Effect of Nitrogen Compounds and Organic Carbon Concentrations on $N_2O$ Emission during Denitrification (탈질에서 질소성분 및 유기탄소 농도가 $N_2O$ 배출에 미치는 영향)

  • The effects of the compounds and concentrations of nitrogenous electron acceptor, the ratio of electron donor/electron acceptor (C/N), and the complexity of electron donor on the emission of $N_2O$ during wastewater denitrification were quantitatively investigated in this study. The higher ${NO_3}^-$ and ${NO_2}^-$ concentrations, the more $N_2O$ emission was observed. ${NO_2}^-$ has strong effect on $N_2O$ emission as it emitted morc $N_2O$ than ${NO_3}^-$, 50 mg/L of ${NO_2}^-$-N gave the highest conversion (9.3%) and yield (9.8%) of $N_2O$ while ${NO_3}^-$-N (50 mg/L) gave 5.6% conversion and 11.0% yield. Lower C/N ratio decreases nitrogen removal efficiency, but it increases the conversion of $N_2O$ because of the incomplete denitrification by the limited organic carbon. When real domestic wastewater is used as the electron donor of the denitrification, $N_2O$ emission is reduced to 1/10 of the emission when single carbon (acetate) is used. It is thought that multiple carbon source utilizes many denitrification pathways and it seems to be helpful for the reduction of $N_2O$ emission.

Electrochemical properties of gel copolymer- electrolyte based on Phosphonium ionic liquid

  • Noble Poly (lithium 2-acrylamido-2-methyl propane sulfonate) and its copolymer with N-vinyl formamide based on trihexyl (tetradecyl) phosphonium acetate [$(C_6H_{13})_3$ P ($C_{14}H_{29}$) $CH_3COO$; $P_{66614}$ $CH_3COO$] and trihexyl (tetradecyl)phosphonium bis(trifluoromethane sulfonyl) amide ([$(C_6H_{13})_3P(C_{14}H_{29})$] [TFSA];$P_{66614}TFSA$) were prepared and analyzed to determine their characteristics and properties. The ionic conductivity of a copolymer based $P_{66614}TFSA$ ionic liquid system exhibits a higher conductivity ($8.9{\times}10^{-5}Scm^{-1}$) than that of a copolymer based $P_{66614}CH_3COO$ system ($1.57{\times}10^{-5}Scm^{-1})$. The charge on the TFSA anion is spread very diffusely through the S-N-S core and particularly in the trifluoromethane groups, and this diffusion results in a decreased interaction between the cation and the anion. The viscosity of $P_{66614}TFSA$ (39 cP at 343 K) and $P_{66614}CH_3COO$ (124 cP at 343 K), which is very hydrophobic, was fairly high. High viscosity leads to a slow rate of diffusion of redox species. The ionic conductivity of copolymer of a phosphonium ionic liquid system also exhibits higher conductivity than that of a homopolymer system. Phosphonium ionic liquids were thermally stable at temperatures up to $400^{\circ}C$.

The Effects of Propolis on Biochemical Parameters and Activity of Antioxidant Enzymes in Broilers Exposed to Lead-Induced Oxidative Stress

  • This study aimed to determine the effects of vitamin C and propolis-supplemented feeds on some blood parameters, lipid peroxidation, and activities of some antioxidant enzymes in broilers exposed to oxidative stress. 360 three-day-old broiler chicks (Ross 308) were randomly divided into four treatment groups each containing 90 animals, including six replicate groups for each treatment. The experimental groups were designated for a 3-42 days period as follows: no supplement to basal ration (Control-Group I); supplement of 500 ppm vitamin C and 200 ppm lead (as lead acetate) to basal ration (Group II); supplement of 1 g/kg propolis and 200 ppm lead (as lead acetate) to basal ration (Group III); and supplement of 200 ppm lead (as lead acetate) to basal ration (Group IV). The highest TG level (86.83 mg/dl) was observed in the lead supplemented group; however, the lowest aspartate aminotransferase (SGOT) level (90.71 IU/L) was observed in the control group (p<0.05). The addition of lead increased the plasma malondialdehyde (MDA) level (p<0.01) compared to other treatments. However, the addition of vitamin C and propolis decreased the plasma MDA level close to control levels. The highest erythrocyte superoxide dismutase (SOD) activity was observed in the lead addition group (p<0.01) while no significant differences were observed for SOD activities of the control, vitamin C +lead, and propolis+lead groups. The plasma reduced glutathione (GSH) activity of the control ($2.30{\mu}mol$/ml) was significantly lower than the lead administered group ($6.20{\mu}mol$/ml) (p<0.01); while this parameter was determined to be similar to other groups. No significant differences were observed between groups for liver GSH activity, but heart GSH activity of the control was significantly higher in comparison to other treatments (p<0.05). To obtain similar antioxidant effects, it is recommend that using propolis (1 g/kg) and vitamin C (500 mg/kg) supplementation in broiler diets may overcome the adverse effects of oxidative stress originating from dietary lead.

