Effect of increasing levels of rice distillers' by-product on growth performance, nutrient digestibility, blood profile and colonic microbiota of weaned piglets

  • Objective: This study was conducted to evaluate the effects of diets containing different wet rice distillers' by-product (RDP) levels on growth performance, nutrient digestibility, blood profiles and gut microbiome of weaned piglets. Methods: A total of 48 weaned castrated male crossbred pigs, initial body weight 7.54±0.97 kg, and age about 4 wks, were used in this experiment. The piglets were randomly allocated into three iso-nitrogenous diet groups that were fed either a control diet, a diet with 15% RDP, or a diet with 30% RDP for a total of 35 days. Chromium oxide was used for apparent digestibility measurements. On d 14 and d 35, half of the piglets were randomly selected for hemato-biochemical and gut microbiota evaluations. Results: Increasing inclusion levels of RDP tended to linearly increase (p≤0.07) average daily gain on d 14 and d 35, and decreased (p = 0.08) feed conversion ratio on d 35. Empty stomach weight increased (p = 0.03) on d 35 while digestibility of diet components decreased. Serum globulin concentration decreased on d 14 (p = 0.003) and red blood cell count tended to decrease (p = 0.06) on d 35, parallel to increase RDP levels. Gene amplicon profiling of 16S rRNA revealed that the colonic microbiota composition of weaned pigs changed by inclusion of RDP over the period. On d 14, decreased proportions of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge, and increased proportions of Prevotellaceae_ge, Prevotella_2, and Prevotella_9 were found with inclusion of RDP, whereas opposite effect was found on d 35. Additionally, the proportion of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge in RDP diets decreased over periods in control diet but increased largely in diet with 30% RDP. Conclusion: These results indicate that RDP in a favorable way modulate gastrointestinal microbiota composition and improve piglet performance despite a negative impact on digestibility of lipids and gross energy.

The importance of NIR spectroscopy in the estimation of nutritional quality of grains for ruminants

  • The production of grain for export and domestic use is one of Australia's most important agricultural industries, and the NIR technique has been used extensively over many years for the routine monitoring of grain quality, particularly moisture and protein content. Because most Australian grain is intended for human food production, the determinants of grain quality for livestock feed, apart from protein, have been largely ignored. However the increasing use of grain for feeding to pigs, poultry, beef cattle and dairy cows has led to an important national research project entitled “Premium Grains for Livestock”. Two of the objectives of this project are to determine the compositional and functional characteristics of grains which influence their nutritional quality for the various classes of livestock, and to adopt rapid and objective analytical tests for these quality criteria. NIR has been used in this project firstly to identify a set of grain samples from a large population of breeders' lines which showed a wide spectral variation, and hence a potentially wide variation in nutritional value. The selected samples were not only subjected to an extensive array of chemical, physical and in vitro analyses, but also were grown out to produce sufficient quantities of grain to feed to animals in vivo studies. Additional grains were also strategically selected from farms in order to include the effect of weather damage, such as rain, drought and frost. In this study to date, NIR calibrations have been derived or attempted, on both ground and whole grains, for in vivo dry matter digestibility (DMD), pepsin-cellulase dry matter disappearance, protein, fat, acid detergent fibre, neutral detergent fibre, starch, in sacco DMD and in vitro assays to simulate starch digestion in the lumen and small intestine. Results so far indicate high calibration accuracy for chemical components (SECV 0.3 to 2.6%) and very promising statistics for in vivo DMD (SECV 1.8, $R^2$ 0.93, SD 7.0, range 61.9 to 92.3, n=60). There appears to be some potential for NIR to estimate some in vitro properties, depending upon the accuracy of reference methods and appropriate sample populations. Current work is in progress to extend the range of grains with in vivo DMD values (a very laborious and expensive process) and to increase the robustness of the various NIR calibrations, with the aim of implementing uniform testing procedures for nutritional value of grains throughout Australia.

