Reduction of Deoxynivalenol in Barley by Chemical Treatments and Malting

  • Fusarium mycotoxin deoxynivalenol (DON) contents and its critical quality parameters were analyzed using two-rowed, six-rowed, and hulless barleys. DON content in the two-rowed Korean barleys was lower than that in the six-rowed barleys. The average DON contents of six-rowed, two-rowed, and hulless barleys were 1.35 ppm, 0.36 ppm, and 0.49 ppm, respectively. The DON content was reduced by 6.2% by sieving, by 6.0% by washing with water and by 18.1-69.8% by treatment with aqueous chemical solutions. Of the reagents investigated, aqueous sodium bicarbonate gave the greatest reduction in the barley DON level. The DON content was reduced to 62.5% of initial level after 3 days of steeping and to 23.1% after 3 days of germination. DON was not detected after steeping barley for 24 hr in 0.1M $Na_2CO_3$ solution with 0.1 % activated carbon.

Production and Characterization of Thermo-alkalotolerant Cyclodextrin Glucanotransferase from Thermo-alkalophilic Bacillus cereus B-13 (고온성이며 호알칼리성인 Bacillus cereus B-13으로부터 내열성, 호알카리성 Cyclodextrin Glucanotransferase의 생산과 특성)

  • To produce a thermostable cyclodextrin by using thermotolerant cyclomaltodextrin glucanotransferase(CGTase), a thermophilic and alkalophilic bacterium isolate, designated B-13 showing the highest CGTase activity was isalated from natural sources and identified as Bacillus cereus B-13 based on the morphological and physiological characteristics, and 16S rRNA sequence. The maximal CGTase activity (130 U/ml) was obtained when Bacillus cereus B-13 was cultured in SYC medium containing 2.0% soluble starch, 1.0% yeast extracts, 1% corn steep liquor and 1% $Na_2CO_3$ (pH 8.5) at $50^{\circ}C$ for 24 h and about 80% of maximal activity was also showed in he culture broth of $60^{\circ}C$ for 18 h. Optimum reaction temperature and pH of the partial purified CGTase for soluble starch were $65^{\circ}C$ and pH 8.5-9.0 respectively. The partial purified CGTase were also stable below $80^{\circ}C$ and pH 5.0-10.0. When 1% soluble starch was digested with the partial purified CGTase, the yield of cyclodextrin was 49%.

  • About 600 microorganisms isolated from soil, marsh, compost, etc. were examined for their flocculating ability in the kaolin suspension and swine wastewater. Among them, K-199 was the best producer of flocculant and was identified to be a species belong to the genus Corynebacterium. The maximum production of the flocculant from Corynebacterium sp. K-199 was observed in culture medium containing 2% glycerol, 0.4% peptone, 0.3% K$_{2}$HPO$_{4}$, 0.1% CaCO$_{3}$, 0.05% NaCl, 0.1% yeast extract, 0.1% MgSO$_{4}$,7H$_{2}$O and initial pH 7.5 when cultured with rotary shaker controlled at 25$\circ$C and 150 rpm. Under the optimal culture condition with jar fermentor, the maximum production was reached to flocculating activity of 780 units/ml after 4 days. From the results of the activity be fully maintained by periodate oxidization, it suggests that the activity is due to the protein.

  • The optimum culture conditions for an antibiotics from Actinomyces sp. were investigated. The optimum composition of medium for antibiotics production was 1% glucose, 1% soybean meal, 0.5% NaCl, 0.1% $CaCO_2$, and the optimum initial pH was 7.0. And the antibiotics showed highest activity when the strain isolated from soil was aerobically cultivated at $28^{\circ}C$ for 72hours under the optimum conditions. A production of the antibiotics from Actinomyces sp. begins at the 36th hours and then reached the maximum at the stationary phase developed at the 72th hours under the optimum conditions.

  • Objectives : This study was designed to investigate the effects of horse bone powder extract on the growth of longitudinal bone in adolescent male rats. Methods: Longitudinal bone growth was measured by fluorescent microscopy. To examine the effects on the growth plate metabolism, the total height of growth plate, the induction of local insulin-like growth factor-1 (IGF-1), and bone morphogenetic protein-2 (BMP-2) were measured. Results: Horse bone powder extract enhanced longitudinal bone growth and total height of the growth plate. Also, it promoted the induction of local IGF-1 and BMP-2 of the growth plate. Conclusions: This study shows that the horse bone powder extract effects longitudinal bone growth in adolescent rats and might be used for both stunted adolescents and inherent growth failure patients.

A Thermal Study of the Harmful Chemical Species of Charcoal and Their Transformation during Combustion (숯의 유해물질과 이들의 연소 중 상변화에 대한 열분석 연구)

  • Charcoal burning in the process of manufacture and ordinary use often release many constituent chemical species. As a result of open burning, the chemical composition as well as the physical properties of original material changes through the modification of surface properties of charcoal. Surface modification could be more responsible toward the outside elements for surface adsorption, it becomes easy to adsorb more toxic elements through surface adsorption. In this study, four kinds of commercially available charcoal were studied against the chemical and thermal stability along with the heavy metals and organic hazardous substances. Thermo gravimetric analysis (TGA) and differential scanning calorimetry, from room temperature to $400^{\circ}C$, were performed to study the weight loss and the changes in the behavior of those substances. According to TGA analysis, about 10% of weight loss was happened before $200^{\circ}C$. It was found that related weight loss of this temperature region may responsible to the gas phase organic matter. Natural charcoal, K1 and C1 show 15% of loss during the reaction heated to $400^{\circ}C$, while the artificial charcoal K2, C2 show the weight loss of about 20% was found. This is consistent with the main organic matter and VOC analysis results shown. Chemical composition based on the x-ray diffraction analysis was carried out. X-ray diffraction analysis reveals the existence of chemical additive in the forms of $Ba(NO_3)_2$, $BaCO_3$, and $NaNO_3$.

