Molecular Mechanism of Pancreatic Bicarbonate Secretion

  • Thanks to recent progress in availability of molecular and functional techniques it became possible to search for the basic molecular and cellular processes that mediate and control $HCO_3{^-}$ and fluid secretion by the pancreatic duct. The coordinated action of various transporters on the luminal and basolateral membranes of polarized epithelial cells mediates the transepithelial $HCO_3{^-}$ transport, which involves $HCO_3{^-}$ absorption in the resting state and $HCO_3{^-}$ secretion in the stimulated state. The overall process of HCO3 secretion can be divided into two steps. First, $HCO_3{^-}$ in the blood enters the ductal epithelial cells across the basolateral membrane either by simple diffusion in the forms of $CO_2$ and $H_2O$ or by the action of an $Na^+-coupled$ transporter, a $Na^+-HCO_3$ cotranporter (NBC) identified as pNBC1. Subsequently, the cells secrete $HCO_3{^-}$ to the luminal space using at least two $HCO_3{^-}$ exit mechanisms at the luminal membrane. One of the critical transporters needed for all forms of $HCO_3{^-}$ secretion across the luminal membrane is the cystic fibrosis transmembrane conductance regulator (CFTR). In the resting state the pancreatic duct, and probably other $HCO_3{^-}$ secretory epithelia, absorb $HCO_3{^-}.$ Interestingly, CFTR also control this mechanism. In this review, we discuss recent progress in understanding epithelial $HCO_3{^-}$ transport, in particular the nature of the luminal transporters and their regulation by CFTR.

Effects of Temperature, Irradiance, and Nutrient Type on the Fragment Growth of Green Tide Alga Cladophora vadorum (녹조 대발생종 금발대마디말(Cladophora vadorum)의 절편 생장에 온도, 조도 및 영양염 종류가 미치는 영향)

  • The green macroalga Cladophora vadorum bloomed along the coast at Sangrok Beach, Buan, South Korea, in September 2015. To elucidate the cause of bloom, the effects of environmental factors on the vegetative growth of adult fragments were examined. Growth experiments were carried out under different combinations of temperatures and irradiances, and with a single factor of nutrients (nitrogen, phosphorus). The maximal growth of C. vadorum was reported under the combination of 25°C and 100 μmol photons m−2s−1. The species grew under a wide range of N and P concentrations. The growth of C. vadorum peaked at 50 μM PO43−, 80 μM NH4+, and 100 μM NO3. Adult fragments formed holdfasts and new branches within 3 days in culture and became adults, showing polarized growth patterns, in 2 weeks. This is the first report showing the development of numerous bladelets from a segment in Cladophora species. The present results indicate that Cladophora blooms appear under growth conditions that are favorable in terms of temperatures, irradiance, and nutrients via fragment growth patterns producing rapid holdfasts and many bladelets.

Morphometric analysis on bone formation effect of $\beta-TCP$ and rhBMP-2 in rabbit mandible (토끼의 하악골에서 $\beta-TCP$ 와 rhBMP-2의 골형성 효과에 대한 형태계측학적 연구)

  • Introduction: This study was to assess the effectiveness of new bone formation and regeneration by using a rhBMP-2 and $\beta-TCP$ as a carrier in rabbits’mandible. Materials and Methods: The mandibles of 36 rabbits were exposed and cortical bone was penetrated for this study. The experimental subjects were divided into 3 groups each 12 rabbits ; control group, experimental group 1, and experimental group 2. Control group had the defect itself without any treatment, in the experimental group 1, $\beta-TCP$P only was grafted, and in the experimental group 2, rhBMP-2 soaked in $\beta-TCP$ was grafted. The rabbits were sacrificed after 1, 2, 3, 4, 6, and 8weeks, and new bone formation area was examined and measured for bone quantitative and qualitative analysis with light, fluorescent and polarized microscopy. Results: In the experimental group 1, new bone formation from the adjacent host bone was made by osteoconduction, and in the experimental group 2, direct new bone formation by osteoinduction of rhBMP-2 as well as new bone formation by osteoconduction of $\beta-TCP$ were observed. Conclusion: rhBMP-2 of experimental group 2 is very effective in the bone formation in early 2weeks and bone remodelling from 3weeks.