The Ethanol Extract of Red Ginseng Enhances Anti-Tumor Effects Using Co60 Gamma Irradiation (홍삼의 에탄올 추출물의 감마선 조사를 통한 항암효과 증대)

  • Red ginseng is one of the most popular traditional medicines in Korea. In this study, we developed a new technique in which ethanol extract of $\underline{r}$ed $\underline{g}$inseng (HRG) was exposed to the $Co^{60}$ gamma radiation ranging from 1~5 kGy. The irradiated HRG (IHRG) were analyzed by high performance liquid chromatography (HPLC) to determine any compositional changes of ginsenosides due to irradiation. No appreciable difference was observed in the HPLC pattern of ginsenosides of HRG. Using MTT assay, the cytotoxicity effect was significantly increased by IHRG compared to HRG. The $LD_{50}$ concentration was $30{\mu}g/mL$ for IHRG-1 (1 kGy), and $15{\mu}g/mL$ for IHRG-5 (5 kGy). The evidences of apoptosis, such as nuclei cleavage and Annexin V staining, were observed in the human prostate cancer PC-3 cells treated with the IHRG. Additionally, the level of reactive oxygen species (ROS) was apparently elevated by IHRG. We also studied the inhibitory effect of IHRG on the growth rate of tumor xenografts in BALB/c male mice. The tumor growth rates were inhibited by 56.9 and 76.1% in mice treated with 10 mg/kg of IHRG-1 and IHRG-5, respectively, compared with control group (21.1%). These results suggest that some biologically active and soluble components in HRG can be more effectively enhancement of anti-tumor effects using irradiation.

Characteristics of Bacterial-Koji and Doenjang(soybean paste) Made by using Bacillus subtilis DJI (Bacillus subtilis DJI을 이용하여 제조된 세균형 코지와 속성된장의 특성)

  • One bacterium with high proteinase production and spore-forming ability was isolated from korean traditional soybean paste(doenjang). The isolated strain was identified as Bacillus subtilis, based on gram-staining, biochemical properties and l6S rRNA gene sequencing, and designated as B. subtilis DJI. Its growth rate was very fast, and it reached its stationary phase within 9 h, and then started to form spores. Bacterial-koji and doenjang were prepared using B. subtilis DJI. Chemical components of the doenjang were determined after 2 months of aging period: amino nitrogen 507 mg%, crude protein 14.3%, crude fat 4.8% and water 54.9%. The composition of total and free amino acids and their ratios of doenjang were changed during the aging period. Among total amino acids in DJI doenjang, glutamic acid, aspartic acid, leucine and arginine were the major amino acids. The fibrinolytic activities of DJI doenjang and traditional doenjangs were 909.7 units/ml and $363.3{\sim}618.6\;units/ml$, respectively. Flavor compounds of DJI doenjang and traditional doenjang were extracted by SDE(simultaneous steam distillation and extraction), and analyzed by GC/MS; DJI doenjang possessed the typically favorable flavor compounds in traditional korean doenjang, with reduced off-flavor compounds.

Morphological, Molecular, and Biochemical Characterization of Astaxanthin-Producing Green Microalga Haematococcus sp. KORDI03 (Haematococcaceae, Chlorophyta) Isolated from Korea

