Digital Chest Radiography에서 방사선량에 대한 Source to Image-Receptor Distance (SID)의 영향 (The Effect of Source to Image-Receptor Distance(SID) on Radiation Dose for Digital Chest Radiography)

  • X선을 이용한 chest radiography는 일반적으로 180 cm의 SID에서 실시되고 있다. digital chest radiography에서 AEC를 적용하고 120 kVp, 320 mA에서 SID를 180 cm부터 340 cm까지 20 cm 단위로 증가시켜 가며 영상의 질과 환자선량의 관계를 알아보았다. chest phantom 영상의 정성적인 영상평가를 위해 VGA를, 정량적인 평가를 위해 SNR을 분석하였다. 선량은 ESAK로 측정하고 effective dose는 PCXMC를 이용하였다. 연구결과 일반적으로 시행되는 SID 180 cm를 기준으로 했을 때, ESAK의 경우 240 cm, 280 cm, 320 cm에서 각각 8.7%, 11.47%, 13.56%의 유의한 감소가 있었다. effective dose의 경우 전신에 대해 2.89%, 4.67%, 6.41%의 감소, 폐에서 5.08%, 6.98%, 9.6%의 감소가 관찰되었다. SNR의 경우 각각 9.04%, 8.24%, 11.46%의 감소가 관찰되었으며 특히, SID 260 cm ~ 300 cm 구간에서 8.03%의 작은 감소가 나타났고 SID 340 cm까지도 5.24로 5이하로 감소되지 않았다. VGA에서는 통계적으로 유의한 차이가 없는 진단적 가치가 높은 영상으로 평가되었다. 따라서 eigital chest radiography에서 SID를 300 cm까지 증가시킴으로 화질의 저하 없이 환자선량을 감소시킬 수 있을 것으로 기대된다.

Subtraction 기법을 이용한 한우 성장 단계 특이 발현 유전자 탐색 (Identification of the Differentially Expressed Genes of Hanwoo During the Growth Stage by Subtractive cDNA Hybridization)

  • 한우의 성장단계 특이발현 유전자를 탐색하기 위하여, 본 연구에서는 유전자의 발현 유무 및 발현정도의 차이를 나타내는 유전자를 분리하는데 있어 가장 강력한 수단으로 알려진 subtractive cDNA hybridization 기법을 이용하여 한우 등심조직으로부터 12개월령 및 24개월령 특이적인 subtractive cDNA library를 제작하였다. 성장단계 특이적인 유전자를 탐색하기 위하여, 6, 12 및 24개월령 cDNA를 사용하여 reverse northern blot 분석을 실시하였으며, 6개월령 cDNA probe에 대하여 특이적인 signal을 나타낸 3개의 clone은 EPV 20, Ca2+ ATPase, 및 TCTP 유전자와 유사성을 나타내었다. 12개월령 cDNA probe에 대하여 특이적인 signal을 나타낸 9개의 cDNA clone은 각각 VCP, HSP 70, aldolase A, MSSK1, GM-2 activator protein, ryanodine receptor, acidic ribosomal phosphoprotein p1, ADP/ATP translocase T1 및 UCP 2 유전자와 높은 homology를 가지고 있었다. 또한 2개의 clone이 각각 12개월령 및 24개월령 cDNA probe에 대하여 특이적인 signal을 나타내었는데, 12개월령 cDNA probe에 대해서만 signal을 나타낸 clone은 ferrochelatase 유전자와 유사하였으며, 24개월령 probe에 대해서만 signal을 나타낸 clone은 ADRP 유전자와 유사하였다. 이상에서와 같이, 본 연구에서 제작한 성장단계 특이적인 subtractive cDNA library를 분석하여 14종의 유전자를 한우 성장단계 특이 발현 후보 유전자로 선정하여 염기서열을 분석하였으며, 이외에도 성장단계에 있어 특이적으로 발현될 것으로 추정되는 cDNA 클론의 염기서열을 분석하였다.