Studies on improvement of manufacturing method of enzymic source for Maggerley(Korea wine) brewing(I) (막걸리의 제조를 위한 효소제의 개발연구 1)

  • It is necessry to develop and strengthen the activity of enzymic source which in low applied for maggerley brewing as an amylolytic and proteolytic starter, recently in this country the active and strong enzymic starter is required for the better brewing and to substitute another starch material for the present wheat flour. In this study, manufacturing method the strong enzymic source have been developed and established with use of raw wheat bran plus fungal strains of Rhizopus sp. and Aspergillus usamii the culture of starter. The results on experimental the activities of enzymic sources (stater) are as following ; 1. Method of making the enzymic source (starter) is to cultivate the strains of Asperguillus orzyae, Asp. kawachii, Asp usamii and Rhizopus sp. in the acid treated raw or heatboiled wheat bran. 2. The saccharogenic pwoer (S.P.) of enzymic source which consisted of raw bran plus fungi and cultured in it is generally stronger than those of heat-boiled bran plus fungi, the strongest power was shown in the culture of Rhizopus plus raw bran, and the next other is in mixture of Asp.usamii and Rhizopus on raw wheat bran. 3. The most strong alpha amylase activity was expressed in the plot of Asp.oryzae on heat-boiled wheat bran, the next was in the culture of Rhizopus nad Aspergillus usamii on raw wheatbran. 4. The most vigourous acidic proteinase activity was expressed in the micture of raw bran plus Asp. usamii and Rhizopus those were independentlu cu;tured before mixing for neutral proteinase activity, it was shown in the mixed culture of Asp. usamii and Rhizopus on raw wheat bran, the msot active alkaline proteinase activity of enzymic source was found in the plot of raw bran material. 5. For poly-preptidase activity in pH 6.5 it is found that the culture of Rhizopus and Asp.usamii on raw bran was most active among them of enzymic sources. 6. Generally, it is concluded that culture of fungi on acid treated raw wheat bran is stronger in its activity than those of heat boiled wheat bran, especially the culture of Rhizopus nad Asp.usamii on raw bran exhibited the most vigorous and non-polarized activity for all aspects, so it is considered to be most desirable enzymic stater in Korean Maggerley brewing and this would be able to substitute brewing material for the present wheat flour because of its strong and wide hand activity of amylolytic and proteolytic action.

In-situ Monitoring of GaN Epilayers by Spectral Reflectance (분광 반사법을 이용한 GaN 박막의 실시간 관찰)

  • An in-situ, real-time monitoring of GaN epilayers grown by low pressure metalorganic chemical vapor deposition system modified for spectral reflectance was performed. Reflectance spectrums from 190~861 nm were observed using p-polarized light with incident angle of $75^{\circ}$. All reflectance spectrums showed interference oscillation caused by multiple reflection within GaN epilayers, and the spectrum from GaN with low crystalline quality showed weak reflectance intensity and much low amplitude of the oscillation because many defects in GaN resulted in light scattering and absorption. Signal variation of reflected light which was selected around strong constructive wavelength range was also observed during $NH_3$ supplying and $NH_3$ cut-off. There was no significant change in signal intensity when $NH_3$ cut-off for 10 sec, but it showed higher intensity when $NH_3$ was cut off for over 30 sec and its intensity kept unchanged. This result indicates that GaN surface was N-terminated during $NH_3$ supplying but Ga-terminated during $NH_3$ cut-off because of high nitrogen equilibrium vapor pressure of GaN, and metallic Ga-terminated surface caused slightly higher reflectance intensity.

  • (La,Sr)CoO3/Pb(Zr,Ti)O3/(La,Sr)CoO3 (LSCO) heterostructures have been grown on LaAlO3 substrates by pulsed laser deposition (PLD) to investigate asymmetric polarization of Pb(Zr,Ti)O3 (PZT) thin flims with different electrode configuration. P-V hysteresis loop of LSCO/PZT/LSCO was symmetric. However, LaCoO3 (LCO_/PZT/LSCO showed a largely asymmetric P-V hystersis loop and large relaxation of the remanent polarization at the negatively poled state, which means that the negatively poled state was unstable. On the other hand, LSCO/PZT/LCO exhibited large relaxation of the positively poled state. The asymmetric behavior of the polarized states implies the presence of an interal electric firld inside the PZT layer. It is suggested that internal electric field is caused by built-in voltages at LCO/PZT and LSCO/PZT interfaces. The built-in voltages at LCO/PZT and CSCO/PZT interfaces were 0.6 V and -0.12 V, respectively.

Clarification of Apple Vinegar by Ultrafiltration and Flux Charcateristics (한외여과를 이용한 사과식초의 청징화와 투과특성)

  • This study was conducted to evaluate the effect of ultrafiltration (UF) process variables on permeate flux and membrane resistance and to clarify apple vinegar for quality improvement. Apple vinegar was clarified in a laboratory ultrafiltration system with hollow fiber membrane made of polysulfone and MWCO 30,000 and 10,000. The permeate flux increased with the increase of flow rate and the optimum pressure was $1.5\;kgf/cm^2$ in this system. The turbidity of clarified apple vinegar treated UF largely decreased. pH and acidity of treated samples showed the same level as those of untreated apple vinegar. The permeate flux continuously declined while the fouling material accumulated on the membrane as the operation time increased. Resistance decreased with lower pressure, which could be explained by expansion of pore size at lower pressure and minor compaction of the polarized layer at lower pressure.