  • A unicellular red microalga was isolated from environmental freshwater in Korea, and its morphological, molecular, and biochemical properties were characterized. Morphological analysis revealed that the isolate was a unicellular biflagellated green microalga that formed a non-motile, thick-walled palmelloid or red aplanospore. To determine the taxonomical position of the isolate, its 18S rRNA and rbcL genes were sequenced and phylogenetic analysis was performed. We found that the isolate was clustered together with other related Haematococcus strains showing differences in the rbcL gene. Therefore, the isolated microalga was classified into the genus Haematococcus, and finally designated Haematococcus sp. KORDI03. The microalga could be cultivated in various culture media under a broad range of pH and temperature conditions. Compositions of the microalgal cellular components were analyzed, and its protein, carbohydrate, and lipid compositions were estimated to be 21.1 ± 0.2%, 48.8 ± 1.8%, and 22.2 ± 0.9%, respectively. In addition, D-glucose and D-mannose were the dominant monosaccharides in the isolate, and its amino acids were composed mainly of aspartic acid, glutamic acid, alanine, and leucine. Moreover, several polyunsaturated fatty acids accounted for about 80% of the total fatty acids in Haematococcus sp. KORDI03, and the astaxanthin content in the red aplanospores was estimated to be 1.8% of the dry cell weight. To the best of our knowledge, this is the first report of an Haematococcus sp. isolated from Korea, which may be used for bioresource production in the microalgal industry.

Comparative Characteristics of Small and Medium-sized Firms in Korea (우리나라 중소기업법인의 업종별 지역별 특성에 관한 연구)

  • Over the past 20 years new and small firms have been identified as significant components of economic strategies for job and wealth creation. Implicit in these strategies has been the search for policies which will increase the supply of new firms, and will encourage established firms' growth. Yet the majority firms spend the whole of their economic life within the small firm sector. The aim of this paper is to suggest a multidimensional approach to the understanding of the comparative characteristics of the small firm by providing evidence as to the factors which describe firms of different sizes, regions and industries. And I discuss in the context of policy options for regional economic development in Korea. The results from the analyses are summarized as follows. The size of the small firms in the sample measured in terms of both number of employees and sales revenue are characterized by regions, export performance and R&D. There is also some indication that growing firms may develop more complex environment. These results are based upon a multi-dimensional analysis of the strategic profile of a sample of small firms using KIS. From this observation of the firms during the data collection period, this is a conclusion that I find intuitively appealing.

Application of Response Surface Methodology in Medium Optimization to Improve Lactic Acid Production by Lactobacillus paracasei SRCM201474 (반응표면분석법을 이용한 Lactobacillus paracasei SRCM201474의 생산배지 최적화)

  • The aim of this study was to establish the optimal medium composition for enhancing L(+)-lactic acid (LLA) production using response surface methodology (RSM). Lactobacillus paracasei SRCM201474 was selected as the LLA producer by productivity analysis from nine candidates isolated from kimchi and identified by 16S rRNA gene sequencing. Plackett-Burman design was used to assess the effect of eleven media components on LLA production, including carbon (glucose, sucrose, molasses), nitrogen (yeast extract, peptone, tryptone, beef extract), and mineral (NaCl, K2HPO4, MgSO4, MnSO4) materials. Glucose, sucrose, molasses, and peptone were subsequently chosen as promising media for further optimization studies, and a hybrid design experiment was used to establish their optimal concentrations as glucose 15.48 g/l, sucrose 16.73 g/l, molasses 39.09 g/l, and peptone 34.91 g/l. The coefficient of determination of the equation derived from RSM regression for LLA production was mathematically reliable at 0.9969. At optimum parameters, 33.38 g/l of maximum LLA increased by 193% when compared with MRS broth as unoptimized medium (17.66 g/l). Our statistical model was confirmed by subsequent validation experiments. Increasing the performance of LLA-producing microorganisms and establishing an effective LLA fermentation process can be of particular benefit for bioplastic technologies and industrial applications.

