은빛까마중 (Solanum elaeagnifolium Cav.), 우리나라 미기록 귀화식물 (Solanum elaeagnifolium Cav. (Solanaceae), an unrecorded naturalized species of Korean flora)

  • 전남 여수시 삼산면 초도에서 우리나라의 미기록 귀화식물 은빛까마중 (Solanum elaeagnifolium Cav.)을 발견하여 보고한다. 이 종의 원산지는 미국남서부와 인접한 멕시코북부이다. 이 식물은 까마중에 근연이지만 식물체 전체가 회백색 성상모로 뒤덮여있고, 길쭉한 잎의 모양, 열매와 꽃의 크기 및 색깔로 쉽게 구분된다. 이종은 현재 유럽 및 지중해지방국가들, 아프리카, 남미, 아시아 및 호주 등의 아열대 및 온대지방에 널리 퍼져있는 잡초로 분류된다. 따라서 우리나라에서도 앞으로 이 종이 확산되는 것에 대한 대책마련이 필요하다.

Antifibrotic Activity of LCC, a Cerebroside of Lycium chinense Fruit, in Bile Duct-Ligated Rats

  • We previously reported that a novel cerebroside, LCC, isolated from the fruits of Lycium chinense (Solanaceae), significantly exerted hepatoprotective activity against both the carbon tetrachloride-induced and galactosamine-induced toxicities in primary cultures of rat hepatocytes. In the present study, we further attempted to determine the effect of LCC on hepatic fibrosis in animal model. Hepatic fibrosis was induced in rats by bile duct ligation/scission (BDL) for a period of 5 weeks. Treatment of BDL rats with LCC significantly reduced collagen deposition and the activities of serum alkaline phosphatase and ${\gamma}$-glutamyl transpeptidase. In addition, the LCC treatment of BDL rats significantly preserved the decreased hepatic glutathione as well as the activities of glutathione reductase and catalase in BDL rats. From the results, it can be speculated that LCC might exert antifibrotic activity in rats with BDL, in part, through the preservation of antioxidant enzymes and hepatic glutathione.

  • Ralstonia solanacearum (Rso) is a causal agent of bacterial wilt in Solanaceae crops worldwide including Republic of Korea. Rso virulence predominantly relies on type III secreted effectors (T3Es). However, only a handful of Rso T3Es have been characterized. In this study, we investigated subcellular localization of and manipulation of plant immunity by 8 Rso T3Es predicted to harbor a nuclear localization signal (NLS). While 2 of these T3Es elicited cell death in both Nicotiana benthamiana and N. tabacum, only one was dependent on suppressor of G2 allele of skp1 (SGT1), a molecular chaperone of nucleotide-binding and leucine-rich repeat immune receptors. We also identified T3Es that differentially regulate flg22-induced reactive oxygen species production and gene expression. Interestingly, several of the NLS-containing T3Es translationally fused with yellow fluorescent protein accumulated in subcellular compartments other than the cell nucleus. Our findings bring new clues to decipher Rso T3E function in planta.