Ca2+/calmodulin-dependent regulation of polycystic kidney disease 2-like-1 by binding at C-terminal domain

  • Polycystic kidney disease 2-like-1 (PKD2L1), also known as polycystin-L or TRPP3, is a non-selective cation channel that regulates intracellular calcium concentration. Calmodulin (CaM) is a calcium binding protein, consisting of N-lobe and C-lobe with two calcium binding EF-hands in each lobe. In previous study, we confirmed that CaM is associated with desensitization of PKD2L1 and that CaM N-lobe and PKD2L1 EF-hand specifically are involved. However, the CaM-binding domain (CaMBD) and its inhibitory mechanism of PKD2L1 have not been identified. In order to identify CaM-binding anchor residue of PKD2L1, single mutants of putative CaMBD and EF-hand deletion mutants were generated. The current changes of the mutants were recorded with whole-cell patch clamp. The calmidazolium (CMZ), a calmodulin inhibitor, was used under different concentrations of intracellular. Among the mutants that showed similar or higher basal currents with that of the PKD2L1 wild type, L593A showed little change in current induced by CMZ. Co-expression of L593A with CaM attenuated the inhibitory effect of PKD2L1 by CaM. In the previous study it was inferred that CaM C-lobe inhibits channels by binding to PKD2L1 at 16 nM calcium concentration and CaM N-lobe at 100 nM. Based on the results at 16 nM calcium concentration condition, this study suggests that CaM C-lobe binds to Leu-593, which can be a CaM C-lobe anchor residue, to regulate channel activity. Taken together, our results provide a model for the regulation of PKD2L1 channel activity by CaM.

C6 Rat Glioma Cell에서 리튬에 의한 성장 억제와 Insulin-like Growth Factor System Components의 발현과의 관계 (Relationships of the Lithium-Induced Growth Inhibition of C6 Rat Glioma Cell to Expression of the Insulin-like Growth Factor System Components)

  • Insulin-like growth factors(IGF)s-I과 -II ligands와 이들의 receptors 및 6종류의 IGF-binding proteins(IGFBPs)로 구성된 IGF systim은 여러 종류의 세포의 생존, 증식 및 분화에 있어서 매우 중요한 역할을 한다. 리튬은 in vitro에서 여러 종류의 세포의 생존과 증식의 조절제로 알려져 있다. 본 연구는 IGF-I, IGF-I receptor 및 IGF carrier로서 주로 IGFBP-3를 발현하는 rat C6 glioma cell에서 LiCl로 유도된 세포 생존 및 증식의 변화와 IGF system components의 발현간의 관계를 구명하고자 수행되었다. 10%혈청을 함유하는 배양액에서 0, 2mM, 혹은 5mM LiCl을 첨가하여 C6 cell을 24시간 배양했을 시 세포의 생존률과 세포 수는 리튬 첨가에 의해 영향을 받지 않았다. 그러나 72시간 배양 했을 때 C6 cell은 명백히 리튬의 첨가수준에 따라 반응하였다. 0, 2m, 5mM LiCl 첨가수준에서 72시간 배양한 C6 cell은 각각 전형적인 세포분열, 세포분열 중지 및 세포사멸 양상을 보였다. 더욱이 사멸돼가는 세포는 reverse transcription-polymerase chain reaction으로 조사한 IGF-I, IGF receptor 및 IGFBP-3의 발현수준이 저하되었다. 흥미롭게도 혈청을 첨가하지 않은 배양조건 하에서 IGFBP-3에 대한 antisense oligodeoxyribonucleotide를 10${\mu}M$ 수준의로 첨가하여 배양했을 때도 표적 mRNA는 물론 세포 수도 줄었다. 종합하자면, C6 cell에서 리튬의 독성 효과의 일부는 이 제제에 의한 IGF system components의 발현 억제 효과에 의해 매개될 소지가 크다. 이러한 관점에서 IGRBP-3는 적어도 이 세포의 정상적인 증식을 위해 꼭 필요한(‘prrmissive') 역할을 할 수 있다는 점을 시사한다.

Expression patterns of innate immunity-related genes in response to polyinosinic:polycytidylic acid (poly[I:C]) stimulation in DF-1 chicken fibroblast cells

