Variation of Decursin Contents of Root Ages in Floral Inhibition Cultured Angelica gigas Nakai (화성억제재배한 참당귀의 년근별 약효성분함량)

  • In order to clarify the contents of active principles in floral inhibition cultured Angelica gigas roots, the amounts of crude extract and the contents of decursin and decursinol angelate in the yearly roots were analyzed and compared with respect to the root age, root part and growth stage. The woody cell, weight and width of pith and cortex were also investigated at different growth stage to observe the developmental characteristics of lignification in the roots as bolting and flowering in normal cultured Angelica gigas. The amount of crude extracts did not differ with plant ages, whereas the contents of decursin and decursinol angelate were differed and the highest in 3 year old roots. The contents of decursin in 1, 2 and 3 year old roots were 3.71, 4.76 and 8.20% and those of decursinol angel ate were 2.84, 3.40 and 5.01%, respectively. The amount of crude extracts, and the contents of decursin and decursinol angelate were the highest in fine roots, followed by the lateral roots and the lowest in the primary roots. On the other hand, the amount of the constituents in the cortex were much higher than those in the pith of the root. The amounts of crude extract, and the contents of decursin and decursinol angelate showed the highest value at the vagetative stage and decreased with development to bolting and blooming stage. Woody cells were accumulated in the pith of the root as advancing growth stage, so that the weight and radius of the pith increased, whereas the relative weight and width of the cortex decreased slightly.

  • This study was intended to find out the effects of the inorganic nutrients in the soil and root on the available constituents contents in Angelica gigas Nakai. The correlation between the contents of decursin and decursinol in roots of the wild and cultivated and the inorganic nutrients contents in the soils and roots was investigated. The contents of decursin in the dried root of the wild was higher than that of the cultivated. The ammonium nitrogen contents in soil and the decursin contents in root had the significant negative correlation, whereas the significant positive correlation was obtained the available phosphorus contents. The significant negative correlation was obtained between the decursin contents and the nitrogen contents of root, and the significant positive correlation between the decursin contents and the contents of phosphorus or potassium in roots.

Development and Validation of Analytical Method for Decursin in Aerial Parts of Angelica gigas Nakai Extract (참당귀 지상부 추출물의 지표성분 decursin의 분석법 개발 및 검증)

  • Method development and validation of decursin for the standardization of Angelica gigas Nakai as a functional ingredient and health food were accomplished. The quantitative determination method of decursin as a marker compound of aerial parts of Angelica gigas Nakai extract (AAGE) was optimized by HPLC analysis using a C18 column ($3{\times}150mm$, $3{\mu}m$) with 0.1% TFA in water and acetonitrile as the mobile phase at a flow rate of 0.5 mL/min and detection wavelength of 330 nm. The HPLC/PDA method was applied successfully to quantification of the marker compound in AAGE after validation of the method with linearity, accuracy, and precision. The method showed high linearity in the calibration curve at a coefficient of correlation ($R^2$) of 0.9994 and the limit of detection and limit of quantitation were $0.011{\mu}g/mL$ and $0.033{\mu}g/mL$, respectively. Relative standard deviation (RSD) values of data from intra- and inter-day precision were less than 1.10% and 1.13%, respectively. Recovery of decursin at 0.5, 1, 5 and $10{\mu}g/mL$ were 92.38 ~ 104.11%. These results suggest that the developed HPLC method is very useful for the determination of marker compound in AAGE to develop a health functional material.

  • Six components were isolated from the $CH_2Cl_2$ fraction of Belamcanda chinensis rhizome by open column chromatography. Their structures were elucidated as ${\beta}-sitosterol$ (1), apocynin (2), decursin (3), iristectorigenin A (4), irigenin (5) and tectorigenin (6) by spectral analysis. Among these compounds, decursin (3) was isolated for the first time from a plant of the family Iridaceae.

  • The anti-oxidant activities of decursinol angelate (1) and decursin (2) isolated from Angelica gigas were investigated. These two coumarins exhibited decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_4-intoxication$ in rats. They also showed increase in anti-oxidant enzyme such as hepatic cytosolic superoxide dismutase (SOD), catalase, and glutathione peroxidase (GSH-px) in $CCl_4-intoxicated$ rats. These results suggest that decursinol angelate (1) and decursin (2) from A. gigas possess not only the anti-oxidant, but also the hepatoprotective activities in rats.

  • Two decursin derivatives were synthesized from decursinol. Compounds 1 and 2 were determined as 7-{3-[4-(2-Methoxy-phenyl)-piperazin-1-yl]-propoxy}-8,8-dimethyl-7,8-dihydro-6H-pyranochromen-2-one (1) and decursinol 3'-O-E-pmethoxycinnamic acid ester (2), respectively and newly reported. Compounds 1 and 2 significantly inhibited AChE activity and ameliorated memory impairment induced in mice by scopolamine (1.0 mg/kg body weight s.c.) as measured in passive avoidance test. We suggest, therefore, that compounds 1 and 2 has both anti-AChE and anti-amnesic activities that may ultimately hold significant therapeutic value in alleviating certain memory impairment observed in Alzheimer's disease.