New Frontiers of Knowledge on Nepalese Plant Science

  • Nepal, with its unique geographical and ecological features due to its abrupt rise in altitude, plays significant role in biological evolution. Existence of numerous wild relatives of the present-day cultivated agricultural crop plants in this small Himalayan nation may serve as a potential source of several yet unidentified desirable genes that are needed for future incorporation in the improvement of cultivated crop plants. This report includes 82 different wild relatives of 41 genera under 19 families of 37 agricultural crops of Nepal(Table 1). It serves as the sample of the glossary of these wild relatives of crop plants in Nepal. Under food grain crop plants of gramineae, leguminoceae and polygonaceae families, 16 different wild species namely wild rices(7 species), wild relatives of wheat plant(3 species), wild arhar(3 species), wild fingermillets(1 species) and wild buckwheat(2 species) have been identified in different parts of the country. Similarly, under vegetable crop plants of Araceae, Amaranthaceae, Crucifereae, Cucurbitaceae, Dioscoreaceae, Labiteae, Leguminosae, Liliaceae, Malvaceae, Polygonaceae, Solanaceae and Umbellifereae, 37 different wild species-wild colocasia(1 species), wild amaranths(3 species), wild leafy vegetables(2 species), wild gourds(3 species), wild cucumber(1 species), wild yams(4 species), wild mints(3 species), wild fenugreeks(4 species), wild pea(1 species), wild beans(3 species), wild garlics(2 species), wild spinach(3 species), wild lady's finger(1 species), wild spinach(3 species), wild eggplants(2 species) and wild carrot(one species) have also been identified. In case of wild relatives of cultivated orchard plants, 11 different wild species namely wild mango(one species), wild banana(one species), wild strawberry(one species), wild pear(one species), wild cherries(2 species), wild apple(one species) and wild grapes(3 species) have been identified, Among 19 different wild species of economic crop plants, five wild species of sugarcane, one species of wild sunhemp, two wild relatives of cotton, three wild relatives of rose, two wild species of tobacco, four wild species of turmeric and two wild species of tea have also been identified. This report includes only sample of the total wild species of the present-day cultivated agricultural crop plants. Further exploration on this economic botany will help the country in cataloging the wild relatives of cultivated crop plants and their future use in crop improvement.

Aqueous extract of Lycii fructus suppresses inflammation through the inhibition of nuclear factor kappa B signal pathway in murine raw 264.7 macrophages

  • Lycii fructus is the fruit of Lycium chinense Miller and is part of the Solanaceae family. Lycii fructus produces various effects such as hypotensive, hypoglycemic, anti-pyretic, and anti-stress activities. Lycii fructus is known to contain betaine, carotene, nicotinic acid, zeaxanthin, and cerebroside. In the present study, the effects of Lycii fructus aqueous extract on lipopolysaccharide (LPS)-induced inflammation in murine raw 264.7 macrophage cells were investigated. In this study we utilized the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, reverse transcriptionpolymerase chain reaction (RT-PCR), Western blotting, and nitric oxide (NO) detection. Lycii fructus aqueous extract suppressed NO production by inhibiting the LPS-induced expressions of inducible nitric oxide synthase (iNOS) and tumor necrosis factor-alpha (TNF-$\alpha$) mRNA and iNOS protein in murine raw 264.7 macrophage cells. Also, Lycii fructus aqueous extract suppressed the activation of nuclear factor-kappa B (NF-${\kappa}B$) in the nucleus. These results demonstrated that Lycii fructus aqueous extract causes an anti-inflammatory effect that was likely produced by the suppression of iNOS expression through the down-regulation of NF-$\hat{e}B$ binding activity.

브라질 전통의약 식물자원의 한의학적 활용가능성 연구 (A Literature Study on the Traditional Herbal Medicine of Brazil)

  • Hanyak, Korean herbal medicine is defined as the herbs understood and explained by traditional Korean medical theories. Considering this definition, there are broad prospective of every species being consilienced and utilized as Korean herbal medicine. Most varied plant species are in tropical regions, and its of these regions posses its own particular traditional medicine. Brazil posses the most varied and abundant plant species and also characteristic traditional medicine, formed by the combination of native indians and immigrants from Africa and Europe. Brazilian traditional medicine are practices by 'garrafeiros', 'raizeiros', or 'curandeiros' and in Caatinga uses herbs in ritual ceremonies. But threatened by modernization, these knowledge may be vanished in a prompt time. Therefore we carried this research in the propose of understanding as well as preserving their traditional medical knowledge. We revised publications about the medical plants and summarized 314 species of 94 families according to repetitive references. The most cited families were COMPOSITAE, LABIATAE, LEGUMINOSAE, RUBIACEAE, SOLANACEAE, UMBELLIFERAE, VERBENACEAE. Also cited major medical efficacies which further medical uses in combinations or modification of current traditional Korean medicine should be studied. This study provides overall prospect of the plants resources of Brazil and their uses. It may serve in the consilience and understanding of varied traditional medicine by traditional Korean Medicine.