  • Polyinosinic:polycytidylic acid (poly[I:C]) can stimulate Toll-like receptor 3 (TLR3) signaling pathways. In this study, DF-1 cells were treated with poly(I:C) at various concentrations and time points to examine the comparative expression patterns of innate immune response genes. The viability of DF-1 cells decreased from 77.41% to 38.68% when cells were treated different dose of poly(I:C) from 0.1 ㎍/mL to 100 ㎍/mL for 24 h respectively. The expressions of TLR3, TLR4, TLR7, TLR15, TLR21, IL1B, and IL10 were increased in dose- and time-dependent manners by poly(I:C) treatment. On the contrary, the expression patterns of interferon regulatory factors 7 (IRF7), Jun proto-oncogene, AP-1 transcription factor subunit (JUN), Nuclear Factor Kappa B Subunit 1 (NF-κB1), and IL8L2 were varied; IRF7 and IL8L2 were increasingly expressed whereas the expressions of JUN and NF-κB1 were decreased in a dose-dependent manner after they were early induced. In time-dependent analysis, IRF7 expression was significantly upregulated from 3 h to 24 h, whereas JUN and NF-κB1 expressions settled down from 6 h to 24 h after poly(I:C) treatment although they were induced at early time from 1 h to 3 h. Poly(I:C) treatment rapidly increased the expression of IL8L2 from 3 h to 6 h with a plateau at 6 h and then the expression of IL8L2 was dramatically decreased until 24 h after poly(I:C) treatment although the expression level was still higher than the non-treated control. These results may provide the basis for understanding host response to viral infection and its mimicry system in chickens.

Protonation and Energetical Investigations of Calix[4]-cyclen-benzo-crown-6 and Its Complexes with Zinc and Copper

  • Protonation constants of calix[4]-cyclen-benzo-crown-6, L in 1X$10^{-2}$ M $Bu_4NCF_3SO_3$ in 40% $CH_2Cl_2/CH_3OH$ at $25^{\circ}C$ determined by potentiometric titration are log $K_1$ = 10.91, log $K_2$ = 10.30, log $K_3$ = 6.24 and log $K_4$ = 2.55. Stability constants for the receptor L complexes with Cu(II) and Zn(II) in 1X$10^{-2}$ M $Bu_4NCF_3SO_3$ in 40% $CH_2Cl_2/CH_3OH$ at $25^{\circ}C$ were determined by UV-VIS spectrometric titration. Stability constants of the CuL and ZnL complexes as log $\beta$ are 4.37 and 3.45, respectively. Stabilization energies for protonations of receptor L, derived from ab initio Hartree-Fock method with 6-31G basis set, are ${\Delta}E_1$ = -290.1, ${\Delta}E_2$ = -205.0, ${\Delta}E_3$ = -124.9 and ${\Delta}E_4$ = -26.9 kcal/mol and complexation energy of ZnL complex is -370.3 kcal/mol.

Expression Profiles of Immune-related Genes in Fluoxetine-treated Human Mononuclear Cells by cDNA Microarray

  • To investigate the effect of fluoxetine, one of selective serotonin reuptake inhibitors (SSRIs), on the immune system, human peripheral blood mononuclear cells (PBMC) were treated with fluoxetine $(10^{-7}\;M)$ for 24 h, and immune-related genes were analyzed by cDNA microarray. Expression of the immunerelated genes such as CD107b (LAMP-2), CD47 receptor (thrombospondin receptor), CD5 antigen-like (scavenger receptor cysteine rich family), copine III (CPNE3), interleukin (IL)-18 (interferon-gammainducing factor), integrin alpha 4 (CD49d), integrin alpha L subunit (CD11a), IL-3 receptor alpha subunit, L apoferritin, and small inducible cytokine subfamily A (Cys-Cys) member 13 (SCYA13) was induced by fluoxetine. This result suggests that fluoxetine may affect the immune system, and provides fundamental data for the involvement of SSRIs on immunoregulation.

흰쥐 자궁에서 Pituitary Adenylate Cyclase-Activating Polypeptide와 수용체 유전자의 발현 (Expressions of Pituitary Adenylate Cyclase-Activating Polypeptide and Its Receptor Gene in the Rat Uterus)

  • 본 연구에서는 흰쥐 자궁에서 pituitary adenylate cyclase-activating polypeptide (PACAP)과 그 수용체 유전자들이 발현되는가와 각각 어떠한 유형의 transcript들이 발현되는가를 조사하였고, 이를 위해 역전사 중합효소 연쇄반응 (RT-PCR)을 시행하였다. 시상하부와 정소에서 공통적으로 존재함이 알려진 PACAP common exon 부위에 해당되는 primer를 사용하여 PCR을 시행한 결과 자궁을 포함한 모든 조직에서 에상대로 297 bp의 band가 확인되었다. 흰쥐 자궁에서 발현되는 PACAP mRNA에 정소 특이적인 exon 1이 포함되는가를 조사하기 위해 정소 특이적인 primer를 사용하여 PCR을 시행한 결과, 정소에서만 예상대로 586 bp의 band가 검출되었고 자궁을 포함한 다른 조직에서는 발견되지 않았다. 흰쥐의 PACAP 수용체 PCR에서는 자궁에서 923 bp와 839 bp크기의 band를 확인할 수 있었으며, 이는 알려진 PACAP type I수용체의 splicing variant 들 가운데 hip-hop (923bp)과 hip- 또는 nop-type (839bp)의 예상 크기와 일치하였다. 인위적으로 성적인 성숙을 유도한 PMSG 주사모델하에서 자궁내 PACAP transcript 수준은 주사 24 시간 후 실험군에서 증가하여 생식주기상 proestus 시기에 해당되는 주사 48 시간까지 증가하였다가 72 시간후에는 control 보다 낮은 수준을 보였는데, 이는 자궁의 PACAP 유전자발현이 생리적으로 조절됨을 나타내는 것이다. 본 연구는 흰쥐의 자궁에서 PACAP과 그 수용체 isoform들의 유전자가 발현됨을 최초로 보고한 것으로 자궁 자체에서 발현되는 PACAP이 autocrine 또는 paracrine하게 자궁의 생리 및 기능 조절을 담당함을 시사한다.