Anti-Tumor Activities of Decursinol Angelate and Decursin from Angelica gigas

  • The in vivo anti-tumor activities of decursinol angelate (1) and decursin (2) isolated from the roots of Angelica gigas were investigated. These two compounds, when administered consecutively for 9 days at 50 and 100 mg/kg i.p. in mice, caused a significant increase in the life span and a significant decrease in the tumor weight and volume of mice inoculated with Sarcoma-180 tumor cells. These results suggest that decursinol angelate (1) and decursin (2) from A. gigas have anti-tumor activities.

  • Diabetes has been one of major health risks in industrialized countries. AMP-activated protein kinase (AMPK) has been focused as a novel therapeutic target for the treatment of metabolic syndromes, because AMPK increases glucose uptake through independent insulin signal pathway. In this study, we investigated the anti-diabetic effect of Angelica gigas Nakai extract (AGNEX), a mixture of decursin and decursinol angelate (53 : 47), decursin and decursinol angelate on blood glucose, glucose transport (GLUT) and AMPK expression levels in streptozotocin (STZ)-induced diabetic rats. To induce diabetes, 50 mg/kg of STZ was injected via i.v. route and AGNEX 2 mg/kg (STZ+AG), decursin 2 mg/kg (STZ+D), decursinol angelate 2 mg/kg (STZ+DA), and metformin 100 mg/kg (STZ+M) were administered orally for 21 days. STZ+DA group showed a significant decrease in fasting blood glucose levels compared to the other groups. Decursinol angelate significantly upregulated expression of glucose transporter 4 (GLUT4) and phosphorylation of AMPK (p-AMPK) in skeletal muscle of rats. In pancreas of rats, decursinol angelate significantly increased expression of GLUT2 through down-regulation of p-AMPK. In addition to the result of pancreatic islets morphology, AGNEX, decursin, decursinol angelate, and metformin treated group recovered ${\beta}$-cell damage by hyperglycemia. These results indicate that decursinol angelate might be a potential anti-diabetic agent and AGNEX could be useful in the treatment of diabetes mellitus.

Effect of Yield and Decursin Content According to the Accumulative Temperature and Seedling Size in Cultivation Areas of Angelica gigas Nakai (지대별 적산온도와 묘 크기가 당귀의 수량 및 Decursin 함량에 미치는 영향)

  • This study was conducted to find out some conditions for optimum cultivation of Angelica gigas Nakai by the investigation of root yield and decursin content from different seeding sizes and accumulative temperature. Accumulative temperature from April to October was $4,309^{\circ}C$ on altitude 100 m, $4,242^{\circ}C$ on 250 m, $3,662^{\circ}C$ on 530 m, $3,435^{\circ}C$ on 730 m, and altitude 530~730 m was less $650{\sim}870^{\circ}C$ than altitude 100 m cultivation areas of A. gigas. Seedling stand rate was increased from 86.4% to more than 90% as accumulative temperature decreases, and was increased in above 7 mm of seedling size, and 10% in non-mulching more than PE film mulching. Yield was increased as accumulative temperature decreases and in PE film mulching as 310.2 kg/10a. Also, Yield was increased in 7~9 mm than seedling diameter 5~7 mm as 313.0 kg/10a. Decursin content of primary roots was increased as altitude rises, ie, as 2.55% on altitude 100 m, 3.33% on 250 m, 5.51% on 530 m, and 6.24% on 730 m. Decursinol angelate content appeared little than decursin content as 1.08% on altitude 100 m, 1.37% on 250 m, 1.99% on 530 m, 2.38% on 730 m, and as altitude was heightened, content was increased.

HPLC Analysis and Extraction Methods of Decursin and Decursinol Angelate in Angelica gigas Roots (당귀(Angelica gigas) 중 Decursin 및 Decursinol Angelate 추출 방법과 HPLC 분석)

  • This paper is intended as an investigation of the analysis of high-performance liquid chromatography and the method of extraction of decursin and decursinol angelate in Angelica gigas roots. There are three kinds of extraction methods: distilled water, 50% EtOH and 100% EtOH. The condition of HPLC was obtained on a reversed-phase column $(Polarity\;dC_{18},\;4.6{\times}250 mm,\;5\;{\mu}m)$ using a phosphate buffer-acetonitrile-sodium lauryl sulfate as the mobile phase. Under these chromatographic conditions, UV detector was 230 nm, column temperature $30^{\circ}C$ and the speed of a current 1.0 ml/min, respectively. The results of extraction with distilled water, 50% EtOH and 100% EtOH in Angelica gigas roots were as follows. The concentrations of decursin and decursinol angelate were 182 and 153 ppm (distilled water), 3,142 and 2,547 ppm (50% EtOH) and 3,341 and 2,778 ppm (100% EtOH). There were high positive correlations between the concentrations of decursin and EtOH (r=0.8928, p<0.01) and decursinol angelate and EtOH (r=0.9009, p<0.01).
