects of consumer's need for uniqueness, whether or not using ALPHA-NUMERIC Brand Name and types of fashion goods upon the consumer's attitude. The experimental design of this study is three-ways complex factors design of 2 (Consumer's Need for Uniqueness : High vs Law) ${\times}$ 2 (Whether or not to use ALPHA-NUMERIC Brand Name : Used brand vs Non-used brand) ${\times}$ 2 (Types of fashion goods: Rational fashion goods vs Emotional fashion goods) The conclusions are as follows. 1. The interaction effect upon a favorite level according to consumer's need for uniqueness, whether or not using ALPHA-NUMERIC brand name and types of fashion goods upon the consumer's attitude is proven significant. For the group where customer's need for uniqueness is low, when ALPHA-NUMERIC brand name is not used, the favorableness of rational fashion good, a parka is lower than that of emotional fashion good, one-piece dress. However, there is no significant difference in favorableness by types of fashion goods when ALPHA-NUMERIC brand name is used. At the group with high need for uniqueness, while there is no significant difference in favorableness when ALPHA-NUMERIC brand name is not used, the favorableness of parka is higher than that of one-piece dress when ALPHA-NUMERIC brand name is used. 2. The interaction effects upon purchase intention according to consumer's need for uniqueness, whether or not using ALPHA-NUMERIC brand name and types of fashion goods are proven significant. For the group where customer's need for uniqueness is low, there is no significant different in the favorableness whether or not ALPHA-NUMERIC brand name is used. On the other hand, the group with high need for uniqueness, if ALPHA-NUMERIC brand name is used, the intention to purchase parka is higher than the intention to purchase one-piece dress.

PDTC Inhibits $TNF-{\alpha}-Induced$ Apoptosis in MC3T3E1 Cells

  • Osteoblasts are affected by TNF-${\alpha}$ overproduction by immune cells during inflammation. It has been suggested that functional $NF-{\kappa}B$ sites are involved in TNF-${\alpha}$-induced bone resorption. Thus, we explored the effect of pyrrolidine dithiocarbamate (PDTC), which potently blocks the activation of nuclear factor $(NF-{\kappa}B)$, on the induction of TNF-${\alpha}$-induced activation of JNK/SAPK, AP-1, cytochrome c, caspase and apoptosis in MC3T3E1 osteoblasts. Pretreatment of the cells with PDTC blocked TNF-${\alpha}$-induced $NF-{\kappa}B$ activation. TNF-${\alpha}$-induced activation of AP-1, another nuclear transcription factor, was suppressed by PDTC. The activation of c-Jun N-terminal kinase, implicated in the regulation of AP-1, was also down regulated by PDTC. TNF-${\alpha}$-induced apoptosis, release of cytochrome c and subsequent activation of caspase-3 were abolished by PDTC. TNF-${\alpha}$-induced apoptosis was partially blocked by Ac-DEVD-CHO, a caspase-3 inhibitor, suggesting that caspase-3 is involved in TNF-${\alpha}$-mediated signaling through $NF-{\kappa}B$ in MC3T3E1 osteoblasts. Thus, these results demonstrate that PDTC, has an inhibitory effect on TNF-${\alpha}$-mediated activation of JNK/SAPK, AP-1, cytochrome c release and subsequent caspase-3, leading to the inhibition of apoptosis. Our study may contribute to the treatment of TNF-${\alpha}$-associated immune and inflammatory diseases such as rheumatoid arthritis and periodontal diseases.

A Facile Synthesis of Disacharides Containing $\alpha$ -Altropyranosidic Linkage by 1, 2-trans Glycosidation of D-allal Derivatives (D-알알 유도체의 1, 2-trans 글리코시드 형성 반응을 이용한 $\alpha$ -알트로피라노시드 결합을 갖는 이당류의 효과적 합성법의 개발)

  • It's necessary to develope a facile methodology forming ${\alpha}$-altropyranosidic linkage, for the synthesis of trisaccharide repeating unit of O-antigenic part of Campylobacter jejuni gram negative bacteria. In this paper, the effective synthesis of disaccharides containing ${\alpha}$-altropyranosidic linkage by 1,2-trans glycosidation of allal derivatives was discussed. 4, 6-O-Benzylidene-3-O-(t-butyldimethylsilyl)-D-allal was treated with DMDO (3,3-dimethy ldioxirane) to yield 1,2-anhydro-4, 6-O-benzylidene-3-O-(t-butyldimethylsilyl)-${\beta}$-D-altropyranose. The reaction of 1,2-anhydro-4, 6-O-benzylidene-3-O-(t-butyldimethylsilyl)-${\beta}$-D-altropyranose with allyl alcohol gave allyl 4, 6-O-benzylidene-3-(t-butyldimethylsilyl)-${\alpha}$-D-altropyranoside quantitatively, and reactions with glucals were also successful to prepare ${\alpha}$-altropyranosodic disaccharides. It is convinced that 1,2-trans glycosidation of allal derivatives should be an attractive choice for preparing oligosaccharides containing ${\alpha}$-altropyranosidic linkages.

Determination of ${\alpha}-lactalbumin$ in Heated Milks by HPLC (HPLC에 의한 열처리된 우유중 ${\alpha}-lactalbumin$의 정량)

  • The ${\alpha}-lactalbumin({\alpha}-la)$ concentration in raw and laboratory-heated milks by HPLC was 1.20 mg/ml (unheated), 1.17 mg/ml ($63^{\circ}C$, 30min), 1.13 mg/ml ($72^{\circ}C$, 15sec) and 0 mg/ml ($100^{\circ}C$, 10min), respectively. Whereas, ${\alpha}-lactalbumin$ concentration ranges of commercial milks were $1.00{\sim}1.02\;mg/ml$ (pasteurized), $0.23{\sim}0.68\;mg/ml$ (UHT-pasteurized) and $0.77{\sim}0.89\;mg/ml$ (UHT-sterilized), respectively. It was supposed that the ${\alpha}-lactalbumin$ content of sterilized milk will be lower than that of UHT milk, but the opposite occurred. This discrepancy would be caused by the different heating system in the milk plants, where indirect UHT-treatment had more heat intensity than direct UHT-processing. The HPLC determination of ${\alpha}-lactalbumin$ may be an indicator to evaluate correctly and rapidly heated milks.