Life Time Prediction and Physical Properties of Chloroprene Rubber Aged by Seawater (클로로프렌 고무의 해수에 의한 물성 변화 및 노화 수명 예측)

  • Herein, life time prediction based on the deterioration of physical properties of chloroprene rubber (CR)aged by heat and seawater was performed. CR samples were experienced an accelerated test at $80^{\circ}C$, $100^{\circ}C$, $120^{\circ}C$ for heat aging, and $40^{\circ}C$, $60^{\circ}C$, $80^{\circ}C$ for seawater aging for 20,000 hrs. The change in tensile strength, maximum elongation,hardness was measured. As a result, the decrease in elongation was a major factor causing failure. The life time estimated using an Arrhenius model was 125 years at $23^{\circ}C$ for thermal aging and 9 years at $23^{\circ}C$ for seawater aging. SEM and elemental analysis reveal that cracks were generated and the content of oxygen was increased for CR agined by seawater. FT-IR spectrum shows the new C-O and C = O bonds were generated by the chemical reaction with seawater. Also, the glass transtion temperature was increased and the thermal decomposition was decreased by seawater aging.

Effect of Supplementation of Fish and Canola Oil in the Diet on Milk Fatty Acid Composition in Early Lactating Holstein Cows

  • This study examined the effects of supplementation of fish oil and canola oil in the diet on milk yield, milk components and fatty acid composition of Holstein dairy cows in early lactation. Eight multiparous early lactation Holstein cows ($42{\pm}12$ DIM, $40{\pm}6kg$ daily milk yield) were fed a total mixed ration supplemented with either 0% oil (Control), 2% fish oil (FO), 1% canola oil +1% fish oil (FOCO), or 2% canola oil (CO) according to a double $4{\times}4$ Latin square design. Each period lasted 3 wk; experimental analyses were restricted to the last week of each period. Supplemental oils were added to a basal diet which was formulated according to NRC (2001) and consisted of 20% alfalfa, 20% corn silage and 60% concentrate. Milk yield was similar between diets (p>0.05), but dry matter intake (DMI) was lower (p<0.05) in cows fed FO diet compared to other diets. Milk fat percentage and daily yield decreased (p<0.01) with the supplementation of fish and canola oil. The daily yield and percentage of milk protein, lactose and solids-not-fat (SNF) were not affected by diets (p>0.05). The proportion (g/100 g fatty acids) of short chain fatty acids (SCFA) decreased and polyunsaturated fatty acids (PUFA) increased (p<0.05) in milk of all cows fed diets supplemented with oil. The proportions of 6:0, 8:0, 10:0 12:0 and 14:0 fatty acids in milk fat decreased (p<0.01) for all diets supplemented with oil, but the proportions of 14:1, 16:0 and 16:1 fatty acids were not affected by diets (p>0.05). The proportion of trans(t)-18:1 increased (p<0.01) in milk fat of cows fed FO and FOCO diets, but CO diet had the highest proportion of cis(c)-11 18:1 (p<0.01). The concentration of t-10, c-12 18:2, c-9 t-11 18:2, 18:3, eicosapentaenoic acid (EPA, 20:5) and docosahexaenoic acid (DHA, 22:6) increased (p<0.05) in FO and FOCO diets in comparison with the other two diets. These data indicate that including fish oil in combination with canola oil significantly modifies the fatty acid composition of milk.

Region Based Image Similarity Search using Multi-point Relevance Feedback (다중점 적합성 피드백방법을 이용한 영역기반 이미지 유사성 검색)

  • Performance of an image retrieval system is usually very low because of the semantic gap between the low level feature and the high level concept in a query image. Semantically relevant images may exhibit very different visual characteristics, and may be scattered in several clusters. In this paper, we propose a content based image rertrieval approach which combines region based image retrieval and a new relevance feedback method using adaptive clustering together. Our main goal is finding semantically related clusters to narrow down the semantic gap. Our method consists of region based clustering processes and cluster-merging process. All segmented regions of relevant images are organized into semantically related hierarchical clusters, and clusters are merged by finding the number of the latent clusters. This method, in the cluster-merging process, applies r: using v principal components instead of classical Hotelling's $T_v^2$ [1] to find the unknown number of clusters and resolve the singularity problem in high dimensions and demonstrate that there is little difference between the performance of $T^2$ and that of $T_v^2$. Experiments have demonstrated that the proposed approach is effective in improving the performance of an image retrieval system.
