Controlled Release of Cyclosporin A from Liposomes-in-Microspheres as an Oral Delivery System

  • The aim of this study was to prepare cyclosporin A-loaded liposome (CyA-Lip) as an oral delivery carrier, with their encapsulation into microspheres based on alginate or extracellular polysaccharide (EPS) p-m10356. The main advantage of liposomes in the microspheres (LIMs) is to improve the restricted drug release property from liposomes and their stability in the stomach environment. Alginate microspheres containing CyA-Lip were prepared with a spray nozzle; CyA-Liploaded EPS microspheres were also prepared using a w/o emulsion method. The shape of the LIMs was spherical and uniform, and the particle size of the alginate-LIMs ranged from 5 to $10\;{\mu}m$, and that of the EPS-LIMs was about $100\;{\mu}m$. In a release test, release rate of CyA in simulated intestinal fluid (SIF) from the LIMs was significantly enhanced compared to that in simulated gastric fluid (SGF). In addition, the CyA release rates were slower from formulations containing the liposomes compared to the microspheres without the liposome. Therefore, alginate-and EPS-LIMs have the potential for the controlled release of CyA and as an oral delivery system.

  • These N-acyl amino acid surfactants is normally produced by reaction of acid anhydride with sodium ${\ell}-glutamate$ hydrolysates under Schotten-Baumann condition i.e., in alkaline aqueous medium. To avoid using fatty acid chlorides, acylations were also carried out with the fatty acids themselves or with their methyl esters, but unfortunately these methods cannot be used in practice, dodecenyl succinic anhydride, was to be studied for their suitability as acylating agents the production if acylated glutamine hydrolysates. The surface activities including surface tension forming power, forming stability and emulsifying power were measured. The experimental results revealed that the products have a good emulsifying power. Thus, there derivatives will be expected to be used an emulsifying agent for O/W type cosmetic emulsion.

Synthesis and Properties of Gemini Type's Diethylene Glycol Dicarboxylates (제미니형 디에틸렌글리콜 디카르복실레이트 류의 합성 및 특성)

  • This study concerned about "Gemini type co-surfactant" which has very interesting properties with new components. They were synthesized by reaction of diethylene glycol monoethyl ether and dicarboxylic acid. The structure could be comfirmed with FT-IR and $^1H$-NMR. Surface active properties such as surface tension, evaluated cmc, cloud point, emulsing power were measured respectively at given conditions. Their surface tensions in the aqueous solution were decreased to 33~35 dyne/cm, which was lower than 39 dyne/cm of SDS, and their cmc values evaluated by surface tension method were $5.0{\times}10^{-1}\sim7.5{\times}10^{-1}$ mol/L. And the emulsifying power was excellent in jojoba oil. All of the synthesized Gemini surfactants possessed good water solubility and their cloud point were $48\sim58^{\circ}C$. As results, Gemini surfactants which were synthesized are expected to be applied as O/W emulsifiers.

  • The action of antioxidants was different depending on the environments where antioxidants were located. Although basic mechanisms of lipid oxidation and antioxidants were related each other, their contribution on the degree of oxidation was different. In thisreview, terminology on antioxidant properties were defined such as antioxidant activity and antioxidant capacities. In addition, antioxidant mechanisms including primary and secondary antioxidants or hydrogen donating or electron transferring antioxidants were introduced. Also, the impact of physical points of view and antioxidant polar paradox were introduced. Depending on the types of food matrice including bulk oil, oil-in-water emulsion (O/W), or solid state, antioxidant actions showed different degree and this point was explained in detail.