  • Agar degrading enzyme-agarase-was purified from the culture fluid of Cytophaga so/ ACLJ-18, by acetone precipitation, DEAE-Cellulose, Sephadex G-100 and CM-Sephadex C25 column chromatographies. The molecular weight of purified agarase was estimated to be 24,700 dalton by SDS-polyacrylamide gel electrophoresis. The optimum pH and temperature for agarase activity were 7.0 and 40$^{\circ}C$, respectively. this agarase was stable in the pH range of 6.5 - 8.0 and 40$^{\circ}C$, and required 0.35M NaCl for optimum activity. And this agarase was inhibited by metal ions such as Ba2+, Cu2+, Co2+, Mn2+, Hg2+, Zn2+, and showed specificity on agar.

Study on the Hollow Fiber Nano-composite Membrane Preparation onto the Porous PVDF Membrane Surfaces using the Interfacial Polymerization (다공성 PVDF 막의 polyamide 계면중합법처리를 통한 나노 중공사 복합막 제조 연구)

  • The composite membranes were prepared on the surface of hydrophobic porous poly (vinylidene fluoride) (PVDF) hollow fiber membranes through the interfacial polymerization. The preparation variables were the concentrations of piperazine (PIP), trimesoyl chloride (TMC) and the contents of polyethylene glyco l (PEG). The separation characterization of the resulting membranes were carried out for aqueous 100 ppm solution of NaCl, $CaSO_4$, and $MgCl_2$ and also mixed 300 ppm solution of NaCl and $CaSO_4$ in terms of the flux and rejection. Both the flux and rejection were the highest when the interfacial polymerization was conducted using TMC. When TMC concentration was 0.1 wt%, the flux and rejection were shown 48.3 LMH ($L/m^2{\cdot}hr$) and 59%, respectively. To improve the flux, the annealing post-treatment and the addition of PEG into piperazine were done. As expected, the overall flux was enhanced while the rejection was reduced.

A Study on Synthesis of Ca and Mg Compounds from Dolomite with Salt Additional React (MgCl2·6H2O) (염 첨가 반응(MgCl2·6H2O)을 이용하여 백운석으로부터 Ca 화합물과 Mg 화합물 합성에 관한 연구)

  • In order to utilize dolomite as a calcium/magnesium compound material, it was prepared highly reactive calcined dolomite(CaO·MgO) using a microwave kiln (950 ℃, 60 min). The experiment was performed according to the standard of the hydration test (ASTM C 110) and hydration reactivity was analyzed as medium reactivity (max 74.1 ℃, 5 min). Experiments were performed with calcined dolomite and salt (MgCl2·6H2O) (a) 1:1, (b) 1:1.5, and (c) 1:2 wt% based on the hydration reaction of calcined dolomite. The result of X-ray diffraction analysis confirmed that MgO of calcined dolomite increased to Mg(OH)2 as the salt addition ratio increased. After the separating reaction, calcium was stirred at 80 ℃, 24 hr that produced CaCl2 of white crystal. XRD results, it was confirmed calcium chloride hydrate (CaCl2·(H2O)x) and CaO of calcined dolomite and salt additional reaction was separated into CaCl2. And it was synthesized with Ca(OH)2 99 wt% by NaOH adding reaction to the CaCl2 solution, and the synthesized Ca(OH)2 was manufactured CaO through the heat treatment process. In order to prepare calcium carbonate, CaCO3 was synthesized by adding Na2CO3 to CaCl2 solution, and the shape was analyzed in cubic form with a purity of 99 wt%.

The Role of Na-K Pump in the Modulation of Vascular Tone in the Rabbit (혈관 긴장도 조절에 미치는 Na-K Pump에 관한 연구)

  • Force development of smooth muscle cells is directly regulated by the concentration of free calcium ions in the sarcoplasm, and the sarcoplasmic concentration of calcium ion can be modulated by electrogenic Na-K pump. The role of Na-K pump on vascular tone was studied in isolated rabbit renal artery. Helical strips of arterial muscle were prepared from left renal arteries. All experiments were performed in $HCO_3^--buffered$ Tyrode solution which was aerated with $3%CO_2-97%\;O_2$ mixed gas and kept at $35^{\circ}C$. In some experiments, rabbit was injected intraperitoneally $18{\sim}24$ hours prior to the experiments, with a large dose(5 mg/kg body wt) of reserpine, in order to eliminate the catecholamines present in intrinsic adrenergic nerve terminate. Treatment used in this experiment that inhibits Na-K pump was the exposure of strips to K-free Tyrode solution. Contractile response to K free Tyrode solution developed slowly and the time required for maximum contracture was $20{\sim}30$ minutes. This K-free contracture was rapidly relaxed by the addition of potassium to the bathing solution. No K-free contracture occurred in a Ca-free Tyrode solution. But contraction developed rapidly when calcium ion was added to the bathing solution after 30 minute exposure of the strip to Ca-free Tyrode solution. This contracture was completely inhibited by Ca-antagonist, verapamil. The K-free contracture was abolished by ${\alpha}-adrenergic$ blocker, phentolamine, as well as by the catecholamine depletion from adrenergic nerve terminals. Even in reserpinized strip, the exogenous norepinephrine-induced contraction in K-free Tyrode solution was rapidly suppressed by the addition of potassium ion. The results of this experiment suggest that K free contracture develops by norepinephrine release from adrenergic nerve terminals, while the relaxation of K-free contracture is induced by the activation of electrogenic Na-K pump.