  • Properties of purified isocitrate lyase from Saccharomycopsis lipolytica ATCC 44601 and MX9-11RX8 temperature-sensitive mutant were investigated. Purified isocitrate lyase from temperature-sensitive mutant was indistinguishable from the wild type enzyme with respect to the isoelectric pH(5.3), the thermostability and Km value for threo-Ds-Isocitrate(about 0.2 mM). When isocitrate lyase induced by acetate minimal medium at 33$^{\circ}C$, MX9-11RX8 mutant did not express enzyme activity but did synthesize polypeptide chain whose electrophoretic mobilities were equal to those of the purified enzymes.

  • The deterioration of cellulosic materials due to the growth of mildew causes enormous loss to us. It is, therefore, necessary to give adequate protectional treatment against mildew spoilage on cellulosic materials. In this study, fourteen chemicals were treated to the strain of Chaetomium globosum A.T.C.C., then phenyl mercuric acetate (P.M.A) was proved to be a most effective fungicide out of various chemicals. Another chemicals, such as Na-pentachlorophenolate, tuget, $\alpha$-naphthol, caprylic acid and orthocidc were also proved to be effective mildew-proofing agents.

Kinetic Study of Thermolysin-Catalyzed Synthesis of N-(Benzyloxycarbonyl)-L-Phenylalanyl-L-Leucine Ethyl Ester in an Ethyl Acetate Saturated Aqueous System

  • The kinetics of the thermolysin-catalyzed synthesis of N-(benzyloxycarbonyl)-L-phenylalanyl-L-leucine ethyl ester (Z-Phe-LeuOEt) from N-(benzyloxycarbonyl)-L-phyenylalanine (Z-Phe) and L-leucine ethyl ester (LeuOEt) in an ethyl acetate saturated aqueous system in a batch operation were studied. The kinetics for the synthesis of Z-Phe-LeuOEt were expressed using a rate equation for the rapid equilibrium random bireactant mechanism. The four kinetic constants involved in the rate equation were determined numerically by the quasi-Newton method so as to fit the calculated results with the experimental data. Within the pH and temperature range examined, the $K_{cat}$ value for the synthesis of Z-Phe-LeuOEt reached a maximum at pH 7.0 and $45^{\circ}C$, whereas the affinity between Z-Phe and thermolysin reached a maximum at pH 6.0 adn $40^{\circ}C$. The inhibitory effect of Z-Phe on the condensation reaction decreased as the pH and temperature decreased. In contrast, they affinity between LeuOEt and thermolysin remained unchanged within the pH and temperature range examined. Therefore, it was concluded that the protonation state of the carboxyl groups. of Z-Phe was more imprtant than that of the amono groups of LeuOEt for the synthesis of Z-Phe-LeuOEt in the present solvent system. The equilibrium yield at pH 6.0 and $30^{\circ}C$ was 8% higher than that at pH 7.0 and $40^{\circ}C$, although the rate was much slower. This result suggested that the affinity between the enzyme and the substrate rather than the overall rate was a more important factor affecting the equilibrium yield, when the peptide synthesis was carried out in a product-precipitation system.

Characteristics of c-axis oriented sol-gel derived ZnO films (C-축으로 정렬된 sol-gel ZnO 박막의 특성)

  • ZnO films were fabricated on p-type Si(100) wafer ITO glass and quartz glass by the sol-gel process using zinc acetate dihydrate as starting material. A homogeneous and stable solution was prepared by dissolving the zinc acetate dihydrate in a solution of 2-methoxyethanol and monoethanolamine (MEA). ZnO films were deposited by spin-coating at 2800 rpm for 25 s and were dried on a hot plate at $250^{\circ}C$ for 10 min. Crystallization of the films was carried out at $400^{\circ}C$~$800^{\circ}C$ for 1 h in air. X-ray diffraction (XRD) analysis, scanning electron microscopy (SEM), UV-vis transmittance spectroscopy, FTIR transmittance spectroscopy and Photoluminescence (PL) spectroscopy measurements have been used to study the structural and optical properties of the films. ZnO films highly oriented along the (002)plane were obtained. In all cases the films were found to be transparent (above 70%) in visible range with a sharp absorption edge at wavelengths of about 380nm, which is very close to the intrinsic band-gap of ZnO(3.2 eV). The low temperature band-edge photoluminescence revealed a complicated multi-line structure in terms of bound exciton complexes and the phonon replicas.