Liquid Crystal Emulsions Containing High Content Ceramides for Improved Skin Barrier Functions

  • In this work, we fabricated liquid crystal (LC) emulsions with fatty alcohol in order to stabilize high content ceramide in cosmetic formulation. We investigated the role of fatty alcohol and surfactant in the formation of higher order structure. As a result, we found that they play a crucial role to form higher order structure. SAXS study shows that ceramide can be incorporated up to 3% in cosmetic formulation with higher order structure and its stability was maintained up to 12 weeks at room temperature. According to WAXS study, the higher order structure can suppress the re-crystallization of ceramide in cosmetic formulation. Finally, we performed in vivo skin barrier recovery test for the damaged skin. LC emulsions with ceramide and O/W emulsions show significant effect in skin barrier recovery at D 1, D 2 and D 6 compared to the untreated condition. While only LC emulsions show significant skin recovery effect at D 14. We expect that LC emulsions are the promising skin carrier to stabilize ceramide and LC emulsions with ceramide can improve the skin barrier function.

Formation of Liquid Crystalline with Hydrogenated Lecithin and Its Effectiveness (수소첨가레시친을 이용한 액정 젤의 형성과 보습효과)

  • This study described about method that forms liquid crystal gel (LCG) by main ingredient with hydrogenated lechin (HL) in O/W emulsion system. Result of stability test is as following with most suitable LCG's composition. Composition of LCG is as following, to form liquid crystal, an emulsifier used 4.0wt% of cetostearyl alcohol (CA) by 4.0wt% of HL as a booster. Moisturizers contained 2wt% of glycerin and 3.0wt% of 1,3-butylene glycol (1,3-BG). Suitable emollients used 3.0wt% of cyclomethicone, 3.0wt% of isononyl isononanoate (ININ), 3.0wt% of cerpric/carprylic triglycerides (CCTG), 3.0wt% of macademia nut oil (MNO) in liquid crystal gel formation. On optimum conditions of LCG formation, the pHs were formed all well under acidity or alkalinity conditions (pH=4.0-11.0). Considering safety of skin, pH was the most suitable 6.0${\pm}$1.0 ranges. The stable hardness of LCG formation appeared best in 32 dyne/$\textrm{cm}^2$. Particle of LCG is forming size of 1-20$\mu\textrm{m}$ range, and confirmed that the most excellent LCG is formed in 1-6$\mu\textrm{m}$ range. According to result that observe shape of LCG with optical or polarization microscope, LCG could was formed, and confirmed that is forming multi -layer lamellar type structure around the LCG. Moisturizing effect measured clinical test about 20 volunteers. As a result, moisturizing effect of LCG compares to placebo cream was increased 36.6%. This could predicted that polyol group is appeared the actual state because is adsorbed much to round liquid crystal droplets to multi-lamellar layer's hydrophilic group. It could predicted that polyol group is vast quantity present phase that appear mixed because is adsorbed to round liquid crystal to multi-lamellar layer's hydrophilic group. This LCG formation theory may contribute greatly in cosmetics and pharmacy industry development.

Development of Antimicrobial Plant Extracts and its Application to Cosmetics (항균활성을 갖는 천연물 개발과 화장품 응용에 관한 연구)

  • This study is focused on finding new natural materials that have antimicrobial activity. We found that 3 plants extracts, Magnolia sieboldii K. Koch (M. sieboldii K. Koch), Rhus chinensis (R. chinensis) and Metasequioa glyptostroboides (M. glyptostroboides), have antimicrobial activities by disk diffusion method. We determined the Minimum Inhibitory Concentation (MIC) of each and found that 0.3 ~ 0.35 % of M. glyptostroboides essential oil and 0.35 ~ 0.4 % of M. sieboldii K. Koch extracts inhibited fungal growth and 0.45 ~ 0.5 % of R. chinensis extracts inhibited bacterial growth. We isolated compounds from extracts and verified what have antimicrobial activity. As a result we found that caryophyllene oxide and caryophyllene isolated from M. glyptostroboides, costunolide and dehydrocostus lactone from M. sieboldii K. Koch and ethyl gallate, ethyl-3-gallate from R. chinensis have antimicrobial activities. In accordance with antimicrobial activity, O/W cosmetic emulsion containing mixture of 3 plants extracts showed preservative efficacy against both bacteria and fungi. Based on the above data we suggest that extracts from M. siebodii K. Koch, R. chinensis, and M. glyptostroboides, replace chemical synthetic preservatives and be applied as a natural preservative.