Beet western yellows virus (BWYV): Aspect of Outbreak and Survey, and First Complete Genome Sequence of a Korea Isolate of BWYV

  • In 2010, foliar symptoms were observed in the paprika leaves in Jinju city, Korea. Beet western yellows virus (BWYV) was identified in paprika by using the large-scale oligonucleotide chip assay. To investigate the occurrence of BWYV, a survey was performed on various crops, including paprika, from 2011 to 2014. Further, the presence of BWYV was consistently verified through literature survey from 2015 to 2017. BWYV infection has been identified in Solanaceae crops (bell pepper, hot pepper, and paprika), various weeds, and green peach aphids and it occurs on a nationwide scale. Cultivation using organic methods involved natural enemies and showed a high BWYV infection rate, which was more than that for conventional cultivation methods in greenhouse. The complete genome sequence of BWYV isolated from paprika was determined for the first time. The genome of the BWYV-Korea isolate consists of 5750 nucleotides and has six open reading frames. Sequence identity results showed maximum similarity between the BWYV-Korea isolate and the BWYV LS isolate (identity > 90%). This study is the first report of BWYV infecting paprika in Korea. The survey revealed that BWYV is naturalized in the domestic ecology of Korea.

Identification of anti-adipogenic withanolides from the roots of Indian ginseng (Withania somnifera)

  • Background: Withania somnifera (Solanaceae), generally known as Indian ginseng, is a medicinal plant that is used in Ayurvedic practice for promoting health and longevity. This study aims to identify the bioactive metabolites from Indian ginseng and elucidate their structures. Methods: Withanolides were purified by chromatographic techniques, including HPLC coupled with LC/MS. Chemical structures of isolated withanolides were clarified by analyzing the spectroscopic data from 1D and 2D NMR, and HR-ESIMS experiment. Absolute configurations of the withanolides were established by the application of NMR chemical shifts and ECD calculations. Anti-adipogenic activities of isolates were evaluated using 3T3-L1 preadipocytes with Oil Red O staining and quantitative real-time PCR (qPCR). Results: Phytochemical examination of the roots of Indian ginseng afforded to the isolation of six withanolides (1-6), including three novel withanolides, withasilolides GeI (1-3). All the six compounds inhibited adipogenesis and suppressed the enlargement of lipid droplets, compared to those of the control. Additionally, the mRNA expression levels of Fabp4 and Adipsin, the adipocyte markers decreased noticeably following treatment with 25 µM of 1-6. The active compounds (1-6) also promoted lipid metabolism by upregulating the expression of the lipolytic genes HSL and ATGL and downregulating the expression of the lipogenic gene SREBP1. Conclusion: The results of our experimental studies suggest that the withasilolides identified herein have anti-adipogenic potential and can be considered for the development of therapeutic strategies against adipogenesis in obesity. Our study also provides a mechanistic rationale for using Indian ginseng as a potential therapeutic agent against obesity and related metabolic diseases.

Comparative Genomic Analysis and Rapid Molecular Detection of Xanthomonas euvesicatoria Using Unique ATP-Dependent DNA Helicase recQ, hrpB1, and hrpB2 Genes Isolated from Physalis pubescens in China