Supplementation Effects of $C_{18:2}$ or $C_{18:3}$ Rich-oils on Formations of CLA and TVA, and Lipogenesis in Adipose Tissues of Sheep

  • The present study was conducted to investigate the supplementation effects of $C_{18:2}$ rich-soybean oil or $C_{18:3}$ rich-perilla oil (7% of total diet, DM basis) for 12 weeks on plasma metabolites, fatty acid profile, in vitro lipogenesis, and activities of LPL and FAS in adipose tissue of sheep. The treatments were basal diet (Control), $C_{18:2}$ rich-soybean oil supplemented diet (SO-D) and $C_{18:3}$ rich-perilla oil supplemented diet (PO-D). All the sheep were fed the diets consisting of roughage to concentrate in the ratio of 40:60 (DM basis). Oil supplemented diets (SO-D and PO-D) slightly increased contents of triglyceride (TG) and total cholesterol (TC), proportions of both cis-9 trans-11 and trans-10 cis-12 CLA and TVA, but lowered (p<0.01) those of $C_{18:0}$ compared to the control diet. No differences were observed in the contents of TG and TC and proportions of fatty acids in plasma between supplemented oils. Oil supplemented diets slightly increased the proportions of cis-9 trans-11 and trans-10 cis-12 types of CLA in subcutaneous adipose tissue of sheep compared to the control diet. The rate of lipogenesis with acetate was higher (p<0.01) for intermuscular- and subcutaneous adipose tissues than that for intramuscular adipose tissue, while that with glucose did not differ among fat locations in sheep fed SO-D. No differences were observed in the rate of lipogenesis between substrates in all fat locations. The rates of lipogenesis with glucose increased only in the intermuscular- (p<0.01) and subcutaneous adipose tissue (p<0.005) compared to those with acetate. The rates of lipogenesis with acetate were the highest in the intermuscular and intramuscular adipose tissue of the sheep fed PO-D. Oil supplemented diets slightly increased the rate of lipogenesis with glucose for all fat locations. Supplementation of oils to the diet numerically increased the fatty acid synthase activity but did not affect the lipoprotein lipase activity in subcutaneous adipose tissue.

HPLC Method for the Determination of Nicorandil in Human Plasma

  • The present study is to determine of sensitive nicorandil analysis method using HPLC and measure the pharmacokinetics parameters (bioavailability, $C_{max}$, $T_{max}$, Ke, $T_{1/2}$) of nicorandil (5 mg, Tab; Choongwae Pharma Corporation). Plasma (500 ul) was mixed with furosemide (internal standard, 500 ug/ml). Detection wavelength was 256 nm. The mixture of 0.01 M ammonium acetate and acetonitrile 80:20 (v/v) was used mobile phase. The HPLC separation was accomplished on ODC reverse HPLC column. The nicorandil was analyzed by a HPLC system, which consists of CAPCELL PAK C18 column (5 ${\mu}$m, 4.6 × 150 mm) and a chromatography data analysis S/W, using a isocratic mobile phase (mixture of 0.01 M ammonium acetate and acetonitrile 80:20 ) at 1.0 ml/min. Its sensitivity, selectivity, accuracy and precision must be adequate for the bioavailabilty study of nicorandil, and the linearity ($r^2$ ≥ 0.9994) of nicorandil was also proved in the range of 0.05 ug/ml . 3 ug/ml. The pharmacokinetic parameters of nicorandil (5 mg) tablets were measured as the follow. AUC: 0.19 ug/ml·hr, $C_{max}$: 0.14 ug/ml, $t_{max}$: 0.58 hr, Ke: 0.11 hr., $t_{1/2\beta}$: 6.76 hrs. This method is simple and sensitive HPLC method using UV detector for determination of nicorandil in human plasma.

Effects of Different Fatty Acids and Levels on the Lipogenesis Capacity and Lipolysis Rate of Broilers In Vitro

  • This study investigated the lipogenesis capacity of hepatocytes and lipolysis rate of adipocytes of broilers as affected by different fatty acids (trial one) and different linoleic acid (C18:2) levels (trial two). Twenty 6-wk old broilers were used; their hepatocytes and adipocytes were isolated for the in vitro study. In trial one, four treatments were tested. The control group in which no fatty acid was added, and the test groups to which were added $300{\mu}M$ of C16:0, C18:1 and C18:2, respectively. For trial two, different levels (0, $300{\mu}M$ and 1 mM) of C18:2 combined to fatty acid-free bovine serum albumin (BSA) were added to the medium. According to results of trial one, added fatty acids significantly reduced the incorporation by hepatocytes of [U,$^{14}C$]glucose into total lipid (p<0.05); the lipogenesis capacity in C18:2 group was the lowest. Although a similar pattern was found with [l,$^{14}C$]acetate, the groups only slightly differed in terms of lipogenesis capacity (p=0.11). In addition, the C18:2 group had a significantly (p<0.05) greater lipolysis rate than the C16:0 and control groups. Results of trial two indicated that C18:2 significantly (p<0.05) reduced lipogenesis capacity both for [U,$^{14}C$]glucose and [l,$^{14}C$]acetate, and markedly stimulated the lipolysis rate (p<0.05), displaying a dose response. Results presented herein demonstrate that C18:2 can reduce lipogenesis capacity and stimulate the lipolysis rate in broilers.