The Preparation and Release Property of Alginate Microspheres Coated Gelatin-cinnamic Acid (젤라틴-신남산 접합체가 코팅된 알긴산나트륨 마이크로스피어의 제조 및 방출 특성)

  • This study is about photosensitive microspheres prepared by coating alginate microspheres with gelatin-cinnamic acid conjugate. Firstly, alginate microspheres was prepared in water-in-oil (W/O) emulsion and then they were coated with gelatin- cinnamic acid conjugate. Herein, gelatin-cinnamic acid conjugate is obtained by the amidation between an amine group of gelatin and a carboxy group of cinnamic acid. Cinnamic acid is widely used as a photo-responsive material easy to dimerize and dedimeriz under UV irradiation at ${\lambda}$ = 254 nm and ${\lambda}$ = 365 nm, respectively. As shown in SEM-EDS, alginate was successfully coated with gelatin-ciannmic acid. By determining the absorbance of coated microspheres at 270nm, the amount of cinnamic acid per microspheres was 0.13/1. The SEM photos showed the size of coated microspheres is around $10{\mu}m$. And the degrees of dimerization and dedimerization were calculated to be 49% and 23% respectively. Then the release of FITC-dextran from the coated micrspheres was studied and release the degree was 42%. As a result, the coated microspheres have potential to be used as a photo-responsive drug carrier to delivery drugs.

Influence of Surface Treatment of SiO$_2$ and Stirring Rate on Fragrant Oil Release Behavior of Poly($\varepsilon$-caprolactone) Microcapsules (실리카의 표면 처리와 교반 속도가 폴리카프로락톤 마이크로캡슐의 향유 방출 거동에 미치는 영향)

  • In this work, the fragrant oil release behavior of poly($\varepsilon$-caprolactone) (PCL) microcapsules containing SiO$_2$ was investigated. The SiO$_2$ was chemically treated in 10, 20, and 30 wt% hydrochloric acid and sodium hydroxide. The acid and base values were determined by Boehm's titration technique and $N_2$/77 K adsorption isotherm characteristics, the specific surface area and total pore volume were studied by BET. The PCL microcapsules containing SiO$_2$ and fragrant oil were prepared by oil-in-water (o/w) emulsion solvent evaporation method. The shape and surface of PCL microcapsules were observed using image analyzer and scanning electron microscope (SEM). The fragrant oil release behavior of PCL microcapsules was characterized using UV/vis. spectra. The average diameters of PCL microcapsules were decreased from 35 to 21 $\mu$m with increasing stirring rate. It was found that in the case of acidic treatment the fragrant oil adsorption capacity and release rate were increased due to the increase of specific surface area and acid value. In the case of basic treatment, the fragrant oil adsorption capacity and release rate were decreased due to the decrease of sp ecific surface area and the increase of acid-base interactions between SiO$_2$-NaOH and fragrant oil with increasing base value of SiO$_2$.

Inorganic-organic nano-hybrid; Preparation of Nano-sized TiO$_2$ Paste Trapped OMC Nano-emulsion and it's Application for Cosmetics (OMC Nano-emulsion을 포집하고 있는 Nano-TiO$_2$-Paste의 합성과 화장품의 응용)

  • Preparations of mesoporous materials using various templates and their applicability have been intensively investigated for many years. We studied on synthesizing mesoporous Ti02 with pores in which sensitive compounds having weak physico-chemical properties such as thermal or UV irradiation and low solubility in solvent are trapped. Prior to trapping OMC in the pores of mesoporous titania, OMC was nano-emulsified in O/W system using Lecithin. Thereafter the OMC was trapped in the pores of mesoporous titania using sol-gel method. Main focus of this work is to prepare OMC-trapped mesoporous titania and to trace the stability and solubility of nano-emulsified OMC in the pores of mesoporous titania, and compared with that of mesoporous silica. OMC-trapped mesoporous Inorganic-Organic hybrid titania showed higher factors in sun protecting and a skin penetration phenomenon was reduced.