  • Ground cherry (Physalis pubescens) is the most prominent species in the Solanaceae family due to its nutritional content, and prospective health advantages. It is grown all over the world, but notably in northern China. In 2019 firstly bacterial leaf spot (BLS) disease was identified on P. pubescens in China that caused by both BLS pathogens Xanthomonas euvesicatoria pv. euvesicatoria resulted in substantial monetary losses. Here, we compared whole genome sequences of X. euvesicatoria to other Xanthomonas species that caused BLS diseases for high similarities and dissimilarities in genomic sequences through average nucleotide identity (ANI) and BLAST comparison. Molecular techniques and phylogenetic trees were adopted to detect X. euvesicatoria on P. pubescens using recQ, hrpB1, and hrpB2 genes for efficient and precise identification. For rapid molecular detection of X. euvesicatoria, loop-mediated isothermal amplification, polymerase chain reaction (PCR), and real-time PCR techniques were used. Whole genome comparison results showed that the genome of X. euvesicatoria was more closely relative to X. perforans than X. vesicatoria, and X. gardneri with 98%, 84%, and 86% ANI, respectively. All infected leaves of P. pubescens found positive amplification, and negative controls did not show amplification. The findings of evolutionary history revealed that isolated strains XeC10RQ, XeH9RQ, XeA10RQ, and XeB10RQ that originated from China were closely relative and highly homologous to the X. euvesicatoria. This research provides information to researchers on genomic variation in BLS pathogens, and further molecular evolution and identification of X. euvesicatoria using the unique target recQ gene through advance molecular approaches.

광의의 미치광이풀속(Scopolia Jacq. s.l., 가지과-Hyoscymeae족)의 잎표피 미세구조와 이의 계통분류학적 중요성 (Leaf epidermal microstructure of the genus Scopolia Jacq. s.l. (Solanaceae-Hyoscymeae) and its systematic significance)

  • 세계산 Scopolia Jacq. s.l.에 포함되는 세 개의 속인 Scopolia s.s., Anisodus, Atropanthe와 근연 군외군으로 선택된 Przewalskia 식물의 잎표피 미세구조를 조사하기 위하여 4속 10종(37개체)의 잎을 광학현미경 및 주사전자현미경을 이용하여 관찰하였다. 연구 분류군의 기공은 향축면과 배축면 모두에 존재하는 양면기공엽 (amphistomatic type)으로 밝혀졌다. 공변세포의 크기는 $18-64{\times}11-48{\mu}m$로 속과 종에 따라 다소 차이를 보이는데, 가장 작은 공변세포는 군외군으로 포함시킨 단형속인 Przewalskia tangutica($24-27{\times}16-17{\mu}m$)이었고, 가장 큰 공변세포는 Anisodus carniolicoides ($62-64{\times}43-48{\mu}m$)에서 나타났다. 기공복합체의 형태는 대부분 불규칙형 (anomocytic:Scopolia s.s., Anisodus taxa:A. luridus, A. carniolicoides, A. acutangulus)과 불균등형 (anisocytic:Anisodus tanguticus, Przewalskia, Atropanthe)이 나타난다. 공변세포의 모양은 대부분 초생달 모양의 타원형이나, Przewalskia tangutica에서는 거의 원형으로 나타난다. 부세포의 수충벽(anticlinal wall)은 대부분 파상형 (undulate)이며, Przewalskia tangutica에서는 다소 아치형을 이루었다. 표피세포의 형태는 거의 불규칙한 다각형 (polygonal)으로 수충벽은 다소 또는 심하게 굴곡을 이루는 undulate/sinuate형이었다. 표피세포의 표면은 왁스층이 대부분의 분류군에서는 잘 발달되지 않았고, 일부의 분류군(e.g., Anisodus tanguticus)에서만 기공 주변으로 발달한 양상을 보였다. Scopolia s.s.와 Przewalskia 분류군에서는 독특하게 다세포로 구성된 'elongate-headed' 선모가 존재하였고, Atropanthe 및 Anisodus의 대부분의 분류군은 모용이 존재하지 않았고, 단지 1종(A. luridus)에서는 단순(simple) 또는 가지를 친(branched:dendritic-type) 비선모가 존재하는 것으로 나타났다. 끝으로 조사된 Scopolia s.l.(Przewalskia 포함)의 분류군내의 속간 그리고 종간의 동정과 식별을 위한 잎의 미세형태학적 형질(기공복합체, 모용, 등)의 계통분류학적 또는 생태학적 가치를 검토하였다.