Signal transduction of C-terminal phosphorylation sites for equine follicle stimulating hormone receptor (eFSHR)

  • Equine follicle stimulating hormone receptor (eFSHR) has a large extracellular domain and an intracellular domain containing approximately 10 phosphorylation sites within the G protein-coupled receptor. This study was conducted to analyze the function of phosphorylation sties at the eFSHR C-terminal region. We constructed a mutant of eFSHR, in which the C-terminal cytoplasmic tail was truncated at residue 641 (eFSHR-t641). This removed 10 potential phosphorylation sites from the C-terminal region of the intracellular loop. The eFSHR-wild type (eFSHR-wt) and eFSHR-t641 cDNAs were subcloned into the pCMV-ARMS1-PK2 expression vector. These plasmids were transfected into PathHunter CHO-K1 Parental cells expressing β-arrestin 2 enzyme acceptor fusion protein and analyzed for agonist-induced cAMP response. The cAMP response in cells expressing eFSHR-t641 was lower than the response in cells expressing eFSHR-wt. EC50 values of eFSHR-wt and eFSHR-t641 were 1079 ng/mL and 1834 ng/mL, respectively. eFSHR-t641 was approximately 0.58-fold compared with that of eFSHR-wt. The maximal response in eFSHR-wt and eFSHR-t641 was 24.7 nM and 16.7 nM, respectively. The Rmax value of phosphorylation sites in eFSHR-t641 was also decreased to approximately 68.4% of that in eFSHR-wt. The collective data implicate that the phosphorylation sites in the eFSHR C-terminal region have a pivotal role in signal transduction in PathHunter CHO-K1 cells, and indicate that β-arrestin is involved in coupling the activated receptors to the internalization system.

아드레날린성 β-수용체에 대한 higemamine의 약리학적 특성 (Pharmacological characteristics of higenamine on adrenergic β-receptors)

  • Higenamine is an Aconiti tuber derived compound whose chemical structure is 1-(4'-hydroxybenzyl)-6, 7-dihydroxy-1, 2, 3, 4-tetrahydroisoquinoline containing catechol ring and tetrahydroisoquinoline nucleus in its own structure, both of which are well known to have agonistic effects on adrenergic receptors. Using guinea-pig atria(rich in ${\beta}_1$-receptor) and treachea(rich in ${\beta}_2$-receptor), we studied pharmacological actions of higenamine on these organs with special interest of its relevancy of ${\beta}$-receptor selectivity. In order to further clarify its pharmacological characteristics, the influncences of pretreatment of reserpine or cocaine were also investigated. The results were summarized as follows : 1. Higenamine had remarkable chronotropic, inotropic and bronchodilator effects in guinea-pig spontaneously beating right atria, left atria and trachea, in dose-dependent manners. 2. All of above actions were blocked competitively by propranolol, which shows nonselectivity of higenamine on ${\beta}$-receptor. $pA_2$ values of propranolol against higenamine were 7.93, 7.76 and 8.46 in guinea-pig right atria, left atria and treachea, respectively. 3. Reserpine pretreatment(5mg/kg, ip, 24h) did not show my decrease in pharmacological actions of higenamine, which suggests higenamine has direct action on ${\beta}$-receptor not via catecholamine release. 4. Cocaine pretreatment$(1{\mu}M)$ had no influence on pharmacological actions of higenamine in contrast with nor epinephrine, which suggests there is no neuronal uptake mechanism of higenamine in the studied organ preparations.