Correlation between Skin Translucency and Scattering Reflection using Miniaturized New Optical Device (피부 투명감 측정 기기의 소형화 및 피부의 확산 반사광과 투명감 사이의 연구)

  • Skin darkness is the source of trouble to many women. Skin darkness is the state that while skin redness and brightness are reduced, skin yellowness is increased. Skin translucency which is measured by skin color, especially brightness is an another expression of skin darkness. Skin brightness is usually expressed by L value of $L^{*} \;a^{*}\; b^{*}$ (CIELAB color space system). However, it is hard to find the relationship between L value and perceptive efficacy such as translucency because the L value is just a factor of evaluation of skin darkness. The skin with high translucency has high scattering reflective light value than low value. In this study, we measured the skin translucency of 20 ~ 30 ages men and women face by both our designed previous device which use polarized light to detect surface and scatter reflective light independently and $Lumiscan^{TM}$ which is improved designed to confirm our new device working ability by calculation of relationship between trnaslucency and scattering reflective light value. The result of this study indicate that there is a high correlation (R = 0.732, p < 0.01) between translucency and scattering reflective light value, and suggest that $Lumiscan^{TM}$ is portable and easy measuring device more that previous device.

REMINERALIZATION EFFECT OF FUJI VII GLASS IONOMER CEMENT (Fuji VII 글래스 아이오노머 시멘트의 재광화 효과)

  • Fuji VII is a glass-ionomer cement specially targeted for early protection in erupting first and second molars. Properties of Fuji VII such as very high level of fluoride release, low viscosity and no need to preliminarily etch the substrate would be useful to erupting molars with primary pit and fissure caries or hypoplastic area for preventive goal or remineralization. The purpose of this study were to evaluate remineralization of Fuji VII glass ionomer cement and to compare with one of other restorative materials such as conventional glass ionomer cement, resin-modified glass ionomer cement, compomer and composite resin. Forty-two extracted human molars were used for this study. All teeth were immersed in demineralizing solution for 48 hours after Class V cavity preparation was made on sound proximal surface. The teeth were randomly divided into six groups and restored with Fuji VII, Fuji II, Fuji II LC improved, F2000, $Filtek^{TM}$ Z250 and control group was unrestored. The middle area with $130{\pm}20{\mu}m$ thickness was separated from specimen using microtome and demineralized area was photographed under polarized microscope. Separated area was relocated to specimen and stored in artificial saliva, After four weeks, changes of demineralized area were observed and compared to them restorated immediately. The results from the this study can be summarized as follows ; 1. Fuji VII, Fuji II, Fuji II LC improved have more prominent remineralization effect than F2000, $Filtek^{TM}$ Z250, control group. 2. No significant differences in remineralization effect are seen between Fuji VII and Fuji II, Fuji II LC improved.

Micromorphological and Mineral Characteristics of the Jang-won Series which have Fragipan in the soil Profile (경반층 토양인 장원통의 미세형태학적 및 광물학적 특성)

  • This study was carry out on a Jang-Won series (fine loamy, mixed, mesic family of typic fragipan) that were established and classified as a fragipan soil in Korea. The morphological, physical, chemical and minerals characteristics of Jang-Won series were studied to determine the genesis of fragipan soils in natural environment. Each sample was analyzed for its physical, chemical and mineralogical characteristics. The particle size distribution of samples was measured using pipette method. Clay minerals were investigated on parallel-oriented specimens of the clay fraction ($<2{\mu}m$) from each horizon, separated by sieving and centrifugation, using X-ray diffraction (XRD) analysis. Micromorphological observations were made on thin sections prepared from soil blocks impregnated with Crystic Resin, cut and ground to less than $30{\mu}m$ in thickness, and finally polished with diamond paste. Most horizons have pH values in the range of fewer than 5.0 and have very low base-saturation values. Their textural classification ranges from silt loam to loam, the lower horizons being the finer. The clay fraction revealed the occurrence of illite, kaolinite, chlorite and vermiculite. The micro-morphological analysis carries out thin sections from each soil profile. The silt concentrations occur as extremely dense and homogenous bands or zones of silt-sized materials, brownish in colour in plane-polarized light and anisotropic in cross-polarized light, surrounding or adhering to skeleton grains. The genesis of fragipan in the Jangweon series assumed composition of clay fraction rather than silt concentration. Therefore, this results suggested an authentic interpretation which Jangweon series is classification as Typic Fragiochrepts.
